Bulletin of The Academy of Military Medical Sciences 2009;33(6):543-545
Objective:To establish a highly sensitive, rapid and selective liquid chromatography mass spectrometry (LC-MS) method for the determination of metoprolol in rat plasma. Methods:A simplified liquid-liquid extraction with acetidin was employed for the sample preparation. The separation was carried out on a Thermo ODS-C_(18)(5 μm,100 mm×2.1 mm).The mobile phase consisted of acetonitrile-methanol-water(20∶20∶60). Propranolol was used as the internal standard. The detection was performed on a liquid chromatography mass spectrometry by selected ion monitoring(SIM) scan mode electrospray ionization(ESI).Results and Conclusion:The range of calibration curve was 0.1-50 ng/ml and the limit of quantification was 0.1 ng/ml. The intra- and inter-day precision RSD was less than 15%.This method is sensitive, simple, rapid and suitable for the pharmacokinetic study of metoprolol.

Successive pathogenic bacteria were examined from liver, spleen, kidney and contents of intestine of diseased stone flounder (Kareius bicoloratus L.) occurring bacterial septicaemia infection caused by Aeromonas salmonicida. Identification of extensive phenotypic information to 6 pure cultures and detection of the mol% G+C ratio of the DNA to representative strains, showed that the examined bacteria belonged to a new morphovar of Acinetobacter junii, and was designated as Acinetobacter junii morphovar I. In addition, serotype, antibiotic sensitivity and pathogenicity of isolates were studied, the results showed that the 6 strains have the same K-antigen and O-antigen, there are no obvious differences in sensitivity and resistance to used 37 antimicrobial agents between strains, and have strong pathogenicity to experimental stone flounder and Bastard halibut.

Objective To evaluate the effects of dexmedetomidine pretreatment on extracellular sig?nal?regulated kinase ( ERK) pathway during acute lung injury in a rat model of liver transplantation. Meth?ods Sixty male Sprague?Dawley rats, weighing 235-250 g, were divided into 4 groups ( n=15 each) u?sing a random number table: sham operation group (group S), liver transplantation group (group LT), low?dose dexmedetomidine pretreatment group ( group LD ) and high?dose dexmedetomidine pretreatment group ( group HD) . In LT, LD and HD groups, the model of orthotopic liver transplantation was estab?lished, and the operation time was about 4 h. Dexmedetomidine 2?5 and 5?0μg·kg-1 ·h-1 were intrave?nously infused for 1 h starting from 1 h prior to clipping the hepatic artery and portal vein in LD and HD groups, respectively. The rats were sacrificed after the end of operation, and the lungs were removed for determination of wet to dry weight ratio ( W∕D ratio) , cell apoptosis and expression of ERK mRNA, ERK, phosphorylated ERK ( p?ERK) , Bcl?2 and Bax in lung tissues and for examination of the pathological chan?ges ( with light microscope) and ultrastructure of lung tissues ( with transmission electron microscope) . The
injured alveolus rate ( IAR) , apoptosis index ( AI) and ratio of Bcl?2 to Bax expression ( Bcl?2∕Bax ratio) were calculated. Results Compared to group S, the W∕D ratio, IAR, AI, expression of ERK?1 mRNA, ERK?2 mRNA, p?ERK, Bcl?2 and Bax and Bcl?2∕Bax ratio were significantly increased in LT, LD and HD groups ( P<0?05) . Compared to group LT, the W∕D ratio, IAR and AI were significantly decreased, the expression of ERK?1 mRNA, ERK?2 mRNA, p?ERK and Bcl?2 and Bcl?2∕Bax ratio were significantly increased, and the expression of Bax was significantly down?regulated in LD and HD groups (P<0?05). Compared to group LD, the W∕D ratio, IAR and AI were significantly decreased, the expression of ERK?1 mRNA, ERK?2 mRNA, p?ERK and Bcl?2 and Bcl?2∕Bax ratio were significantly increased, and the ex?pression of Bax was significantly down?regulated in group HD ( P<0?05) . The pathological changes of lung tissues were significantly attenuated in LD and HD groups as compared with group LT, and in group HD as compared with group LD. Conclusion The mechanism by which dexmedetomidine pretreatment mitigates cell apoptosis during acute lung injury is related to activation of ERK pathway in a rat model of liver trans?plantation.

AIM:To investigate the pathogenesis of aplastic anemia ( AA) through the expression of P 53 and P21, and the telomere length in the bone marrow mononuclear cells (BMMNC) of patients with AA.METHODS: The BMMNC were collected from 60 cases of AA, including 38 non-severe aplastic anemia (NSAA) and 22 severe aplastic ane-mia (SAA), and 25 healthy controls. The mRNA expression of P53 and P21, and the telomere length were detected by real-time quantitative polymerase chain reaction (RT-qPCR), while the protein expression of P53 and P21 was determined by Western blot. The correlations among P53, P21 and telomere length were analyzed .Hematopoietic components of bone marrow were measured by bone marrow biopsy .CD34 +cells proportion in nuclear cells were assessed by flow cytometry . RESULTS:Telomere length in the patients with AA , including NSAA and SAA , was significantly lower than that in con-trol group (P<0.05).The same results of the hematopoietic components of bone marrow and the CD 34+cells proportion in nuclear cells were observed .Telomere length in SAA group was shorter than that in NSAA group ( P<0.05 ) , while the hematopoietic components of bone marrow and the CD 34 +cells proportion in nuclear cells also showed the same results . The expression of P53 and P21 at both mRNA and protein levels in the AA patients , including NSAA and SAA , was signi-ficantly higher than that in control group (P<0.05).The expression of P53 and P21 in the SAA patients was higher signi-ficantly than that in the NSAA patients ( P<0.05 ) .No correlation between the expression of P 53 or P21 and telomere length was found . There was significant positive correlation between the expression of P 53 and P21 ( P <0.05 ) . CONCLUSION:Telomere length, P53 and P21 may be involved in the pathogenesis of AA by inhibiting the proliferation and differentiation of hematopoietic stem cells , and triggering cell apoptosis .
