Objective To determine the value of vascular indication of sexual excitation response (VISER) in the diagnosis of erectile dysfunction (ED). Methods Fifty three cases of ED underwent PGE1 intracavernous injection,VISER and color duplex ultrasonography.The comparison of the results was performed. Results Forty three cases were in Porst Grade 3, whose average cavernous pressure and carvenous pressure index were 144.29 mmHg and 1.24,respectively.Of them,42 cases were of normal penile vascular function,the remaining one was of venous ED;10 cases were in Porst Grade 2, whose average cavernous pressure and carvenous pressure index were 87.76 mmHg and 0.81,respectively.There were significant differences in the average cavernous pressure and carvenous pressure index between the two groups ( P
3.Preliminary Isolation and Purification of Spermatogonial Stem Cells

Objective To obtain a simple method to isolate and purify spermatogonia stem cells. Methods The testis of newborn mouse were digested by modified two step digest method, and the type A spermatogonia cells were separated by different adhesive time with Sertoli cells. Results High concentration (95%) purified type A spermatogonia cells were acquired, and the spermatogonia transplanting experiment demonstrated the efficiency of the method.Conclusions This method is easy and efficient to isolation and purification of spermatogonial stem cells.
4.Diffusion tensor MR imaging in the assessment of cognitive functions in children with periventricular leukomalacia

Objective To investigate the values of diffusion tensor MR imaging(DTI)and fiber tractography(FT)in the assessment of cognitive functions in children with periventricular leukomalacia (PVL),to facilitate to understand the mechanism of PVL children's cognitive functions.Methods Forty six children with PVL and 16 age-matched volunters were recruited.Developmental quotient(DQ)of all cases was recorded.According to the DQ scores,patients were divided into 3 groups:mild in 11 patients,moderate in 19,and severe in 16.DTI was performed in all cases and fractional anisotropy(FA)values were be calculated.Non-paired t test was used to compare the FA values of major white matter fibers between patients group and controls group.One-way ANOVA was used to compare the FA values among 4 groups.Results Compared with controls,all 46 patients with PVL showed a significant mean FA reduction in left corticospinal tract(CST)(0.476 ±0.064 vs.0.531 ±0.064),bilateral anterior limb of internal capsula (ICAL)(left 0.357 ±0.050 vs.0.405 ±0.081,right 0.370 ±0.040 vs.0.405 ±0.036),posterior limb of internal capsule(ICPL)(left 0.483 ±0.044 vs.0.546 ±0.091,right 0.485 ±0.046 vs.0.547 ±0.083),arcuate fasciculus(AF)(left 0.367 ± 0.069 vs.0.433 ± 0.097,right 0.363 ± 0.064 vs.0.421 ±0.091),posterior thalamic radiation(PTR)(left 0.390 ± 0.059 vs.0.459 ± 0.067,right 0.382 ± 0.047 vs.0.446 ± 0.064),anterior CG(ACG)(left 0.362 ± 0.056 vs.0.423 ± 0.057,right 0.345 ± 0.056 vs.0.417 ± 0.080),superior longitudinal fasciculus(SLF)(left 0.323 ± 0.050 vs.0.426 ±0.102,right 0.341 ± 0.056 vs.0.416 ± 0.092)and splenium of the corpus callosum(SCC)(0.535 ±0.090 vs.0.606 ±0.060)(t =2.037-3.745,P <0.05).The severity of cognitive impairment companied with decrease of the FA values of all fibers.The FA values of bilateral CST,ICAL,ICPL,left AF,bilateral PTR,ACG,SLF and SCC were significantly different among different groups(F =3.021-7.146,P < 0.05).Conclusions The damage widespread of white matter bundles is found in children with PVL DTI and FT can provide details in the assessment of cognitive impairment in children with PVL.
5.Effect of Irbesartan and Triptolide combination on the level of urine protein in patients with diabetic nephropathy at high altitude area

