Chinese Journal of Organ Transplantation 2003;0(05):-
Objective To investigate whether the signaling pathway of Akt/protein kinase B (PKB)is involved in the pathogenesis in the early stage of chronic allograft nephropathy(CAN). Methods The kidneys of Fisher(F344)rats were orthotopically transplanted into Lewis recipients. Animals were harvested respectively at 4th,8th,12th and 16th week after transplantation for renal function and histological examination. The expression of p-Akt and MMP-9 was detected by immuno- histochemical assay, and that expression of p-Akt mRNA by RT-PCR.Results At the 16th week after kidney transplantation, the levels of 24-h proteinuria and serum creatinine in transplant rats were higher significantly than those in LEW controls and F344 controls. The expression of p-Akt protein and mRNA was up-regulated,and there was a significantly negative correlation between the expression of p-Akt and the quantity of smooth muscular cells(SMCs)in arteriolar wall, the quality of tubuloin- terstitial infiltrated mononuclear cells and also MMP-9 level. Conclusion The up-regulation of p-Akt played an important role in the pathogenesis such as proliferation and migration of SMCs,infiltrated mononuclear cells in tubulointerstitial in CAN,and may be the signaling pathway leading to the up- regulation of MMP-9 expression in the early stage of CAN.
8.Analysis of ocular biometric parmeters in hyperopia preschool children
International Eye Science 2019;19(11):1936-1939
AIM:To measure and analyze the relationship between ocular biometric parameters and refractive status in hyperopia preschool children.
METHODS: Two hundred and three cases(405 eyes)of hyperopia were included in this study, who were diagnosed in Northwest Women and Children Hospital from January 2016 to December 2018.According to different mean spherical equivalent(SE)diopter, the children were divided into mild hyperopia, moderate hyperopia and high hyperopia groups though the retinoscopy after cycloplegia. Anterior chamber depth(ACD), lens thickness(LT), vitreous depth(VITR)and axial length(AL)were measured by A-scan. The value of K1, K2 were got by computer refractor. And analyze the relationship between ocular biometric parameters and refractive.
RESULTS: The mean anterior chamber depth for hyperopia preschool children was 3.08±0.38mm, the lens thickness was 3.91±0.34mm, the vitreous depth was 14.53±1.85mm, the axial length was 21.45±1.01mm and the average cornea K was 43.34±1.70D; There were statistically significant differences in AL and VITR among the three hyperopia groups(P<0.05). There were statistically significant difference in LT between low hyperopia and moderate hyperopia, moderate hyperopia and high hyperopia(P<0.05). There were statistically significant difference in ACD between low hyperopia and moderate hyperopia(P<0.05). No difference in K was found across the three groups(P>0.05); SE was negatively correlated with AL and ACD(P<0.01), but had no correlation with ACD, LT and K(P >0.05).
CONCLUSION: The change of AL plays an important role affecting the refractive state of preschoolers with hyperopia ametropia, which is manifested as higher the hyperopia diopter, the shorter AL and the shallower vitreous cavity, which are co-involved in the change of refractive state, while the hyperopic degree had no correlation with ACD, LT and K. The axial refractive error is the main refractive error in preschoolers.

Using universal primer Tyl-copia retrotransposon RT, the conserved reverse transcriptase domain of about 260 bp was amplified by RT-PCR from the Dendrobium officinale which induced by 100 micromol x L(-1) abscisic acid (ABA), indicating these retrotransposons activated by 100 micromol x L(-1) ABA. The amplicons were recovered and cloned, then sequenced and analyzed by related bioinformatics software. Forty-two Ty1-copia like retrotransposon RT transcriptionally activated were obtained with high heterogeneity. The length of these sequences varied from 247 to 266 bp, and was rich in AT and homology ranged from 46.3% to 98.9%. The same to Ty1-copia like retrotransposon RT of genome, different c/s-acting regulatory elements induced by stress conditions and the starting transcription signals, corresponding to CAAT box, TATA box conserved sequences and some other regulatory elements. The c/s-acting regulatory elements induced by stress conditions of reverse transcriptase transcriptionally activated of Tyl-copia retrotransposons were significantly increased than that of Ty1-copia like retrotransposon RT of genome. When being translated into amino acids, fifteen sequences presented stop codon mutation, nineteen sequences presented frameshift mutation, and all sequences presented conserved sequence "SLYGKQ" mutation. Five categories were identified through phylogenic analysis after alignment analyses of their amino acid sequences, and with Ty1-copia like retrotransposon RT of genome having low homology, which indicated that reverse transcriptase transcriptionally activated of Ty1-copia retrotransposons which induced by ABA had Significantly differences with Ty1-copia like retrotransposon RT of genome.

China Journal of Chinese Materia Medica 2014;39(2):209-215
Using universal primer Ty1-copia retrotransposon RT,43 Ty1-copia like retrotransposon RT with high heterogeneity, stop codon mutation and frameshift mutation were amplified by PCR from genomic DNA of Zhejiang Lin'an (C15) and Yunnan Guangnan (A39) of Dendrobium officinale. The length of these sequences varied from 260 to 266 bp, and was rich in AT and consistency ranged from 47.1% to 97.7%. Different c/s-acting regulatory elements induced by low temperature, heat, light, all kinds of plant growth regulating substances and the starting transcription signals, corresponding to CAAT box, TATA box conserved sequences and some other regulatory elements. When being translated into amino acids, ten sequences presented stop codon mutation, five sequences presented frameshift mutation, and thirty-seven sequences presented conserved sequence "SLYGKQ" mutation. Six categories were identified through phylogenic analysis after alignment analyses of their amino acid sequences, and with other plants (eg. Triticum aestivum, Eleocharis quinqueflora) having high homology, which indicated that horizontal transmission of retrotransposon occurred among the plants in the past.
