1.A method of medical image registration based on frequency domain and time domain

To resolve the problems of relativity translation variant and relativity rotation variant existed in the two medical images, combining the removing characteristics and rotating characteristics in image signal Fourier transform, a method of medical image registration combining with frequency domain and time domain is put forward based on the well-known Fourier property. The method decouples the estimation of rotation and translation. It determines rotation parameters based on amplitude spectrum correlation in frequency domain and estimation translation parameters based on intensity value correlation in time domain. The experimental results of nuclear medical image registration and multi-modality medical image registration show that this method is a very robust and fast registration method with high accuracy.
2.Protective effect of schisandrin B on cerebral ischemia reperfusion injury of rats and its mechanisms

Objective:To investigate the protective effect of schisandrin B (SchB)on the cerebral ischemia reperfusion injury of the rats and the influence in HSPA12B/PI3K/Akt signaling pathway,and to explore the mechanisms.Methods:130 SD rats were divided into sham group,cerebral ischemia reperfusion injury model group (model group),low dose of SchB group (SchB 3 mg· kg-1 ,SchB1 group),middle dose of SchB group (SchB 10 mg·kg-1 ,SchB2 group)and high dose of SchB group (SchB 30 mg·kg-1 ,SchB3 group)(n=26).The rats in sham group didn’t plug lines;the rats in model were used to establish ischemia reperfusion models;the rats in SchB1 ,SchB2 and SchB3 groups were firstly pretreated with different doses of SchB for 7 d,and then they were used to build cerebral ischemia reperfusion injury models.The nerve dysfunction of rats was evaluated by neurologic deficit score.The cerebral edema was detected by measuring the content of water in brain tissue.The morphological changes of brain tissue were observed by toluidine blue staining.The levels of nuclear factor kappa B (NF-κB), tumor necrosis factor-α(TNF-α),interleukin-1 (IL-1)and interleukin-6 (IL-6)in the brain tissue were detected by ELISA.Western blotting method was used to detect the protein expression levels of heat shock protein A12B (HSPA12B ), serine-threonine kinase (Akt ) and phosphorylation serine-threonine kinase (p-AKT ). Results:Compared with sham group,the neurologic deficit score of rats in model group was significantly increased (P <0.01),and the content of water in brain tissue was increased (P < 0.01 );the brain tissue structure was loosened,and the mesenchyme appeared edema;the NF-κB,TNF-α,IL-1,and IL-6 levels were increased (P <0.01),and the expression levels of HSPA12B and p-Akt proteins were decreased (P <0.01).Compared with model group,the neurologic deficit scores of the rats in SchB1 ,SchB2 ,and SchB3 groups were decreased (P <0.01),and the contents of water in brain tissue of the rats in SchB2 and SchB3 groups were decreased (P <0.05);the edema of nerve cells was alleviated,and the cavities were reduced;the NF-κB,TNF-α,IL-1,and IL-6 levels were decreased (P <0.05 or P <0.01),the expression levels of the HSPA12B protein in SchB2 ,and SchB3 groups were increased (P <0.05),and the p-Akt protein expression levels of the rats in SchB1 ,SchB2 ,and SchB3 groups were increased (P <0.01).Conclusion:SchB could protect the cerebral ischemia reperfusion injury of rats,its mechanism may be related to regulating the HSPA12B/PI3K/Akt signaling pathway and inhibiting the inflammatory reaction damage to the nerve cells of reperfusion.
3.Explore the effectiveness of melformin sustained release tablets in treatment of patients with abnormal glucose tolerance

Objective To evaluate the effectiveness of melformin sustained release tablets in treatment of patients with abnormal glucose tolerance. Methods 78 cases with abnormal glucose tolerances were selected and divided into two groups. Patients in treatment group took melformin sustained release tablets, while patients in control group took normal therapy. The impaired glucose tolerance (IGT) and body mass index (BMI) were observed and compared. Results The blood glucose level of OGTT 2 h in treatment group was (7.61±1.04)mmol/L, while it was only (9.96±1.08) mmol/L in control group. The BMI of treatment group was 23.71±3.41, while it was 29.53±3.72 in control group. The glycosylated hemoglobin was (5.79±0.41)%, while it was (6.23±0.52)%in control group. The fasting blood glucose in treatment group was (5.34±0.39)mmol/L and in control group was (8.16±0.48)mmol/L. The differences of those indicators between two groups were signiifcant. Conclusion Melformin gains excellent clinical effects in treating patients with abnormal glucose tolerance. It can adjust blood glucose, improve glucose tolerance, and decrease blood glucose level of OGTT 2 h and BMI effectively.
4.Pilot Study on the Relationship of LPPCN with Neovascularization and Prognosis of Melanoma

