2.Progress on the study of mechanism of the direct action of TCM bioactive components on ovarian cancer.

Both the morbidity and mortality of ovarian cancer, among malignant tumors ot temale genital organs, are quite high. The traditional therapeutic methods on ovarian cancer are surgical operation, chemotherapy, radiotherapy and combination of these methods mentioned above. In recent years, some components of traditional Chinese medicine, such as genistein, semen Coicis, phytosterols, curcumin, quercetin, ginsenoside, etc. have been found to exert anticancer actions of inhibiting proliferation and inducing apoptosis of cancer cells, increasing the sensitivity of patients to chemotherapeutic agents in viva and/or in vitro, the mechanisms involve such aspects as inhibiting activity of key enzymes in cell metabolism, affecting gene expression, antioxidation, and inhibiting tumor angiogenesis, etc. As an adjuvant therapeutic means, the bioactive components of traditional Chinese medicine have broad future of clinical application.

Objective:To study the effect of total saponins on cerebral blood flow and vascular resistance in anesthetized dogs.Methods:Thirty hybrid dogs in either sex,with a body weight of(11?1.5)kg,were evenly randomized into 5 groups: negative control group(saline 5 ml/kg,ig),positive control(nimodipine 300?g/kg,iv),and 3 groups treated with total saponins (low-dose group[10 mg/kg,ig],middle-dose group[30 mg/kg,ig],and high-dose group[60 mg/kg,ig]).The dogs were anes- thetized with intravenous pentobarbital sodium(30 mg/kg).The right common carotid artery was exposed to measure the cere- bral blood flow,cerebral vascular resistance,blood pressure and heart rate using the MFV-3200 electromagnetic flow meter and MPA-3000 bioelectricity signal-amplifier.Results:Compared with negative control,cerebral blood flow was significantly in- creased in animals treated with asparagus root saponins(30 and 60 mg/kg,ig)during 5 and 120 min after drug administration (P
4.Congenital Vaginal Atresia: A Report of 39 Cases in a Regional Obstetrics and Gynecology Hospital

To investigate the clinical course and management of congenital vaginal atresia.This retrospective analysis included patients with congenital vaginal atresia treated from March 2004 to August 2014 at the Obstetrics and Gynecology Hospital of Fudan University.Thirty-nine patients were included in this study.Their average age was 16.87±2.2 years when they came to our hospital.Totally,51% of the patients had isolated congenital vaginal atresia with a normal cervix,whereas the others had either cervical atresia or imperforate hymen.The primary presenting signs and symptoms included primary amenorrhea (71.8%),periodic abdominalgia (41.0%),abdominal pain (36.0%),dyspareunia (10.3%),menstrual disorders (5.1%),and pelvic mass (5.1%).Ultrasound and magnetic resonance imaging (MRI) were effective inspection methods for the screening of urogenital tract-associated anomalies.Vaginoplasty mainly included simple vagina reconstruction with insertion of a mold (n=22) and split-thickness skin grafting (n=4).In 64% of surgical patients,normal menstrual bleeding was achieved.Four of the patients subsequently became pregnant and delivered at term.Primary amenorrhea,periodic abdominalgia and abdominal pain are the main reasons for the post pubertal patients to visit doctors.Surgical methods can successfully provide these patients an opportunity for subsequent conservative management,can result in normal menstrual bleeding,resolve cyclic pelvic pain,and provide some potential for fertility.
5.Research Progress of MicroRNA in Early Detection of Ovarian Cancer.

