1.Pathogenic Bacteria Resistance to Disinfectants in Hospital Infection

Hospital infection has become a global public health problem. The strain of anti-disinfectant, same as the antibiotic-resistant strain, will lead to the failure of the disinfection of the hospital, which will cause hospital infection. The bacterial resistance to disinfectants and resistance mechanism is not clear. Most of the studies focused on the cell research, the molecular research, non-physiological mechanism of resistance, in recent years, the selection pressure, outflow pump were studied. Currently, disinfectants abuse, improper handling and inadequate amount of disinfectant were considered as the main cause of the resistance development.
2.Radiotherapy induced microRNA expression in the plasma of breast cancer patients

Objective To investigate the expressions of miR-210-3p,miR-221-3p,miR-21-5p and miR-150-Sp in the plasma of breast cancer patients before and after radiotherapy in order to establish reliable early biomarkers of non-uniform radiation injuries.Methods Blood samples were collected from 13 patients before radiotherapy (0 Gy) and 24 h after radiotherapy of 2,10,20,30 Gy.The miRNAs in the blood plasma were detected with qRT-PCR.Results The levels of miR-210-3p,miR-221-3p,miR-21-5p had no significant difference among different dosage groups after radiotherapy(P > 0.05).There was no significant difference between the expression of miR-150-5p before and after 2 Gy radiotherapy (P > 0.05).While the relative level of miR-150-5p gradually decreased to 0.808,0.605,0.565 (x2 =18.76,P < 0.05) with increased accumulative dosage of 10,20,and 30 Gy,respectively.In addition,the miR-150-5p expression levels had no relationship (P > 0.05) with situations (positive or negative group) of human epidermal growth factor receptor 2 (HER2),estrogen receptor (ER) and progesterone receptor (PR) in the breast cancer cells.Conclusions Ionizing radiation could reduce the expression of miR-150 in the plasma of breast cancer patients in a dose-dependent manner.
3.Bacteria Contamination on Hands of Medical Staff in Tianjin and Their Antibiotic Resistance

OBJECTIVE To investigate and analyze hand carrier state of medical staff in Tianjin and the antibiotic resistance of the pathogens.METHODS Bacteria or pathogens were isolated from the samples using a sterilized cotton swab on the whole hand before hand washing or hand disinfection,and drug sensitivity test was performed.RESULTS In 16.72% of all samples(50/299),there were 64 kinds of bacteria or pathogens isolated.A.baumannii,Staphylococcus aureus and A.lwoffii were the most common bacteria,40.6% resistant to more then 3 kinds of drugs.Four Meticillin-resistant S.aureus(MRSA) strains were detected out.CONCLUSIONS The high rate of hand carrying bacteria and resistance in Tianjin should pay attention to.
4.Effects of radon and its progeny on the expression and mutation of p53 in lung tissues of mice

Objective To explore the effect of radon and its progeny on the expression and mutations of p53 in lung tissue of mouse model. Methods Apoptosis was detected by terminal deoxynucleotidy transferase-mediated dUTP-biotin nick end labeling. The expression of p53 gene was analyzed by immunohistochemistry, Western blot and realtime-PCR. PCR-SSCP was used to detect the mutation of p53 in lung tissues. Results Compared with those in the control group, the apoptotic index were increased significantly in 30 WLM and 60 WLM groups( t = 18.11, -10.30,P < 0.05 ). The p53 protein was increased significantly ( t = -11.08, P < 0.05; t = - 7.00, P < 0.0 ) ) in 30 WLM and 60 WLM groups. The mutation of p53 gene was not detected in lungs of radon-exposure mice. Conclusions Lung and bronchus might be the targets of radon and its progeny, and p53 gene plays an important role in the progression of radon-induced lung injury.
5.Effect of interference of SNCG gene on radiosensitivity of breast cancer T47D cells

