1.An epidemiological survey of trachoma in primary and secondary students in Keshiketeng of Chifeng city in Inner Mongolia

Background Trachoma is a common infectious eye disease,and its incidence is associated with region,economy and sanitary condition.To survey the prevalence status of an area is very important for the prevention and treatment of trachoma.Objective Aim of this study was to survey the prevalence of trachoma in primary and secondary school students of Chifeng Keshiketengqi,and to investigate the relationship between the prevalence of trachoma with age,gender,ethnicity and living environment.Methods This was a cross-sectional study.According to cluster random sampling method,2067 students were drawn from 10 primary and secondary schools,including 1061male and 1006 female.The age of students ranged from 6 to 17 years old,with the average age (12.53 ± 2.31)years.In the taken samples,Han students were 1768,and Mongolian students were 299;the agricultural and pastoral students were 1209,and the urban students were 858.The general life factors associated with trachoma were surveyed by questionnaire.Trachoma was diagnosed and graded based to 1987 WHO Simplified Trachoma Grading Standards.Eye examinations included visual inspection,eyelids,conjunctiva and cornea neovascularization under the slit lamp microscope,and the clinical data were recorded and collected.The results were statistically analyzed by SPSS13.0 software.Results According to the sampling proportion of 10.05%,2153 students were included and 2067 students participated in this study with the response rate 96%.Trachoma was diagnosed in 972 students with a morbidity 47.0%.The prevalence rate of trachoma was increased with the ageing,and a statistically significant difference was seen among the 6-8 years group,9-11 years group,12-14 years group and 15-17 years group (x2 =11.043,P =0.011),and the prevalence of trachomatous infiltration was significantly higher than that of trachomatous folliele (x2=11.493,P<0.05).The prevalence rate of trachoma in Han and Mongolian students was 45.8% and 54.5%,showing a significant difference between them (x2 =6.228,P =0.013).In addition,statistically significant differences also were found in the prevalence rate of trachoma between the pastoral areas students and urban students (56.1% vs.39.7%,x2=30.226,P =0.000),but no significant difference was seen between male and female (47.0% vs.46.9%,x2 =0.000,P =0.995).Conclusions The trachoma prevalence rate is relatively higher in the primary and secondary students in Chifeng Keshiketeng.These results offer a basis for the prevention and treatment of trachoma.
2.Hair roots induction and culture of Withania somnifera and its withanolide A synthesis.

Withanolide A is a biologically active secondary metabolite occuring in roots and leaves of Withania somnifera. In the present study, adventitious roots from leaf explants of W. somnifera were induced for the production of withanolide-A by Agrobacterium tumefaciens strain C58C1 to obtain hair roots. Hair roots induction rate reached 30%. The withanolide A was determined by HPLC in different hair roots lines and different parts of W. somnifera. The average content of withanolide A in all hair roots lines were 1.96 times as high as that in wild-plant, the concentration of withanolide A in hair roots (1.783 mg x g(-1) dry weight) were 1.51 times as high as the roots of wild W. somnifera (1.180 mg x g(-1) dry weight), respectively. It is possible to obtain withanolide A from hair roots culture of W. somnifera.

4.Application of nursing intervention in non-anesthesia textile fiber laryngoscope inspection

Objective To explore the application effect of nursing intervention under non-anesthesia textile fiber laryngoscope inspection. Methods Selects 1000 textile fiber laryngoscope inspection patients,divides into the experimental group and control group each 500 individuals randomly, the control group carries out the convention operation sequence, and the experimental group does not carry on the nasopharyngeal throat anesthesia before inspection. The psychological suggestion, music nursing, touching applies in textile fiber laryngoscope inspection were implemented under non-anesthesia in textile fiber laryngoscope inspection and observe its function effect. Results Experimental group in textile fiber laryngoscope inspection process,diastolic pressure and heart rate change obvious lightly in control group, the difference has statistics significance (t =-2.164,-2.282,P <0.05), tense sense of fear and disgusting vomitive degree also remarkable lightly in control group, the difference has statistics s.ignificance (x2 =9.06,24.9 ,P <0.01),the inspection defeat case number obviously is also short in the control group. Conclusions The nursing intervention can help the textile fiber laryngoscope to inspect the patient to sharpen the adaptive comprehensively, complete the inspection smoothly under the non-anesthesia, after having avoided the anesthesia, the body produces each kind of ill response, is providing the user-friendly service, establishes protects the trouble relations harmoniously,enhances the patient degree of satisfaction aspect to have the important meaning.
5.Study on the micropermeability of resin-dentin bonding interfaces with ethanol-wet bonding technique

