1.Experimental study on the change in intracellular glycogen in ovarian cancer after all-trans retinoic acid treatment

Objective To investigate the effect of all-trans retinoic acid on the change in intracellular glycogen in ovarian epithelioma cell line in vitro and ovarian epithelial carcinoma in nude mice in vivo. Methods COC2 cells were treated with all-trans retinoic acid in 1, 5, 10 and 30?mol/L drug concentrations for different length of time, and then intracellular glycogen and LDH were determined by biochemistry assay. Morphologic changes were observed with light and electron microscopy. CAOV3 tumor-bearing nude mice were treated with intragastric injection of the same drug in a dose of 2mg/(kg?d) for four weeks. The tumor samples were harvested thereafter for pathological study with histochemical and immunohistochemical staining, and also with electron microscopy. Results Intracellular glycogen was significantly increased, while LDH level was lowered after the cell line was treated with 5~10?mol/L of all-trans retinoic acid, and apoptosis of cancer cell occurred after using 30?mol/L of the drug. These changes were also observed in CAOV3 cells of tumor-bearing nude mice. Conclusion Our results suggest that treatment with all-trans retinoic acid resultin an increase in intracellular glycogen and decrease in LDH level both in COC2 cells in vitro and in CAOV3 tumor-bearing nude mice in vivo, and the suppression of tumor cell proliferation may be attributed to retarded intracellular metabolism.
2.THE CHARACTERISTICS OF STRESS AND DISPLACEMENT IN HUMAN TEMPOROMANDIBULAR JOINT DURING CENTRIC OCCLUSION

In order to study the stress distribution and displacement characternstic of the temporomandibular joint (TMJ) during centric occlusion, using the three dimensional nonlinear finite element model of normal human TMJ developed from in vivo with simulation of the contact relationship within TMJ, the stress and the maximum displacement of various structures in the TMJ were calculated and analyzed during centric occlusion. Furthermore, we separately accounted the mechanical properties of various structures in human TMJ and three dimensional muscle force vectors of their masticatory muscles measured and analyzed by use of the fusion of cranio jaw facial CT and MR image with the multiresolution method based on wavelet pyramid. The results showed that there were significant differences in the region and value of stress distribution on the surface of the disc, condyle and articular fossa during centric occlusion, in which the biggest stress was distributed in the condyle, the second in the articular fossa, and the smallest in the disc. During centric occlusion, the disc, condyle and articular fossa moved upperly, posteriorly and medially with the biggest displacement on the condyle, the secondary one on the disc and the smallest one on the fossa. It suggested that it is clinically feasible to systematically and rationally calculate and analyze the stress distribution and displacement character, and the results were satisfying
3.Trends of colorectal cancer incidence in Xuhui District of Shanghai (2001-2011)

Objective To investigate the trends of colorectal cancer incidence in Shanghai Xuhui District from 2001 to 2011.Methods Total 3042 cases of colon cancer and 1958 cases of rectal cancer were collected from the Shanghai Cancer Reporting System.The results were age-adjusted according the census data of 2000 as the standardized incidence.Results The standardized incidence rate of colorectal cancer in Xuhui District rose from the 21.83/100 000 in 2001 to 27.35/100 000 in 2006 and increased by 25.29% ; from 2009 to 2011 the incidence rate was stabilized.The ratio of colon to rectal cancer was 1.55:1,and the incidence of colon cancer was significantly higher than rectal cancer.The incidence rate of male and feinale colorectal cancer increased with age,especially at > 45 y group accounting for 98.70% and 98.37% of total incidence rate respectively.The ratio of male to female in colon cancer incidence rate was 0.98∶ 1,while that of rectal cancer was1.25∶ 1.Conclusion The incidence of colorectal cancer in Xuhui District from the last 11 years is significantly higher than of national level.More effective preventive measures should be taken.
4.Comparison of the determination results of potassium by blood-gas analyzer and dry chemical analyzer

