2.Effect of coloring agent on the color of zirconia.

PURPOSE: The aim of this study was to evaluate the effect of two types of coloring agents and the number of application on the color of zirconia. MATERIALS AND METHODS: Monolithic zirconia specimens (15.7 mm × 15.7 mm × 2.0 mm) (n = 33) was prepared and divided into 11 groups. Each experimental group was coded as a1-a5, w1-w5 according to the type of coloring agent and number of application. Specimens with no coloring agent applied were set as control group. The color difference of specimen was measured by using double-beam spectrophotometer, and calculated color difference (ΔE*(ab)), translucency parameter (TP). All data was analyzed with two-way ANOVA, multiple comparison Schéffe test, Pearson correlation and linear regression analysis. RESULTS: As the number of application increased, values of CIE L* was decreased, but values of CIE b* was increased in both coloring agents. However, there was no significant difference on values of translucency parameter. The color difference range of each group was 0.87 ΔE*(ab) to 9.43 ΔE*(ab). CONCLUSION: In this study, type of coloring agent and the number of application did not affect the color difference of zirconia.

4.Sensitivity and Usefulness of VE1 Immunohistochemical Staining in Acral Melanomas with BRAF Mutation.

BACKGROUND: Acral melanomas are known to have a low frequency of BRAF mutation, in contrary to higher KIT mutation. Recently, VE1 immunostaining was reported to have a good correlation with BRAF mutation status. OBJECTIVE: We aimed to evaluate the clinicopathological features of BRAF-mutated acral melanomas and validate the correlation of the VE1 immunohistochemical stains in those cases. METHODS: The clinical features (age, sex, anatomical site), and histopathological characteristics of 41 patients with acral melanoma were evaluated. We performed a next-generation sequencing to detect BRAF mutation status. We also determined the correlation of VE1 immunohistochemical staining with BRAF mutation status. RESULTS: Among 19 acral melanomas with BRAF mutation, common histopathological subtype was acral lentiginous melanoma (8/19, 42%) and nodular melanoma (8/19, 42%) and superficial spreading melanoma (3/19, 16%) followed. VE1 immunostaining results were positive in all 15 cases with BRAF V600E mutation (sensitivity 100%), and negative in 4 cases of BRAF non-V600E mutation. However, VE1 immunostaining was negative in all 22 patients with BRAF wild-type. CONCLUSION: VE1 immunostaining had a good correlation with BRAF V600E mutation status.

5.Application of Differex(TM) system and FTA(R) technology to separation of sperm DNA from mixed stains.

Specimens from sexual crimes are generally mixed stains consisted of sperm cells(from suspect) and virginal cells(from victim). We have combined two new methods - Differex(TM) system and FTA(R) technology- to overcome shortcomings of method that has been used before to separate sperm DNA from mixed stains. This methods have shown additional benefits and similar quality than using the only Differex(TM) system to the experiment. The result of our experiment represents the possibility that Differex(TM) system and FTA(R) technology would be useful methods for DNA analysis related to sexual crimes because this system can save time, labor and contamination for experiments.

6.A Case of Eccrine Chromhidriosis.

Chromhidriosis or the production of colored sweat is a rare clinical finding. Several dyes, when injected intravenously, have been shown to be secreted in eccrine gland, but in most cases the sweat is colored in skin surface, resulting from the contamination by dyes, pigment from microorganisms, or other chemicals. We report a case of eccrine chromhidriosis which revealed blue-green spots on palms, soles, buttocks, and back.

7.Thin - Layer Chromatography Search for Disperce Yellow 3 and Disperce Orange 3 in 60 Brownish Stockings.

Thin-layer (uni-dimensional and bi-dimensional) chrornatography studies were performed on 60 brownish stockings from 12 companies in Korea to search for Disperse Yellow 3 and Disperse Orange 3, which are known as common sensitizers in textiles. The results were as follows : 1) Mixture of various-colored dyes were being used to reveal the brownish hue in the stockings. 2) TLC demonstrated the presence of Disperse Yellow 3 in 53 stocking. 3) TLC demonstrated the presence of Disperse Orange 3 in 3 stockings, Although lack or more sophisticsted tests such as spectroseopic, infra red or nuclear spectra identification. These results suggest a strong clue to the presence of Disperse Yellow 3 and Disperse Orange 3 in the commercial stocking now being used in Korea.

8.Expressions of MIB-1, p53 and CEA in Endocervical Glandular Lesions.

