1.Study of the bone-like apatite's depositing induced by collagen I with its mechanism.

The collagen I was made with the dialysis method of enzymolysising the pig skin, and the static deposition in vitro of calcium phosphate was comparative studied by X-ray diffraction (XRD) and infrared spectroscopy (FTIR) under the condition of pH7. 4, Ca/P 1.67 and whether adding the collagen I into the system. Then the chemical composition of the sedimentary product and the diversification of the collagen I 's IR and Raman spectra (RS) before and after the mineralization were analyzed. The results showed that,under the physiological pH condition that there was not any collagen I, though Ca/P reached up to 1.67, the sedimentary product was CaHPO4 x 2H2O yet, however, after adding collagen I into the system, the bone-like apatite was deposited, which proved that collagen I indeed had the effects on the inducing of the bone-like apatite's mineralization in vitro; there was obviously mutual coordination action between collagen I and its mineralization product--bone-like apatite, which caused that amide peak I red-shifted, amide peak II and amide peak III decreased significantly or disappeared on the IR of collagen I, which maybe was the mechanism that how collagen I induced the depositing of the bone-like apatite.

2.An autopsy specimen study of benign hyperplastic nodules in the peripheral zone of the prostate.

OBJECTIVETo investigate the correlation and anatomic association of benign hyperplastic nodules in the peripheral zone (PZ) with those in the transition zone (TZ) of the prostate, and to compare the histological components of the two kinds of nodules.
METHODSWe obtained benign hyperplastic nodules specimens from the PZ and TZ by autopsy, measured the distance between the outer surface of the nodules and the inner gland, observed the integrity of the surgical envelope of the prostate, and determined the histological components of the two kinds of nodules by HE staining, immunohistochemistry and automatic quantitative image analysis.
RESULTSThe surgical envelope of the prostate was integrated and the distance between the nodules of the PZ and the outer surface of the inner gland was about 2.5 to 5 mm ([3.9 +/- 0.8] mm), with no signs of anatomic connection in between. The stromata and epithelia in the nodules accounted for (69.32 +/- 8.35)% and (16.08 +/- 5.36)% in the PZ and (74.58 +/- 8.95)% and (15.82 +/- 6.41)% in the TZ.
CONCLUSIONBenign hyperplastic nodules may originate from the PZ of the prostate and not correlate with the inner gland hyperplasia in the TZ, but with no statistical difference between the histological components of the two kinds of nodules.

3.Major constituent proteins in donkey hide and their interaction.

OBJECTIVETo analyze the constituent proteins in donkey hide, the key ingredient for Ejiao, an important traditional Chinese medicine for the blood-related conditions, in hope to eventually decipher the biochemical mechanism behind Ejiao's prominent medicinal efficacy.
METHODTwo methods were employed to extract proteins in donkey skin. One used TriPure isolation reagent to extract the total proteins in donkey skin. Another used 1% sodium dodecyl sulfate (SDS) to heat the sample at 100 degrees C overnight. And then sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and capillary HPLC were used to analyze the component of proteins.
RESULTThere are not only collagen alpha1 (I) and collagen alpha2 (I), but also serum albumin in donkey skin. The content is over 25% in total proteins with the method of TriPure isolation reagent. The content of donkey serum albumin is up to 20% with the method of 1% SDS heating. And two bands, molecular weight are nearly 200 kDa,were found on 7.5% SDS-PAGE. Extracted these proteins to analyze with capillary HPLC, they were found to be the complex products of collagen and serum albumin of donkey.
CONCLUSIONDonkey serum albumin is a main protein component in the hide, which is a clue to expose is the effect of Ejiao on blood.

4.The effect of the Sanqi qisodium hyaluronate gel on the collagen of epidural scar after rabbits laminectomy.