Objective To analyze the comparability of potassium results detected by blood-gas analyzer and dry chemical analy-zer .Methods Dry chemical detection system was used as comparison method (X) ,and blood gas analyzer systems was used as the experimental method (Y ) .Arterial blood samples collected from ICU newborns were detected respectively by the two methods .If SE of medical decision level was less than or equal to 1/2 TEa decided by CLIA′88 ,the results was acceptable .Results There was a linear correlation in potassium concentrations of the two detection systems(r=0 .976 ,P<0 .01) .But among three medical decision levels ,the SE of the two detection systems only acceptable at 3 .0 mmol/L .Conclusion The potassium concentration of blood gas analyzer is lower than that of dry chemistry analyzer .The potassium concentration of dry chemistry analyzer should be taken as a reference to diagnose and clinical treatment .
5.Analgesic sites of total alkaloids in Papaver nudicaule

Objective To study the analgesic sites and mechanism of total alkaloids in Papaver nudicaule (TAPN). Methods The sites of analgesic action of TAPN were observed by adopting Formalin test and two different routes of intracerebroventricular (icv), intrathecal (ith) in mice and rats and comparing ED 50 of TAPN in writhing reaction in mice induced by acetic acid by ip and iv administration. Results TAPN significantly lowered pain score of the early and late phase of Formalin response and was more sensitive in the late phase. Inhibition of TAPN at the same dose by ip administration in writhing reaction in mice induced by acetic acid was stronger than that by iv. The ED 50 value of TAPN at any time by ip administration was lower than that by iv administration. By means of central microinjection, TAPN by icv administration induced remarkable analgesic effect in mice, but TAPN by ith administration had no significant analgesic effect. Conclusion The sites of analgesic action of TAPN are mainly at periphery. TAPN has also central analgesic effect, and the site may be at the level above the spinal cord.
6.Experimental study on establishing tumor bearing nude mice model with different ovarian adenocarcinoma cell lines

Objective:To establish the best way of tumor-bearing nude mice model with epithelial ovarian adenocarcinoma cell lines. Methods:CAOV3 (wall-attached cell) and COC2 (suspended cell) cell lines were transplanted subcutaneously to the nude mice by injection with different cell concentration or tissue block implantation. Results:The rate and time of tumor formation in the two cell lines were not related to cell concentration in the range from 5?10 6/0.2 ml to 6?10 7/0.2 ml. Time of tumor formation was earlier(especially in the manner of tissue block implantation),the successful rate of tumor formation was higher and the speed of tumor growth was relatively retarded in the nude mice bore CAOV3 cell line's tumor than that in COC2 cell line's. But once tumors were formed in nude mice bearing COC2 cell line, the mice died too fast to make experiment because of very quick speed of tumor growth. Conclusion:Wall-attached cell line, tissue block implantation with trocar may be the best way to establish tumor-bearing nude mice model in epithelial ovarian adenocarcinoma cell lines.
7.Role of endometrial and subendometrial blood flows measured by three-dimensional power Doppler ultrasound in the prediction of endometrial receptivity during IVF-ET treatment

Objective To investigate the role of endometrial and subendometrial blood flows measured by transvaginal three-dimensional(3D) power Doppler ultrasound in the prediction of endometrial receptivity during in vitro fertilization-embryo transfer (IVF-ET) cycles. Methods One hundred and twenty patients undergoing IVF-ET cycles were enrolled in this study. Transvaginal 3D power Doppler ultrasound examination was performed on the day of human chorionic gonadotropin injection to determine endometrial thickness,endometrial volume, vascularization index(VI), flow index(FI) and vascularization flow index (VFI) of endometrial and subendometrial regions by experienced sonograpbers. The patients were divided into pregnant group and non-pregnant group according to pregnant outcome. The data between the two groups were compared and analysed. Results There were no significant differences( P >0.05) between the pregnant group and non-pregnant group in endometrial thickness, endometrial volume. Endometrial and subendometrial VI,FI, VFI were higher in the pregnant group than those in the non-pregnant group, and statistical comparison had significant difference( P <0.05). Conclusions Endometrial and subendometrial blood flows measured by transvaginal 3D power Doppler ultrasound can be as a reliable indicator to assess endometrial receptivity and to predict the pregnancy outcome.
8.Effect of 308 nm laser irradiation on Treg cells and Th17 cells in guinea pigs modelling vitiligo

