2.Analysis of clinical characteristics and follow-up of patients with panniculitis

Objective To explore the clinical characteristics, treatment and outcomes of patients with panniculitis. Methods Clinical data of patients with panniculitis admitted to our hospital were analyzed and followed up. The disease was classified into two sub-types: cutaneous type and systemic type. The clinical profile and treatment as well as the outcomes were compared. Results The mean follow up time of 61 in-patients with panniculitis was 49.2 months (range: 2~216 months). Remission could be achieved in 17 patients, recurrent episodes happened in 26 patients. Eighteen patients turued out to be diffuse connective tissue disease (CTD), hematological disorders, malignant tumors and tuberculosis during follow-up. The age at sympton onset was 33±17 years old in 43 patients with primarily panniculitis which eutaneoua sub-tye was 12 patients and systemic sub-tye was 31 patients. The male: female ratio was 1:15. Patients with systemic sub-type were associated with liver (25 patients), spleen (23 patients), respiratory system (11 patients), kidney (12 patients), and circulatory (3 patients) system involvement. Nine patients with systemic sub-type presen-ted white blood cell decrease while none of in the cutaneous sub-type. The difference was significant (P=0.044). Biopsy of the lesions had shown nuclear dust in 16.3% of primarily panniculitis patients. Conclusion Pannieulitis can associate with systemic rheumatic diseases, malignant tumors, hematological disorders and tuberculosis. The main systemic involvements with systemic sub-type are liver and spleen, and may have whiteblood cell decreasing. Some patients could achieve remission when treated with corticosteroids and/or immunosuppressive drugs. However,the majority of patients will have recurrences when corticosteroids is tapered.
3.Study progress of TCM syndrome of type 2 diabetes mellitus

We summarized the researches of syndrome differentiation for type 2 diabetes mellitus (T2DM) in this article. According to the constitution feature of obesity and non-obesity and analysis of researches of different medical scholars, we put forward that the easy-suffering constitutions were the basis of the evolution and development of syndromes of T2DM, and the different inducements were the conditions of that courses. The persons whose constitutions were of qi-deficiency of spleen and kidney and excessive stomach often appeared a obese physique, and the people whose constitutions were of yin-deficiency of both the liver and kidney usually showed a non-obese shape. Affected chronically by the inducements, the evolution and development of the syndromes had its own rules respectively corresponding to the different constitutions as the diseases go on. Therefore, to integrate the constitution, disease stage and pathological change with specific zang-fu organs can better grasp its rules of syndrome differentiation.
4.Clinical study on treatment of senile severe pneumonia by Qingre-Tongfu enema combined with non-invasive ventilation with syndrome of excess of Yangming fushi

