1.Model of B immunoblastic lymphomas in the Hu-PBL-SCID mice.

OBJECTIVETo constitute a model of B immunoblastic lymphomas in the Hu-PBL-SCID mice.
METHODSThe SCID mice were reconstituted by intraperitoneal injection (i.p.) of 5 x 10(7) human lymphocytes from Epstein-Barr virus (EBV) seronegative individuals. After one week, the SCID mice were inoculated with EBV by i.p. injection, and subjected to the investigation of whether there was any tumor in the abdomen of such SCID mice four weeks later. The characteristics of the found tumor was observed by the methods of Hematoxylin-eosin (HE) stain, immunohistochemical staining and polymerase chain reaction (PCR).
RESULTSCompared with the control groups, all the EBV-infected Hu-PBL-SCID mice had abdominal solid tumors [(32 +/- 12.5) mm3] developed, often located in the liver. HE staining and immunohistochemical staining showed the tumors were human B cell lymphomas. EBV DNA could be detected in the tumors by the PCR.
CONCLUSIONSThe model of B immunoblastic lymphomas in the Hu-PBL-SCID mice is successfully constituted, and may well be useful to the human tumor immunological study.

2.Repair of canine segmental mandibular defects by using autogenous bone marrow stromal cells and?-tricalcium phosphate

Objective To repair segmental mandibular defects with autogenous bone marrow stromal cells(BMSCs)and?-triealcium phosphate.Methods Isolated BMSCs were in vitro expand- ed.A 3 cm-long segmental mandibular defect was created at right mandible in 12 canines,of which de- fects in six canines were repaired with BMSCs and?-tricalcium phosphate(?-TCP)and that in other six cases repaired with?-TCP,which was used as control.The engineered bone was evaluated by X-ray, CT,DXA,gross and histological examination,immunohistochemistry and biomechanical test 4,12,26,32 weeks after operation respectively.Results In induced BMSCs,histochemistry showed AKP activity. Oral X-ray showed obvious callus formation 4-26 weeks after operation in experimental group but minimal bone formation in control group.At 32 weeks after operation,gross observation,X-ray and CT demonstra- ted well bony-union in experimental group but bony-nonunion in control group.DXA indicated that the bone density of experimental group was significantly higher than that of control group.Biomechanical test revealed no statistical difference upon mechanical strength of mandibula between experimental group and normal group.Conclusions Canine segmental mandibular defects can be well repaired with the tissue- engineered bone generated by autogenous osteogenic BMSCs and?-TCP scaffold.
3.Pancreatic cancer cell-secreted exosomes promote apoptosis of β cells via activation of mitochondrial apoptotic pathways

AIM:To investigate the effects of exosomes secreted by pancreatic cancer cells on the viability and function of βcells and the possible mechanism .METHODS:ExoQuick-TC kit was used to extract exosomes in the super-natants of mouse pancreatic cancer Pan 02 and MPC-83 cells, and the extracted exosomes were identified by transmission electron microscopy.Fluorescence-labeled exosomes were incubated with mouse insulinoma MIN 6 cells for 48 h to detect whether exosomes secreted by pancreatic cancer cells were uptaken by MIN 6 cells.MTT and glucose-stimulated insulin se-cretion ( GSIS) assays were conducted to examine cell viability and insulin secretion of MIN 6 cells after incubating with ex-osomes.The expression of miR-204 and Bcl-2 mRNA in MIN6 cells was detected by qPCR .The protein expression of Bcl-2, Bax, caspase-3 and cytochrome C (Cyt-C) in MIN6 cells was determined by Western blot .RESULTS:The results of transmission electron microscopy showed that both Pan 02 cells and MPC-83 cells secreted exosomes , and Pan02 cells secre-ted more.The co-incubation results of fluorescence-labeled exosomes and MIN6 cells confirmed that MIN6 cells were able to ingest large amounts of exosomes secreted by pancreatic cancer cells .The results of MTT and GSIS assays showed that the viability and the level of high glucose-stimulated insulin secretion of MIN 6 cells in exosome treatment group significantly decreased compared with nontreatment group (P<0.01).The results of qPCR showed that the exosomes secreted by pan-creatic cancer cells were rich in miR-204, and the mRNA expression of Bcl-2 in MIN6 cells was significantly down-regula-ted by exosome incubation ( P<0.01) .The results of Western blot showed that the protein expression of Bcl-2 in the MIN6 cells treated with exosomes was significantly down-regulated (P<0.05), and the protein levels of Bax, cleaved caspase-3 and Cyt-C in exosomes treatment group were significantly up-regulated ( P<0.01 ) .CONCLUSION: Pancreatic cancer cells secrete exosomes .The exosomes secreted by pancreatic cancer cells are ingested by βcells, and reduce the viability and insulin secretion of βcells.The mechanism may be related to the increase in exosomal miR-204 in the βcells.In-creasing miR-204 may inhibit the expression of Bcl-2 and promote the activation of mitochondrial apoptosis in βcells.
4.Epidemiological study on risk factors of hepatitis E in Yantai, Shandong province