Objective To assess the efficacy of 308 nm excimer laser irradiation for the treatment of guinea pig model of vitiligo and its immunoregulatory effect on Treg cell and Th17 cell.Methods Vitiligo was induced in guinea pigs by hydroquinone bleaching,and they were randomly divided into an experimental group and a control group.There was also a normal control group.The experimental group was treated with a 308 nm excimer laser at the nidus of vitiligo; the two control groups were given no intervention.After 8 weeks of treatment,leukasmus of the different groups were compared.The mRNA expression of Foxp3 and IL-17 were measured by real-time fluorescence quantitative (RTFQ-PCR) and the expression of Foxp3 and IL-17 in the lesions were measured by immunohistochemistry staining.Results The effectiveness rate was 100% in the experiment group.The results of RTFQ-PCR showed that the mRNA expression of Foxp3 in the experimental group and the vitiligo control group was significantly higher than the healthy control group (all P < 0.05),being 0.33 ± 0.03 and 0.02 ±0.07 respectively,as was the mRNA expression of IL-17,being 0.21 ± 0.05 and 0.94 ± 0.06 accordingly; and the mRNA expression of IL-17 in the experimental group was significantly lower than the vitiligo control group(P < 0.05).The immunohistochemical staining showed that Foxp3 and IL-17 protein was scattered in the lesions of the experimental group,while they were crowded in those of the vitiligo control group.Conclusion Laser irradiation might contribute to a therapeutic effect on vitiligo through modulating the balance of Treg and Th17 cells,at least in guinea pigs.
9.Rapid Determination of N-Methylcarbamoyl Adduct in Hemoglobin of Workers Exposed to N,N-Dimethylformamide by Ultra High Performance Liquid Chromatography-Mass Spectrometry

A rapid analytical method was developed for the determination of N-methylcarbamoyl adduct in Hemoglobin of workers exposed to N,N-dimethylformamide by ultra high performance liquid chromatography-mass spectrometry ( UPLC/MS/MS). About 0. 1 g of hemoglobin sample, 40 μmol/L of 3-methyl-5-isopropylhydantoin (MIH) as the internal standard and 4. 75 mL of HCl-acetic acid (2∶1, V/V) were added in the centrifuge tube, and mixed for 3 min. Then the tube was heated in boiling water bath for 1h. After cooling down, 200 μL of the mixture and 600 μL of formic acid-acetonitrile (1%) were added into 96-well extract plate. The vacuum pump pressure was controlled to make the sample collection elute within 2-4 min.The purified collection was transferred into the sample vial, and 3-methyl-5-isopropylhydantoin ( MVH ) as degration product of N-methylcarbamoyl adduct was quantified by UPLC/MS/MS in multiple reaction monitoring ( MRM ) by internal standard method. A good linear relationship was obtained in the MVH concentration range of 0 . 01-1 . 0 μmol/L with the correlation coefficient of 0 . 999 . The recovery of added MVH in the blank sample was 97 . 3% and the relative standard deviation was 1 . 7%. The limit of detection (LOD) was 0. 01 μmol/g. This method was proved to be fast and efficient.
10.Determination of Atractylodin in Essential Oil of Jingfukang Preparations and Stability in Different Solutions

Objective To establish a method for determination of atractylodin in essential oil of Jingfukang preparations by GC and analyze its stability in different solutions. Methods Using HP-5 quartz capillary column (30 m×0.32 mm), programmed temperature was conducted with the FID as detector. The content of atractylodin was determined both at room temperature and 4 ℃ in 20 days. Results The linear range of atractylodin was 4.172-41.72 μg/mL (r=0.999 7). The stability of atractylodin in essential oil of atractylodes was better than in anhydrous alcohol solution or in essential oil of Jingfukang preparation. Conclusion This method is suitable for the quality control of atractylodin in essential oil of Jingfukang preparations. Low temperature is helpful to the stability of atractylodin. The storage period of essential oil of Jingfukang preparations should not be too long.