OBJECTIVEThis review aimed to update the progress of microRNA (miRNA) in early detection of ovarian cancer. We discussed the current clinical diagnosis methods and biomarkers of ovarian cancer, especially the methods of miRNA in early detection of ovarian cancer.
DATA SOURCESWe collected all relevant studies about miRNA and ovarian cancer in PubMed and CNKI from 1995 to 2015.
STUDY SELECTIONWe included all relevant studies concerning miRNA in early detection of ovarian cancer, and excluded the duplicated articles.
RESULTSmiRNAs play a key role in various biological processes of ovarian cancer, such as development, proliferation, differentiation, apoptosis and metastasis, and these phenomena appear in the early-stage. Therefore, miRNA can be used as a new biomarker for early diagnosis of ovarian cancer, intervention on miRNA expression of known target genes, and potential target genes can achieve the effect of early prevention. With the development of nanoscience and technology, analysis methods of miRNA are also quickly developed, which may provide better characterization of early detection of ovarian cancer.
CONCLUSIONSIn the near future, miRNA therapy could be a powerful tool for ovarian cancer prevention and treatment, and combining with the new analysis technology and new nanomaterials, point-of-care tests for miRNA with high throughput, high sensitivity, and strong specificity are developed to achieve the application of diagnostic kits in screening of early ovarian cancer.

6.Effects of extracts of Ginseng and Ginkgo biloba on hippocampal acetylcholine and monoamines in PDAP-pV717I transgenic mice.

OBJECTIVETo study the effects and its possible mechanism of Naoweikang (NWK), a composite of ginseng and ginkgo extracts, on hippocampal neurotransmitters in APP transgenic mice.
METHODSP-DAPPV717I transgenic mice were taken as the model of Alzheimer's disease (AD) and be treated with different doses of NWK (31 mg/kg and 62 mg/kg) respectively by gastrogavage once per day for 12 weeks. Contents of hippocampal acetylcholine (ACh), monoamine neurotransmitters and their metabolites were determined with high performance liquid chromatography.
RESULTSCompared with nontransgenic mice, the levels of ACh and 5-HIAA in hippocampus of transgenic mice lowered significantly (P < 0.01), while 5-HT increased significantly (P < 0.05), and the levels of norepinephrine and dopamine increased by 14.6% and 17.7%, respectively. After 12-week administration, the ACh level increased significantly in the two NWK treated groups (P<0.05), and the 5-HT level in the high dose NWK treated group decreased (P<0.05), as compared with those in the untreated transgenic mice.
CONCLUSIONNWK shows a significant regulatory effect on the activities of hippocampal acetylcholine and monoamine system, especially the cholinergic and 5-HT systems, in APP transgenic mice, which might be one of its mechanisms in improving learning and memory of AD model, and therefore, NWK might exert certain curative effect on AD.

7.Effect of Sailuotong capsule on mitochondrial dynamics in focal cerebral ischemia/reperfusion rats.

To observe the protective effect and mechanism of Sailuotong capsule in focal cerebral ischemia/reperfusion. The 90 min middle cerebral artery occlusion (MCAO) reperfusion model was established. The expressions of dynamin-related protein 1 ( Drp1) and optic atrophy 1 (Opa1) were tested by Western blot. The transmission electron microscope was used to observe the changes in the mitochondrial ultra-structure. The pathological morphological changes were observed through the HE staining. The immunohistochemical method was used to test Drp1 and Opa1 expressions. Sailuotong capsule (33, 16.5 mg x kg(-1), ig) can inhibit the abnormal mitochondrial fission and fusion in the cortical area on the ischemia side and the mitochondrial fission gene expression and promote the mitochondrial fusion gene Opa1 expression, so as to alleviate the energy metabolism disorder caused by ischemia/reperfusion. Sailuotong capsule can inhibit the abnormal mitochondrial dynamics in peri-ischemic regions and maintain the normal morphology of mitochondria, which may be the mechanism of Sailuotong capsule in promoting the self-recovery function in the ischemic brain region.

8.A network meta-analysis of the short-term efficacy of five chemotherapy regimens based on cisplatin and fluorouracil for esophagogastric junctional adenocarcinoma.