Objective: To study the relationship of LPPCN with neovascularization and to analyze its clini-copathologic significance, in an effort to find a new target for anti-vascular therapies. Methods: Sixty-eight ma-lignant melanoma specimens were analyzed to observe the distribution of LPPCN and to examine the expres-sion of CD105 and TGFβ1 using immunohistochemistry. The distribution of vasculogenic mimicry (VM) was observed by immunohistochemical and histochemical double staining of CD31 and PAS. Results: (1) The tu-lines and networks. Of the 68 cases of melanoma, 55.89% (38/68) were recognized as having LPPCN. (2) In malignant melanoma specimens, the rate of vasculogenic mimicry density (VMD) and microvessel density (MVD) labeled by CD105 in LPPCN-positive group were higher than those in LPPCN-negative group, with sig-nificant differences (P<0.05). VMD and MVD were positively-correlated with the density of LPPCN. The posi-tive expression of TGFβ1 in LPPCN-positive group was higher than that in LPPCN-negative group and its ex-pression in the regions of LPPCN was obviously higher than that in circumambient tumor cells, with a signifi-cant difference (P<0.05). The expression of TGFβ1 was positively correlated with MVD labeled by CD105. (3) There was no relationship between LPPCN and gender, age, site, tumor embolus, lymph node metastasis or distant metastasis (P>0.05), but LPPCN was correlated with tumor size, mitosis figure count and Breslow depth (P<0.05). Kaplan-Meier survival analysis showed the survival rate of patients with LPPCN was lower than that of patients without LPPCN, with a statistical significance (P<0.05). The presence of LPPCN indicat-ed poor prognosis. Conclusion: LPPCN exists in malignant melanoma and is associated with VM and angio- genesis. Some tumor cells undergoing LPPCN have a spacial foundation for VM and angiogenesis. LPPCN can be an index for the evaluation of the prognosis of melanoma patients.
5.Prediction for hemorrhagic transformation risk after intravenous thrombolysis in acute ischemic stroke patients in different therapeutic windows: comparison of 5 scoring systems

Objective To compare the predictive value of 5 scoring systems for hemorrhagic transformation risk after intravenous thrombolysis in patients with acute ischemic stroke (AIS) in different therapeutic windows.Methods A single-center and retrospective study was performed for 243 AIS patients who underwent intravenous thrombolysis using recombinant tissue plasminogen activator (rt-PA) in different therapeutic windows in our department during January 2014 and December 2016.Five scoring systems,including HAT model (hemorrhage after thrombolysis),MSS model (multicenter stoker survey),GRASPS model (glucose at presentation,race,age,sex,systolic blood pressure at presentation,severity of stroke at presentation),SEDAN model (baseline blood sugar,early infarct signs,hyperdense cerebral artery sign on admission CT,age,NIHSS on admission),and SITS model (safe implementation of thrombolysis in strokemonitoring study) were used to evaluate the risks for hemorrhagic transformation.The relationships between the 5 scoring systems and incidence rate of hemorrhagic transformation were analyzed among the patients in different therapeutic windows.The predictive values of the 5 scoring systems were compared using the areas (AUC) under the receiver operating characteristic (ROC) curve.Results When the AIS patients were treated by intravenous thrombolysis within 3 h,the AUC of GRASPS and HAT models were 0.698 and 0.619,respectively,higher than those of the other 3 systems.When the therapeutic window was between 3 to 4.5 h,HAT model and SEDAN model had highest AUC (0.719,0.744) than the other 3 systems (P <0.05).When the windows were >4.5 ~6 h,the HAT model had the highest AUC (0.676).Conclusion The 5 scoring systems show better predictive value for hemorrhagic transformation after intravenous thrombolysis.For the therapeutic window within 4.5 h,HAT model presents best predictive value than the other 4 scoring systems.
6.Forkhead or winged helix transcription 3 and interleukin-17 expression in colon cancer and clinical significance