Objective To explore the role of γ-synuclein(SNCG) siRNA in the radiosensitivity of breast cancer T47D cells.Methods SNCG siRNA was synthesized according to the coding sequence of SNCG mRNA and then transiently transferred into T 47D cell with lipofectamine .The expression of SNCG gene and protein was detected by RT-PCR and Western-blot, respectively.Cells were divided into three groups, SNCG siRNA interference group , negative control group and blank control group , which were irradiated with different doses of 60 Coγ-rays.Cell radiosensitivity was evaluated by colony formation assay , cell proliferation was assayed by CCK-8 kit, and the protein expressions of phosphorylated-AKT and mTOR were detected by Western blot assay .Results Compared with blank control cells , the expressions of SNCG gene and protein in the SNCG siRNA transferred T 47D cells were efficiently diminished .Cell colony formation results showed that , under 4, 6, 8 Gy irradiation, the cell survival of siRNA transfection group was lower than that of control group (t=5.449, 8.882, 21.503, P<0.05).CCK-8 experiments showed that the cell proliferation abilities of siRNA group at 24, 48, 72 h after 6 Gy irradiation were lower than those of control group (t=5.603, 4.839, 6.115, P<0.05).In addition, after 6 Gy irraddaition, the AKT and mTOR phosphorylation levels in the siRNA group were more obviously reduced compared with blank groups , but the total AKT and mTOR had no changes .Conclusions Transfection of SNCG siRNA can enhance the radiosensitivity of breast cancer cells probably by inhibiting p -AKT signal pathway .
6.Evaluation of vascular complications after living donor liver transplantation with dynamic contrast-enhanced MR angiography

Objective To observe the value of dynamic contrast-enhanced MR angiography (DCE MRA) with gadobenate dimeglumine in evaluation of vascular complications after living donor liver transplantation. Methods Thirty-four consecutive patients were examined with MR after living donor liver transplantation. First, 1 ml gadobenate dimeglumine was injected in vein to infer the time of gadobenate dimeglumine reaching abdominal artery performing testbolus. Then a scan with three-dimensional T1-weighted fast low-angle shot (3D-FLASH) sequence was performed. Four phases in series from arterial period were scanned and every phase paused 10 s to obtain images of the arterial, portal venous and hepatic venous systems. The original and maximum intensity projection (MIP) reconstructed images, categorized vessel visualization on a five-point scale and observed stoma of hepatic artery, portal venous and hepatic venous inferior vena cava, diameter and display of surrounding vessels were observed. The results were compared with those of digital subtraction angiography (DSA), ultrasound and clinical data. Results Overall vessel visualization assessment demonstrated good or very good ratings for the majority of patients. Among all 34 patients, hepatic artery stenosis was found in 4, portal vein stenosis in 6, portal vein thrombosis was detected in 2, while middle hepatic veins stenosis was detected in 1 patient, among whom 10 patients were confirmed with DSA, 4 with surgery, the others with ultrasound or follow-up. Conclusion Gadobenate dimeglumine DCE MRA is a highly accurate, noninvasive tool for evaluation of vascular complications after living donor liver transplantation, may be regarded as the first choice in postoperative evaluation.
7.The immunoregulation and clinical effect of Yupingfeng capsule combined with Seretide on patients with cough variant asthma

Objective To detect the immunoregulation and clinical effect ofYupingfeng capsule combined with Seretide on patients with cough variant asthma (CVA).Methods CVA Patients were randomly divided into the Seretide control group (n=54) andYupingfeng capsule combined with Seretide group (n=54). Seretide group received inhaled Seretide. Combined traditional Chinese medicine group received Seretide and Yuping Feng capsule. Two groups were treated for 12 weeks. The IL-17, IL-10 and IL-6 expression was detected by ELISA analysis. The clinic effect rate and adverse events were compared.Results After treatment, compared with the Seretide group, the expression of IL-17 (18.72 ± 4.26 ng/mlvs. 26.17 ± 5.58 ng/ml;t=2.462,P<0.05) and IL-6 (21.58 ± 4.12 ng/mlvs. 30.66 ± 6.27 ng/ml;t=2.523,P<0.05) were significantly lower in combined traditional Chinese medicine group than that in Seretide group; and IL-10 (15.56 ± 2.74 ng/mlvs. 12.25 ± 2.81 ng/ml;t=2.244, P<0.05) was significantly higher in combined traditional Chinese medicine group. The daytime (1.12 ± 0.26 vs.1.42 ± 0.33,t=2.283) and night time cough score (1.24 ± 0.28vs. 1.52 ± 0.37,t=2.291) in combined traditional Chinese medicine group was significantly lower than that in Seretide group (P<0.05). The clinic effect rate (92.6%vs. 77.8%,χ2=2.438) in combined traditional Chinese medicine group was significantly higher than that in Seretide group (P=0.037).ConclusionYupingfengcapsule combined with Seretide can decrease IL-17 expression and increase IL-10 expression to inhibit inflammatory reaction in CVA patients, and showed significantly higher clinical effect rates.
8.Retraction of dental unit waterlines in Tianjin City

