Objective To construct the KU80 inhibition cell model by RNAi in U2OS cell and to explore the relationship between the Ku80,telomeres and radiosensitivity in telomerase-negative tumor cells. Methods U2OS cells were transfected with the recombinant plasmids of pshRNA-K80 by the lipofectamine, and the stable transfected cell clones were selected by G418.After the selection, the cells were collected and analyzed by the flow cytometry.RT-PCR and Western blot were used to measure the expression of Ku80 and Real-time PCR was used to detect the length of telomeres. The radiosensitivity of U2OS was determined by clone formation array. Results The transfection efficiency of the positive cell clones detected by the flow cytometry was (83.23 ± 7.63) %.The inhibition rate of the Ku80 gene transcription in the cell group with recombinant plasmid was(68.09 ± 1.16)% and the inhibition rate of the Ku80 protein expression in the same group was (11.03 ± 2.45) %.The results of Real-time PCR showed that the telomere length of the cell group with recombinant plasmid (1.07 ± 0.07) was significantly shorter than that of the control group (4.42 ± 1.30,F =38.58,P < 0.05) and that of the empty plasmid group (4.11 ±0.84,F =38.58,P < 0.05).Compared to the control group, the telomere length of the empty plasmid group did not changed(4.42 ±0.84 vs.4.11 ±0.84).U2OS cells with Ku80 expression suppressed had lower SF2 than that of the control cells (F =1089.61,P <0.05),and resulted in the SER of 1.47.Conclusions The Ku80 inhibition cell model in telomerase-negative U2OS cell line is successfully established which has the shorter telomere length, and is more sensitive to radiation.Telomere shortening caused by pshRNA-of Ku80 is likely to be one of the mechanisms of radiosensitization in this kind of cell model.
9.Long-term stability of orthodontic treatment out of retention.
Chinese Journal of Stomatology 2002;37(3):216-218
OBJECTIVETo investigate the correlative factors influencing the stability of orthodontic treatment out of retention.
METHODSPre-treatment, post-treatment and 2 approximately 8 years out of retention study models of 74 cases with malocclusion were assessed by the PAR (Peer Assessment Rating) index.
RESULTS(1) Out of retention, the relapse rate was 13.01% in weighted PAR total scores and the relapse rate was 6.38%, 15.56%, 28.86%, 22.41% in alignment, overjet, overbite and transverse buccal occlusion respectively. (2) The rate of great improvement was decreased from 77.03% after treatment to 64.86% out of retention. The PAR score increased in 53 cases (71.62%), unchanged in 6 cases (8.11%) and reduced in 15 cases (20.27%) out of retention. (3) The weighted total scores in the Begg appliance were higher than that in Edgewise out of retention (P < 0.05). (4) The alignment and PAR total scores out of retention were higher in class II malocclusion than in class I malocclusion (P < 0.05). (5) The weighted PAR total scores post-retention were higher in unilateral extraction than in other groups (P < 0.01).
CONCLUSIONS(1) There was a trend that teeth relapsed to original position out of retention. The relapse rate was greater in alignment, overjet, overbite and transverse buccal occlusion. (2) The stability out of retention was not related to patient's age and sex, but was related to the types of appliance, malocclusion and extraction.

Chinese Journal of Radiation Oncology 2021;30(5):518-522
Novel cancer immunotherapy can treat tumors through regulating innate immunity and adaptive immune system. cGMP-AMP synthase (cGAS) is a key regulator of innate immune response to both exogenous and endogenous DNA. After recognizing the cytoplasmic DNA, cGAS produces the second messenger cyclic GMP-AMP (cGAMP), which subsequently combines with the adaptor STING (also known as MITA, MPYS and ERIS) to mediate innate immunity by inducing the production of type I interferons and inflammatory cytokines. Recent studies have revealed that the cGAS/STING signaling pathway can be activated by tumor-derived DNA and by-products of genomic instability and affect the incidence and development of tumors, which plays a critical role in the natural antitumor immunity across cancer types and immune checkpoint blockade therapy. In this article, current understanding of cGAS/STING signaling pathway in tumors was summarized, the pivotal role in tumor immunity and radiotherapy was highlighted, and the potential targeted or alternative therapy of this signaling pathway was reviewed.