The primary purpose of this study was to explore the short-term efficacy of different cisplatin and fluorouracil-based chemotherapy regimens in the treatment of patients with esophagogastric junctional adenocarcinoma (EGJA) using a network meta-analysis (NMA). Randomized controlled trials (RCTs) related to chemotherapy regimens based on cisplatin and fluorouracil for EGJA were included from the PubMed, EMBASE and Cochrane Library electronic databases (from inception to June 2016). Direct and indirect evidence were combined to calculate the pooled odds ratio (OR) and its 95% confidence interval (95% CI) as well as to draw the surface under the cumulative ranking (SUCRA) curves. This NMA finally enrolled ten eligible RCTs with the following five regimens: cisplatin plus fluorouracil (cisplatin+fluorouracil), cisplatin+fluorouracil-based chemotherapy (cisplatin+fluorouracil+docetaxel/epirubicin/irinotecan), fluorouracil-based chemotherapy (fluorouracil+docetaxel/doxorubicin/methotrexate/irinotecan), cisplatin-based chemotherapy (cisplatin+docetaxel/epirubicin/irinotecan/capecitabine/s-1) and other drug-based chemotherapy (docetaxel/irinotecan/capecitabine). These results revealed that compared with a cisplatin+ fluorouracil-based chemotherapy regimen, the fluorouracil-based chemotherapy regimen had a lower overall response rate (ORR) and partial response (PR) for EGJA patients (ORR: OR=0.43, 95% CI=0.22–0.86; PR: OR=0.46, 95% CI=0.23–0.91). Cluster analyses suggested that the cisplatin+fluorouracil-based chemotherapy regimen had the best short-term efficacy for EGJA in terms of the complete response (CR), PR, ORR, stable disease (SD) and progression disease (PD). Our results indicated that cisplatin+fluorouracil-based chemotherapy regimens may have the best short-term efficacy in the treatment of EGJA.

9.Effect of bushen yiqi recipe on bioactivity behavior of human cytotrophoblast of early pregnancy.

OBJECTIVETo investigate the effect of Chinese drugs for tonifying Shen on the bioactivity, cell proliferation, invasion and differentiation of human cytotrophoblast of early pregnancy.
METHODSThe human early pregnant cytotrophoblast was cultured in vitro, and treated with saline (as blank control) and drug containing serum in different concentrations (5%, 10% and 20%) respectively. The changes of morphology, proliferation and invasive capacity of cells were examined by scanning electron microscopy, MTT method, flow cytometry, Transwell invasive assay at 24 hrs, 48 hrs and 72 hrs after treatment.
RESULTSAfter cells being cultured with drug containing serum, the cytotrophoblast became abundant in microvilli, with more and prolonged pseudopodia. The cell absorbency in 490 nm wave length increased significantly (P < 0.01), cells of sub-G1 and G2/M phase obviously decreased and that of S phase increased (P < 0.01), and the cells penetrated through PET membrane in each visual field significantly increased (P < 0.01).
CONCLUSIONChinese herbs for tonifying Shen could promote the proliferation and invasive capacity of cytotrophoblast and might influence its differentiation.

10.A novel mutation of the KIT gene in a Chinese family with piebaldism.

BACKGROUNDHuman piebaldism is a rare autosomal dominant condition characterized by congenital white forelock and depigmented patches of skin, typically on the forehead, anterior trunk and extremities. Mutations in the KIT gene have been proposed to be responsible for the underlying changes in this disorder. The aim of this study was to identify gene mutation in a Chinese family with piebaldism.
METHODSA Chinese family with piebaldism presenting with white forelock and large depigmented skin macules on the abdomen, arms and legs was collected. DNA was isolated from peripheral blood of the family members. The encoding exons with flanking intron regions of the KIT gene were analyzed by polymerase chain reactions (PCR) and direct DNA sequencing. Besides, DNA extracted from 100 ethnically matched population individuals was as controls.
RESULTSA heterozygous missense mutation c.2590T > C was identified in the patients of the family. This mutation converted a serine residue to proline (p.Ser864Pro). The mutation was not found in their unaffected family members or normal controls.
CONCLUSIONA novel missense mutation c.2590 T > C was found and it might play a significant role in the piebaldism phenotype in the family.
