1.Curcumin Antagonizes Rotenone-induced Injury of PC12 Cells by Antioxidant Activity

Objective To investigate the cytoprotection of curcumin against rotenone(Ro)-induced injury and the molecular mechanisms underlying in PC12 cells.Methods The insulted model of PC12 cells was established with Ro.Cell viability was determined using MTT reduction assay.The content of reactive oxygen species(ROS)was determined by the method of DCFH-DA staining.Chromatometry was used to measure the total activity of SOD,DCFH-DA staining to measure the level of intracellular ROS,and flow cytometry to assay the apoptosis rate.Results 0.5 and 1.0 μmol/L curcumin significantly decreased the inhibitory rate of Ro on the growth of PC12 cells for 24 h as compared with the Ro group(P<0.01).0.5 and 1.0 μmol/L curcumin significantly ameliorated the changes in morphology of PC12 cells,increased the activities of intracellular SOD as compared with the Ro group(P<0.05),decreased the production of intracellular ROS and inhibited the apoptosis of PC12 cells induced by Ro for 24 h as compared with Ro group(P<0.01).Conclusion Curcumin can resist Ro-induced cytotoxicity probably by the mechanism of scavenging intracellular ROS and increasing the activity of antioxidase.
2.Metabolic characteristic and insulin resistance in newly diagnosed type 2 diabetes patients with nonalcoholic fatty liver disease

Objective To investigate metabolic characteristics and insulin resistance in newly diagnosed type 2 diabetes pa-tients with nonalcoholic fatty liver disease .Methods Two hundred patients with newly diagnosed type 2 diabetes mellitus including 140 with nonalcoholic fatty liver disease (NAFLD) (NAFLD group) and 60 without NAFLD (non-NAFLD group) were recruited. Metabolic characteristic were measured and recorded .homeostasis model assessment insulin resistance ( HOMA-IR) index and HOMA-C were used to assess insulin resistance .Results were compared between two groups .Results Compared with non-NAFLD group, body mass index (BMI), fasting blood glucose (FBG),alanine aminotransferase (ALT),gamma-glutamyl transferase (GGT), uric acid ( UA) , triglyceride ( TG) , low density lipoprotein cholesterol ( LDL-C) ,fasting insulin ( FINS) , fasting C peptide ( FCP) , gly-cated hemoglobin a1c (HbA1c), HOMA-IR, and HOMA-C were significantly higher in the NAFLD group ( P <0.05).Logistics re-gression analysis showed that BMI , TG, GGT, and UA were risk factots for NAFLD ( P <0.05, OR =1.82, 1.53, 1.37, and 1.09 ) .Conclusions Compared patients without NAFLD , newly diagnosed type 2 diabetes patients with NAFLD present more meta-bolic disturbance, and severely insulin resistance.Obesity and the increased level of postprandial UA , triglyceridemia, and GGT might increase the risk of NAFLD in the patients with newly diagnosed type 2 diabetes mellitus .

Objective To explore the electronystagmography (ENG) changes and their clinical significance in aged patients with vertebro-basilar insufficiency (VBI). Methods Sixty aged patients with VBI were selected as experimental group for testing of visuo-oculomotor system reaction, spontaneous nystagmus, caloric test and positional nystagmus. Objective To investigate the inhibitory effects of exogenous WAF 1 S gene on human laryngeal cancer Hep 2 cell line, and to explore the potential use of WAF 1 S in gene therapy for laryngeal cancer. Methods A eukaryotic expression vector containing 2 1kb human full length WAF 1 S cDNA was transfected into human laryngeal cancer Hep 2 cell line by using lipofectamine. Expression of exogenous WAF 1 S gene was detected by dot blot hybridization. By using Western blot and confocal microscope, expression of p21 protein was quantitatively analyzed in situ . The growth state of transfected Hep 2 cell was determined by flow cytometry and MTT. Results It was found by dot blot hybridization that WAF 1 S gene could express in Hep 2 cell. The expression of the exogenous p21 gene in Hep 2 cells was markedly higher than that in the control group. It was confirmed with flow cytometry that WAF 1 S gene could induce apoptosis of laryngeal cancer Hep 2 cell line, and the progression of cell cycle was arrested at G 1 phase. Conclusion Laryngeal cancer cells could be arrested at G 1 /S phase and the growth of the cells could be significantly suppressed by exogenous WAF 1 S gene