Objective To evaluate the micropermeability on bonding hydrophobic adhesive to dentin with ethanol-wet bonding under simulated pulp pressure.Methods Twenty-four intact human third molars were used in the study.After the enamel of occlusal surfaces was removed,the molars were randomly divided into six groups.Adper Scotchbond Multi-Purpose was used in the control group; in the experimental groups,the dentin surfaces were saturated with ethanol for 20 s ( group 1 ),1 min ( group 2 ),2 min ( group 3 ),3 min ( group 4 ) or with a series of increasing ethanol concentrations before application of hydrophobic adhesive ( group 5 ).All the bonding procedures were done under simulated pulp pressure.After 24 hours,micro-tensile bond strength test were performed on the specimens.Bonding interfaces were observed under laser scanning confocal microscope (LSCM) after the pulp chamber were filled with a water-soluble fluoroprobe rhodamine B for 3 hours.Results Compared with the control group[(38.14 ± 4.97 ) MPa],bond strengths in group 1 [(21.02 ± 7.23 ) MPa]and group 2 [( 29.64 ± 3.81 ) MPa]were statistically lower ( P > 0.05 ),while bond strngth in group 3 [( 38.40 ± 5.03 ) MPa],group 4 [( 37.26 ± 4.68 ) MPa]and group 5[(40.12 ±5.95) MPa]were similar to the control group (P<0.05).The images taken by LSCM showed that with extension of ethanol-wet time,the deposition of fluorescent dye in hybrid layer and along the dentinal tubules decreased gradually.Especially in group 5,only spare fluorescent dye deposition could be detected in the hybrid layer.Conclusions Dentin saturated with ethanol for more than 2 min before bonding hydrophobic adhesive to dentin could provide favorable bond strength and decreased the micropermeability of bonding interfaces under simulated pulp pressure.
6.The difference of IL-28B polymorphisms between hepatitis C patients with and without cryoglobulinemia.

OBJECTIVETo determine whether patients infected with chronic hepatitis C (CHC) show a differential distribution profile of IL-28B polymorphisms according to the presence of concomitant cryoglobulinemia.
METHODSSixty-two consecutive CHC patients were enrolled in the study between December 2008 and December 2010. All patients received combination therapy of pegylated interferon alpha-2a (weekly, 180 g, subcutaneous injection) plus ribavirin (daily, 10to15 mg/kg body weight, oral) for 48 weeks, with individualized dosage adjustments according to the patient's clinical situation. Cryoglobulins were detected visibly by separation of cryoprecipitates in patient serum samples. Three IL-28B SNPs (rs8099917, rs12979860, and rs12980275) were detected by sequencing. Response to treatment was assessed by measuring serum levels of HCV RNA by quantitative PCR at baseline (prior to treatment initiation), during treatment (4 and 12 weeks after treatment initiation), end of therapy (48 weeks after treatment initiation), and post-treatment (24 weeks after end of therapy). The significance of between-group differences were assessed by the Chi-square and Fisher's exact tests.
RESULTSCryoglobulinemia was detected in 43.5% (27/62) of the CHC patients and showed a female bias (59.3% vs. males: 34.3%, P = 0.05). Compared to CHC patients without cryoglobulinemia, the CHC patients with cryoglobulinemia showed significantly higher levels of HCV RNA at baseline (5.64+/-1.20 vs. 6.37+/-0.67, P less than 0.05) but lower frequencies of the IL28B rs8099917 TT genotype (94.3% vs. 63.0%, P = 0.002), rs8099917 T allele (97.1% vs. 81.5%, P = 0.003), and rs12979860 C allele (94.3% vs. 83.3%, P = 0.048). CHC patients with cryoglobulinemia and having the rs8099917 TT, rs12979860 CC, or rs12980275 AA genotype achieved a higher rate of sustained virological response.
CONCLUSIONCryoglobulinemia in CHC patients is associated with a differential distribution of IL-28B polymorphisms, and certain polymorphisms may be related to anti-viral treatment response.