Objective To survey the retraction of dental unit waterlines (DUWLs)in medical institutions,and evaluate the influencing factors for retraction of DUWLs.Methods In May-November 2014,dental chair units (DCUs)in medical institutions in Tianjin City were sampled through systematic random sampling method,retrac-tion volume of DUWLs was detected by independently made detector,the relevant influencing factors were investi-gated.Results A total of 58 DCUs in 30 medical institutions in 10 districts(counties)of Tianjin were investigated, the average retraction volume of DUWLs was (103.60 ±117.85 )mm3 ,the qualified rate of retraction volume of DUWLs was 48.28 %(28/58);length of utilization of DCUs was positively correlated with DUWLs retraction (r=0.52,P <0.001).There were no significant difference in retraction volume of DUWLs among medical institutions in different districts(counties),levels of medical institutions,DUC brands,and modes of DUC water supply(all P >0.05 ).Conclusion The qualified rate of retraction volume of DUWLs is low,routine maintenance of DUCs needs to be intensified,especially the long-term used DUCs,contaminatin of DUWLs due to the invalidity of retraction valve should be prevented.
9.Changes of lymphocytes and regulatory T cell subset of mice influenced by 60 Co γ-ray irradiation

Objective To analyze the influence of ionizing radiation on the lymphocytes and its regulatory T cells in mice.Methods Mice were administered with whole body irradiation of γ-rays at different doses,and lymphocytes were separated from thymus and spleen,then the number of total cells were counted and the percentages of CD4 + T and CD4 + FOXP3 + CD25 + Treg lymphocytes were analyzed by using FACS.Results The lymphocyte numbers in thymus and spleen decreased in dosedependent manner and reached to the minimum at 4 d after irradiation (F =118.08,144.01,P < 0.05).Exposure to higher dose(more than 1 Gy) decreased Treg number time-dependently in thymus,however increased it in spleen.On the contrary,exposure to lower dose (less than 0.75 Gy) increased Treg number in thymus.Besides,the percentage of Treg cells increased dose-dependently(in thymus,F =5.16,89.44,3.01,P < 0.05 ; in spleen,F =52.02,32.13,27.45,P < 0.05).Conclusions The radiation responses of lymphocytes and their Treg subpopulation vary with the different doses.Treg cells are resistant to high dose irradiation,however,their differentiation could be induced by low dose irradiation.In addition,the different time-dependent responses of lymphocytes and their subpopulation to ionizing radiation indicate the difference of lymphocyte maturation,differentiation and emigration.
10.Expression of JNK/SAPK in lung and bronchus of radon-exposed mice

Objective To study the expression of JNK/SAPK(c-Jun NH2-terminal kinase/stress activated protein kinase)in lung and bronchus of radon-exposed mice.Methods Male BALB/c mice were exposed to radon and its progeny with the cumulative dose of 0.02,30 or 60 working level month(WLM),respectively.The expression levels of JNK/SAPK in lung and bronchus were determined with Real-Time PCR,Western blot and immunohistochemistry methods.Results The JNK mRNA levels in lung tissues of mice exposed to radon of 30 and 60 WLM were higher than those of the control by 3.56 and 2.96 times,respectively.The relative expression levels of JNK and phospho-JNK proteins were higher than those of the control by using Western blot and immunohistochemistry methods.Condusiom Expose to the radon and its progeny might activate the JNK/SAPK intracellular signaling pathway.