OBJECTIVESTo study the effects of Sanqi qisodium hyaluronate gel on collagen-I and collagen-III expression in the process of rabbits' epidural scar formation after operation.
METHODSNinety-six white rabbits with 6-month-old, half males and half females, weighted from 2 to 2.5 kg, which were randomly divided into normal saline group (A), Sanqi group (B), qisodium hyaluronate group (C) and Sanqi qisodium hyaluronate gel group (D). The laminectomy of rabbits were performed in group A, B, C, D, the duras were surrounded with normal saline, Sanqi liquid, qisodium hyaluronate and Sanqi qisodium hyaluronate gel respectively. Animals of each group were killed at 1, 2, 4, 8 weeks after operation. Use Masson staining for histological observation of collagen, and in situ hybridization staining for the analysis of collagen-I and collagen-III expression.
RESULTSIn the Masson staining, Sanqi qisodium hyaluronate gel group was more regular than the control group in the shape of collagen texture. As to the expression of collagen-I, and Sanqi qisodium hyaluronate gel group was lower than normal saline group, the Sanqi group and qisodium hyaluronate group at 4 weeks after using medicine (P < 0.01); while the Sanqi qisodium hyaluronate gel group was higher than normal saline group, Sanqi group and qisodium hyaluronate group in the collagen-III expression (P < 0.01).
CONCLUSIONSSanqi qisodium hyaluronate gel could improve collagen's arrangement of the rabbit's epidural scar after operation, reduce its rigidity and increase flexibility.

5.Quantification of type I and III collagen content in normal human skin in different age groups.

OBJECTIVETo quantify the content of type I, III collagen and their ratio in normal human skin of different age, and to explore the regulation of changes.
METHODSThe normal human skin specimens were obtained from 6 spontaneously aborted fetus and 56 burn patients of different ages, including infants (newborn -3 years), pre-school group ( > 3, < or =7 years), adolescent group ( >7, < or = 18 years), youth and middle age group ( > 18, < or = 50 years), and elderly group ( > 50 years), were studied. The total collagen content were determined by hydroxyproline method. The contents of type I, Ill collagen and their ratio were examined by immunohistochemistry.
RESULTSThe total collagen content decreased along with increase in age, and it was highest in fetus [(543 +/- 13) microg/g]. The ratio between type I and Ill collagen increased along with increase in age. The content of type III collagen was highest in fetus [(278 +/- 7) microg/g], and it decreased along with increase in age. The content of type I collagen content was [(265 +/- 7) microg/g] in fetus, and it was increased slightly in infant and pre-school groups, then decreased along with advance in age.
CONCLUSIONDecomposition of type III collagen in normal human skin may exceed its synthesis after birth immediately, leading to its reduction. Synthesis of type I collagen in normal human skin is dominant before 8 years old, and it shows an opposite tendency afterwards.

6.Effect of Capparis spinosa on fibroblast proliferation and type I collagen production in progressive systemic sclerosis.

OBJECTIVETo investigate the effects of ethanolic extract from Capparis spinosa (ECS) on the fibroblast proliferation and type I collagen production in normal and progressive systemic sclerosis (PSS).
METHODCellular activity was determined by the MTT method. Apoptosis was detected by flow cytometry analysis of Annexin V-stained cells. The expression levels of type I collagen messenger RNA and protein were analyzed by RT-PCR and western blot analysis.
RESULTECS could significantly inhibit the proliferation of fibroblast and reduced the expression of alpha2 (I) collagen mRNA and type I collagen protein in PSS in a dose-and time-dependent manner. ECS did not affect the proliferation of fibroblast and expression of type I collagen mRNA and protein in normal human. ECS could counteract the harmful effects on fibroblast by H2O2.
CONCLUSIONECS can effectively inhibit the fibroblast proliferation and type I collagen production in PSS.

7.Effect of Panax notoginseng saponins on human kidney fibroblast.

