1.Correlation between growth, proliferation and apoptosis of leukemia cell lines K562/AO2 and bone marrow mesenchymal stem cells in children with leukemia

BACKGROUND:Leukemia comes from the damage to bone marrow microenvironment, and changes in bone marrow microenvironment lead to some effects on biological characteristics and behavior of leukemia cel s.
OBJECTIVE:To explore the effect of bone marrow mesenchymal stem cel s from children with leukemia on K562/AO2 cel line growth, proliferation and apoptosis.
METHODS:K562/AO2 cel s were cultured alone in vitro, or co-cultured with bone marrow mesenchymal stem cel s from leukemia children. The living cel s in the two groups were counted using trypan blue staining, and the cel growth curves were described. PI single staining method was used to detect the cel cycle, and cel apoptosis was detected by V/PI Annexin method in the two groups.
RESULTS AND CONCLUSION:After single suspension culture, the cel s showed no marked changes within the first 3 days, but began to proliferate fast at 4 days and reached the proliferation peak at 6 days. Co-cultured cells showed a gentle growth curve with no presence of obvious proliferation peak. Compared with the co-culture group, the proportion of G0/G1 phase cel s was significantly lower, but the proportion of S phase cells was significantly higher in the single culture group (P<0.05). The number of cells in the G2/M phase showed no significant difference between the two groups (P>0.05). All these findings indicate that co-culture with bone marrow mesenchymal stem cel s from leukemia children inhibits the proliferation of K562/AO2 cells through G0/G1 phase arrest, and also resists apoptosis in K562/AO2 cells.
2.Clinical Observation of Pidotimod Combined with Montelukast Sodium in the Treatment of Children with Bronchial Asthma

OBJECTIVE:To observe clinical efficacy and safety of pidotimod combined with montelukast sodium in the treat-ment of children with bronchial asthma. METHODS:120 children with bronchial asthma were randomly divided into control group and observation group,with 60 cases in each group. Both group were given routine treatment as aerosol inhalation of glucocorticoid and anti-infective treatment. Control group was additionally given Montelukast sodium chewable tablet 5 mg orally,qd,at bed-time;observation group was additionally given Pidotimod capsule 0.4 g,qd,on the basis of control group. Both group received treatment for consecutive 3 months. Clinical efficacies of 2 groups were observed as well as IL-4,IFN-γ,IgE,T lymphocyte sub-set and lung function indexes of 2 groups before and after treatment. The occurrence of ADR were compared between 2 groups. RE-SULTS:The effective rate of observation group was 88.3%,which was significantly higher than that of control group (61.7%). There was no statistical significance in IL-4,IFN-γ,IgE,T lymphocyte subset and lung function indexes between 2 groups before treatment(P>0.05). After treatment,IL-4,IFN-γ,IgE,CD4+,CD4+/CD8+ and lung function of 2 groups were improved signifi-cantly,and the observation group was significantly better than the control group,with statistical significance(P<0.05). There was no statistical significance in the incidence of ADR between 2 groups(P>0.05). CONCLUSIONS:Pidotimod combined with monte-lukast sodium is effective for children with bronchial asthma,and can significantly relieve airway inflammation,enhance immunity and improve lung function with good safety.
3.Influence of using health belief model among chronic prosatitis patients on their ability of serf-control

Objective To know the influence of using health belief model among chronic prosatitis patients on their ability of self-controL Methods Divided 96 patients with chronic prosatitis into the observation group and the control group randomly,there were 48 cases in each group. Routine health education was used in the control group,while the health education according to health belief model was used in the observation group. Compared the ability of self-control between the two groups. Results Before the health education,there was no significant difference about the ability of self-control between the two groups,while after 3-month follow-up,the difference was significantly. Conclusions Health education based on health belief model can effective promote the ability of self-control in patients with chronic prosatitis.
5.Effects of short-term enriched environment on the hippocampal formation and the myelinated fibers in the hippocampal formation of mid-aged female rats

