Weanling female rats were fed 12% lard diet supplemented with 2 g cholesterol and 60 mg cholic acid per 100 g diet for 52 days. Blood cholesterol levels were then analyzed for individual rat. Animals were divided into 4 groups according to their blood cholesterol levels and body weights. One group of animals, regarded as negative control, was killed immediately. The other 3 groups were fed 12% level of synthetic oil, lard or soybean oil diet respectively without cholesterol and cholic acid supplementation for 3 weeks. During the 3 weeks of rehabilitation, the following parameters were measured: body weight and serum cholesterol levels, every week; food intakes, every other day; high density lipoprotein cholesterol (HDL-C) levels at the beginning and the end of the rehabilitation period; fecal steroids excretion, including neutral and acid steroids from the 2nd to the 4th day of rehabilitation; liver weights, liver fat contents and liver cholesterol levels after animals were killed. The results showed that no significant effects on body weights, serum cholesterol levels, liver weights and liver fat contents of different kinds of dietary fat were observed during the rehabilitation period of hyperch-olesterolemia. But synthetic oil, as well as soybean oil diet caused a lower liver cholesterol levels, and higher levels of both serum HDL-C and fecal neutral steroids.

BACKGROUND: Clinical observation demonstrates that accelerated fracture healing or lower limb heterotopic ossifications always occur in patients with paraplegia. It indicates that peripheral nervous system may play an important role in fracture healing process.OBJECTIVE: To observe bone histomorphometery parameter, callus formation and biochemical change during the process of fracture healing of unilateral lower limb denervated tibia.DESIGN: Self-control animal experiment.SETTING: Tianjin Hospital.MATERIALS: Totally 36 six-month-old healthy male Wistar rats, with mean body mass of 210 g, were used in this experiment.METHODS: This experiment was carried out at Animal Experimental Center of Tianjin Hospital from March 2001 to March 2004. Denervated tibia fracture model and innervated tibia fracture model were made in the same rat. Animals were executed under anaesthetic status at week 2 and week 4 after fracture. Bilateral tibias were chosen to take radiografts.Biomachamical strength was measured and non-decalcification sections were prepared to perform bone histomorphometery observation.MAIN OUTCOME MEASURES: ① Comparison of wet weight of bilateral tibias and callus of rats between two groups after fracture. ②X-ray plain film scoring. ③ Biomechanical testing of tibial samples. ④ Histomorphological observation of fracture healing RESULTS: ① Wet weight of bilateral tibia and callus of rats in denervated group was much higher than that in innervated group at weeks 2 and 4 after fracture [(0.94±0.15) vs (0.76±0.14) g, (1.06±0.26)vs (0.81±0.10) g,P < 0.05]. ②In X-ray plain film scoring, callus formation was significantly increased in denervated group (P < 0.01). ③In biomechanical testing of three-point bending of tibial sample, callus intensity was significantly lower at weeks 2 and 4 after fracture in denervated group than in innervated group[ (9.88±8.49)vs ( 16.62±13.38 ) N, ( 12.77±7.55 )vs (20.19±10.60) N,P < 0.05]. ④Bone histomorphometery showed that compared with innervated group, mineralized bone trabecula width of denervated group was significantly reduced (P < 0.05), osteoid width was increased , osteoclast index and bone absorption area were significantly increased (P < 0.05), and there were no significant difference of fibroblast index and bone formation area between two groups; Compared with innervated group, mineralized deposition rate in the denervated group was significantly reduced (P < 0.05), the mature time of osteoid was elongated (P < 0.05).CONCLUSION: Peripheral nervous system may play an important role during early and middle period of fracture healing. Intact innervation is essential for normal fracture healing.

The effects of synthetic oil used as dietary fat source on the body weight, organ weight, liver and serum lipids of rats during the hypercho-lesterolemic inducing period were investigated, and comparison was made with lard and soybean oil diets.Weanling albino rats were fed first with stock diet for 3 weeks. Bloods were collected from tails, and serum cholesterol contents were determined. Based upon the body weight and serum cholesterol level, animals were then divided into 5 groups. Four groups were fed respectively with synthetic oil, synthetic oil plus linoleic acid, lard and soybean oil diets, all were supplemented with 1% cholesterol. The, fifth group served as control, was fed with soybean oil diet without cholesterol. Body weights and serum lipids (cholesterol and triglyceride) were measured at intervals during the experiment. Organ weights and liver lipids (total fat, cholesterol and triglyceride) were determined at the end of the experiment. The results showed that there was marked sex difference between male and female rats, that is, synthetic oil diet caused significant lower body weights and lower serum cholesterol levels to female but not to male rats.Cholesterol inducing diets apparently increased the liver weights of all animals, yet fat accumulation only occured in those livers of rats fed with lard and soybean oil diets. The liver fat contents of animals of both synthetic oil and synthetic oil plus linoleic acid groups were at normal level. This experiment reveals that when dietary cholesterol intake is high, the use of synthetic oil as a dietary fat source for maintaining a normal lipid metabolic condition in the body seems beneficial.

Objective To explore the anti-inflammatory effect of antiflammin-2 (AF2) and recombinant peptide sequence 2(R2) on acute lung injury of mouse. To observe the expression of clara cell 16000 protein (CC16) and surfactant protein A (SP-A) in the lung of mouse inoculated with lipopolysaccharide (LPS) and the impact of AF2,R2,and glucocorticoids(hydrocortisone,HC) may have on the expression of the CC16 and SP-A in the lung of mice with acute lung injury. Methods Balb/c mice were inoculated with LPS (5 mg/kg) by intraperitoneal injection to set up ALI mice model. Mice weighed from 15 g to 16 g were grouped into control group, model group and treated groups respectively treated with AF2, R2 or HC. Mice in the control group were injected with physiological saline solution, while mice in the other four groups were inoculated with LPS to induce acute lung injury. Then animals in the treated groups were treated with AF2, R2 or HC each on a dose of 2 mg/kg also through intraperitoneal injection, while those of the control group and the model group, were given equivalent physiological saline solution as a placebo. The respiratory rate of all of these animals were recorded 6 hours after the injection. And at the time point of 12 hour, all the mice were sacrificed for a preparation of the whole lung tissue for the sake of a pathological investigation ,or for extractions of RNA for a semiquantitative analysis of the expression of CC16 and SP-A within the lungs. Results (1) An obvious attenuation of the respiratory rates of the three treated groups were observed when comparing with that of the mice in the model group without any anti-inflammatory treatment. (2) Remarkable extenuation of the extent of intra-alveolar and intersticial hemorrhage and infiltration of inflammatory cells were observed within the treated groups comparing with that of the model group. (3) An attenuate expressions of CC16 or SP-A were observed in the model group, while obvious uptrend of CC16 expression was observed in AF2 treated groups and increase of SP-A expressions were found in R2 and HC treated groups. Conclusion The anti-inflammatory effect of the peptide, AF-2 or R2, has been conformed on ALI mice model induced by LPS.