Objective:To investigate the effect of Qingre-Tongfu enema combined with noninvasive positive pressure ventilation on the ventilation function of elderly patients with severe pneumonia. Methods:A total of 60 elderly patients with severe pneumonia with syndrome of excess of Yangming fushi from September 2017 to August 2019 in the respiratory department of the First People’s Hospital of Dongcheng District, Beijing were randomly divided into two groups, 30 cases in each group. The control group was treated with western medicine to control infection and noninvasive positive pressure ventilation. The treatment group was treated with Qingre-Tongfu enema on the basis of the control group. The Traditional Chinese Medicine Syndrome score (TCMSSS), Acute Physiology and Chronic Health Evaluation Ⅱ(APACHE-Ⅱ) and Clinical Pulmonary Infection Score (CPIS) were compared before and after treatment, The Pneumonia Severity Index (PSI) was compared. Serum procalcitonin (PCT) was detected by upconversion luminescence immunoassay, CRP was detected by double antibody sandwich ELISA, WBC was detected by automatic hematology analyzer; partial pressure of carbon dioxide (PaCO 2), oxygen saturation (SaO 2), partial pressure of oxygen (PaO 2) were detected by blood gas analyzer, oxygenation index (OI) and respiratory rate (RR) were calculated. The complications were investigated and the clinical efficacy was evaluated. Results:The total effective rate was 96.7% (29/30) in the treatment group and 73.3% (22/30) in the control group, with significant difference between the two groups ( χ2=4.706, P=0.030). After treatment, the levels of serum PCT, CRP and WBC in the treatment group were significantly lower than those in the control group ( t=15.359, 25.784 and 13.460, respectively, P<0.01); after treatment, the levels of SaO 2 [(93.18 ± 3.79)% vs. (88.78 ± 5.56)%, t=3.584], PaO 2 [(86.81 ± 4.01) mmHg vs. (80.01 ± 4.76) mmHg, t=5.975], OI [(285.53 ± 15.05) mmHg vs. (227.65 ± 12.37) mmHg, t=16.272] in the treatment group significantly were higher than those in the control group ( P<0.01); PaCO 2 [(43.28 ± 6.84) mmHg vs. (48.83 ± 7.66) mmHg, t=-2.956], RR [(22.00 ± 3.79) times/min vs. (26.30 ± 3.73) times/min, t=-4.434] in the treatment group were significantly lower than those in the control group ( P<0.05). After treatment, the scores of TCMSSS, APACHE-Ⅱ, CPIS and PSI in the treatment group were significantly lower than those in the control group ( t=41.310, 11.035, 8.399, 5.752, P<0.01). The treatment group in the course of antibiotics, mechanical ventilation time, hospital stay were significantly shorter than the control group ( P<0.01). Conclusion:Qingre-Tongfu enema combined with noninvasive positive pressure ventilation can improve the clinical symptoms of elderly patients with severe pneumonia, reduce inflammatory reaction, shorten the time of mechanical ventilation and antibiotic treatment, and improve the clinical efficacy.
5.Radiation-induced autophagy in colon cancer cells and its effect in radiation responses

Objective To observe the autophagy induced by heavy ion and X-ray radiations in SW480 cells and its effect in radiation responses.Methods The cellular ultrastructure was observed with a transmission electron microscope (TEM).The autophagy vesicles were labeled by monodansylcadaverin (MDC) fluorescence.The levels of LC3 which were induced by radiation and rapamycin were measured by Western blot assay.Cell proliferation was detected by MTT assay.The invasion of SW480 colon cells was measured with a Transwell.Cell wound healing assay was used to observe the migration of SW480 colon cells.Results Heavy ion and X-ray radiation could induce autophagy in SW480 cells in a dose dependent manner i.e., the autophagy level increased along with irradiation dose (F =458.526, P < 0.05) , but this induction of autophagy was inhibited by chloroquine.Rapamycin could also induce autophagy in SW480 cells and it had a synergistic effect with irradiation (F =189.393, P < 0.05).The abilities of invasion, migration and proliferation of SW480 cells were reduced by irradiation and the combination treatment of irradiation and rapamycin (F =194.692, 629.917, 302.903, P < 0.05) , but they were enhanced by chloroquine (F =194.692, 629.917, 302.903, P < 0.05).Conclusions Ionization radiation causes cell death by inducing autophagy.
6.Therapeutic Effects of Artemisia Argyi Ferment Substance on Systemic Candida Albicans Infection

