1.CONSTRUCTION OF RECOMBINANT RETROVIRAL VECTOR CARRYING RAT IGF-1 GENE

Objective To establish a recombinant retroviral vector containing insulin-like growth factor-1(IGF-1) gene and to provide the basis for the application of IGF-1 in treating nervous system diseases such as stroke.Methods The plasmid pcDNA3.1-IGF-1 was cut by EcoR I/Xho I,and subcloned to retroviral vector pLXSN,resulting in the recombinant plasmid(pLXSN-IGF-1.)The recombinant IGF-1 expression vector was evaluated by using enzyme cutting and sequencing.By the Lipofectamine 2000,pLXSN-IGF-1 was transferred to packaging cell line-pA317.Culture supernatant of these cells was detected for titration of the recombinant virus.Results The two fragments from recombined IGF-1 eukaryotic expression vector by EcoR I and Xho I represented 400?bp and 6.0?kb by agarose electrophoresis,and PCR showed positive fragment which was about 400?bp long,and sequence analysis showed the same sequence as expected.The cell line pA317-IGF-1 was established,and average virus titer of the recombinant virus in the culture supernatant was about 6.5?10~5 CFU/ml.Conclusion A recombinant retroviral containing IGF-1 gene was successfully constructed.

4.Clinical significance of serum CEA, CA125 and CA72-4 in gastric carcinoma with peritoneal metastasis

Background and purpose: Peritoneal metastasis of gastric cancer is mainly discovered in the ad-vanced cancer. Nonetheless, the clinical applicability of each tumor biomarker in peritoneal metastasis of gastric cancer is still ambiguous. Therefore, this study investigated the diagnostic value and clinical significance of CEA, CA125 and CA72-4 in gastric carcinoma patients with peritoneal metastases. Methods: A total of 108 gastric carcinoma patients with peritoneal metastases from Jan. 2008 to Dec. 2013 were studied. All patients were diagnosed by imaging, operations and pathological examination, and also received intravenous or intraperitoneal chemotherapy. Serum tumor markers such as CEA, CA125 and CA72-4 were determined during diagnosis and before each chemotherapy. The diagnostic sensitivity of single marker and combined detection with 2 or 3 markers were analyzed. The correlations among the serum tumor markers and clinical pathological factors, chemotherapeutic effects and survival time were analyzed. Results: Positive rates of CEA, CA125 and CA72-4 were 20.4%, 46.3% and 45.4% in gastric cancer patients with peritoneal metastases, respectively. For these patients, the positive rates of CEA/CA125, CEA/CA72-4, CA125/CA72-4 and CEA/CA125/CA72-4 were 54.7%, 52.8%, 69.5% and 79.6%, respectively. The combined detection of 3 tumor markers was much better than single marker detection (P<0.05). Positive rates of CEA, CA125 and CA72-4 were correlated with the ECOG scale (P<0.05). Positive rate of CA125 was associated with ascites (P<0.001), while positive rate of CA72-4 was associated with ovarian metastasis (P<0.05). Median survival time of patients with positive rates of CEA, CA125 and CA72-4 was significantly lower than that of the patients with normal levels of these markers (P<0.05). Compared with pre-treatment, the levels of all three tumor markers significantly declined after three cycles of chemo-therapy (P<0.05). The decline in CA125 level after chemotherapy was significantly correlated with decreased amount of ascites (P<0.05). The tumor markers turned negative after 3 cycles chemotherapy in patients with positive markers upon initial diagnosis, their survival was significantly prolonged (P<0.001). Conclusion: Combined detection of serum CEA, CA125 and CA72-4 can significantly promote diagnostic rate of gastric cancer with peritoneal metastasis, and may be helpful in evaluating chemotherapeutic effects and predicting prognosis.
6.Correlation of NADPH oxidase subunit p22phox gene C242T polymorphism with left ventricular hypertrophy in Chinese Korean hypertensive patients

Objective To investigate the relation between the NOSP gene C242T polymorphism and Chinese Korean nationality EH patients with LVH. Methods 108 Chinese Korean controls and 231 Chinese Korean EH patients were enrolled in the research.NOSP gene C242T polymorphism was analyzed by PCR. EH patients were divided into EH and EH+LVH group,all of them were evaluated with CDUS for left ventricular mass index. Results The distribution of NOSP gene C242T polymorphism showed no significant difference compared with controls and EH group.There was significant difference in distribution of CC vs CT+TT genotype between EH and EH+ LVH patients.CC genotype is more dangerous for EH patients combined with LVH than CT and TT genotypes. Conclusions The NOSP gene C242T polymorphism has no relation with EH in Korean nationality people. The NOSP gene C242T polymorphism has relation with EH+LVH; and the CC genotype is a susceptible gene for the Korean EH patients with LVH.
7.Changes of CGRP and NPY expression in tissue-engineered bone with fascia flap

Objective To evaluate the expressive levels of calcitonin gene related peptide (CGRP) and neuropeptide Y (NPY) in tissue-engineered bone with fascia flap in vivo. Method A segmental bone defect 1.5cm in length was made at the both radius of 12 healthy New Zealand rabbits. The defects were repaired by implantation in two ways: for left radius, engineered bone with fascia flap was implanted (served as experiment group), and for right radius only engineered bone was implanted (served as control group). 3, 6 and 12 months after implantation, the expressions of CGRP and NPY in the new bone were determined with immunohistochemistry and semi-quantified using image analysis software. Objective To analyze the problem existing in the process of health records informatization con-struction and explore solution on the basis of situation of health record informatization in Yanta district of Xi'an city. And to provide bases for acceleration of residents'health records informatization construction in Xi'an.Methods Send 10 investigators to study and investigate the hospital information management system of 10 full-time public universities in Yanta district.Results There were a total of 25 711 faculties in 10 full-time public universities and 19 809 facul-ties had established electronic files,the filing rate was 77.04% A.On the whole,it seemed that the teaching staff in the filing rate(78.91%)was higher than that of the retired workers filing rate(73.78%),the two groups had statisti-cally significant difference(χ2 =88.164,P <0.05)B.Conclusion Xi'an Yanta district university faculty of electron-ic health records has exceeded the national average level,but there are still many problems need to be improved.We should strengthen government investment,the establishment of the region's unified information management standards, increase publicity and education efforts,and actively create the region's resource information sharing platform.
9.The effect of cognitive behavior therapy on PANSS of schizophrenia: a Meta-analysis

Objective To assess systematically the effect of (cognitive behavior therapy,CBT) on PANSS of schizophrenia.Methods Relative literature was searched by computer at home according to the inclusion and the exclusion criteria,which were analyzed by RevMan5.2 software,and literature selection and repetition were used according to the Note Express software.Results A total of 11 studies were brought into the study.According to the different time of intervention,the results showed that,the cognitive behavior therapy intervention continued for 2 months,I2 was 80%,the statistical result of random effects model analysis was [SMD=-0.64,95%CI (-1.14~-1.13),P<0.05]; the cognitive behavior therapy intervention continued for 6 months,I2 was 64%,the statistical result of random effects model analysis was [SMD=-0.40,95%CI (-0.71,-0.08),P<0.05]; the cognitive behavior therapy intervention continued for 12 months,the statistical result of fixed effects model analysis was[MD=-5.65,95%CI(-6.20,-5.09),P<0.01].The replacement of statistical models for the sensitive analysis showed that the result was stabile and reliable.Conclusions The cognitive behavior therapy may reduce PANSS of patients with schizophrenia,But because the influence of some factors,the time of intervention is limited,so the result of this research is that the best intervention time for clinical symptoms improvement is from 6 months to 1 year.
