1.Distribution and Drug Resistance of Isolates from Blood Culture Samples

OBJECTIVE To enhance the positive rate of the blood culture in order to make pathogenic diagnosis actually and quickly and conducting usage of antimicrobial agents in clinic.To value the clinical applied circumstance of BacT/Alert 3D automated blood culture system.METHODS The 3728 blood cultures were detected by BacT/Alert 3D automated blood culture system,and bacterial susceptibility test was conducted on all isolates using Kirby-Bauer methods with CLSI standards.To statistically analyze the examined time,the positive rate and variety and drug resistance for all kinds of pathogens.RESULTS The blood culture positive rate was 6.0% in the 3728 blood cultures with 222 strains of bacteria.The false positive rate was 0.2% and the false negative rate was 0.4%.The positive rate of blood culture was 0.89% in 12 hours,2.01% in 18 hours,and 3.51% in 24 hours.Among all the 222 isolates,29.7% were Enterobacteriaceae,the drug resistant rates to amikacin,ceftazidime,ciprofloxacin,and imipenem were 13.6%,36.3%,42.4%,and 3.0%,respectively;20.3% were Staphylococcus,the drug resistant rates to erythromycin,levofloxacin,cefoxitin,and vancomycin were 75.6%,33.3%,68.9%,and 0,respectively;11.7% were Enterococcus,the drug resistant rates to errythromycin,levofloxacin,fosfomycin,and vancomycin were 96.2%,80.8%,23.1%,and 0,respectively;11.3% were non-fermented bacilli,the drug resistant rates to amikacin,ceftazidime,ciprofloxacin,and imipenem were 32.0%,52.0%,32.0%,and 32.0%,respectively;12.6% were fungi.CONCLUSIONS The pathogens in the blood specimens are more wider in distribution and more higher in drug resistance rates than before.BacT/Alert 3D automated blood culture system can be an important tool for the blood culture,it can provide the diagnostic help quickly for the clinic and increase the positive rates in blood culture.It can not only shorten the check-up time,but also be more quick and more exact.
2.Effects of sevoflurane on development of posttraumatic stress disorder in rats and the role of hippocampal GSK-3β

Objective To evaluate the effects of sevoflurane on the development of posttraumatic stress disorder (PTSD) in rats and the role of hippocampal glycogen synthase kinase-3beta (GSK-3β).Methods Forty male Sprague-Dawley rats,weighing 250-300 g,aged 2 months,were randomly divided into 4 groups (n =10 each):control group (group C) ; PTSD group (group P) ; sevoflurane group (group S) ; lithium chloride (LiC1) + sevoflurane group (group LS).Establishment of PTSD model:the rats received 15 footshocks (1 mA,1 s shocks with a variable intershock interval of 240-480 s) in room A on the first day.On the second day,the rats received 1 footshock after an adaptation of 192 s in room B where its context was completely different from room A,and then they were removed from room B 32 s later.The rats were placed in room B and observed for 512 s on the third day.Group C did not receive any treatment on the first day in room A,while the other three groups received 15 footshocks in room A.In addition,0.8% sevoflurane was administrated while training in group S,and group LS received intraperitoneal LiCl 100 mg/kg at 30 min before entering room A,and then the other treatments were similar to those previously described in group S.The ratio of freezing was recorded in room B on the second and third days.Four rats in each group were randomly sacrificed at 2 h after training in room A on the first day and their hippocampal tissues were taken to detect the expression of GSK-3β and phosphorylated GSK-3β (p-GSK-3β).Results Compared with group C,the ratio of freezing was significantly increased on the third day and the p-GSK-3β expression was up-regulated in P and LS groups,and no significant change was found in the parameters mentioned above in group S.Compared with group P,the ratio of freezing was significantly decreased on the third day and the p-GSK-3β expression was down-regulated in group S,and no significant change was found in the parameters mentioned above in group LS.Compared with group S,the ratio of freezing was significantly increased on the third day and the p-GSK-3β expression was up-regulated in group LS.There was no significant difference in the ratio of freezing on the second day and expression of p-GSK-3β between the four groups.Conclusion Sevoflurane can inhibit the development of PTSD in rats,and enhancement of hippocampal GSK-3β activity may be involved in the mechanism.
3.Cluster analysis on hypocrellin A content in anamorphs of Shiraia bambusicola.

OBJECTIVEBy examining the colony growth rate, colony texture, substrate mycelium color and separation method of 40 anamorph strains of Shiraia bambusicola to investigate the relationship between these indexes and content of hypocrellin A.
METHODIn UV-spectrophotometry and microscopic observation were applied in the experiments. The results were analyzed by the clustering and variance analysis.
RESULTSPSS clustering analysis showed that the colony characters of S. bambusicola anamorph strains had significant differences with hypocrellin A content.
CONCLUSIONIn the tested conditions, substrate mycelium color of S. bambusicola anamophs has the most distinct correlation with the yield of hypocrellin A.