Objective To investigate the therapeutic effects and mechanism of Artemisia argyi ferment substance on systemic Candida albicans infection. Methods The model of systemic Candida albicans infection was established in immunosuppressed mice. The model mice were randomly divided into the model control,Artemisia argyi ferment substance( AAFS) at different doses(100,200,and 400 mg·kg-1 )and fluconazole group(20 mg·kg-1 ),30 mice in each. Mice in each treatment group were given therapeutic drugs by gavage for 5 consecutive days,twice daily. The survival of mice was determined 21 days after the model was set up. The serum levels of IFN-γand IL-2 were determined by ELISA. The proliferation activity of T lymphocyte in the spleen was detected by MTT assay. The number of living fungi in liver and kidney tissues was counted. Results Compared with the model control,AAFS at middle and high doses and fluconazole significantly increased the survival rate of mice,the serum levels of IFN-γand IL-2,and the proliferation activity of T lymphocyte in the spleen,but decreased the number of living fungi in tissues(P〈0. 01). Compared with low dose AAFS,middle and high doses of AAFS and fluconazole showed significantly different effect on each index(P〈0. 05 or P〈0. 01),but there was no difference among these groups(P〉0. 05). Conclusion AAFS at 200-400 mg·kg-1 has inhibitory effects on systemic Candida albicans infection in mice,the mechanism of which is related to increasing the proliferation of T lymphocyte in spleen and the levels of IFN-γand IL-2 in serum.
7.Effect of hydrogen sulfide on oxidative stress in hypoxic pulmonary hypertension

Objective:To study the modulatory effect of hydrogen sulfide (H2S) on oxidative stress in the development of pulmonary hypertension induced by hypoxia. Methods:Twenty male Wistar rats were randomly divided into control group (n=6), hypoxic group (n=6) and hypoxia+NaHS group (n=8). Hypoxic challenge was performed everyday for 21 days. NaHS solution was injected intra-peritoneally everyday before hypoxic challenge for rats in the hypoxia+NaHS group. After 21 days of hypoxia, the mean pulmonary artery pressure(mPAP) was measured by cardiac catheterization. The weight ratio of right ventricle to left ventricle+septum [RV/(LV+SP) ] was also measured. The lung homogenates were assayed for total antioxidant capacity(T-AOC), superoxide dismutase (SOD), oxidized glutathione (GSSG), reduced glutathione (GSH), malondiadehyde(MDA) and hydroxy radical(?OH), and the SOD mRNA levels were assayed by real time polymerse chain reaction. 8.Mining prognostic marker of glioma based on weighted gene co-expression network analysis

Objective:To identify effective biomarkers for glioma patients.Methods:The mRNA expression profiles of 464 glioma patients with complete clinical follow-up information were downloaded from the Chinese Glioma Genome Atlas (CGGA). Weighted gene co-expression network analysis (WGCNA) was used to identify gene modules related to World Health Organization (WHO) grading of glioma, and univariate and multivatiate Cox regression analysis were performed to identify gliomas survival-related genes.Results:In weighted gene co-expression analysis, the module Brown was significantly positively correlated with glioma WHO stage ( r=0.55, P<0.05). In univariate analysis, five genes (TAGLN2, IGFBP2, METTL7B, ARAP3, PLAT) that were most significantly associated with clinical prognosis were selected for multivariate survival analysis, and the prognosis model was established to calculate the risk score. The receiver operating characteristic curve (ROC) confirmed that the risk score had high accuracy in predicting the 1-, 3-, 5-year survival rate of glioma patients. The above survival analysis results were verified in the Cancer Genome Atlas (TCGA) database. Conclusions:We use mRNA expression profiles to establish prognostic markers for gliomas to assess the overall survival of patients with glioma.
9.Effect of high quality nursing intervention for herpes simplex virus keratitis

Objective To explore the effect of high quality nursing intervention for herpes simplex keratitis.Methods A total of 92 patients with herpes simplex keratitis treated in our hospital were selected as study subjects,and were divided into control group and observation group according to the random number table method,with 46 cases in each group.Both groups were treated with routine therapy,and the observation group was additionally given high quality nursing intervention.The disappeared time of herpes simplex keratitis symptoms,adverse reactions and reoccurrence condition were compared between the two groups.Results After the intervention,the observation group was significantly higher than the control group in eyesight (P < 0.05);The disappearance time of rash,eye damage and pain in the observation group were significantly shorter than that in the control group (P < 0.05);The adverse reactions rates of the observation group was significantly lower than that of the control group (P < 0.05).Conclusion High quality care intervention can effectively improve the patient's vision,shorten disappearance time of symptoms,and reduce adverse reactions for herpes simplex keratitis patients.
