Journal of Practical Stomatology 1996;0(02):-
Objective:To investigate the root canal anatomy of one-rooted mandibular second molars. Methods:The root canals of 18 one-rooted mandibular second molars were visualized on radiographs and clinical examination, some specimens were cut at the mid-root level and observations were made on the coronal root section. Teeth with C-shaped canal system were categorized by using a modification of Melton classification.Results:12 molars exhibited C-shaped canals.2 molars were classified as category Ⅰ,5 as Ⅱ and another 5 cases as Ⅲ.Conclusions:C-shaped canals in mandibular second molars vary in canal number and shape.

Objective To explore the relationship between single nucleotide polymorphisms (rs12953-717) of SMAD7 and susceptibility of non-small cell lung cancer (NSCLC) in Chinese Han population. Methods A single nucleotide polymorphisms (rs12953717) from SMAD7 was detected via Sequenom system in 528 NSCLC cases and 762 healthy controls.Data was statistically analyzed by unconditional Logistic regression method. Results rs12953717 had significant differences between non-small cell lung cancer patients and the controls.Compared with CC/TT (CC combined with TT) genotype, the adjusted odds ratio for the CT genotype was 4.107 (95% CI:3.206 ~ 5.260,P =0.000 1).Smokers had a 2.004 odds ratio (95 % CI =1.583 ~ 2.537,P =0.000 1) of NSCLC compared with the controls. There was a 10.074-fold increased risk of NSCLC among the subjects with CT genotype and smokers. Conclusion The polymorphism of rs12953717 may have relation with risk of NSCLC.Heterozygote (CT) is a susceptibility genotype of NSCLC.Smoking is one of the risk factors of NSCLC.Smoking and CT genotype have synergistic effects on NSCLC susceptibility.

Objective: To investigate the roots and canals morphology o f extracted maxillary first premolars. Methods: The root morphol ogy and the root canals of 115 extracted maxillary first premolars were visualiz ed on radiographs taken in the mesio-distal direction, then the teeth were cut transversely and root sections were examined, the root canal systems was analyse d with Vertucci’s classification. Results: 74(64%) teeth were w ith one root, 41(36%) with two roots, all teeth with two roots had two canals a nd each canal with one apical foramen,〔TypeⅠ(1)〕.39% of the single-root teet h demonstrated one canal, whereas 61% of the single-root teeth had two canals, 〔TypeⅡ(2-1), TypeⅣ(2) and TypeⅤ(1-2)〕.Conclusions: The roots of maxillary first premolars possesses a variety of canal system types.

Chinese Journal of Clinical Laboratory Science 2006;0(02):-
Objective The aim of this study was to establish a new method for genotyping ATP7B Arg778Leu gene mutation that does not require RFLP PCR or sequencing.Method 4-primer amplification refractory mutation system (ARMS)-PCR was performed to screen the Arg778Leu mutation in 47 unrelated Wilson's disease (WD) patients and 30 unrelated healthy controls.Direct sequencing was used to confirm the specific amplification products. Results PCR products were visualized on agarose gel electrophoresis.Among the 47 WD patients,4 were homozygous and 14 were heterozygous for this mutation. The total mutation rate was 38.3% (18/47).The results of direct sequencing completely consisted with the results of 4-primer ARMS-PCR.Conclusions The ATP7B Arg778Leu gene mutation is a hot spot for the research of Chinese WD patients.4-primer ARMS-PCR is a fast convenient and accurate method for typing mutation in high throughput population screening. This approach can be used to detect other point mutations.
6.Application of microfluidic chip technology in pharmaceutical analysis:A review
Journal of Pharmaceutical Analysis 2019;9(4):238-247
The development of pharmaceutical analytical methods represents one of the most significant aspects of drug development. Recent advances in microfabrication and microfluidics could provide new approaches for drug analysis, including drug screening, active testing and the study of metabolism. Microfluidic chip technologies, such as lab-on-a-chip technology, three-dimensional (3D) cell culture, organs-on-chip and droplet techniques, have all been developed rapidly. Microfluidic chips coupled with various kinds of detection techniques are suitable for the high-throughput screening, detection and mechanistic study of drugs. This review highlights the latest (2010–2018) microfluidic technology for drug analysis and dis-cusses the potential future development in this field.

BACKGROUND:How to promote wound healing is always the research focus of the surgical physicians in the clinic. Recombinant human epidermal growth factor (rhEGF) can effectively promote wound healing. However, as a biological agent, it is easy to be decomposed under normal temperature.
OBJECTIVE:Toexplorethe mechanisms underlying the promotion of wound healing in the back of rabbits by bletila carrying exogenous rhEGF.
METHODS:Model rabbitswith ful-thickness skin defects in the back were treated with bletila carrying exogenous rhEGF (combined treatment group), bletila, rhEGF, or saline (control group).
RESULTS AND CONCLUSION:The time of wound healing was the shortest (P< 0.05) and the wound healing rate was the highest (P< 0.05) in the combined treatmentgroup. On postoperative days 3 and 10, newly formed granulation tissue, capilaries, and colagenous fibersshowed by hematoxylin-eosin staining staining and Masson staining and the strongest immunoreactivity of vascular endothelial growth factor determined by immunohistochemical staining were found in the combined treatment group (P< 0.05). These findings confirm that bletila carrying exogenous rhEGF promotes wound healing by accelerating the forming of granulation tissue, new-born capilaries, and colagenous fibers, and the effects are superior to either of them alone.

Objective:To study the protective effect of Ginkgo biloba L.extract(EGB) on PC12 cell damage induced by rotenone and 1-methyl-4-phenylpyridium(MPP +). Methods: PC12 cells damage model was induced with different concentrations of rotenone and MPP + and were treated with EGB . Then cell morphology was observed; survival rate and ultracellular dopamine (DA) level were determined by MTT assay and ELISA assay, respectively. Results: MPP + -induced damage was attenuated by pre- or co-treatment with EGB at 0.35,0.70 and 1.40 mg/ml, while rotenone-induced damage was attenuated with EGB only at 1.40 mg/ml.The DA concentrations of extracellular at 2 mmol/L MPP + (6.875?0.201) ng/ml and 10 ?mol/L rotenone (5.321?0.167) ng/ml increased to ( 7.595?0.139 ) ng/ml (P

Objective To investigate the effect of autophagy on histone-mediated apoptosis of human proximal tubular endothelial cells(HK-2).Methods To investigate the effect of histones on the autophagy and apoptosis,HK-2 cells were treated with increasing concentrations of histones. The rate of apoptosis and the expressions of autophagy-related protien LC3Ⅱ and Beclin1 in HK-2 cells were detected by using flow cytometry and immunoblotting assay, respectively. To further confirm the effect of autophagy on apoptosis of HK-2 cells, cells were incubated with histones after one hour pretreatment with 10 mmol/L 3-MA,a pharmacological inhibitor. The rate of apoptosis and the activity of caspase-3 of HK-2 cells were detected separately by using flow cytometry and immunoblotting assay. Results Histones significantly enhanced apoptosis of HK-2 cells in a dose-dependent manner, with the increased expressions of LC3Ⅱ and Beclin1.Blockage of autophagy by 3-MA significantly increased the apoptosis of HK-2 cells and the activity of caspase3.Conclusion Autophagy in proximal tubules protects against apoptosis induced by histones, with potential value in acute kidney injury (AKI).
