1.Pay attention to the implementation and management of the laboratory medicine research projects

In recent years,with increasing investment on laboratory medicine scientific research in China,the project management,implementation,evaluation system and quality improvement become more and more important for research managers and executives.In this paper,the main problems of project execution and implementation and accordingly proposed some constructive suggestions were analysed,in the aspects of the leading role of principal investigator,periodical discussion,peer guidance,scientific progress management,effective using of project funds and the establishment of appropriate reward and punishment mechanisms.It is hoped that these suggestions can be used to achieve success in laboratory medicine research project.
2.Study on effect of lycorine in inducing apoptosis of pulmonary carcinoma cell A549.

To explore the effect of lycorine in inducing apoptosis of pulmonary carcinoma cell A549 and its mechanism. In the study, pulmonary carcinoma cell A549 were taken as the experimental subject and processed with different concentrations of lycorine (0, 0.5, 1.0, 2.0, 4.0 and 8.0 μmol x L(-1)). The MTT method was used to observe the cell proliferation. The apoptosis rate of A549 cells was determined by Annexin FITC/PI double staining. The microplate reader was used to detect the activities of Bcl-2, Bax and p53. The changes in mitochondrial membrane potential were measured by the flow cytometry. The expressions of apoptosis-related factors Bcl-2, Bax, p53 and Survivin were determined by Real-time PCR. The results showed that lycorine significantly inhibited the proliferation of A549 cells (P < 0.05), induced the apoptosis on A549 cells (P < 0.05), increased the activities of Bax and p53, reduced Bcl-2 activity and mitochondrial membrane potential, and notably changed the gene expressions of Bcl-2, Bax, p53 and Survivin (P < 0.05). In conclusion, lycorine can induce the apoptosis of A549 cells and be applied to treat pulmonary carcinoma. Its mechanism may be related to the activation of relevant factors in Bcl-2 signaling pathway.

3.Pedicle screw fixation through paraspinal approach for thoracolumbar fractures:a follow-up of correction effect and biocompatibility

BACKGROUND:Different operation approaches can be selected during the repair of thoracolumbar fractures. Traditional posterior median approach wil cause great wound on patients, and easily induces some adverse consequences. OBJECTIVE: To compare the effects and biocompatibility of conventional posterior median approach and paraspinal approach fixation for treatment of thoracolumbar fractures. METHODS: A retrospective analysis was performed on clinical data of 53 cases of thoracolumbar fracture in the Shaanxi Province Nuclear Industry 215 Hospital from December 2012 to December 2013. They were divided into two groups according to approach method. The control group (n=27) received pedicle screw placement fixation through conventional posterior median approach. The observation group (n=26) underwent pedicle screw placement fixation through paraspinal muscle approach. After repair, they were folowed up for 12 months. Imaging results, pain score and perioperative relevant indexes were observed and compared in both groups. RESULTS AND CONCLUSION: Imaging examination was conducted before repair, immediately after repair and during final folow-up in both groups. The height of vertebral anterior border and kyphosis correction effect were good after different therapies in both group, but no significant difference was detectable between the two groups (al P > 0.05). These findings suggest that height of vertebral anterior border and kyphosis correction effect were identical between the two groups. Pain visual analog scale score was significantly lower in the observation group than in the control group at 24 hours and 3 days after repair and during final folow-up (al P < 0.05). Perioperative relevant indexes were analyzed in both groups. Intraoperative bleeding amount, drainage and bed time after repair were observed in the observation group, which showed significant advantages as compared with the control group (al P < 0.05). These results confirmed that compared with the traditional posterior median approach surgery, minimaly invasive pedicle screw placement through paraspinal muscle approach for thoracolumbar fractures can obtain better repair effects and biocompatibility.
4.Expression of glycosylphosphatidylinositol-anchored protein in lymphocytes and the subsets of healthy individuals