ObjectiveTo observe the effect of irbesartan and triptolide combination on the level of urine protein in patients with diabetic nephropathy at high altitude area.MethodsFifty patients with diabetic nephropathy (24-hour urine protein excretion over 1.0 g and serum creatinine level below 265.2 μmol/L) at high altitude area were randomly divided into two groups,the control group were treated with irbesartan 150 mg/d for three months,and the treatment group received irbesartan 150 mg/d combined with triptolide 40 mg/d for three months.24-hour urine protein concentration,arterial pressure,liver function and renal function were measured before and after the treatment.Results After three months' treatment,the levels of 24-hour urine protein and arterial pressure were significantly lower in both control and treatment group (P < 0.01 ).Twenty-four hour urine protein in treatment group were reduced from ( 8.34 ± 1.29) g before treatment to (6.42 ± 0.95 ) g after treatment ( t =5.994,P < 0.001 ).Twenty four-hour urine protein in control group were reduced from (8.57 ± 0.53 )g before treatment to (7.10 ± 0.79 )g after treatment( t =7.730,P < 0.001 ).Systolic pressure in treatment group were reduced from ( 152.04 ± 18.80)mm Hg before treatment to ( 131.24 ± 10.56)mm Hg after treatment(t =4.817,P < 0.001 ) ; Diastolic pressure in treatment group was reduced from (93.60 ± 11.36 )mm Hg before treatment to ( 82.68 ± 7.30) mm Hg after treatment ( t =4.053,P < 0.001 ).Systolic pressure in control group were reduced from ( 151.20 ± 10.17 ) mm Hg before treatment to ( 130.00 ± 10.10 ) mm Hg after treatment(t =7.396,P < 0.001 );Diastolic pressure in treatment group were reduced from (92.76 ± 7.03 )mm Hg before treatment to (84.20 ± 7.56)mmHg after treatment (t =4.147,P < 0.01 ).No statistic differences were observed in liver function and renal function before and after the treatment ( P > 0.01 ).Conclusion Irbesartan and triptolide combination can reduce 24-hour urine protein to a certain extent and donot adversely affect liver function and renal function in patients with diabetic nephropathy at high altitude area
6.Mouse testicular gene expression pattern differences between spermatogonial stem cell proliferative and differential stages

Objective To detect the mouse testicular gene expression pattern differences be-tween spermatogonial stem cell (SSC) proliferative and differential stages and study the molecular reg-ulation mechanism in SSC proliferation and differentiation. Methods With the interval of 24 days, male Kunming mice were injected intraperitoneally with two doses of busulfan (10 mg/kg) to establish spermatogenesis regeneration models. 36 k Mouse Genome Array was used to detect the differential gene expression profiles between the stages of SSC proliferation and differentiation. Bioinforrnsties analysis was conducted in GO (gene ontology) and KEGG (Kyoto Encyclopedia of Genes and Ge-nomes) pathway to describe the potential roles that may play in spermatogonial stern cells behavior regulation. Results Nine hundred and eleven differential expression genes were identified by gene arrays in mice testes, consisting of 608 up-regulated and 303 down-regulated in SSC proliferation stage and SSC differentiation stage. The differential expression genes were classified by their biological process, molecular function and cellular component, respectively. Alterations with statistical signifi-cance (P<0.05)appeared in 84 KEG(;signal pathways, including Notch and Wnt signaling pathways which had been proved to be important for stem cell maintenance. Fifty-six differential expression genes were selected as genes related to stem cells, among which 40 genes were up-regulated, including some stem cell biomarkers(such as Cd9, StraS, hgbl-, Oct4 and Thyl)and some growth factors(such as Fgf2, Pdgfa and Csfl). Conclustion The regulation of SSC proliferation and differentiation involves inmany differentially expressed genes in various signal pathways. This study provides a molecular basis for the elucidation of the molecular mechanism behind self-renewal and differentiation of spermatogonial stem cells.
7.Clinical analysis of influencing factors of fluid extravasation in patients with calyceal calculi treated by ureteroscopic holmium laser lithotripsy

Objective To explore the underlying clinical factors and precautionary measures of fluid extravasation in patients with calyceal calculi treated by ureteroscopic holmium laser lithotripsy.Methods A retrospective review was made on clinical records of 138 patients with calyceal calculi receiving retrograde ureteroscopic holmium laser lithotripsy from May 2005 to March 2009. The relevance was studied between the occurance of fluid extravasation complications and various clinical factors using x2 test and binary Logistic regression. The clinical factors included patients' sexes, age groups (<30 years, 30-50 years, >50 years), history of treatment (ESWL or open surgery) for upper urinary tract calculi, preoperative upper urinary tract infection, intraoperative placement of ureteral catheter and the length of procedure duration (< 50 min, 50-80 min, > 80 min). Results Fluid extravasation complications occurred in 24 patients. The sexes and age groups were irrelevant to the occurance of fluid extravasation complications; while history of ESWL or open surgery and preoperative infection in upper urinary tract, without intraoperative ureteral catheter placement and long duration of procedure were responsible for the higher rates of the fluid extravasation complications.Conclusion Reasonable selection of patients and timing of operation, regular intraoperative ureteral catheter placement and control the length of procedure duration help to reduce fluid extravasation during retrograde ureteroscopic lithotripsy.
8.Experimental study on the effect of free radical on kidney reperfusion injury caused by infrarenal abdominal aorta occlusion in rat model

