1.Radix Astragali injection at appropriate concentrations enhances surface adhesion of bone marrow stromal cells

BACKGROUND:Several studies have demonstrated that many kinds of Chinese herbs for benefiting vital energy and enriching the blood secrete some cytokines by influencing bone marrow stromal cel s to promote the differentiation and proliferation of hemopoietic stem cel s or exert effects by promoting the adhesion of bone marrow stromal cel s and hemopoietic stem cel s.
OBJECTIVE:To investigate the effects of Radix Astragali injection on the expression of intercel ular adhesion molecule 1 and vascular cel adhesion molecule 1 on the surface of mouse bone marrow stromal cel s.
METHODS:Mouse bone marrow stromal cel s were isolated by adherent culture of whole bone marrow. The morphology of mouse bone marrow stromal cel s were observed using inverted phase microscopy, hematoxylin-eosin staining, optical microscopy and transmission electron microscopy. The optimal concentration at which Radix Astragali injection exhibited the strongest effects on promoting the proliferation of bone marrow stromal cel s was detected using MTT assay. Expression of intercel ular adhesion molecule 1 and vascular cel adhesion molecule 1 on the surface of mouse bone marrow stromal cel s was detected using flow cytrometry after intervention by Radix Astragali injection.
RESULTS AND CONCLUSION:Inverted phase microscopy and optical microscopy showed that bone marrow stromal cel s adhered to the wal of culture flask, exhibited a shuttle-shaped or irregular appearance with processes. Through transmission electron microscopy, organel es were abundant, such as rough endoplasmic reticulum, secretory vesicle, and mitochondria. Radix Astragali injection at 400 and 600 mg/L significantly promoted the proliferation of mouse bone marrow stromal cel s (P<0.05) and there was no significant difference in the cel proliferation between these two concentrations. Radix Astragali injection at 600 mg/L increased the expression of intercel ular adhesion molecule 1 and vascular cel adhesion molecule 1 on the surface of mouse bone marrow stromal cel s. These findings suggest that Radix Astragali injection at appropriate concentrations can increase the surface adhesion of bone marrow stromal cel s, thereby improving hemopoietic microenvironment.
2.Effect of monosialotetrahexosy-1 ganglioside on autophagic neuronal death in rats with spinal cord injury

Objective To investigate effect of monosialotetrahexosy-1 ganglioside (GM1) on the autophagic neuronal death induced by spinal cord injury.Methods Ninety SD rats were randomly divided into sham group,isotonic saline group and GM1 group,with 30 rats per group.Spinal cord injury at T10 segment was induced by Allen method in the isotonic saline group and GM1 group.Expression of endogenous LC3 was detected by florescence microscope.Ratio of LC3-Ⅱ and LC3-Ⅰ,and expression of Beclin-1 were detected by Western blot.Results LC3 was significantly up-regulated in the isotonic saline group,as compared to the sham group (P <0.05).Also,an up-regulation of LC3 and a decline of autophagic body formation were observed in GM1 group,as compared to isotonic saline group (P <0.05).Expressions of LC3-Ⅱ and Beclin-1 and ratio of LC3-Ⅱ to LC3-Ⅰ were significantly increased in the isotonic saline group,as compared to the sham group (P < 0.05).Expressions of LC3-Ⅱ and Beclin1 and ratio of LC3-Ⅱ to LC3-Ⅰ significantly were decreased in the GM1 group,when compared to the isotonic saline group (P < 0.05).Conclusion GM1 inhibits autophagic neuronal death after spinal cord injury and hence exerts protective effect on the neurons.
3.EFFECT OF PHYTIC ACID ON EXPRESSIONS OF APOPTOSIS ASSOCIATED PROTEIN FAS、BAX AND CASPASE-3 IN SGC-7901 CELLS

Objective:To explore the effect of phytic acid on apoptosis of SGC-7901 cells and its mechanisms. Method:The inhibiting action of phytic acid on SGC-7901 cells was examed by MTT assay,and the effect on apoptosis of SGC-7901 cells was examined by DNA gel electrophoresis,and the expressions of Fas,Bax and caspase-3 protein were detected by Western blot method. Results:From MTT assay,we observed that phytic acid inhibited the growth of SGC-7901 cells in dose and time dependent manner. Typical DNA ladder was found during gel eletrophoresis. Caspase-3 can be activated by phytic acid and the expressions of Bax and Fas protein in phytic acid group were higher compared with control group,and increased in a dose-dependent manner. Conclusion:Phytic acid can induce apoptosis of SGC-7901 cells which may be correlated to apoptosis associated protein Fas,Bax and caspase-3.
4.Clinical value of helfet test in the diagnosis of meniscus injury

