1.The effect and mechanism of epithelial growth factor receptor (EGFR)in inducing vasculogenic mimicry formation

Objective To investigate the effect and mechanism of EGFR in inducing vasculogenic mimicry (VM)formation.Methods Immunohistochemistry was used to detect the expression of EGFR protein and CD34-PAS double staining was used to detect the VM in gliomas paraffin blocks.The migration and VM formation of U87 cell in hypoxic and normoxic groups were detected by transwell and three-dimensional culture.Then we used Western blot to detect the expressions of EGFR,AKT and PI3K protein.Results The positive rate of EGFR expression in VM positive patients was 94.4%,which was significantly higher than that in VM negative patients (71.8%).In hypoxic group,the abilities of migration and VM formation were significantly higher those in normoxic group.The expressions of EGFR,AKT and PI3K were higher in hypoxic group.Conclusion VM formation can be induced by the activation of EGFR/AKT/PI3K signaling pathway.EGFR is very important for VM formation.
2.A Case Control Study of Behavioral Problems in Children with Attention Deficit Hyperactivity Disorder

Objective: To analyse the behavioral problems of children with attention deficit hyperactivity disorder.Methods: 70 boys fulfilled ADHD criteria of DSM-IV were further divided into 3 subtypes: predominantly inattentive(PI,44 cases),predominantly hyperactive-impulsive(HI,6 cases) and combined type(CT,20 cases).Compare the behavioral problems of subtypes of ADHD and 63 normal children.Results: The incidence of behavioral problems in boys with ADHD(48.57%) was significantly higher than that in control group(12.70%),and significant differences in incidence of behavioral problems were found among the three subtypes of ADHD.ADHD CT subtype had the highest rate of behavioral problems.PI subtype had more fissility than HI;CT and HI subtypes had more aggressive behaviors,and HI had more delinquence than PI and CT.Conclusion: Boys with ADHD had more behavioral problems than normal ones.CT subtype had the highest rate.The characteristics of behaviors were different among three subtypes.
3.Inhibitory effects of phosphorothioate-modified antisense VEGF oligodeoxynucleotides formulated in cationic liposome on angiogenesis and metastasis of lung cancer

Objective:The study aims to investigate whether phosphorothioate-modified antisense vascular endothelial growth factor oligodeoxynucleotides(VEGF ASODN)micro-encapsulated in cationic liposome could inhibit angiogenesis and metastasis of lung cancer.Methods:The phosphorothioate-modify VEGF ASODN, VEGF SODN(sense oligodeoxynucleotides) and VEGF MODN(mismatch oligonucleotides)micro-encapsulated in cationic liposome were added into in vitro culture of Lewis lung carcinoma(LLC ) cells,respectively. Expression of VEGF protein was detected by immunohistochemistry staining. And the inhibitory effects on bovine aortic endothelial cell proliferation by conditioned media obtained from LLC cells treated with ODN were observed. 40 mice of Lewis lung cancer models were established and subsequently were randomized into 4 groups: control group, ASODN group, SODN group,and MSODN group. Liposome, ASODN, SODN or MODN were given by twice a week for 4 weeks,respectively.The weights of subcutaneous tumors were measured. The rates of lung metastasis were detected, while the microvessel density(MVD) in tumor mass was detected by imuunohistochemistry staining. Peak systolic flow velocity(PS) and resistance index(RI) were measured with a sonographic scanner.Results:ASODN downregulated the expression of VEGF in LLC cells at the level of protein in vitro. The conditioned media obtained from LLC cells treated with ASODN significantly inhibited the proliferation of bovine aortic endothelial cells. 5.Study on Communication and Adaptive Function of Children with Asperger Syndrome

Objective:To investigate communication and adaptive function of children with Asperger Syndrome (AS).Methods:The adaptive function of two groups,123 with AS and 23 normal, were investigated with Adaptive Ability Scale for Children; Intelligence were measured with WISC; Communication behaviors of AS children were also investigated. Results:Significant differences were found between the two groups in total score of adaptive abilities and in domains of socialization, communication and self-administration; Abnormal communication behaviors were found in AS children。Conclusion: AS children demonstrated abnormal communication behaviors and deficits in social adaptive abilities.Early diagrosis and intervention can reduce children's adaptive trouble after entering school.
6.Research advances in differential diagnosis of benign and malignant ascites

Differential diagnosis of benign and malignant ascites is of a great value in determining etiology, evaluating prognosis, and determining therapy regimens. There are many diagnostic methods for ascites, but up to now, no method can achieve high sensitivity and specificity and meet the requirements of clinical economics and feasibility at the same time. Therefore, searching for an ideal method for differential diagnosis of ascites has become an important topic of current research. This article describes the clinical value of diagnostic methods, such as molecular biological detection, cytological examination, combined determination of tumor markers, and routine tests, in the differential diagnosis of benign and malignant ascites, analyzes the diagnostic efficiency of related biomarkers, and points out that combined determination of various parameters has an increased value in the differential diagnosis of benign and malignant ascites.
7.Research on multiplication of the H9N2 subtype avian influenza virus in large-scale microcarrier-based MDCK cell culture system