Objective To study the expression of glycosylphosphatidylinositol(GPI) anchored protein in lymphocyte subsets of healthy individuals. Methods Flow cytometry and two/three color McAbs were used to detect CD59 expression in granulocytes, lymphocytes and subsets. Results In 50 samples, granulocytes mainly showed a single population of cells strongly positive for CD59, the percentage of CD59 expression was (98.6?1 5)%. Lymphocytes had a small population of weakly positive and negative for CD59, positive cells being (90 0? 14.9)%. CD3 +T cells had a similar profile as that of total lymphocytes, but there were more weakly positive /negative cells and less strong positive cells. As compared with CD3 +T cells, CD4 +T cells had a larger population of strongly positive for CD59, positive cells being (92.9?8.1)%; while CD8 +T cells had a smaller population of strongly positive, positive cells being (74.6?9.1)%. CD59 expression of activated T cells was between that of CD3 +and CD8 +T cells. CD45RA +and CD45RO +T cells both had a similar profile like that of CD3 +T cells. CD20 +B cells showed a similar profile to that of granulocytes, positive cells being (96.3?0 6)%. Conclusions Population of low or negative expression of CD59 among the lymphocytes of healthy controls was related to T cells, especially CD8 +T cells. B cells had uniform positive expression of CD59, and could be a candidate for detecting PNH.
5.Effects of dexmedetomidine on serum inflammatory factor and oxidative stress response in septic rats

Objective To investigate the effects of dexmedetomidine on serum inflammatory factor and oxidative stress response in septic rats.Methods Thirty healthy male SD rats,aged 10-14 weeks,weighing 250-300 g,were randomly divided into 3 groups( n =10 each): sham operation group (group S),sepsis group (group CLP) and sepsis + dexmedetomidine group (group CLP + D).Sepsis was induced by cecal ligation and puncture in groups CLP and CLP + D.Group CLP + D received intravenous infusion of dexmedetomidine at 10 μg· kg- 1 ·h- 1 from the end of operation until dead or 12 h after operation.Groups S and CLP received equal volume of normal saline at 1 ml·kg-1 ·h-1.Arterial blood samples were taken from 5 rats in each group before (basline) and at 1,6 and 12 h after operation for determination of serum IL-6,IL-10,SOD and MDA levels.The survival rate within 12 h after operation was recorded.Results Compared with group S,serum IL-6,IL-10 and MDA concentrations at 1,6 and 12 h after operation were increased,while serum SOD activity at 1,6 and 12 h after operation and survival rate were decreased in group CLP ( P < 0.05 ).Compared with group CLP,serum IL-6 and MDA concentrations at 6 and 12 h after operation were decreased,while serum SOD activity at 12 h after operation and survival rate were increased in group CLP + D ( P < 0.05 ).Conclusion Dexmedetomidine can increase the survival rate of septic rats by inhibiting inflammatory factor release and oxidative stress response.
6.Comparison of single-plane and biplane area-length methods for right ventricular volume calculation

Objective To compare the relative accuracy of single-plane and biplane area-length methods for determining right ventricular (RV) volume and ejection fraction (RVEF) both in vitro and in vivo studies. Methods The in vitro study consisted of 15 human right ventricular casts and the in vivo study consisted of 45 patients. RV volume by single-plane area-length method was determined by the product of RV area from apical four-chamber view and the distance from RV apex to tricuspid annulus. RV volume by biplane area-length method was defined as 2/3?(the product of RV area from apical four-chamber view and the distance from pulmonary valve to tricuspid valve). The actual volume of RV casts was determined by water displacement method. RV angiography was used to determine RV volumes and RVEF in the in vivo study. Results RV volumes calculated from both single-plane and biplane methods correlated closely with RV cast volumes in the in vitro study (r= 0.87- and 0.91-, respectively; both P 0.05-), but RV volume determined by single-plane method significantly overestimated angiographic volume (P

MicroRNAs (miRNAs) are small noncoding regulatory RNAs ranging in size from 17 to 25 nucleotides which participate many physiological and pathological processes.MicroRNA could also stably exist in peripheral blood,and the detection of circulating microRNA is of great significance for disease diagnosis and prognosis.As so far,there is no unified method and standard for detection of microRNA,which is the major reason for discrepant results between different studies.However,circulating microRNA,as a new disease detection biomarker,has stable properties,convenient detection and high accuracy features,so it till has an important potential value in clinical applications.
8.The Protective Effects of Butylphthalide on Amyloid β Peptide_(25-35) Induced Apoptosis in PC-12 Cells