[Objective]To study the accuracy of Helfet test in the diagnosis of the knee meniscus tear.[Method]From Apr.2007 to Oct.2007,52 cases of the lateral meniscus tear and 23 cases of the medial meniscus tear were checked-up with Helfet test,McMurray test ,KS test,and arthroscope comparatively.[Result]The Helfet Test was positive in most of the lateral meniscus tear cases.The consistency with McMurray test and arthroscope was lateral meniscus tear,especially to bucket-handle and Horizontal tear high respectively.The rate of accuracy to lateral menicus was 81.4%.The rate of accuracy to bucket-handle and Horizontal tear were 89.6% and 87.2% respectively.[Conclusion]The "Helfet" test is sensitive to the knee lateral meniscus tear.It is fast,simple,reliable,reproducible,positive for diagnose of lateral meniscus tear,and is worth of being used widely.
5.Changes of Syndromes and Therapeutic Characteristics in the Patient of Steroid-dependent Asthma during Steroid Withdrawal

Changes of syndromes and therapeutic feature of the patients with steroid-dependent asthma(SDA)during steroid withdrawal were investigated.It was held that the patient presented pathological changes from deficiency of Yin and hyperactivity of evil fire before steroid withdrawal to deficiency of Yin and Yang during steroid withdrawal to deficiency of kidney-Yang after steroid withdrawal,with the syndrome of stasis of phlegm and blood throughout the whole process.Therefore,a new scheme for weaning the steroid with the TCM three-step sequential therapy was established according to the changes of syndromes,i,e., nourishing Yin and blood,clearing away excessive heat and eliminating stasis of phlegm and blood before steroid withdrawal; nourishing Yin and blood,inhibiting pathogenic wind,replenishing Qi and Yang,activating blood circulation,resolving phlegm and lowering the adverse rise of Qi to relieve asthma during steroid withdrawal;warming kidney-Yang and adjusting Qi and blood,and regulating functional activity of Qi after steroid withdrawal.Because the three-steep sequential therapy was designed based on the dynamic change of SDA during steroid withdrawal,it conformed with the whole clinical process of the occurrence,development and changes of SDA than any other single way,and reflected the essential characteristics of the pathological changes.

9.Clinical Application of Nano-hydroxyapatete/collagen Composite

Objective To observe the repairing effects of bone after nano-hydroxyapatite/collagen (nHAC)being implanted in and to study its prospect in clinical application.Methods Nano-hydroxyapatite/collagen composite was developed by biomimetic method. The material was used in clinical operation,the healing effects of bone defects were assessed by methods of radiography.Results The composite showed some features of natural bone in both composition and microstructure.After implanting,the patients were not hyperpyrexial,did not have extravasate and immunoreaction,the bone defects were healing well.Conclusion The composite can serve as one of the promising materials for bone repairing.
10.Multi-slice CT evaluation of glenoid bone loss in patients with recurrent anterior shoulder dislocation

ObjectiveTo evaluate multi-slice CT (MSCT) in glenoid bone loss of patients with recurrent anterior shoulder dislocation.Methods MSCT findings of 108 patients with recurrent anterior shoulder dislocation and 12 with single anterior shoulder dislocation were retrospectively studied.The incidence,degrees and locations of glenoid bone losses were recorded.The incidence was analyzed with Fisher exact test.The maximum length,depth and proportion were compared with Wilcoxon rank sum test.ResultsGlenoid bone loss was detected in 91.7% (99/108)patients with recurrent anterior shoulder dislocation.The proportion of glenoid bone loss was ( 16.0 ± 6.0)%,and the central locations of glenoid bone loss were from 2: 20 to 4: 25 ( mean 3: 20).Sixty-two percent (67/108) patients had bony Bankart lesions in which 58.2% (39/67) bony fragments were free and 41.8% (28/67) were adherent to the anterior border of the glenoid cavity.Seventy-five percent (9/12) patients with single shoulder dislocation had anterior glenoid bone loss,and the proportion of glenoid bone loss was ( 15.2 ± 7.1 ) %.There were no statistical differences of the incidence (P =0.100) and proportion of glenoid bone loss ( P =0.453 ) between the recurrent and single anterior shoulder dislocation.ConclusionsAnterior glenoid bone loss is common in patients with recurrent anterior shoulder dislocation.