Objective To observe the expression of forkhead or winged helix transcription 3 (Foxp3) in colon cancer tissue and paracancerous tissue,and detect the level of serum interleukin (IL)-17 in colon cancer patients and healthy human,to explore the changes of regulatory T cell (Treg cell) and Th17 cell in the process of occurrence and development of colon cancer.Methods The expression of Foxp3 in 56 patients with colon cancer and paracancerous tissue and 15 cases with normal colon tissue was measured by immunohistochemical SP method and the level of serum IL-17 was determined by enzyme-linked immunosorbent method.Results The level of serum IL-17 in colon cancer tissue was higher than that in normal colon tissue [(9.1 ± 2.3) ng/L vs.(6.2 ± 1.5) ng/L],and there was significant difference (P =0.007).Foxp3 positive cell number in colon cancer tissue was more than that in normal colon tissue and paracancerous tissue (24.1 ± 6.4 vs.2.7 ± 1.1 and 8.7 ± 2.3),paracancerous tissue was more than normal colon tissue,and there was significant difference (P < 0.01).The level of serum IL-17 in colon cancer tissue with TNM Ⅲ was higher than that in TNM Ⅰ-Ⅱ [(8.5 ± 2.1) ng/L vs.(5.4 ± 0.9) ng/L],Foxp3 was more than that in TNM Ⅰ-Ⅱ (25.8 ± 6.2 vs.18.2 ± 4.4),and there was significant difference (P< 0.01).There was no significant difference in the level of serum IL-17 between middle-high differentiated adenocarcinoma and low differentiated adenocarcinoma [(9.4 ± 1.1) ng/L vs.(8.9 ± 1.8) ng/L] (P > 0.05).Foxp3 in middlehigh differentiated adenocarcinoma was more than that in low differentiated adenocarcinoma (26.8 ± 5.5 vs.17.2 ± 3.2),and there was significant difference (P < 0.01).Conclusions Detection of IL-17 and Foxp3 can provide a new way of targeting therapy for colon cancer.IL-17 levels and Foxp3 expression are closely related to the immune status of local tumor tissues,the joint detection is benefitial to the further understanding of the patients with tumor immune state,provide basic information for tumor immunotherapy.
7. Differential diagnosis of mass-like polyps and tumors in nasal cavity and paranasal sinus with quantitative parameters of dual-energy CT iodine map

Objective: To explore the value of quantitative parameters of dual-energy CT arterial iodine map for differentiating nasal and paranasal sinus mass-like polyps from tumors. Methods: Dual-energy CT data of 80 patients with nasal sinus masses confirmed by surgical pathology were retrospectively analyzed. According to pathological results, the patients were divided into polyp group (n=26), benign tumor group (n=24) and malignant tumor group (n=30). Arterial phase iodine concentration (IC), normalized iodine concentration (NIC) and iodine map CT value (Overlay value) of lesions were measured, calculated and compared among 3 groups. Then patients in benign tumor group and malignant tumor group were combined into tumor group (n=54). ROC curves of each quantitative parameters of arterial phase iodine map for differentiating nasal and paranasal mass polyps and tumors were drawn, and the relative diagnostic efficacy were evaluated. Results: The arterial phase IC, NIC and Overlay value of benign tumor group (H=-4.13, -2.80, -4.00) and malignant tumor group (H=-7.02, -5.75, -6.12) were all higher than those of polyp group (all P<0.05). The arterial phase IC and NIC in malignant tumor group were higher than those in benign tumor group (H=-2.60, -2.73, both P<0.05), while Overlay value showed no statistical difference between benign and malignant tumor groups (H=-1.85, P=0.20). AUC of arterial phase IC (0.95) for differentiating nasal and paranasal mass polyps and tumors was larger than that of NIC (0.85) and Overlay value (0.91). Taken 1.15 mg/ml as the diagnostic threshold, the diagnostic sensitivity, specificity and Youden index was 81.50%, 96.20% and 78.00%, respectively. Conclusion: Arterial phase iodine-related quantitative parameters of dual-energy CT have certain application value in differential diagnosis of nasal and paranasal sinus mass-like polyps and tumors.
8.Evaluation of photodynamic therapy of skin cancers with partial differential alpha-aminolevulinic acid.

