Objective To evaluate the influence of psychological intervention on pregnancy rate of in vitro fertilization-embryo transfer.Methods Using the key words,including in vitro fertilization-embryo transfer and psychological,the data were retrieved from CNKI,CBM,VIP,Wanfang and PubMed.The quality of eligible studies was evaluated by two reviews independently and Meta-analysis was conducted on studies.Results Totally thirteen Chinese studies and three foreign studies were included.Conclusions Psychological intervention using in the process of in vitro fertilization-embryo transfer can improve the pregnancy rate,which is worthy of clinical application.

Objective To explore the prevalence of qnr gene in gram-negative bacteria isolated from Renmin Hospital of Wuhan University. Methods qnr gene was detected by PCR in non-duplicate E.coli (n=129), E.cloacae (n=10) and K.pneumoniae (n=29) isolates.PCR technique was used to amplify the gene encoding type I integrons.Kirby-Bauer disk diffusion method was used to test the in vitro suscepti- bility of the isolates to 16 antimicrobial agents.ESBLs were investigated according to phenotypie screening and confirmatory test recom- mended by NCCLS.MIC values of ciprofloxacin were determined for qnr gene positive strains.Results qnr gene were identified in 6 clinical isolates including 5 strains of E.coli and I E.cloacae, qnr gene was not found in K.pneumoniae isolates.The 6 qnr-positive clinical iso- lates were positive for type I integrons.All the 6 strains were susceptible to imipenem but resistant to many other drugs tested.Only 2 of the E.coli isolates were susceptible to ciprofloxacin.Conclusions In Hubei Province, the prevalence of qnr is confirmed.The serious situa- tion of multidrug-resistance in clinical isolates with qnr gene requires careful monitoring of qnr-carrying isolates.

Hepatic ischemia-reperfusion injury(IRI)is a critical problem of liver surgery,especially when comes to liver transplantation.Presently.there are no effective measures for diagnosis,prevention and therapy of IRI,as the mechanisms of IRI still remain unclear.This review summarizes several new hepatic ischemia-reperfusion markers related to cell signal transduetion pathway.including transcription factor STAT,HIF-1 and PPARs,transmission factor MAPK,membrane receptor TLR4 and PARs.and iNOS.Animal studies have indicated that IRI was ameliorated by activating or blockading these markers,which might serve as targets for diagnosis,prevention and therapy of IRI.

Total mRNA was extracted from lymphocytes separated from the peripheral blood of allergic patients, and then variable region of heavy chain (VH) and variable region of light chain (VL) cDNA library were constructed by RT-PCR. Human scFv templates for rabbit reticulocyte lysate ribosome display were assembled by primers and linker peptide (Gly4Ser)3. mRNA bound in antibody-ribosome-mRNA complexes was recovered using in-situ single primer RT-PCR, and three rounds of anti-IgE scFv DNA were enriched. The target DNA fragments were double enzyme digested and ligated into plasmid pET22b (+), followed by transformation in E. coli Rosseta (DE3). Positive clones were screened using clone PCR, Dot blotting and antigen ELISA. The correct lengths of VH (400 bp) and VL (710 bp) PCR products were obtained. The expected 1,000 bp ribosome display templates were also observed in agarose gel electrophoresis. After three rounds of ribosome display target sequences were effectively enriched, leading to a library of 10(13) members. Antibodies with the highest ELISA value for IgE were generated in the strain pET-IgE-6. A human anti-IgE scFv library was successfully constructed as described herein. Ribosome display using single primer in-situ RT-PCR as the recovery procedure effectively enriched target sequences. Anti-IgE scFv with high affinity and specificity were identified. The prepared human anti-IgE scFv fragment might be self-developed to a lead drug for treating asthma. Our study provides an alternative method for rapid discovery of human antibodies of therapeutic importance.

Objective To prepare a new type of micropore porcine acellular dermis matrix with the aid of laser (LPADM),and to validate the healing effect of the LPADM through the phrase Ⅰ composite transplanting on the back of the full- thickness skin defects in SD rats.Methods In vitro,the allogeneic fibroblasts were separately cultured with the LPADM (LPADM group) or the non-pore PADM (non-pore LPADM group),while fibroblasts cultured by pure medium were used as control.After culture of 1 day,3 days and 5 days,the contents of IL-10,IL-6,TGF-β1,LN,VEGF expressed by fibroblasts were determined by double-antibody sandwich ELISA method.In vivo,the phrase Ⅰ transplantations of LPADM graft with split-thickness autologous skin were carried on the backs of the full-thickness cutaneous defects of SD rats.The healing condition was observed and analyzed by histological tests.Results The differences of the absorbance value between the LPADMgroup,PADM group and control group in each day were not statistically significant (F=0.050-1.763,P＞0.05).The transplantation of LPADM graft with split-thickness autologous skin graft resulted in high rate of surviving without signs of rejection 3 weeks later.After 1-month of transplantation,the regenerated skin was well enough to be lifted without any serious scars.Conclusions The phrase Ⅰ transplantation of LPADM graft with split-thickness autologous skin graft can accelerate the healing process of full-thickness skin wounds with high biological safety.

