1.The Study on Problem of the Prosthesis Fitting in the Non-ideal Stump of the Lower Limb Amputee

The prosthesis fitting were discussed in this paper for 48 un-reoperation of 91 cases the non-ideal stump of the lower limb amputees hospitalized in the past six years. The author suggests that silicon socks,the supplementary hang band,a combinative use of the prosthesis and orthosis,adjustment aligment of the prosthesis provide a good method for prosthesis fitting of the non-ideal stump of the lower limbamputees. It was able to wearing a good prosthesis and bring the compensatory functions into full play.

Objective T Iymphocytes were considered to be activated and involved in the ischemia-reperfusion injury during lung transplantation.Carbon dioxide pneumoperitoneum was shown to have inhibitory activity on the immune system.This study was designed to_investigate the effects of the effects of the therapeutic hypercapnia on the T Iymphocytes of rats in which ischemia-reperfusion injury occurred during lung transplantation.Melhods Sixteen Wismr rats weighed 300 to 400 g were randomized into control group(8 rats) or therapeutic group (8 ras)after transplantaion.Animals in both grotups were Oven inluded nitrogen(50%)and oxygen N2+(50%) at baseline. Animats in the control groap were given irked nitrogen (50%)and oxygen(50%)throughout the experiment ,and that in the thera-peutic group were given mixed gas which was composed of nitroged(40%),oxygen(60%)and carbon dioxide in appropriate concentra-tion to keep arterial partial pressure of carbon dioxide (PaCO2)at 80-100 mm Hg and FiO2 at 50%after reperfusion.All of the ani-mals were observed for 90 minutes after reperfusion.Mean arterial pressure(MAP) and arterila partial pressure of oxygen(PaO2) were recorded at baseline and every 15 minutes during the period of reperfusion.The expression of CD3,CD4 and CD28 in the peripheral blood was,examined,and the concentrations of Ifn-у,IL-2,IL-4 and IL-1O in the homogenate were measured after the experiment. 3.Compatibility Rules of Traditional Chinese Medicine in Prevention and Treatment of Scleroderma Based on Data Mining

This study was aimed to analyze compatibility rules of Chinese drugs and prescription design in the pre-vention and treatment of scleroderma. The database of scleroderma treated by Chinese medicine was set up. And then, association rules were used to analyze the prescription compatibility rules. The results showed the highest fre-quency of drugs in the treatment of scleroderma were Astragalus, Angelica sinensis and Salvia. The tonifying and re-plenishing medicinal, blood-activating and stasis-resolving medicinal and exterior-releasing medicinal were with the highest frequency. The commonly used formula in the treatment of scleroderma was Y ang-He decoction. For herbal groups, occurrence of the highest was the combinations of Ramulus Cinnamomi-Astragalus and Carthamus tinctorius L.-Astragalus. Three kinds of herbals were commonly coupled, such as Polygonum multiflorum Thunb. and Caulis Spatholobi with Salvia, P. multiflorum Thunb. and R. Cinnamomi with Salvia, P. multiflorum Thunb. and Astragalus with Salvia, prepared Radix Rehmanniae and Codonopsis with C. tinctorius L., prepared Radix Rehmanniae and Codonopsis with R. Cinnamomi, Codonopsis and Radix paeoniae rubra with R. Cinnamomi. Four kinds of herbals such as R. Cinnamomi, Glycyrrhiza uralensis Fisch., Astragalus and Codonopsis were commonly coupled. It was con-cluded that compatibility rules of Chinese medicine in the prevention and treatment of scleroderma laid the founda-tion for herbal medicine selection and prescription design.

5.Angiogenesis in acute myocardial infarction rats after vascular endothelial growth factor 121 gene therapy

BACKGROUND: Previous studies suggest that vascular endothelial growth factor 121 may be an optimal target gene for thetreatment of acute myocardial infarction.OBJECTIVE: To investigate effect of direct myocardial injection of adenovirus recombinant human vascular endothelial growthfactor 121 gene (Ad-hVEGF121) on myocardial infracted rat heart structure, function and angiogenesis.METHODS: Totally 78 male SD rats were randomly divided into the sham-surgery (n=18), acute myocardial infarction (n=24),Ad-VEGF121 (n=19) and normal saline (n=17) groups. Among them, left anterior descending coronary arteries of the latter threegroups were ligated to prepare acute myocardial infarction models and rats were randomly selected to receive Ad-hVEGF12 ornormal saline via three points in the cardiac muscle at the 10-15 minutes after ligation. The chest was exposed without ligation inthe sham-surgery group.RESULTS AND CONCLUSION: At 2 weeks after injection, cardiac ultrasound showed that, compared with the sham-surgerygroup, the number of new capillaries, body weight and left ventricular mass / body weight of the acute myocardial infarction,Ad-hVEGF121 and normal saline groups were obviously increased (P < 0.05 or P < 0.01), especially those received transfectedrAd-hVEGF12, had higher density of blood capillaries than those of the normal saline and acute myocardial infarction groups.However, there were no obviously differences between each group in infarct size, cardiac structure or functions. The directmyocardial injection of Ad-VEGF121 can significantly promote the formation of new blood vessels within the myocardium.
6.Expression of Disabled-1 in human breast cancer cells and its role in cell cycle

