1.Effect of triiodothy ronine on neurogenesis in subventricular zone after bilateral common carotid artery occlusion in adult rats

Objective To investigate the effect of triiodothyronine (T3) on neurogenesis in after bilateral common carotid artery occlusion in adult rats. Methods Fifteen adult male Sprague-Dawley (SD) rats were randomly divided into sham operation, 2-vessel occlusion (2VO) and T3 intervention groups (n = 5 in each group ). A chronic cerebral ischemia model was established by the bilateral common carotid artery permanent ligation. Seven days after modeling 5-bromo-2'-deoxyuridine (BrdU) was injectied intraperitoneally and the new proliferating cells were labeled. The BrdU positive cells in SVZ were observed using immunofluorescence staining.Results The numbers of BrdU positive cells in the sham operation, 2VO, and T3 intervention groups were 174. 8 ± 18. 5, 325.0 ± 51.0, and 499. 4 ± 30. 8, respectively. There were significant differences among all the groups (F = 101.68, P < 0. 001 ). The numbers of BrdU positive cells in the T3 intervention group were significantly higher than those in the 2VO and sham operation groups (all P <0. 001 ). The numbers of BrdU positive cells in the 2VO group were also significantly higher than those in the sham operation group (p <0. 001). Conclusions Thyroid hormones may promote neural progenitor cell proliferation in SVZ in adult 2VO rats.
2.Efficacy of cisplatin against human ovarian cancer cells when combined with p53 gene therapy

Objective:To explore the effects of wild type p53 gene on biologic behavior of human ovarian cancer cell line SKOV 3 and the efficacy of cisplatin against the cancer cells when combined with p53 gene therapy.Methods:A human wild type p53 gene recombinant eukaryotic expression vector was introduced by lipofectin mediated gene transfection into SKOV 3 cells which does not express endogenous p53.The clones obtained were observed for their biological behavior and the colony formation when exposed to cisplatin.Results:The exogenous wild type p53 gene expressed stably in the cells.The growth rate and colony formation of these transfected cells were suppressed significantly by exogenous p53 gene and the percentage of phase G 1 cells increased.There was an increasement in the sensitivity to cisplatin of the transfected cells.Conclusion:Exogenous wild type p53 gene can suppress the growth of human ovarian cancer cells and increase the efficacy of cisplatin against the cells.
3.Research on relationship between abnormal regulation of histone acetylation and depression in rats

Aim To study the role of histone acetyla-tion and its involvement in the depression-like behav-iors of rats induced by chronic unpredictable stress (CUS ). Methods Thirty male Sprague Dawley (SD ) rats were randomly divided into control group and model group.The method of solitary condition with CUS for consecutive 28 days was used to establish the rat depression model.The open-field test (OFT)and the forced swimming test (FST)were used to evaluate the depressive behaviors of rats;the real time PCR was used to detect the change of HDAC2 mRNA, and Western blot was used to determine the protein expres-sions of H3,H4,acH3 and acH4 in the prefrontal cor-tex and hippocampus of rats.Results Model group showed obvious depression-like behaviors with decrea-sing locomotive activity in OFT (P <0.01 )and in-creasing immobility time in FST (P<0.01),up-regu-lating the mRNA and protein expression of HDAC2 (P<0.0 1 ),and down-regulating the protein expression of acH3 and acH4 (P<0.01)in the prefrontal cortex and hippocampus significantly,compared with control group.Conclusion The mechanism of depressive be-haviors of rats induced by CUS may be associated with down-regulating the level of histone acetylation modifi-cation.
4.Survey on Anxiety and Depression Status in Junior Standardized Training Nurses and Exploration of the Countermeasures for Humanistic Care

Objective:To understand the anxiety and depression status of junior standardized training nurses and its influencing factors,to explore the humanistic care strategies for anxiety and depression,and thus to provide the basis for improving the psychological state of the nurses and the content of standardized training.Methods:Across-sectional questionnaire survey were conducted among 161 standardized training nurses in one Three-A hospital using Self-rating Depression Scale (SDS) and Self-rating Anxiety Scale (SAS).Results:The positive rate of depression and anxiety in 161 standardized training nurses were 51.60% and 34.70%,respectively.The coarse average scores of depression and anxiety were (39.73 ± 8.55) and (37.21 ± 8.64),respectively,which were higher than Chinese norm (33.46 ± 8.55) and (29.78 ± 6.46) (P < 0.05).Family burden,clinical profes-sional knowledge and skills and work environment had influences on depression;family burden and the differences of rotation departments had influences on anxiety.Conclusion:The formulation of content of standardized train-ing should consider the different characteristics of rotation department.We should create a good working atmo-spherefor standardized training nurses,improve the training of professional knowledge and skills on the basis of paying attention to humanistic care,strengthen the training of psychological quality and psychological counseling.
5.Research progress on indoleamine 2,3-dioxygenase 1 (IDO1) inhibitors