Objective To study the prevalent characteristics and risk factors of viral hepatitis E in Yantai and the relative for strategy on viral hepatitis E control in the area. Methods Data on viral hepatitis E incidence reported by the Notifiable Infectious Disease Reporting System in 2005-2009 was analyzed. 2028 persons were chosen for hepatitis E virus (HEV) antibody detection by enzyme linked immunosorbent assay method. RT-nPCR method was applied to obtain the sequence of HEV in HEV cases. A case-control study was used to identify the risk factors of HEV infection.Results The distribution of HEV cases was sporadic in Yantai, and the annual incidence rate was 5.70/100 000, with spring as the prevalent season. Farmers were the main population involved and with those over the age of 40. Regional distribution was mainly along the coastline. Data from the sequential analysis showed that gene type of local patients was type 4 and healthy person whose HEV-IgM was positive was type 1. Finding from the case-control study suggested that eating seafoods,living condition in the households and unhealthy habits played important roles in the infection of HEV.Results from multiple logistic regression showed that eating seafood, with eat-out history, drinking alcohol and un-boiled water were the main risk factors in the infection of HEV. Conclusion The level of HE in Yantai will maintain in a high level, but there is no chance of epidemic outbreak of HE in large range. Population of incidence mainly concentrates on farmers.
5.Species analysis of fungi isolated in a laboratory from patients with mycoses during 1960-2006

Objective To analyze fungal isolates from patients with superficial fungal infections during 1960-2006.Methods Fungal strains isolated from patients with superficial (mucocutaneous and cutaneous)fungal infections and identified in the Medical Mycology Clinical Laboratory,Department of Dermatology and Venereology,Union Hospital,from 1960 to 2006 (data from September 1991 to July 1992 were unavailable),were subjected to a classification and statistical analysis.Clinical samples for mycological examination were taken from outpatients or inpatients of different departments in hospitals of Hubei province and surrounding areas.Morphological,physiological and biochemical methods were applied for species identification.Results A total of 11 989 Candida strains were isolated,which belonged to 23 species and 16 genera.They fell into 3 groups,i.e.,dermatophytes,Candida and yeasts (including Malassezia),and non-dermatophyte moulds.Since 287 strains of moulds were suspected to be contaminating fungi,11 702 residual isolates were analyzed.Of the analyzed isolates,Candida species (5642/11 702,48.2% )and dermatophytes (5279/11 702,45.1% )predominated,followed by yeasts (449/11 702,3.8%) and Malassezia species (332/11 702,2.8%).The most frequently isolated species was Trichophyton rubrum (3865/11 702,33.0%),Candida albicans (3110/11 702,26.6% ) and non-albicans Candida species (2532/11 702,21.6% ).Dermatophyte strains were mostly isolated from lesions of smooth skin with an exception of palmoplantar and interdigit regions (1787/5279,37.7%).The most common dermatophyte species was Trichophyton rubrum,followed by Trichophyton violanceum.Candida was mainly isolated from mucous membrane lesions (4099/5642,72.7%),with Candida albicans being the predominant species.Conclusions Candida species and dermatophytes predominate in patients with superficial fungal infections during 1960-2006,with Trichophyton rubrum being the most common species.
6.Study of ATP7A gene in a Chinese family with Menkes disease

Objective To analyze the A TP7A gene mutations in 2 related Chinese patients with Menkes disease (MD) and other members of the family and their hereditary features. Methods Two patients were clinically diagnosed as having MD. All 23 exons and exon-intron boundaries of ATP7A gene were polymerase chain reaction (PCR)-amplified and directly sequenced for genomic DNA extracted from the peripheral blood of both 2 patients and other members of the family; healthy controls were employed too. The mutations were proved by fluorescence quantitative PCR. Results Gross deletions from exon 2-12 were found in these 2 patients,respectively; their mothers,grandmother and aunt with normal phenotype carried those heterozygous mutations in the same site of A TP7A gene.Conclusions The 2 patients with MD are identified by gene and gross deletions from exon 2-12 are reported.
7.Relationship between DNA degradation and postmortem interval of corrupt corpse.