BACKGROUND: Endocervical glandular lesions include glandular atypia (GA), endocervical glandular dysplasia (EGD), adenocarcinoma in situ (AIS), and invasive adenocarcinoma (IA). The diagnosis of malignant glandular lesions is occasionally difficult to distinguish from benign mimickers, and the morphologic features of EGD remain unsettled. METHODS: Immunohistochemical stains for MIB-1, p53 and CEA were performed on 81 cases of paraffin-embedded endocervical glandular lesions including 22 IA, 15 AIS, 15 EGD, 13 GA, 8 microglandular hyperplasia (MGH) and 8 tubal metaplasia (TM). RESULTS: The MIB-1 labelling index of IA was 59.68%, 69.53% for AIS, 26.60% for EGD, 16.03% for benign. p53 overexpression was noted in 4 (18%) cases of IA, 3 (20%) of AIS, but none of EGD and benign lesions. It was Interesting to note that one case of MGH showed p53 staining in low intensity. Diffuse strong cytoplasmic CEA positivity was present in all of IA and AIS, whereas seven (47%) of 15 EGD and 12 (41%) of 29 benign lesions showed focal cytoplasmic CEA positivity. There were significant differences in MIB-1 and CEA immunostainings among the adenocarcinoma, EGD, and benign glandular lesions. Adenocarcinoma was closely related to p53 overexpression, although occurring in a low percentage of the cases. CONCLUSION: MIB-1 immunostaining can be useful in differentiating among endocervical adenocarcinoma, endocervical glandular dysplasia and benign glandular lesions. p53 overexpression might be helpful in the diagnosis of adenocarcinoma.

9.Evaluation of Automatic Acid-Fast Bacilli Stainer AT-2000F.

BACKGROUND: Sputum smear microscopy is rapid, economic, and useful to detect patients with transmittable tuberculosis, albeit laborious. We aimed to evaluate the usefulness of an automated acid-fast bacilli stainer, which had been developed for lowering the labor and maintaining or increasing the staining quality. METHODS: One hundred sputum samples including some known positive smear specimens which were selected from clinical specimens requested for smear and culture for mycobacteria at Pusan National University Hospital, were used for evaluation. Auramine/rhodamine fluorescent acid-fast stainings were performed manually or by using the automated stainer, AT-2000F (Dagatron, Ilsan, Korea). Ziehl-Neelsen stain was also performed simultaneously. RESULTS: Concordance rate between automated and manual fluorescent stains was 98.0% and that between automated fluorescent and manual Ziehl-Neelsen stains was 88.0%. In all discordant cases, the automated stains showed one-grade higher results compared to the respective manual fluorescent or Ziehl-Neelsen stains. With the automatic stainer, all staining procedures were processed automatically except for slide loading and unloading. The process time was reduced by a half, and the slide-to-slide or day-to-day variations of staining quality were reduced compared with the manual fluorescent stain. CONCLUSION: Acid-fast bacilli stain using automated stainer AF-2000F can reduce the processing time, labor, and variations of staining quality, and enhance or maintain the detection of positive smears.

10.Comparative Study for METHODSfor Diagnosing Onychomycosis.

BACKGROUND: Onychomycosis is very common nail problem, so an in xpensive, quick and sensitive test is essential for screening nail specimens. Recently, there is a report of new method for diagnosing onychomycosis u.;ing KOH treated nail clippings which vri then crushed and finally stained with periodic acid-Schifft(PAS) stain (KONCPA). OBJECTIVE: We evaluate the susefulness of the new methods using KOH treated nail clippings and nail debris which were then crushed and finally stained with FS (KONPA) or chlorazol black E(KONBE), for the diagriosis of onychomycosis. METHODS: We compare different methods for diagnosing onychonycisis such as KOH stains, fungal cultures, histologic evaluation, SEM, KONCPA, KONBE, and KOHJPA. RESULTS: KONPA was proved to be more effective indentifying uiigal hyphae in comparison with conventional KOH nail scraping preparation, fungal culture, and FONBE. The positive rates of each method were 74%, 46%, 43%, and 63%, respectively. Also, KENPA proved to be more rapid and easy to perform in cotnparison to the histologic evaluation of tiail clippings and SEM. CONCLUSION: KONPA is a sensitive, quick, and readily available teled for use in clinical settings in cases that are highy siispected to be onychomycosis clinicaly, but show negative results using conventional methods