Indoleamine 2,3-dioxygenase 1 (IDO1) is the rate-limiting enzyme in the degradation of tryptophan to kynurenine. IDO1 is highly expressed in some tumor tissues. IDO1 can deplete tryptophan in tumor microenvironment, inhibit T cell function, and mediate the immune escape of tumor cells. Thus, IDO1 is considered a potential target of tumor immunotherapy. Currently, there are several IDO1 inhibitors in clinical research studies. The mechanism of IDO1-mediated tumor immune escape and the structure of IDO1 inhibitors are summarized in this review.
6.Reliability and validity of the Chinese version of the Anticoagulation Knowledge Tool in patients with atrial fibrillation

Objective:To translate the Anticoagulation Knowledge Tool(AKT) into Chinese,and to test its reliability and validity in patients with atrial fibrillation.Methods:After obtaining the authorization of the original author, the Brislin cross-cultural translation method was used to translate from the original, back translate and cross-cultural adaptation of AKT to form the Chinese version of AKT. The reliability and validity were tested in 147 patients with atrial fibrillation from January to June in 2020 in Ruijin Hospital, Shanghai Jiaotong University School of Medicine.Results:The Chinese version retained 20 items of the original questionnaire,with the item content validity index of 0.857-1.000 and the average scale content validity index of 0.85. The Cronbach alpha coefficient was 0.812 and test-retest coefficient was 0.781.Conclusions:The Chinese version of AKT has been proved to be valid and reliable, which can be used to assess anticoagulation knowledge of patients with atrial fibrillation.
7.TNF-αmediated activation of IDO pathway involved in mechanism of depression in rats induced by CUS

Aim To investigate the relationship be-tween the depression-like behaviors of rats induced by chronically unpredictable stress( CUS) and the protein expression of tumor necrosis factor alpha( TNF-α) , in-dole-2 , 3-dioxygenase ( IDO ) and 3-hydroxyl amino acid oxygenase( HAAO) . Methods Thirty male Spra-gue Dawley( SD) rats were randomly divided into con-trol group and model group. CUS plus solitary condi-tion were used to establish the depression model. The open field test ( OFT ) and the force swimming test ( FST ) were used to evaluate the depression-like behaviors of rats ;Western blot was used to determine the protein expressions of TNF-α, IDO and HAAO in the prefrontal cortex and hippocampus of rats. Results Model group rats showed obvious depression-like be-haviors with increasing immobile time in FST ( P <0. 01) and decreasing locomotive activity in OFT( P<0. 01 ) , and up-regulating the protein expression of TNF-α, IDO and HAAO in the prefrontal cortex and hippocampus significantly ( P <0. 05 ) , compared with control group rats. Conclusion The depression-like behaviors of rats induced by CUS may be associated with the activation of IDO-HAAO pathway mediated by TNF-α.
8.Isolation and Characterization of a Temperature-sensitive p-Nitrophenol Degradation Related Genes Mutant Strain

A temperature-sensitive mutant strain was isolated after transposon mutagenesis with Tn5 and named MT54.PCR was carried out with primers designed according to the sequence of transposon,the PCR products showed that the MT54 carried transposon in the genome. The mutant grew well at 30℃in minimal medium(MM)containing p-nitrophenol(PNP)as sole carbon source,while it cannot grow at 37℃in the same medium,NO_2.detection results also proved that.Comparing the degradation rate of PNP and hydroquinone of MT54 and DLL-E4 at different temperature,it was speculated that the mutant site locate in the PNP degradation related genes.
9.Buyang Huanwu decoction promotes neuroblast migration from subventricular zone via inducing angiogenesis after ischemia.

OBJECTIVETo study the effect of Buyang Huanwu decoction (BYHWD) inducing angiogenesis on the neuroblast migration from the subventricular zone and its mechanisms after focal cerebral ischemia.
METHODThe middle cerebral artery occlusion (MCAO) was performed to mice for 30 minutes to establish the model. The rats were divided into sham group, model group, BYHWD group and endostatin group. BYHWD (20 g x kg(-1), ig) and endostatin (10 μg, sc) were administered 24 h after ischemia once a day for consecutively 14 days. At 14 d after ischemia, the density of micro-vessel and the number of neuroblasts in the ischemia border zone were determined by immunofluorescence staining. The mRNA and protein expression of cell-derived factor-1 (SDF-1) and brain-derived neurotrophic (BDNF) were examined by real-time PCR and Western blot.
RESULTCompared with the model group, BYHWD significantly increased the density of micro-vessel and the number of DCX positive cells in the ischemia border zone (P < 0.01), and significantly increased the SDF-1 and BDNF mRNA and protein expression (P < 0.01). Compared with BYHWD group, endostatin significantly reduced the density of micro-vessel and the number of DCX positive cells in the ischemia border zone (P < 0.01), as well as the SDF-1, BDNF mRNA and protein expression (P < 0.01).
CONCLUSIONBYHWD could promote the neuroblast migration from the subventricular zone via inducing angiogenesis after cerebral ischemia, the mechanism may be correlated with up-regulating the expression of SDF-1 and BDNF.