Objective To investigate the protective effects of dl-3-n-butylphthalide(NBP)on amyloid β peptide 25-35(Aβ25-35)-induced apoptosis in PC-12 cells.Methods Cultured PC12 cells were divided into 6 groups:normal group,Aβ25-35 treated model group,0.1,1.0,10,100 μmol/L NBP pretreatment groups.MTT assay was employed to analyze the PC12 cell viability.The ultrastructural changes of neuronal mitochondria were viewed under transmission electron microscope.In order to observe the effects of oxidative stress,MDA and SOD activities were detected by spectrophotometry.Results NBP pretreatment could significantly prevent the cell viability induced by Aβ25-35(P 0.05).Pretreatment with 10 μmol/L NBP could significantly inhibit the viability decrease induced by Aβ25-35(P 0.05).Compared with the cells in the model group,the number and morphology of neuronal mitochondria changed distinctly in the NBP pretreated cells.The activity of SOD in the NBP pretreated cells was obviously higher than that of the cell in the model group,while MDA activity had opposite result.Conclusion NBP could protect the mitochondria in Aβ25-35 induced apoptosis by inhibiting the MDA activity and activating the SOD activity.
9.Meta analysis of the level of plasma D-dimer in patients with ovarian cancer

Objective To evaluate the correlation between plasma D dimer levels and ovarian cancer, as well as its pathologic staging.Methods According to the inclusion and exclusion criteria , articles published about the correlation between plasma D dimer levels and human ovarian cancer were searched by means of PubMed , Embase, CNKI and WanFang Data from their inception to December 31th , 2013.In addition , references′list of the included articles and their related citations in Pubmed were also reviewed for additional articles.After quality assessment of all studies , Meta-analysis and ANOVA were performed with RevMan 5.2 and SAS 9.1 for statistic analysis.Results A total of 13 articles were identified, including 679 cases of ovarian cancers , 685 cases of benign controls and 477 cases of healthy controls.Meta-analysis revealed that:(1) Plasma D dimer levels in ovarian cancer groups were higher than that in benign control groups ( Z =8.61, P <0.000 01 ) and healthy control groups ( Z =6.76, P <0.000 01);(2) Plasma D dimer levels in benign control groups were higher than that in healthy controls (Z=3.15, P=0.002); (3) Plasma D dimer levels in advanced ovarian cancer FIGO (Ⅲ-Ⅳ) were higher than that in early cancer FIGO (Ⅰ-Ⅱ) (Z=3.38, P=0.000 9);(4)But there were no difference between relapse groups and remission groups (Z=1.19, P=0.23).ANOVA revealed that:(1) There were significant difference of D dimer levels among ovarian cancer groups or benign groups , healthy control groups; ( 2 ) Significant increasing of D dimer levels was revealed by Pairwise ANOVA from healthy controls, benign controls to ovarian cancer.Conclusions Plasma D dimer levels were high in patients with ovarian cancer , especially in FIGO (Ⅲ-Ⅳ) ovarian cancer.No difference of D dimer levels were investigated between relapse phase and remission phase.
10.Relationship between augmentation index and severity of coronary artery disease

Objective To explore whether augmentation index is an independent risk factor for coronary heart disease or not,and the relationship between augmentation index and coronary artery disease severity.Methods In the study,245 subjects who were hospitalized between Dec.2003 and Dec.2004,were classified into 4 groups and their clinical and biological characteristics and the summary of the scores in each of the eight segment and aortic pressure waveform in ascending aorta were recorded respectively.Results The larger the augmentation index was,the more the number of coronary stenosis vessels was.Linear regression analysis indicated that augmentation index was significantly correlated with grade of coronary stenosis.When augmentation index was more than 45%,the sensibility and specificity was 91.5%and 92.6%respectively.In the logistic regression model,augmentation index was the independent risk factor for coronary heart disease,and the odds ratio of coronary heart disease was 1.893.The 95%CI was 1.421~2.521.Conclusion Augmentation index is an independent risk factor for coronary heart disease and a predictor of angiographic coronary artery disease severity.