Objective To study the effect of flee radical on kidney repedusion injury caused by infra-renal abdominal aorta occlusion in rat model and its possible mechanism. Methods Forty-two healthy Wis-ter rats were randondy divided into 6 groups as following ( n = 7) : the control group( sham group) ; simply isehemia 5 h without reperfusion( group I) ; 2 hours reperfusinn following ischemia 5 h ( I/R 2 h), 4 honrs reperfusion following isebemia 5 h ( I/R 4 h), 8 hours reperfusinn following ischemia 5 h( I/R 8 h) and 12 hours reperfusion following ischemia 5 h ( I/R 12 h). In each group the rats were killed to obtain samples of blood and kidney at the specified time points. The contents of BUN, Cr, MDA, SOD in blood and in renal homogenate were measured in each group. We observed the morphological changes of kidney and muscles of lower limb by light microscope. Results BUN level of serum in model group I, I/R 2 h, I/R 4 h, I/R 12 h were higher obvionsly than those of control group, which were maximal in I/R 4 h, then decreased. MDA level of plasma in model group I, I/R 2 h, I/R 4 h, I/R 8 h, I/R 12 h groups were higher obvionsly than those of control group, which were maximal in I/R4 h group, then decreased. SOD level of plasma in model I/R 4 h, I/R 8 h groups were lower obviously than those of control group; SOD level of renal homogenate in model group I, I/R 2 h, I/R 8 h, I/R 12 h groups were higher obviously than those of group I/R 4 h, which were minimal in I/R 4 h group, then increased. By light microscope: The injury degree of kidney and muscles of lower limb in ischemia group was slight, the injury degree of repedusion group was severer than ischemia group. Conclusion The kidney repedusion injury caused by infrarenal abdominal aorta occlusion in rat model might be concerned with the increase of lipid peroxidatian damage after ischemia-repedusion in-jury of lower limbs.
9. Oxidative stress is involved in the apoptosis of human breast cancer MCF-7/ADR cells induced by valdecoxib

OBJECTIVE: To study the effect of selective COX (cyclooxygenase)-2 inhibitor valdecoxib on the growth of human breast cancer MCF-7/ADR(MCF-7/adriamycin) cells. METHODS: MTT assay was used to observe the effect of drugs on the growth of cells. Flow cytometry and Hoechst 33258 dye were used to detect apoptosis of MCF-7/ADR cells. The levels of GSH and GSSG were detected by kit; Laser confocal microscopy was used to detect the levels of ROS. RESULTS: Valdecoxib significantly inhibited the growth of human breast cancer MCF-7/ADR cells and induced apoptosis of the cells. Caspase 3 inhibitor Ac-DEVD-CHO and caspase inhibitor Z-VAD-FMK antagonized the inhibitory effect of valdecoxib on MCF-7/ADR cell growth. Valdecoxib significantly decreased GSH/GSSG ratio and increased the level of ROS, and antioxidant V-acetylcysteine antagonized the inhibitory effect of valdecoxib on MCF-7/ADR cell growth. CONCLUSION: The apoptosis of human breast cancer MCF-7/ADR cells induced by valdecoxib is associated with increase of ROS.
10.Optimization of Extraction Process for Paeoniae radix Alba by Central Composite Design-Response Surface Methodology

Objective To optimize the extraction process of Paeoniae radix alba. Methods Extraction rates of paeoniflorin and total glucosides from Paeoniae radix alba and yield of dry extract were served as research indexes. Extraction process was optimized by central composite design-response surface methodology. Results Optimum condition of extraction technology was to extract 2 times with 9-fold of 85.00% ethanol for 100 min each time. Results of the three batches of confirmatory test were accordant with the predicted values. Conclusion The observed and predicted values were close to each other,and the optimum formulation by CCD-RSM was successful. The established model has a reliable predictability. The optimum extraction process is reasonable,stable and feasible.
