Intracranial aneurysm often result from congenital defects of regional cerebral artery wall and increased endovascular pressure. Its occurrence and development are associated with several risk factors including genetics, smoking, hypertension, cerebral atherosclerosis and vasculitis. With the application of gene chip technology for the past few years, the research of pathogenic genes associated with intracranial aneurysms was further deepened, and some susceptibility genes closely associated with the occurrence of intracranial aneurysms have been found, and this suggests that intracranial aneurysm may be a polygenic disease caused by the combined action of multiple genes.

Objective To evaluate the radioprotective effects of Quercetin (QN) on 6 Gy X-ray irradiation-induced immune dysfunction and toxicity in hepatic tissue in rats. Methods 40 adult rats were randomly divided into 4 groups. Group Ⅰ was injected intraperitoneally with saline solution for 7 consecutive day sand served as control group. Group Ⅱ was daily injected with QN (40 mg/kg) for 7 consecutive days. Group Ⅲ was irradiated with a single dose of 6 Gy X-ray. Group Ⅳ received a daily injection of QN (40 mg/kg) for 7 consecutive days, and 1 h after the last injection rats were irradiated with a single dose (6 Gy) X-ray irradiation. The animals were sacrificed after 24 h. Lymphocyte transforming rate was measured with MTT method, and CD+4 T, CD+4 T and CD+8/CD+8 T were measured with flow cytometry method. Oxidative conditions in liver were measured with malondialdehyde (MDA), reduced glutathione hormone (GSH), supernxide dismutase (SOD) andglutathione peroxidase (GSH-Px) activities kits. HE staining was used to observe the general condition of rat's liver. Results Lymphocyte transforming rate, CD+4 T, CD+8 T and CD+8/CD+8 T in rats of Group Ⅳ were all higher than those in rats of Group Ⅲ ( F = 8.455,22.644, 18.911, P < 0.01 ). MDA content in the Group Ⅳ rat's liver was lower than that in the Group Ⅲ ( F = 10.059, P < 0.01 ) and antioxidant enzymes SOD, GSH-Px activities were higher than those in Group Ⅲ (F = 23.688,186.046,19.788, P < 0.01 ). The capillary of the hepatic lobules dilated and congested obviously in portal area, involving infiltration of polymorphonuclear leukocytes in the Group Ⅲ, while QN improved this change apparendy. Conclusions Pretreatment with Quercetin improved the irradiated rat's immune functions and protected the irradiated rats from oxidative stress to some extent.

Basic & Clinical Medicine 2006;0(08):-
Objective To find an ideal method of DNA isolation from blood and especially from clotted blood and to minimize the volume of blood collected for laboratory and clinical tests. Methods DNAs were isolated from antiagglutinated and agglutinated blood samples from auricular veins of 30 healthy subjects. The DNAs of these samples were obtained by a nonenzymatic, nontoxic procedure optimized by us and determinated by agarose gel electrophoesis and PCR. Results The yields of DNA isolated from clotted blood and antiagglutinated blood were (40.2?8.86)mg DNA/L and (39.1?10.2)mg DNA/L, and purities were 1.87?0.11 and 1.92? 0.12. The DNAs that we isolated from all samples had high molecular weight and by PCR the dimorphism of ALU alleles of the 8th intron of t-PA was easy to be obtained, so they were complete and reliable. Conclusion This method is rapid, easy, efficient and nontoxic for isolation of DNA from clotted and fresh blood and meets requirements for clinical testing and molecular biology study.

Lectin from green marine algae Enteromorpha intestinalis (EIL) was isolated by extraction with PBS, followed by ammonium sulfate fraction and a combination of ion-exchange chromatography on DEAE-52 fiber and gel filtration on SephadexG-200. Lectin showed single band on both PAGE and isoelectrofocusing electrophoresis. Its isoelectric point was 8.9 and molecular weight was 17783 tested on SephadexG-200. EIL displayed some agglutination activities with Chlamydomonas and erythrocytes of rabbit, common carp, crucian carp and human (A, B, AB, O), but the maximum agglutination was with crucian carp. The hemagglutinating activity for rabbit erythrocyte could not be inhibited by D-fructose, D-galactose, glucose, sucrose, mannose, lactose, bovine-thyroglobulin, egg albumin and gamma-globulin. The hemagglutinating activity of lectin would be showed at pH 4.0~10.2, and more potent at pH 6.9~8.0. The hemagglutinating activity was not affected at 90℃ for 1h, so it could be said that the lectin had high heat resistance.

Objective To study the relationship between the stenosis content of carotid artery and the degree of homocysteine (Hcy) by analyzing the level changes of Hcy in ischemic stroke patients. Methods One hundred and thirty patients of carotid artery stenosis with ischemic stroke were divided into four groups based on the degrees of carotid artery stenosis determined by ultrasound Doppler flow imaging:the slight stenosis group (32patients),the moderate group (35 patients),the severe group (33 patients) and the total occlusion group (30 patients).Meanwhile,30 healthy people were chosen as the control group. The levels of serum homocysteine (Hcy),total cholesterol (TC),triglyceride (TG) and low density lipoprotein (LDL) of each group were measured, and the results were compared with those of the control group. Results It was found that the serum Hcy level in ischemic stroke group were significantly higher than that in the control group (P<0.05),whereas the variations ofTC,TG,HDL and LDL levels between the ischemic stroke group and the control group could be neglected (P>0.05).An obviously increased order of serum Hcy levels was observed as the following, the control group, the slight stenosis group, the moderate group, the severe group as well as the total occlusion group. The difference between every two groups was statistically significant (P<0.05).The level of serum Hcy was positive correlated to the content of carotid artery stenosis (r=0.835,P<0.05).However,no correlation was found between the carotid artery stenosis and each level of TC,TG and LDL (r=0.031,0.012,0.018,respectively,P>0.05).Conclusions The serum Hcy level is closely related to the degree of carotid artery stenosis, and high level Hcy is suggested to be one of major risk factors for ischemic stroke.