7.Correlation between fibrinogen level and cerebral infarction.

OBJECTIVETo investigate the correlation between plasma fibrinogen level and cerebral infarction (CI) as well as the difference of fibrinogen among subtypes of CI.
METHODSA case-controlled study was conducted with 131 cases of CI and 148 controls. Plasma fibrinogen levels were detected by the Clauss method.
RESULTSHigh fibrinogen level (3.09 +/- 0.94 g/L) was correlated with CI (OR = 2.47, 95% CI: 1.51-4.04, P < 0.005) at the onset stage of the disease. Persistent high fibrinogen level (3.14 +/- 0.81 g/L) at 6-month after stroke onset was detected and correlated with CI (OR = 4.34, 95% CI: 1.80-10.51, P = 0.001). Higher fibrinogen level was correlated with total anterior circulation infarction (TACI), partial anterior circulation infarction (PACI), and posterior circulation infarction (POCI) (OR = 4.008, P < 0.001). Higher fibrinogen level was correlated with extracranial atherosclerosis (OR = 3.220, P < 0.05, but not with intracranial atherosclerosis.
CONCLUSIONFibrinogen level may be a risk factor of CI and probably correlates with subtypes of CI and distributions of atherosclerosis.

Insulin-like growth factor binding-protein-7 (IGFBP7) was obtained from our previous colonic adenocarcinoma (CRC) and normal mucosa suppression subtraction hybridization (SSH) cDNA libraries. By RT-PCR and immunohistochemistry, we found that IGFBP7 was overexpressed in CRC tissue compared to normal tissue. However, our in vitro experiments performed in 10 CRC cell lines showed that IGFBP7 expressed only in SW480 and Caco2 cell lines, which implied an underlying reversible regulatory mechanism. Using methylation-specific PCR (MSP) and bisulfite sodium PCR (BSP), we found that its expression was associated with DNA hypomethylation of exon1. This was further supported by the in vitro study which showed restored IGFBP7 expression after demethylation agent 5-aza-2'-deoxycytidine treatment. Correlation analysis between IGFBP7 expression and prognosis indicated that overexpression of IGFBP7 in CRC tissue correlated with favourable survival. Investigation of the functional role of IGFBP7 through transfection studies showed that IGFBP7 protein could inhibit growth rate, decrease colony formation activity, and induce apoptosis in RKO and SW620 cells, suggesting it a potential tumor suppressor protein in colorectal carcinogenesis. In conclusion, our study clearly demonstrated that IGFBP7 plays a potential tumor suppressor role against colorectal carcinogenesis and its expression is associated with DNA hypomethylation of exon 1.

9.Effect of liposomal transfection of cyclin A antisense oligodeoxynucleotide (ASON) on HL-60 cell proliferation and apoptosis.

OBJECTIVETo explore the effect of liposomal transfection of cyclin A antisense oligodeoxynucleotide (ASON) on HL-60 cell proliferation and apoptosis.
METHODSBy liposomal transfection, cyclin A ASON was co-cultured with HL-60 cells, the cell growth curve was determined by MTT assay and cell apoptosis electron-microscopy in situ cell apoptosis detection kit (POD), the protein and mRNA of cyclin A and bcl-2 were measured by FACS and RT-PCR, the role of cyclin A ASON in the development of leukemia was tested by the tumor formation in nude mice.
RESULTS(1) In the cyclin A ASON liposomal transfection group (group A), the proliferation of HL-60 cell was significantly inhibited as compared to those in cyclin A ASON group (group B) (68.9% vs 24.8%) (P < 0.01). (2) The expressions of cyclin A and bcl-2 of group A were significantly lower than those in the control group (1.1% vs 38.8%, P < 0.01; 21.9% vs 65.0%, P < 0.01, respectively), and the DNA ladder and apoptosis body was displayed. (3) In group A, the rate of tumor formation in nude mice was lower, the time for tumor formation was longer and the volume of tumor was smaller than those in control group.
CONCLUSIONLiposomal transfection of cyclin A ASON can inhibit in vitro proliferation of leukemia cells and induce in vivo apoptosis of the tumor cell, which might provide a new target for gene therapy.
