Objective To study the mechanism of myocardical aging. Methods Wistar rats used were divided aging group (22 months old) and adult group (7 months old). Qualitative morphological changes of organelles of myocardium were observed by TEM. Quantitative morphological changes of organelles of myocardium were determined by stereological method, Ca 2+ regulation of organelles of myocardium were analysed by EDS. Results Compared with adult group, aging group:(1) The nuclei were indented, myofibril were arranged irregularly, intercalated disk were separated, mitochondria and sarcoplasmic reticulum were swelled, lipofuscin and residual body were increased. (2) The volume of myocardium not occupied by myocyte were increased, the volume density of mitochondria and sarcoplasmic reticulum were reduced, specific surface of outer membrance of mitochondria inter membrane plus cristae of mitochondria and the membrane of sarcoplasmic reticulum were also reduced. (3) Ca 2+ in myofibril and mitochondria were increased, but Ca 2+ in sarcoplasmic reticulum were decreased. Conclusion The contractility of aging myocardium were declined, the morphological changes and Ca 2+ regulation of mitochondria and sarcoplasmic reticulum might be directly related to myocardical aging.

AIM: To assess the effect of RNAi on suppressing the exogenous reporter gene expression in mammalian neurons, and explore the effect of siRNA quantitation on interference efficiency.METHODS: Exogenous green fluorescent protein(GFP) expression vector was transferred into neuro-2a cells, and then the small interference RNA targeting GFP mRNA(siGFP) synthesized by transcription in vitro at three different concentration was used in this experiment.RESULTS: The results showed that the neuro-2a cells can be transfected efficiently and siGFP can inhibit GFP expression greatly.CONCLUSION: RNAi can be applied into mammalian neurons successfully. The research on siRNA quantitation will provide technique support for studying the gene function of neurons in the future.

(1)H-NMR and (13)C-NMR assignments of 12-oleanene-3,11-dione (compound 1) were completely described for the first time through conventional 1D NMR and 2D shift-correlated NMR experiments using (1)H-(1)HCOSY, HMQC, HMBC techniques. Based on its NMR data, the assignments of 28-hydroxyolean-12-ene-3,11-dione (compound 2) were partially revised.

Objective To compare the perioperative outcomes of laparoscopic liver resection (LLR) versus open liver resection (OLR) for hepatocellular carcinoma (HCC).Methods A total of 89 HCC patients undergoing liver resection between January 2012 and November 2016 were enrolled.Nonparametric tests were employed to compare the clinicalpathological characters and preoperative outcomes. Results No significant difference was observed in clinicalpathological features and postoperative morbidity.LLR group had shorter hospital stay (Z =4.642,P <0.01),lower serum ALT level in 1st,3rd and 5 day (Z =2.157,3.089,2.384,all P <0.05) and AST level in 1st-and 3rd-day postoperatively (Z =2.688,2.566,all P <0.05).The growth rate in serum total protein (TP) and albumin (ALB) postoperatively is higher for LLR group (y =2.348 4x + 51.696 vs.y =0.902 9 + 35.532),(y =1.539 9x + 29.68 vs.y =0.732 9x + 30.406).Conclusion LLR allows quicker liver function recovery and shortens patients' postoperative hospital stay.

Objective:To investigate the effect of blocking BRCAA1 gene expression with siRNA on the proliferation of tumor cell line MCF-7 and Rb gene expression. Methods:RNAi was employed to specifically knock down BRCAA1 expression.MCF-7 cells were transfected with complexes constructed with lipids and chemically synthesized Pre-designed anti-BRCAA1 siRNAs.The total RNA was isolated and reversely transcribed after 48 h.The expressions of BRCAA1 and Rb mRNA were determined by Real-Time PCR.Results:Compared with negative control, transfected MCF-7 cells had a 42.3% decrease in expression of BRCAA1 mRNA and an 11.1% increase in Rb mRNA expression. The inhibitory rate of MCF-7 cells proliferation was(81.6?6.1)%.Conclusion:There may be some antagonistic effect between BRCAA1 gene and Rb gene in proliferation of tumor cells, which provides a potential target for anti-tumor gene therapy.

Objective To evaluate the effect of Eucommia on hyperlipidemia and related indexes in rats, and provide animal data useful for the clinical experimental studies on hyperlipidemia. Methods Seventy-two healthy male SD rats were used in this study. One group of 12 rats fed with normal diet was chosen as normal control group, and other 60 rats were fed with high fat diet for two weeks to generate rat models of hyperlipidemia.48 of the hyperlipidemic model rats were taken and divided randomly into 4 groups, including model group, high dose Eucommia, moderate dose Eucommia, and low dose Eucommia groups. The last three groups were gavaged different dose of Eucommia, respectively.Druing this period, the other groups except the normal control group were fed with high fat diet continuously. The levels of serum TC, TG, LDL-C, and HDL-C of rats were measured on day 30 and 45.Results The serum levels of TC and LDL-C of the rats in the model group were obviously higher than those in the normal control group. The rat models of hyperlipidemia were established successfully. The three dose groups had a tendency of lowing blood lipid after 30 days.At 45 days, the levels of serum TC and LDL-C in the low and high dose groups were lower than those in the model group (P<0.01, P<0.05), (P<0.01, P<0.01).TG in the high, moderate and low dose groups were lower than that in the model group (P<0.01, P<0.01, P<0.01), but the level of the serum HDL-C was not significantly lower than that in the model group (P>0.05, P>0.05, P>0.05).Conclusions Eucommia in a dose of 0.43 g/kg, 0.86 g/kg and 1.71 g/kg administered for 30 days have a tendency to reduce the level of serum TC, TG, and LDL-C.When Eucommia is administered in a dose of 0.43 g/kg, 1.71g/kg and 3.42 g/kg for 45 days, it shows an adjuvant hypolipidemic effect.

Journal of Biomedical Engineering 2013;30(3):627-634
Masking therapy can make patients accustom to tinnitus. This therapy is safe and easy to implement, so that it has become a widely used treatment of curing tinnitus. According to surveys of tinnitus sounds, cicada sound is one of the most usual tinnituses. Meanwhile, we have not hitherto found published papers concerning how to synthesize cicada sound and to use it to ameliorate tinnitus. Inspired by the human acoustics theory, we proposed a method to synthesize medical masking sound and to realize the diversity by illustrating the process of synthesizing various cicada sounds. In addition, energy attenuation problem in spectrum shifting process has been successfully solved. Simulation results indicated that the proposed method achieved decent results and would have practical value for the future applications.
