Objective To explore the role of Kupffer cells in high fat diet induced hepatic insulin resistance. Meth-ods Eighty-four C57BL/6J mice were divided into two groups: normal chow (NC) group (7 subgroups) and high fat diet (HFD) group (7 subgroups). Glucose tolerance tests were performed at feeding time of 1,2,4,8,12 and 16 weeks in one NC group and one HFD group. The mice livers at the same feeding time were obtained in other 6 NC groups and 6 HFD groups respectively. The expression levels of IRS1-ser307, Akt-ser473, S6K1-thr389 and p-JNK were detected by Western blot as-say. The values of MCP-1,TNF-α,IL-1βand IL-10 mRNA were examined by qRT-PCR. Results Compared with NC group, the impaired glucose tolerance was found from the first week in HFD group (P < 0.05). The hepatic expressions of IRS1-ser307, S6K1-thr389 (4-16 weeks ) and p-JNK(2-16 weeks ) increased and Akt-ser473(8-16 weeks )decreased in HFD group than those of NC group. The same results were gained by analysis of protein relative expression (all P<0.05). The hepatic pro-inflammatory factor MCP-1,TNF-αand IL-1βmRNA expressions were higher in HFD group than those in NC group during 2-16 weeks (all P < 0.01). The anti-inflammatory factor IL-10 mRNA was significantly lower in HFD group than that in NC group during 4-16 weeks (all P<0.01). Conclusion High fat diet maybe play a role in the hepatic insulin resistance by stimulating M1 Kupffer cells to secrete pro-inflammatory factor and inhibiting M2 Kupffer cells to se-crete anti-inflammatory factor.

Objective To establish a method for the determination of forsy thin in Baohe Oral Liquid by HPLC.Methods The determination was carried out on a Diamonsil C18 column, with mobile phase of acetonitrile-water(29∶71) at room temperature. The flow rate was 1.0mL?min-1 and the detective wavelength at 277 nm.Results The calibration curves was linear in the range of 0.068~ 0.340 ? g(r=0.9996).The average recovery rate of tested sample was 99.7 %(RSD=2.1 %).Conclusion The method was specific, accurate and precise. It can be used for the quality control of Baohe Oral Liquid.

so higher in diabetic patients 4 h after the meal (all P<0. 05). Positive correlation existed between serum triglycerides and white blood cell counting, neutrophils, and high-sensitivity C-reactive protein(r were between 0.268 and 0.548, all P<0.05).

Objective:To investigate the correlation between multiple parameters of islet function evaluation and atherogenic index (AIP) in patients with type 2 diabetes mellitus (T2DM).Methods:A cross-sectional study was conducted to observe 216 T2DM patients hospitalized in Tianjin Medical University Chu Hsien-I Memorial Hospital in 2019. They were divided into non atherogenic phenotype group (N Group, AIP<0.06) and atherogenic phenotype group (A group, AIP≥0.06) with AIP=0.06 as the cut-off point. The general clinical indexes and evaluation indexes of oral glucose tolerance test (OGTT), such as insulin action index (IAI), quantitative insulin sensitivity check index (QUICKI), homeostasis model of insulin resistance index (HOMA-IR), homeostasis model assessment-β (HOMA-β), C peptide insulin resistance index (HOMA IR-CP) and C peptide islet function index suitable to diabetes patients (HOMA islet-CP DM) were compared between two groups. Pearson correlation analysis and multiple stepwise regression analysis were performed. Results:Compared with the N group, the very low density lipoprotein-cholesterol (VLDL-C), ln (HOMA IR-CP) and ln (HOMA islet-CP DM) in A group were higher ( P<0.01), while ln (QUICKI) was lower ( P<0.05), with statistical significance. AIP was positively correlated with VLDL-C ( r=0.765), ln (HOMA-IR) ( r=0.257), ln (HOMA-β) ( r=0.189), ln (HOMA IR-CP) ( r=0.418) and ln (HOMA islet-CP DM) ( r=0.377, P<0.01), and negatively correlated with IAI ( r=-0.145, P<0.05) and ln (QUICKI) ( r=-0.254, P<0.01). Multiple stepwise regression analysis showed that VLDL-C and ln (HOMA IR-CP) were independent influencing factors of AIP in type 2 diabetic patients ( P<0.01) . Conclusions:AIP is positively correlated with insulin resistance and negatively correlated with insulin sensitivity. VLDL-C and HOMA IR-CP are independent predictors of AIP.