OBJECTIVETo study the effect of Panax notoginseng saponins (PNS) on cell proliferation, type I collagen secretion and integrin beta 1 expression in human kidney fibroblast (KFB).
METHODSKFB were cultured and stimulated by PNS in vitro. The cell proliferation, type I collagen secretion and integrin beta 1 expression in KFB were measured by methyl thiazolyl tetrazolium (MTT), enzyme-linked immunoabsorbent assay (ELISA) and flowcytometry respectively.
RESULTSWithin its optimal concentration range and effective time range, PNS could obviously inhibit the proliferation, type I collagen secretion and integrin beta 1 expression (all P < 0.05) in human KFB.
CONCLUSIONPNS would possibly be an effective drug for renal interstitial fibrosis prevention and treatment.

8.Comparison between heparin-conjugated fibrin and collagen sponge as bone morphogenetic protein-2 carriers for bone regeneration.

Bone morphogenetic protein-2 (BMP-2) is used to promote bone regeneration. However, the bone regeneration ability of BMP-2 relies heavily on the delivery vehicle. Previously, we have developed heparin-conjugated fibrin (HCF), a vehicle for long-term delivery of BMP-2 and demonstrated that long-term delivery of BMP-2 enhanced its osteogenic efficacy as compared to short-term delivery at an equivalent dose. The aim of this study was to compare the bone-forming ability of the BMP-2 delivered by HCF to that delivered by clinically utilized BMP-2 delivery vehicle collagen sponge. An in vitro release profile of BMP-2 showed that HCF released 80% of the loaded BMP-2 within 20 days, whereas collagen sponge released the same amount within the first 6 days. Moreover, the BMP-2 released from the HCF showed significantly higher alkaline phosphatase activity than the BMP-2 released from collagen sponge at 2 weeks in vitro. Various doses of BMP-2 were delivered with HCF or collagen sponge to mouse calvarial defects. Eight weeks after the treatment, bone regeneration was evaluated by computed tomography, histology, and histomorphometric analysis. The dose of BMP-2 delivered by HCF to achieve 100% bone formation in the defects was less than half of the BMP-2 dose delivered by collagen sponge to achieve a similar level of bone formation. Additionally, bone regenerated by the HCF-BMP-2 had higher bone density than bone regenerated by the collagen sponge-BMP-2. These data demonstrate that HCF as a BMP-2 delivery vehicle exerts better osteogenic ability of BMP-2 than collagen sponge, a clinically utilized delivery vehicle.

This study aims to investigate the effect of total flavones of Fructus Chorspondiatis (TFFC) on the mRNA and protein expression of collagen type I and III of rat cardiac fibroblasts (CFs) induced by angiotensin II (Ang II), and explore its anti-myocardial fibrosis molecular mechanism. Neonatal rat CFs were prepared from Sprague-Dawley rats (1-3 d after birth). The expression of collagen type I and III mRNA and protein were measured by RT-PCR and Western blotting, respectively. The study showed that stimulation of neonatal rat CFs with 100 nmol.L-1 of Ang II for 72 h resulted in a significant increase of the expression of collagen type I and III mRNA and protein. The changes on the expression level were blocked by TFFC. The results demonstrated that TFFC can inhibit myocardial fibrosis induced by Ang II in rats, which is probably associated with the collagen type I and III mRNA and protein levels up-regulated by Ang II, and TFFC was shown to decrease the expression levels of collagen type I and III mRNA and protein.

10.Effects of 5F from Pteris semipinnate L on growth of human pathological scars in nude mice.

OBJECTIVETo investigate the effect of 5F from Pteris semipinnate L on the growth of human pathological scar in nude mice.
METHODS5F from Pteris semipinnate L was administered at different doses in nude mouse models bearing human pathological scars. The morphology, histology, tumor growth factor-beta1 and type I collagen content of the scar tissues were examined after the administration.
RESULTSAdministration of 5F significantly reduced the volume of the implanted pathological scars in the nude mouse models, and histologically, the scar tissue exhibited a transition to the normal scar architecture with decreased TGF-beta1 and type I collagen content.
CONCLUSION5F could effectively inhibit the growth of pathological scars in nude mice.