ObjectiveTo investigate the effects of short-term enriched environment on the hippocampal formation and the myelinated fibers in the hippocampal formation of mid-aged female rats. Methods Twenty 14-month female SD rats were randomly divided into 10 enriched environment (EE) rats and 10 standard environment (SE) rats. EE rats were reared in enriched environment and SE rats were reared in standard environment for 4 months. Then, five rats were randomly selected from each group. The spatial learning capacity was assessed with Morris water maze. The hippocampal formation and the myelinated fibers in the rat hippocampal formation were quantitatively investigated with transmission electronic microscopy technique and stereological methods. Results Short-term enriched environment enhanced the spatial learning capacity of the mid-aged female rats. The total length and total volume of the myelinated fibers in the hippocampal formation of the EE rats was significantly increased by 43.3% and 47.4%, respectively, when compared to the SE rats. There was no significant difference in the hippocampal volume and the mean diameter of the myelinated fibers in the hippocampal formation between two groups. The increase of the total length of the myelinated nerve fibers in the hippocampal formation was mainly due to the increase of the myelinated fibers with small diameter. Conclusion Short-term enriched environment had significant effects on the spatial learning capacity and the myelinated fibers in the hippocampal formation of middle-aged female rats.
6.Short-term enriched environment increases the total length of the myelinated nerve fibers in the hippocampal formation of mid-aged male rats

After the Morris water maze test, the total volume of the hippocampal formation and the myelinated nerve fibers in the hippocampal formation were quantitatively estimated with transmission electronic microscopy technique and stereological methods. Results There was not significant difference in the spatial learning capacity between enriched group and standard group. The total volume of the hippocampal formation of enriched rats was not significantly increased by 4.6% when compared with that of standard rats. The total volume, total length and mean diameter of the myelinated nerve fibers in the hippocampal formation of enriched rats were significantly increased when compared with those of standard rats. Conclusions Four-months enriched environment significantly affected the myelinated fibers in the hippocampal formation of mid-aged male SD rats. The present results might provide an important theoretical basis for searching the ethology strategy to delay the progress of brain aging in the future.
7.Effects of running exercise on the hippocapal formation and the myelinated nerve fibers in the hippocampal formation of mid-aged rats

ObjectiveTo investigate the effects of exercise on the hippocampal formation and the myelinated nerve fibers in the hippocampal formation of middle-aged rats. MethodsTen 14-month female SD rats were randomly divided into exercise group and sedentary group.Rats in the exercise group were forced to run on a treadmill for 4 months. After 4 months, spatial learning capacity of two group rats was tested using the Morris water maze.Then, the hippocampal formation and the myelinated nerve fibers in the hippocampal formation were quantitatively estimated using transmission electronic microscopy and stereological techniques. Results Treadmill running enhanced the spatial learning capacity of the rats. The volume of hippocampal formation and the total length of the myelinated nerve fibers in the hippocampal formation were significantly increased after 4 months exercise.However,there was no significant difference in the total volume of the myelinated fibers in the hippocampal formation between the two groups.The absolute distributions of the total length of the myelinated fibers in the hippocampal formation of two groups indicated that the exercise-induced increase of the total length of the myelinated nerve fibers in the hippocampal formation was mainly due to the increase of the myelinated fibers with small diameter. Conclusions Four months running exercise remarkably influence the spatial learning capacity,hippocampal formation and the myelinated fibers in the hippocampal formation of the middle-aged famale SD rats. The present results reveal a potential mechanism for the fact that exercise might improve brain function.
8.Associated factors of early abortion among singleton pregnancies after assisted reproductive technology