OBJECTIVE:
To study the relationship between DNA degradation and postmortem interval of corrupt corpse.
METHODS:
By determining the marrow DNA content with histochemical technique and image analysis.
RESULTS:
The content of marrow DNA decreased gradually with prolongation of postmortem interval, and it evencould be detected till 14 days after death.
CONCLUSION
There was a linear relationship between the degradation rate of the nuclear DNA and postmortem interval.

8.Effect of continuous nursing support on breast cancer patients with radical mastectomy

Objective To investigate the effect of the continuous care support intervention on breast cancer patients with radical mastectomy.Methods Totals of 107 patients undergoing elective radical mastectomy were randomly divided into intervention group ( n =54 ) and control group ( n =53 ).Both of two groups of patients were treated on the basis of conventional care according to the doctors' advice before and after surgery.The intervention group received the continuous individual nursing intervention model,included preoperative interactive support intervention,postoperative pain assessment support intervention and rehabilitation support intervention after discharge. Intervention effects of two groups were observed and compared. Results The anxiety,positive response and negative response of two groups patients were no significant differences before intervention (P>0.05).After intervention,the scores of anxiety,positive response and negative response in intervention group respectively were (35.28 ± 5.43 ),(39.82 ±4.03 ) and (25.61 ± 5.12),and that of control group were (42.73 ± 6.72),(33.15 ± 5.62) and (31.75 ± 6.38),and the difference was statistically significant ( t =6.313,7.065,5.495,respectively; P < 0.01 ).The duration of postoperative different period pain score,first off the bed time,sleep time,upper extremity swelling and hospitalization days were statistically significant between two groups (P <0.01 ).And the postoperative recovery state of upper limb function after six months,and life quality score after one year in intervention group were significantly better than that of control group ( P<0.01 ).Conclusions Continuous individualized specialized nursing intervention support model can promote physical function in patients with early recovery and improve quality of the prognosis life and the treatment outcome.
9.Influence of different catheterization times on patients with time selecting operation

Objective To explore the effects of difierent preoperative catheterization times to blood pressure,heart rate and occurrence rate of catheter-related bladder discomfort during postoperative emergence peiled among patients with time selecting operation.Methods 90 patients were randomly divided into three groups,30 patients in each group:the routine catheterization group(group A),the catheterization after premeditation for 15 minutes before anesthesia(group B),and the catheterization after anesthesia(group C).Physiological parameters of subjects(including systolic pressure,SBP;diastolic pressure,DBP;heart rate,HR)Were monitored pre-operatively,intra-operatively and post-operatively;catheter-related bladder discomfort was evaluated at Oh.1h,2h and 6h after operation.Results Blood pressure and heart rate at three measurement times(pre-operatively,intra-operatively and post-operatively)in group A had statistically significant meanings,respectively(P<0.05).There was no difference in group B and group C(P>0.05).The incidence of catheter-related bladder discomfort was higher in group C than that in group A and B(P<0.05).and the difference had obviously statistical meaning(P<0.05).Conclusions Group B (catheterization after premeditation for 15 minutes before anesthesia)has the least effects of blood pressure,heart rate and catheter-related bladder discomfort on patients with time selecting surgery.So catheterization after premedication for 15 minutes before anesthesia is the choice with less influence to the patients.
10.Investigation and nursing of mental state in intrauterine artificial insemination patients

Objective To investigate the psychologic status of irritability, depression and anxiety in patients who underwent intrauterine insemination (IUI) and to provide mental nursing care. Methods This study included 120 infertile patients who underwent IUI treatment from August 2008 to September 2009 in infertility clinics. All patients were divided into two groups randomly and assessed with the Irritability,Depression and Anxiety Scale (IDA). The 60 patients in control group were treated according to the standard IUI procedure, while the patients in study group were given mental nursing additionally. IDA scores before and after psychological intervention and clinical pregnancy rate were compared between the two groups. Results The IDA scores decreased obviously after mental nursing in study group (P <0.05 ). The clinical pregnancy rate per cycle was much higher in study group than that in control group ( P < 0.05). Conclusions IDA can be applied to the psychological investigation of IUI patients. Efficient mental nursing care is beneficial to the success of IUI.