Objective:To explore the expressions of Disabled-1 (Dab1 )in human breast epithelial cells and breast cancer cells,and to clarify its role in cell cycle.Methods:Real-time PCR was used to analyze the Dab1 mRNA expressions in breast epithelial cells MCF-10A and breast cancer cells MCF-7,BT-549,and MDA-MB-231. The Dab1 protein expressions in those cells were tested by Western blotting method. The BT-549 cells at logarithmic growth phase were divided into control,pKH3,and pKH3-Dab1 groups;the cell cycle was investigated by flow cytometry.Results:The Real-time PCR results showed that the Dab1 mRNA expression levels in MCF-7 cells (0.504 ± 0.037),BT-549 cells (0.302 ± 0.027),and MDA-MB-231 cells (0.330 ± 0.031 )were reduced compared with MCF-10A cells (0.998±0.020)(P <0.05).The Western blotting results showed that the Dab1 protein expression levels in breast cancer cells MCF-7 (0.134±0.014),BT-549 (0.076±0.01),and MDA-MB-231 (0.074±0.005)were reduced compared with MCF-10A cells (0.227±0.021)(P <0.05).Compared with control group and pKH3 group,the cell cycle in pKH3-Dab1 group was inhibited at G1 phase detected by flow cytometry analysis. Conclusion:The expression of Dab1 is down-regulated in breast cancer cells,and the over-expression of Dab1 can inhibit the cell cycle at G1 phase.
7.Relationship Between the Central Hypotensive Effect of Acupuncture or Melatonin and the Changes of Medullary Amino Acid Neurotransmitter

To investigate whether changes of amino acid neurotransmitter releases in the rostral ventrolateral medulla (rVLM) were related to acupuncture and to the antihypertensive effect of melatonin (Mel) microinjected into the anterior hypothalamic area (AHA) of rats with stress-induced hypertension (SIH), as well as to compare and analyze the relationship between the both antihypertensive mechanisms of acupuncture and of Mel in the AHA. Methods: Animal model of SIHR was made by electric foot shocks combined with noises. Electroacupuncture (EA) was used and Zusanli acupoint of both side was selected. The technique of drug microinjection into the brain was used to observe the change of blood pressure (BP), and synchronously, brain microdialysis was performed for collecting dialysate samples, and then the concentration of amino acid neurotransmitters in the dialysate samples was determined by high performance liquid chromatography combined with fluorescent detection (HPLC-FD). Results: After the animal received stress treatment, the BP elevated, synchronously, the release of glutamate (Glu) in the rVLM increased, and when EA was performed, the elevated BP of the rats with SIH decreased,simultaneously, the release of Glu in the rVLM decreased also. After Mel was microinjected of into the AHA of the SIHR, the elevated BP attenuated, meanwhile, the release of Glu decreased,and those of γ-aminobutyric acid (GABA) and taurine (Tau) increased in the rVLM.Administration ofbicuculline, an antagonist of GABAA receptor, into the rVLM prior to microinjection of Mel into the AHA could partially block the depressor effect of Mel in the AHA. Conclusion: The decrease in the release of Glu in the rVLM contributes to the antihypertensive effect of both acupuncture and Mel in the AHA in the rats with SIH, and the increase in the release of GABA and Tau in the rVLM is also important to Mel in the AHA.
8.Effect of Chronic Alcohol Consumption on Left Ventricular Myocardial Collagen Content and Diastolic Function in Rats