OBJECTIVETo use partial differential -aminolevulinic acid induced photodynamic therapy (ALA-PDT) increasingly in treating skin cancers and other diseases in many countries and to explore the efficacy of ALA-PDT for skin cancers in China.
METHODSEighty-eight patients, including 34 cases of basal cell carcinoma (BCC), 32 cases of squamous cell carcinoma (SCC), two cases of basal-squamous cell carcinoma (BSCC), one case of verrucuous carcinoma, nine cases of Bowen disease, two cases Paget disease of the nipple and eight cases of extramammary Paget disease, were treated by the partial differential alpha-aminolevulinic acid induced photodynamic therapy first in China from 1997 to 2000.
RESULTSAll BCC, including 11 cases of superficial lesions and 29 solid lesions, achieved complete reaction (CR) by 1-4 times of the ALA-PDT. Except one patient with adenoid SCC (grade III), all SCC (grade I and grade II) patients achieved complete remission by 3-6 times of ALA-PDT. All Bowen diseases achieved complete reaction by 1-4 times. Although for Paget diseases it could not cure the disease simply by ALA-PDT, it could control the symptoms. The recurrence rates were 11% (4/34) for BCC, and 22% (7/32) for SCC by following up 1-3 years after the therapy. The continuous therapy is still effective.
CONCLUSIONSALA-PDT is an effective, non-traumatic treatment for patients with BCC, SCC, Bowen and Paget diseases. It is especially suitable for older and weaker patients or those who are not tolerable to other therapies. It also has a unique advantage for tumors in specific anatomical areas. It is a new alternative modality for skin cancer therapies.

9.Identification of differentially expressed genes in different anomalies of Glycyrrhiza uralensis by cDNA-AFLP method.

OBJECTIVETo understand the molecular mechanism in genetic variation within species, and identify the differentially expressed genes among 8 anomalies of Glycyrrhiza uralensis under the same genetic background and cultivated environment.
METHODThe differentially expressed genes in roots of different anomalies of G. uralensis in vigorous growing stage were identified by cDNA-AFLP and confirmed by reverse-Northern hybridization. Their functions were inferred through bioinformatics method.
RESULTSFourteen differentially expressed genes were identified, among which, the function of 6 were unknown, and the rest were involved in antibacterium, regulating metabolism, enhancing the heat resistance ability, increasing the nitrogen fixation ability, and other bioprocesses.
CONCLUSIONFor the first time, the differentially expressed genes were cloned from the different anomalies of G. uralensis. This provided the basis for the screening of fine anomalies and varieties, and the research of functional genomics.

10.Comparative study of HPV infective genotypes distribution in tissues of cervical cancers and cervical intraepithelial neoplasias

Objective To compare the genotypes distribution of human papillomavirus (HPV ) infection in tissues of cervical cancers and cervical intraepithelial neoplasias (CIN ) and its clinical significance .Methods The polymerase chain reaction (PCR) and the gene-chips technique were utilized for the detection of 23 kinds of HPV genotypes in the tissue specimens from 192 cases of cervical intraepithelial neoplasia (CIN) and 85 cases of cervical cancers .And the related data of all subjects were analyzed .Results In 192 cases of CIN ,the total positive rate of HPV was 82 .29% (158/192) ,the positive rate of single genotype infection was 46 .88% (90/192) and the positive rate of multiple genotypes infection was 35 .42% (68/192);In 85 cases of cervical cancers ,the to-tal infection rate of HPV was 88 .24% (75/85) ,the positive rate of single genotype infection was 65 .88% (56/85) and the positive rate of multiple genotypes infection was 22 .35% (19/85) .Conclusion PCR combined with the gene-chips technique can be used in the detection of the tissue samples of cervical lesions ,once detection can detect 23 kinds of HPV genotypes with high sensitivity and strong specificity ,which has very important significance to the prevention and treatment of cervical cancer and precancerous lesions and the their vaccine research .
