AIM: To explore the effects and mechanism of interleukin-2 (IL-2) on the positive effect of isoproterenol (ISO) in the isolated rat cardiomyocytes. METHODS: Enzymatically isolated cardiomyocytes were used. Peak twitch amplitude and maximal velocity of shortening/relaxation (?dL/dt_~max ) in the isolated cardiomyocytes were recorded with a microscope coupled to a charge-coupled device camera and [Ca~2+ ]i transients were determined with a fluorometric ratio method by using Fura-2/AM as Ca~2+ indicators. RESULTS: ① ISO increased the peak twitch amplitude and ?dL/dt_~max of the isolated cardiomyocytes. Perfusion for 15 min with IL-2 at 2?103 U/L, which had no effect at all, attenuated the enhancing effect of ISO on the peak twitch amplitude and ?dL/dt_~max . ② ISO increased the [Ca~2+ ]i transients of the single ventricular myocytes in a dose dependent manner and the corresponding EC_~50 values of ISO was (0.12?0.01) ?mol/L. Perfusion for 15 min with IL-2 at 2?103 U/L, which had no effect on the ~[Ca~2+ ]i transient at all, attenuated the enhancing effect of ISO and the corresponding EC_~50 was (0.44?0.06) ?mol/L. ③ The electrically induced [Ca~2+ ]i transient was significantly increased by pretreatment with 20 mg/L cholera toxin for ~12 h. The elevation of the [Ca~2+ ]i transient induced by cholera toxin was significantly attenuated by 2?103 U/L IL-2. ④ Forskolin (1 ?mol/L), the activator of adenyl cyclase, significantly increased the electrically induced [Ca~2+ ]i transient, which was attenuated by IL-2 at 2?103 U/L. CONCLUSION: IL-2 inhibits the positive effect of isoproterenol in the isolated single ventricular myocytes, in which Gs protein and adenyl cyclase are involved. [

OBJECTIVE:
To explore the severity index of Meniere's disease during acute phase by a statistical method of principal component analysis (PCA).
METHOD:
Ninety-five patients with Meniere's disease in the acute phase who saw the doctor with 24 hours after onset were included in this study and their clinical data were analyzed retrospectively. The subjective symptoms of hearing loss (X1) and ear stuffy plug (X4) were evaluated by VAS scoring system, and the impact of vertigo (X2) and tinnitus (X3) on the patients were evaluated by DHI and THI score. The principal component analysis (PCA) method was used to analyze the quantified data and construct a synthetic evaluate function of subjective symptoms.
RESULT:
(1) The quantitative results of subjective symptoms in patients with Meniere's disease were as follow: VAS score (X1) of hearing loss was 0-91 points, with an average of 46.23 ± 18.80, DHI score (X2) of vertigo was 8-98 points, with an average of 49.66 ± 15.67, THI score (X3) of tinnitus was 10-100 points, with an average of 47.53 ± 17.44, and ear VAS score (X4) of stuffy plug feeling was 0-82 points, with an average of 21.55 ± 27.54. (2) The eigenvalue of principal components Z1, Z2 and Z3 were 1.876, 0.984 and 0.703 respectively, and the variance contribution were 46.898%, 24.592% and 17.574% respectively. (3) The constructed synthetic evaluate function of the disease was as follow: The Meniere's disease severity index ƒ = 0.213ZX1 + 0.398ZX2 + 0.370ZX3 + 0.455ZX4.
CONCLUSION
The method of PCA for the subjective evaluation of symptoms in Meniere's disease can be constructed as a model of comprehensive evaluation system, which may provide relatively comprehensive information of clinical original variables included in the four main symptoms, reflecting the severity of the disease.