Objective:To investigate the effect of chronic alcohol consumption on both left ventricular myocardial collagen and diastolic function in rats,and their relationship thereof.Methods:Twenty-four male Wistar rats were randomly divided into 2 groups:control group(n=12)and ethanol group(n=12).The changes in cardiac diastolic function were evaluated by echocardiography and tissue Doppler imaging(TDI).The value of myocardial hydroxyproline content was determined by hydroxyproline reagent kit.The expressions of collagen Ⅰ and collagen Ⅲ mRNA were detected by RT-PCR analysis.Results:It was found that mitral E and mitral annulus Ea were decreased,mitral annulus Aa was increased,and isovolumic relaxation time(IVRT)was prolonged in the ethanol group compared with those in control group(P<0.05).The value of Ea/Aa ratio was greater than 1 in control group and less than 1 in ethanol group(P<0.01).It was found that myocardial hydroxyproline content,collagen Ⅰ,collagen Ⅲ mRNA expression and their ratio significantly increased in ethanol group compared with those in the control group(P<0.01).There was positive correlation between hydroxyproline content,collagen Ⅰ,collagen Ⅲ mRNA expression,and collagen Ⅰ /collagen Ⅲ mRNA ratio with IVRT(P<0.05),and negative correlation between hydroxyproline content,collagen Ⅰ,collagen Ⅲ mRNA expression,and collagen Ⅰ /collagen Ⅲ mRNA ratio with the Ea/Aa ratio(P<0.01).Conclusion:Chronic ethanol consumption can induce increase in left ventricular myocardial collagen synthesis and impairment in diastolic function in rats.Left ventricular diastolic dysfunction correlates with increase in myocardial collagen synthesis positively.
9.Correlation of survival time and C-reactive protein after artificial tracheal replacement in rabbits

BACKGROUND:C-reactive protein has been shown to rapidly increase during the occurrence of inflammation and tissue injury, and can indicate the degree of inflammatory reaction.
OBJECTIVE:To analyze the correlation between survival time and C-reactive protein in rabbits after transplantation of polytetrafluoroethene artificial trachea with support ring.
METHODS:The cervical trachea of rabbits was replaced by polytetrafluoroethene artificial trachea with support ring. Survival time of the rabbit, and the changes in serum C-reactive protein at 1-7 days after transplantation were observed. Linear regression was used to assess the univariate association between serum C-reactive protein and survival time.
RESULTS AND CONCLUSION:The linear correlation was observed between changes of serum C-reactive protein and survival time in rabbits with artificial trachea replacement operation. C-reactive protein levels in rabbits with<13 days of survival time were increased and positively associated with the number of days after transplantation. However, C-reactive protein levels in rabbits with>13 days were decreased and negatively associated with the number of days after transplantation. In rabbits with positive correlation and negative correlation, the median survival time and 95%confidence interval (CI) were respectively 10 days (95%CI 8.614-11.386 days) and 27 days (95%CI 23.970-30.030 days). The survival rate in negative correlation group was significantly higher than positive correlation group (x2=29.364, P<0.01). Results suggested that the prolonged survival time of rabbits after artificial trachea replacement operation was related to the decreased concentration of serum C-reactive protein.
10.Diagnostic value of lower limber Short-latency somatosensory evoked potential and F wave in proximal diabetic neuropathy

Objective To explore the diagnostic value of tibial nerve short-latency somatosensory evoked potential( SLSEP)and F wave in proximal diabetic neuropathy.Methods Excluding stroke,waist cervical vertebra sickness and nerve-muscle diseases caused by other diseases,there were clinical signs of neuropathy but normal nerve conduction EMG(Ⅰ group)32 cases,there weren't clinical signs of neuropathy normal nerve conduction EMG(Ⅱ group)30 cases and abnormal nerve conduction EMG(Ⅲgroup)30 cases.Tibial nerve SLSEP and F wave were measured in 92 patients and 30 normal persons.Results The highest whole abnormality rate of tibial nerve SLSEP's and F wave's all parameters was the sub-group (there were neuropathy symptom lower limbs n=43)of Ⅰ group(97.7% (42/43)).In Ⅰ group Fdur,NI3CV,N24CV, N13-N24CV,N9-N24CV were significantly different from the control group (q value:5.887 and 6.780,-4.568 and-5.062,-6.799 and -6.905,-5.978 and -5.609,-5.433 and -5.190, P<0.01);The abnormality rate of Fdur and N13-N24CV (25.0%(15/60),23.3% (14/60))in Ⅱgroup were significantly different from the control group (0,1.7%(1/60))also (x2=17.143,12.876, both P=0.0050).In Ⅲ group,except for N9-N24CV,N13 and N24 wave amplitude other parameters'abnormality rates were significantly different from the control group,and the abnormality rate of N9CV was the highest (76.7%(46/60)).Conclusion(1)SLSEP can provide information of proximal sensory nerve segment.(2)SLSEP and F wave can improve the diagnosis rate of proximal diabetic neuropathy,and the diagnostic value is particularly significant to the diabetes patients who have clinical signs of neuropathy but normal nerve conduction EMG.