To explore the regularity for the multiplication of avian influenza virus subtype H9N2 in large-scale microcarrier-based MDCK cell culture system, and to determine the optimal proliferation conditions. H9N2 subtype of avian influenza virus was inoculated into the MDCK cell growing on 24 well plate, and the HA titers of virus at different time were detected in the conditions of different infectious doses,different concentrations of TPCK- trypsin and different pH. The optimal conditions were determined. Then the H9N2 subtype avian influenza virus was grown in microcarrier-based MDCK cell in 250mL and 5L roller bottles. It was demonstrated that high viruse yield with a hemagglutination unit of 9 log2(1:512) could be obtained under the optimal conditions of multiplication . The result indicated the H9N2 subtype avian influenza virus could be produced in microcarrier-based MDCK cell in a large-scale culture system with a high virus yield and demonstrates the feasibility of the development of mammalian cell-based in influenza vaccine in microcarrier culture systems.
8.Analysis of Abnormal Sensory Behaviors of Autism Children

Objective: To explore the characteristics of sensory behavior of autism children. Methods: 93 autism children between age 2 to 12, including 87 boys and 6 girls, were investigated based on Harrison's Sensory Behavior Scale. Results: Abnormal visual and auditory responses are quite common among autism children. Hyposensitivity to pain and overusing of smelling are less common. Significant differences were detected in auditory filtering, unusual visual response, hyposensitivity to pain and smell sensitivity between mild and severe groups. Conclusion: Autism children present varied sensory abnormalities. Severe autism children had significantly more sensory symptoms than the mild autism. The mechanisms for this deserves further exploring.
9.T type calciumchannel activated by brain injury induces neural stem cell proliferation

ABSTRACT:Objective To investigate the mechanism and induction of T type calcium channel on neural stem cells after brain injury .Methods Adult mice brain injury model was established and divided into control ,sham operation ,surgery ,and surgery+mebefradil groups .Neural stem cells were separated from the subventricular zone (SVZ) and identified .Moreover ,we analyzed the results using MTT and neural stem cell sphere counting after adding different doses of mibefradil in culture medium ,respectively .Then we calculated the half maximal inhibitory concentration (IC50) of mibefradil on neural stem cells .Finally ,we analyzed the expression of Cav3 .2 in SVZ and the protein expressions of Cav3 .2 ,Cyclin A and caspase‐3 in neural stem cells by Western blot .Results In vivo , neural stem cell proliferation was increased in surgery group compared with that in control and sham‐operation groups .The proliferation of neural stem cells in surgery + mibefradil groups was significantly decreased compared with that in surgery group after mibefradil‐induced inhibition of T type calcium channel protein . In vitro , the formation of neural stem cell sphere was significantly inhibited after adding mibefradil .The cell growth ratio was significantly decreased when the concentration of mibefradil was above 5μmol/L .A values in 5 μmol/L ,10 μmol/L and 20μmol/L groups were significantly lower than those in control group (P<0 .05) .IC50 was 8 .93μmol/L .The protein expressions of Cyclin A and Cav3 .2 were inhibited while that of caspase‐3 was increased after mibefradil treatment .Conclusion Neural stem cell proliferation was enhanced by activating T type calcium channel after brain injury .
10.Current status of nonalcoholic fatty liver disease and related risk factors: an analysis of an enterprise in Xi′an

ObjectiveTo investigate the current status of nonalcoholic fatty liver disease (NAFLD) in an enterprise in Xi′an, China, and to provide a scientific basis for further studies on NAFLD in Shanxi. MethodsA total of 415 employees who underwent health examination in The First Affiliated Hospital of Xi′an Medical University from June to July, 2014 were enrolled. All the patients underwent physical examination, abdominal ultrasound, and serological examination, and completed related questionnaires. The t-test was applied for comparison of continuous data, and the Pearson correlation analysis was applied for correlation analysis. ResultsOf all the 415 patients, 86 (20.7%) had NAFLD, consisting of 70 patients (16.9%) with mild NAFLD and 16 patients (3.8%) with moderate NAFLD. The population with an age of 50-58 years had the highest prevalence rate (30.8%, 16/52). Of all the 86 patients with NAFLD, 57 (66.3%) had obesity, and 76 (88.4%) had abdominal obesity. The employees with moderate NAFLD had a significantly higher alanine aminotransferase (ALT) level, a significantly higher aspartate aminotransferase (AST) level, a significantly larger abdominal circumference, and a significantly higher body mass index (BMI) than those with mild NAFLD (t=-4.140, -3.512, -4.405, and -5.051, respectively, all P<0.05). In the employees with NAFLD, BMI and abdominal circumference were correlated with AST level (r=0.283 and 0.271, both P<0.05) and ALT level (r=0.325 and 0.316, both P<0.05). Most employees in this enterprise had a bad eating habit, a sedentary lifestyle, and a lack of physical exercise. ConclusionThis enterprise has a high prevalence rate of NAFLD. BMI and abdominal circumference of patients with NAFLD have certain clinical values in predicting the degree of fat deposition in the liver, and are correlated with AST and ALT levels.