Objective To explore the correlation of posttraumatic growth with social support in patients with gastrointestinal neoplasms at the end of life.Methods Cluster random sampling was used to select 280 patients with gastrointestinal neoplasms who were predicted to survive no longer than 6 months in 10 first-class hospitals in Beijing and Tangshan,and they were investigated by Posttraumatic Growth Inventor (PTGI) and Social Support Rate Scale (SSRS).Results The total score of PTGI (61.13 ± 16.78)and the scores of appreciation of life (23.71±7.34),personal strength(10.67±3.55),new possibility(10.07±2.11),relating to others(12.54±5.14),spititual change(10.89±4.35)were higher than median value.The total score of SSRS(41.21±8.48) and objective support(10.27±2.75) were higer than the normal level((39.05±6.35),(8.42±2.27)) (P＜0.05).The appreciation of life was positively correlated with subjective support and the total score of SSRS (r=0.25,r=0.26,P＜0.05),personal strength was positively correlated with objective support,subjective support and the total score of SSRS(r=0.24,r=0.29,r=0.23,P＜0.05),new possibility was positively correlated with subjective support(r=0.21,P＜0.05),relating to others was positively correlated with objective support and the total score of SSRS (r=0.26,r=0.26,P＜0.05),spititual change was positively correlated with objective support and subjective support (r=0.28,r=0.31,P＜0.05).The total score of PTGI was positively correlated with subjective support and the total score of SSRS (r=0.29,r=0.29,P＜0.05).Conclusion Patients with gastrointestinal neoplasms at the end of life experience a certain extent of posttraumatic growth and high level of social support.Posttraumatic growth is positively correlated with social support.

Objective To explore the relationship between the expression levels of excision repair cross complementation group 1(ERCC1), breast cancer susceptibility gene 1(BRCA1), thymidylate synthase (TS) mRNA and clinicopathological features, prognosis in advanced colorectal cancer, and the correlation between the expression levels of ERCC1 and BRCA1. Methods The expression levels of ERCC1, BRCA1 and TS mRNA of postoperative paraffin embedded tissue were tested by real-time quantitative reverse transcription polymerase chain reaction (RT-PCR) in 49 advanced colorectal cancer cases. The results were analyzed by χ2 test of the correlation between the expression levels and clinicopathological characteristics. Patients were followed up by clinic or telephone. The prognosis was analyzed by small sample Kaplan-Meier survival analysis and Log-rank time series analysis, and P<0.05 was statistically significant. Results The expression level of ERCC1 mRNA in patients with colorectal carcinoma had no obvious correlation with the clinical and pathological characteristics such as gender, age, tumor location, lymph node metastasis, distant metastasis, CEA and differentiation degree (P>0.05). The expression level of BRCA1 mRNA had no significant correlation with the above clinical and pathological features (P>0.05) except distant metastasis (P=0.030) and differentiation degree (P=0.002). The expression level of TS mRNA had no significant correlation with the above clinical and pathological features (P>0.05) except distant metastasis (P=0.003). The expression level of ERCC1 and BRCA1 mRNA obviously correlated (P=0.002). The 1 year overall survival rate was 95.92%(47/49);the 2 year overall survival rate was 83.67%(41/49);and the 3 year overall survival rate was 73.47%(36/49). Overall survival and progression-free survival time in ERCC1 mRNA low expression group (47.8, 41.0 months) was higher than that in ERCC1 mRNA low expression group (27.3, 20.0 months) respectively (P=0.001, P=0.001). Overall survival and progression-free survival time in BRCA1 mRNA low expression group (43.7, 42.7 months) was higher than that in BRCA1 mRNA high expression group (29.3, 25.1 months) respectively (P=0.009, 0.006). Overall survival time in TS mRNA low expression group (39.8 months) was higher than that in BRCA1 mRNA high expression group (25.2 months). Conclusions The expression level of ERCC1 mRNA is not correlated with its clinical and pathological characteristics, but with its biological characteristics. BRCA1 and TS levels are correlated with invasion and metastasis. Low levels of ERCC1 and BRCA1 expression have a better prognostic effect on platinum based first-line chemotherapy for advanced colorectal cancer, and they are correlated. Low level of TS also has longer disease-free survival. Three joint detection could be used as a prognostic factor for colorectal cancer chemotherapy.

Objective To examine the retina of healthy adults,hypertensive and diabetic patients by integrated backscatter(IBS), in order to offer the reference for clinical diagnosis and differential diagnosis in the various of ocular fundus affection. Methods Retinal IBS of 120 healthy adults,40 hypertensive and 40 diabetic patients was analyzed by HP Sonos 5500. Results IBS% of retina increased with age, and it had positive correlation with age(r= 0.82 ). IBS% of hypertensive group was higher than that of healthy adult group(P 0.05 ). Conclusions IBS% of healthy adults′ retina increases with age; the retinal IBS% of the hypertensive and the diabetic patients are higher than that of the healthy adults.