Objective To investigate the relative risk factors for early abortion among singleton pregnancies after assisted reproductive technology (ART) treatment. Methods A retrospective analysis was performed on 1636 singleton pregnancies, including 196 early abortion cases and 1195pregnancies with live birth after exclusion of those lost cases during follow-up, or complicated with uterine deformity, or oocyte receptor, or late abortion, or incomplete medical record, following in vitro fertilization(IVF)/ intracytoplasmic sperm injection (ICSI) treatment and the risk factors of early abortion were investigated. The early abortion rate was also compared between fresh IVF/ICSI group and frozen embryo transfer (FET) group (n=386). Results Multivariate Logistic regression analysis indicated that elder women (OR= 1. 143,95%CI: 1. 096-1. 196) and patients with polycystic ovarian syndrome (OR = 4. 309,95 % CI : 2. 564-7.243) were risk factors of spontaneous early abortion,and high mean score of transferred embryos (MSTE) (OR = 0. 808, 95% CI: 0. 717-0. 912) and endometrial triple-lined pattern on the day of human chorionic gonadotropin (hCG) administration (OR=0. 431, 95% CI: 0. 243-0.764)were protective factors. Significant difference were found in the maternal age [(32.22±4. 10) yrs vs (30.28±3. 66) yrs],the duration of infertility [(5. 90±4.26) yrs vs (5.20 ± 3. 32) yrs], basal serum follicle-stimulating hormone (FSH) level [(6. 35 ±2.30) mIU/ml vs (5.95±2.12) mIU/ml], number of transferred embryos (2. 31±0. 51) vs (2. 18±0.49), serum estradiol level on the day of hCG administration [(2467. 1 ± 1588. 8) pg/ml vs (2934. 5 ±1785.2) pg/ml] and MSTE (7.03 ±1.35 vs 7.74 ± 1.25) between the abortion group and livebirth group (all P<0. 05). The spontaneous abortion rate was higher in the FET group than in the fresh embryo transfer group [17. 36%(67/386) vs 13.02% (213/1636), χ2 =4. 296, P=0. 023].Conclusions Women at elder age, or with long duration of infertility, high basal FSH level,polycystic ovarian syndrome, low MSTE, non-triple-lined pattern of endometrium on the day of hCG administration are at risk of spontaneous early abortion in pregnancies after ART. The rate of spontaneous abortion is higher in FET group than in fresh IVF/ICSI group.
9.Skin adnexal tumor with multipote ntial differentiation complicated by nevus spilus: a case report

A 41-year-old female patient developed round,bright yellow patches on the left calvarial region without obvious precipitating factors 40 years prior to the presentation,which gradually grew to form plaques with age.Two years prior to the presentation,nipple-like lesions appeared in the calvarial and temporal region with an erythematous and wet surface; concurrently,black masses developed in the left temporal region and gradually enlarged with central ulceration but no subjective symptoms.At about 1 year of age,pitchy macules developed on the light tan patches located on the left jaw,posterior and anterior neck,trunk and upper limbs,and gradually increased in quantity and size with the involvement of the homolateral dorsal hand and gradual appearance of papules.Skin examination revealed two well-marginated,indurated,bright red neoplasms sized 3 cm × 2 cm and 2 cm × 1 cm respectively,with erosive and cauliflower-like surface; black or pink papules were scattered between these neoplasms.There was a ring-shaped black mass sized 1.5 cm × 1.5 cm in the left temporal region with central ulceration.Pitchy tough macules and papules were observed on the light tan patches located in the left cheek,lower mandible,posterior and anterior neck,protothorax,shoulder and back,upper limbs and dorsal hand.Based on the histopathology of multiple lesions,the cauliflower-like lesions on the head were diagnosed as syringocystadenoma papilliferum,the yellow plaques as syringocystadenoma papilliferum complicated by sebaceous adenoma,the black proliferative lesions in the temporal region as trichoblastoma accompanied by basal cell epithelioma,the black papuloid lesions and brown maculopapuloid lesions on the lower mandible as nevus spilus.The patient was diagnosed with skin adnexal tumor with multipotential differentiation (syringocystadenoma papilliferum,sebaceous adenoma,trichoblastoma and basal cell epithelioma)accompanied by nevus spilus.
10.Sex differences of short-term enriched environment on myelinated nerve fibers in the hippocampal formation of middle-aged rats

Objective To investigate the sex differences of the myelinated nerve fibers in the hippocampal formation of mid-aged standard and enriched rats. Methods Twenty 14-month Sprague-Dawley (SD) rats, 10 female and 10 male rats, were randomly divided into enriched group and standard group. Enriched rats were reared in enriched environment and standard rats were reared in standard environment for 4 months. Then, the total volume of the hippocampal formation and the myelinated nerve fibers in the hippocampus were quantitatively estimated by transmission electron microscopy and stereological methods. Results The total volume and mean diameter of the myelinated nerve fibers in the hippocampal formation of standard female rats were significantly bigger than those of standard male rats (P0.05). The mean diameter of the myelinated nerve fibers in the hippocampal formation of enriched female rats was significantly smaller than that of enriched male rats (P0.05). Conclusion The sex differences of the myelinated nerve fibers in the hippocampal formation of mid-aged standard and enriched rats are significantly different, which indicates the response to the enriched environment in the mid-aged male and female rats is different.
