Objective To compare the growth property of the stem cells taken from different brain regions at the same developmental stage. Methods Mice embryos at the same development stage were isolated under sterile conditions, cortex, striatum, diencephalon, mid-hind brain and spinal cord were collected and pooled separately, after single-cell suspension obtained, different regions' cell suspensions were seeded in FGF supplemented serum-free culture medium. Followed the neural stem cell clone(neurospheres) fromation, immuno-cytochemistry method was utilized to identify the cell characteristics, all these clones were passaged under same conditions, clone formation and cell migration were observed under phase-contrast microscope. Results In the FGF added serum-free medium, neural cells experienced a large scale death within 48h after being seeded, then few single cells began to proliferate and formed the floating cell clones in the medium. These clones (neurospheres) could form new clones when seeded as single cell suspension. If these clones were seeded on poly-orithine, they could differentiate into neurons and glia cells. Compare the clone formation and cell migration, we found that: cortex, striatum, diencephalon all could form floating clones with different rate, the cortex formed clones at the highest rate, striatum and diencephalon at lower rate; few neurospheres formed from cortex adhered to the culture plate substrate and few cells were found migrating out from the adhered clones, striatum and diencephalon derived neurospheres adhered the plate more easily, and there were apparent cell migration. Mid-hind brain and spinal cord formed clones at the lowest rate, floating clones were scarce, and the clones adhered to the substrate readily, there were large amount of cell migrating out from these adhered clones. Conclusion Neural stem cells could proliferate and be passaged in vitro in serum-free medium, and they could be induced to differentiate under certain conditions into major cells types of CNS, there were differences in clone forming rate and cell migration between neural stem cells derived from different CNC regions, nonetheless they were at the same development stage, this may reflect that, in some degree, these cells can keep some of their region-specified developmental intrinsic property in vitro.

Objective:To annalyze the constitution rule and pathogenesis of Diabetic Peripheral Neuropathy (DPN) by data mining technology, so as to provide a theoretical reference for improving the therapeutic effect of TCM on treating DPN.Methods:343 cases of DPN patients dianosed with four TCM diagostic methods chosen from the Department of Endocrinology of Affiliated Hospital of Traditional Chinese Medicine of Guangzhou Medical University from January 2016 to January 2020 were collected. Statistical analysis software was used to conduct frequency analysis, correlation analysis and cluster analysis, and explained with TCM theory. Results:Among the 343 cases, 143 cases were male and 200 cases were female; Symptoms such as limb numbness, thirst, dry mouth, frequent urination at night, limb pain and so on occur more frequently; The main tongue images include thin white tongue, dark and red tongue or light red tongue; the main pulse conditions include string, fine and deep pulse. Numbness, thirst, dry mouth, thin and white fur, frequent nocturia and so on were closely related with other diseases. Four types of syndrome can be recombined by cluster analysis, which are blood stasis obstructing collaterals syndrome; kidney deficiency and blood stasis syndrome; stomach heat injuring Yin syndrome; spleen deficiency and phlegm dampness syndrome; heat toxin accumulation and syndrome of lingering pathogen due to deficient vital qi.Conclusion:The root of the disease is Yin deficiency with dry heat as the sympotom. Blood stasis runs through the whole process of the disease. The function of spleen and stomach should not be ignored in treatment.

BACKGROUND:Recent studies have shown that research on naringin anti-osteoporosis mostly stays in in vitro and in vivo experiments.Understanding the mechanism of related signaling pathways and the expression of related proteins and some specific genes is an important way to deeply understand naringin anti-osteoporosis.At present, traditional Chinese medicine has been confirmed to have a significant role in anti-osteoporosis.Naringin is one of the main active ingredients in Rhizoma Drynariae. Its effectiveness and mechanism of action against osteoporosis have been gradually recognized by scholars, and its clinical and basic research has been gradually emphasized. OBJECTIVE:To analyze and summarize the research progress of naringin in anti-osteoporosis in vitro and in vivo, thereby providing some ideas for the next step to study its related mechanism of action. METHODS:The relevant literatures included in CNKI and PubMed database were searched with the Chinese search terms of"naringin, osteoporosis, traditional Chinese medicine compound, pathogenesis, signaling pathway, bone marrow mesenchymal stem cells, osteoblasts, osteoclasts"in Chinese and English, respectively. The corresponding criteria were established according to the research needs, and finally 69 articles were included for review. RESULTS AND CONCLUSION:Naringin blocks the increase in the number of osteoclasts and adipocytes, the decrease in the number of osteocytes and osteocalcin(+)cells induced by fructose-rich diet, and promotes the secretion of Sema3A from osteoblasts and osteocytes, thereby enhancing local bone formation and inhibiting osteoclast production by activating the Wnt/β-catenin pathway.Naringin is an important way to induce autophagy of osteoblasts, but autophagy-related proteins participate in osteoblast differentiation and bone formation.Lack of autophagy in osteoblasts reduces mineralization and leads to an imbalance in the number of osteoblasts and osteoclasts, which results in bone loss and decreased bone density. The composite scaffold loaded with naringin can be used as a necessary carrier for bone defect repair and has excellent bone repair properties.Naringin can also accelerate the growth of new bone tissue by increasing the local contents of bone morphogenetic protein 2 and vascular endothelial growth factor.Naringin can regulate bone metabolism and inhibit oxidative stress via ERK,PI3K/Akt and Wnt signaling pathways to improve osteoporosis, which can play a good role in preventing and controlling the disease. However, the depth and breadth of the relevant research is insufficient.Based on the mechanism of the current study,we should investigate the specific mechanisms by which naringin regulates different pathways and inter-pathway interactions in the future, which will be beneficial to the multifaceted development of naringin used in the treatment of osteoporosis..