Objective To investigate distribution and antimicrobial resistance among nosocomial pathogens from 13 teaching hospitals in China in 2009. Methods Non-repetitive pathogens from nosocomial BSI, HAP and IAI were collected and sent to the central lab for MIC determination by agar dilution method.WHONET5.6 software was used to analyze the data. Results A total of 2 502 clinical isolates were collected. The top three pathogens of BSI were Escherichia coli [27. 1% (285/1 052 )] , coagulase-negutive staphylococcus [12. 6% ( 133/1 052)] and Klebsiella pneumoniae [10. 8% ( 114/1 052)]. The top three pathogens of HAP were Acinetobacter baumannii [28. 8% (226/785)], Pseudomonas aeruginosa [16. 1% (126/785)] and Klebsiella pneumoniae [14.6% (115/785 )] . The top three pathogens of IAI were Escherichia coli[31.0% ( 206/665 )], Klebsiella pneumonia [11.3% ( 75/665 )] and Enterococcus faecium [10. 8% (72/665)]. Against Escherichia coil and Klebsiella spp. , the antimicrobial agents with higher than 80% susceptibility rate included imipenem and meropenem (98. 1%-100% ), tigecycline (95.3%-100% ), piperacillin-tazobactam ( 88.6% -97. 1% ) and amikacin ( 88. 3% -92. 5% ). Against Enterobacter spp. , Citrobacter spp. and Serratia spp. , the susceptibility rates of tigecycline were 93.5% -100% whereas the value of imipenem and meropenem were 92.9% -100%. Other antimicrobial agents with high activity included amikacin ( 85.2% -96. 7% ), pipcracillin-tazobactam ( 82.4% -96.4% ), cefepime ( 79. 6% -96. 7% ) and cefoperazonc-sulbactam (78. 7%-90. 0% ). Polymyxin B showed the highest susceptibility rateagainst Pseudomonas aeruginosa ( 100% ), followed by amikacin ( 81.9% ) and piperacillin-tazobactam (80.1% ). Polymyxin B also showed the highest susceptibility rate against Acinetobacter baumannii (98. 8% ), followed by tigecycline (90. 1% ) and minocycline (72. 0% ). The incidence of carbapenemresistant Acinetobacter baumannii was 60. 1%. The MRSA rate was 60. 2% and the MRSCoN rate was 84. 2%. All Staphylococcus strains were susceptible to tigecycline, vancomycin, teicoplanin and linezolid except for one isolate of Staphylococcus haemolysis with intermediate to teicoplanin. Two Enterococcus faecalis isolates which were intermediate to linezolid and one Enterococcus faecium isolate which was resistant to vancomycin and teicoplanin was found in this surveillance, while the MICs of tigecycline against these three isolates were 0. 032-0. 064 μg/ml. Conclusions Tigecycline, carbapenems, piperacillin-tazobactam,amikacin and cefepime remain relatively high activity against nosocomial Enterobacteriaceae. Pseudomonas aeruginosa exhibite high susceptibility to polymyxin B, while Acinetobacter baumanni shows high susceptibility to polymyxin B and tigecycline. Tigecycline, vancomycin, teicoplanin and linezolid remain high activity against nosocomial gram-positive cocci.

China Pharmacy 2019;30(23):3221-3226
OBJECTIVE: To study the improvement effect of Shenrong bunao capsule on learning and memory ability of Alzheimer’s disease (AD) model mice, and to investigate its mechanism. METHODS: Totally 72 mice were randomly divided into blank control group (normal saline), model group (normal saline), Piracetam tablets group (positive control,0.80 g/kg),Shenrong bunao capsule high-dose,middle-dose and low-dose groups (1.92, 0.96, 0.48 g/kg), with 12 mice in each group. Except that blank control group was given constant volume of normal saline subcutaneously. Other groups were given D-galactose (150 mg/kg) subcutaneously and sodium nitrite (50 mg/kg) intraperitoneally every day to induce AD model. At the same time, they were given relevant medicine intragastrically, once a day, for consecutive 60 d. 1 h after last administration, Morris water maze test was used to measure escape latency and times of crossing the platform within 90 s. HE staining was used to observe pathological changes of cerebral cortex in mice. Immunohistochemistry was used to detect the expressions of TNF-α, NF-κB p65, PI3K and Akt in cerebral cortex of mice. RESULTS: Compared with blank control group, escape latency was prolonged significantly (P<0.01), and the times of crossing the platform within 90 s was decreased significantly in model group (P<0.01). The neurons in cerebral cortex was damaged obviously, and the number of intact neurons was decreased significantly (P<0.01). The protein expressions of TNF-α, NF-κB p65, PI3K and Akt in cerebral cortex were increased significantly (P<0.01). Compared with model group, except that there was no statistical significance in escape latency, protein expressions of TNF-α, NF-кB p65, PI3K and Akt in Shenrong bunao capsule low-dose group (P>0.05), above indexes of other administration groups were improved significantly (P<0.05 or P<0.01). CONCLUSIONS: Shenrong bunao capsule can improve the learning and memory ability of AD model mice, and its mechanism may be related to the inhibiting the protein expressions of TNF-α,NF-κB p65, PI3K and Akt in cerebral cortex region and relieving inflammation injury so as to protect cranial nerve.