7.Effects of angiogenesis on the development of endometrial carcinoma

ObjectiveTo study the effects of angiogenesis on the development of endometrial carcinoma. Methods Hysterectomy specimens were stained immunohistochemically by the marker of factor Ⅷ-related antigen for endothelium vessels in normal controls, patients with endometrial hyperplasia and with endometrial carcinoma. Results The microvascular density(MVD) in tumour increased gradually from normal endometrium to endometrial hyperplasia and to endometrial carcinoma(P<0.01). MVD correlated with the mvometrial invasion, histologic grades and the stages.ConclusionMYD in endometrial hyperplasia increases. MVD can be used as a prognostic factor.
8.Bioinformatics screening of up-regulated protein kinases and their inhibitors in nasopharyngeal cancer

Objective To screening up-regulated protein kinases and their inhibitors in order to provide new targets for molecular thera-py of nasopharyngeal cancer.Methods GEO database and SAM software were employed to screen the up-regulated protein kinase gene in nasopharyngeal cancer.Based on DAVID database,the regulating functions of kinases were identified.The inhibitors of up-regulated kinase genes were identified by Selleckchem database.Literature mining was used to screen the potential anti-cancer drugs.Results Totally 2360 differentially expressed genes including 21 up-regulated protein kinases (CHEK1,CHEK2,PRKDC,AURKA,VRK2,STK17A,MELK,NU-AK1,TRPM7,MASTL,AXL,BUB1,BUB1B,CDK4,TTK,CDC7,CASK,AKT3,TBK1 and PBK)were identified in the whole genome profi-ling (Fold Change≥2,P <0.05).The results of function analysis showed the up-regulated genes were enriched in 10 function terms such as‘protein amino acid phosphorylation’‘phosphorylation’‘phosphate metabolic process’‘mitotic cell cycle’‘cell cycle phase’‘regulation of cell cycle’,and so on.The Selleckchem database analysis showed there were 9 up-regulated protein kinases equipped with 51 inhibitors which were proved already.The results of literature mining showed that 18 inhibitors of them had a few studies (less than 10 literatures)in cancer terms,and there was a potential to become new drugs to treat nasopharyngeal cancer.Conclusion A total of 21 up-regulated protein kinases were identified,and they might promote the nasopharyngeal carcinoma by regulating functions such as the cell-cycle control pathway.Their ki-nase inhibitors may have a potential role in anti-cancer treatment,which provided a new target point for molecular therapy of nasopharyngeal cancer.
9.Efficacy andβcell function improvement of Linagliptin in combination with Metformin in newly diagnosed T2DM patients

Objective To explore the efficacy and safety of Linagliptin in combination with Metformin in newly diagnosed T2DM patients. Methods A total of 140 newly diagnosed T2DM patients were enrolled in this study. All the patients were admitted in the Endocrinology and Metabolism department of our hospital from January 2013 to October 2013. Random number table method was used in patients’ selection. All the patients were divided into two groups :Linagliptin plus Metformin group (Linagliptin group ,n=70) and Glipizide plus Metformin group (Glipizide group ,n=70) ,and followed up for 12 weeks. Efficacy and safety of treatment were compared between the two groups. Results The difference of FPG ,2 hPG and HbA1 c did not reach the statistical significance between the two groups at baseline treatment (P>0.05). FPG ,2 hPG and HbA1 c were significantly lower after treatment compared with baseline in both groups (P<0.05). FC-P ,2 hC-P ,HOMA-β,HOMA-IR ,IAI and adiponectin level at baseline were similar between the two groups ( P> 0.05 ). All these indicators were improved after treatment in both groups (P< 0.05) ,and were significant better in Linagliptin group than in Glipizide group. Adverse events rate were significantly higher in Glipizide group than in Linagliptin group (18.57%vs 5.71% ,χ2 = 5.423 ,P= 0.020 ). Conclusion The efficacy of Linagliptin in combination with Metformin in newly diagnosed T2DM patients was similar with Glipizide plus Metformin. The β cell function improvement was better and APN level was higher after Linagliptin treatment.