4.Investigations on the effect of measuring value transfer for human serum samples assigned by the reference laboratory network

Objective To investigate the effect of measuring value transfer for human serum samples assigned by the reference laboratory network on improving the trueness of seven enzyme activities in clinical laboratories,such as ALT,AST,GGT,LDH,CK,AMY and ALP.Methods Depending on the medical imtitutions at all levels contacted by 5 reference laboratories in North China,South China,East China and Southwest China,the corresponding clinical laboratory measuring value transfer/traceability network was established.The frozen human serum samples with good interehangeability and standard material characteristics,including calibrator,sample 1 and sample 2,were provided by Beijing Aerospace General Hospital,and were assigned by 5 reference labotatories in four regiom.These samples were sent to 48 clinical laboratories.These clinical laboratories measured sample 1 and sample 2 according to their standard operating procedures,and then measured.the two samples again after adjusting their measurement system by using the supplied calibrator.The changes of trueness of detection results in these laboratories were evaluated according to the WS/T 403-2012 standard,and the changes of consistency for ALT and AST before and after measuring value tramfer were investigated.Results The results of AMY,ALP,GGT,CK and LDH calibrator,sample 1 and sample 2 assigned by the established network were 138.7 U/L,278.5 U/L and 68.3 U/L,265.3 U/L,94.5 U/L and 134.4 U/L,195.8 U/L,89.0 U/L and 158.9 U/L,393.7 U/L,260.0 U/L and 645.3 U/L,and 302.0 U/L,250.0 U/L and 452.7 U/L,respectively.The percentages of sample 1 and sample 2 which met the bias requirements of the WS/T 403-2012 standard before measuring value transfer for AMY,ALP and GGT were 65.9% and 61.0%,76.6% and 78.7%,and 66.7% and 70.8%,respectively,while after measuring value transfer,they were 89.2% and 83.8%,86.7% and 80.0%,and 85.4% and 91.7%,respectively.The percentages of sample 2 which met the bias requirements of the WS/T 403-2012 standard before measuring value transfer for CK and LDH were 64.6% and 58.3%,respectively,while after measuring value trander,they were 93.5% and 84.8%,respectively.The coefficients of variation (consistency) of sample 1 and sample 2 for ALT and AST before measuring value tramfer were 12.9% and 11.3%,and 10.2% and 8.9%,respectively,while after measuring value transfer,they were 9.3% and 8.2%,and 5.6% and 5.9%,respectively.Conclusion The calibration of routine measurement systems based on the measuring value transfer for human serum samples assigned by the reference laboratory network may improve the comparability of 7 enzyme actvities measurement results in chnical laboratories at all levels obviously,which deserves to be further spread.
5.Analysis of age-period-cohort model for incidence and mortality of digestive tract tumors in China

Objective This study analyzed the incidence and mortality trends of four types of digestive tract tumors in China from 1990 to 2019,including gastric cancer,esophageal cancer,colorectal cancer,and liver cancer,in order to provide scientific basis for the formulation of prevention and control strategies for digestive tract tumors in China.Methods The Global Burden of Disease Study 2019(GBD 2019)database was used to analyze the trends of standardized incidence and standardized mortality of digestive tract tumors in China by the Joinpoint regression model.The age-period-cohort model was used to explore the age-period and cohort effects on the incidence and mortality of digestive tract tumors.Results The incidence order of digestive tract tumors in 2019 was stomach cancer(43.09/100,000),Colon and rectum(42.74/100,000),esophageal cancer(19.55/100,000)and liver cancer(14.80/100,000).The death order was gastric cancer(29.64/100,000),Colon and rectum(18.40/100,000),esophageal cancer(18.09/100,000)and liver cancer(13.20/100,000).After eliminating the influence of population age structure,the standardized in-cidence and standardized mortality of liver cancer,gastric cancer and esophageal cancer from 1990 to 2019 showed a decreasing trend year by year(AAPC liver cancer incidence=-3.1%,AAPC gastric cancer incidence=-0.07%,AAPC esophageal cancer inci-dence=-1.5%;AAPC liver cancer death=-3.4%,AAPC gastric cancer death=-1.9%,AAPC esophageal cancer death=-1.8%)(P<0.001),and the standardized incidence and standardized mortality of Colon and rectum showed an increasing trend year by year(AAPC Colon and rectum incidence=3.1%,AAPC Colon and rectum death=1.1%)(P<0.001).The results of age-period-cohort model showed that the incidence and mortality of gastric cancer,esophageal cancer and Colon and rectum were increas-ing with age,reached the peak in 85+group,80-84 group and 85+group,respectively.The incidence and mortality of liver cancer peaked in the 55-59 group and then decreased.The risk of Colon and rectum showed an increasing trend year by year,and the risk of death showed a first increasing trend and then decreasing trend.The incidence and mortality of liver cancer showed a decreasing trend followed by an increasing trend,while the overall incidence and mortality of gastric cancer and esophageal cancer showed a decreasing trend year by year.The earlier birth cohort of esophageal cancer and liver cancer,the higher risk of morbidity and mortality,while the later birth cohort of Colon and rectum,the higher risk of morbidity and mortality.The risk of gastric cancer showed the fluctuation.Conclusion The disease burden of digestive tract tumors in China is still heavy in China,especially the standardized incidence and standardized mortality of Colon and rectum are still increasing year by year.It is necessary to implement targeted prevention and treat-ment measures to effectively reduce the disease burden of digestive tract tumors.
6. Expression of Tim-3 on natural killer cells in patients with acute myeloid leukemia and its clinical significance

Objective:
To investigate the expression characteristics of Tim-3 on natural killer (NK) cells of peripheral blood in patients with acute myeloid leukemia (AML) and its clinical significance.
Methods:
Peripheral blood was obtained from 39 patients with newly diagnosed AML before intervention, with peripheral blood from 28 cases of healthy volunteers collected as normal control. Using CD3, CD56 and Tim-3 as markers, expression levels of Tim-3 on the peripheral blood NK cells were detected by immune fluorescence labeling and flow cytometry.
Results:

7. CD34, VEGF and endothelin-1 expression in intrahepatic metastasis of hepatocellular carcinoma

Objective:
To explore the roles of CD34, VEGF and endothelin-1 in hepatic sinusoid capillarization and the intrahepatic metastasis of hepatocellular carcinoma.
Methods:
Data of 10 cases of hepatocellular carcinoma without intrahepatic metastasis were compared with those 10 cases of hepatocellular carcinoma with intrahepatic metastasis . Liver cancer tissues and paracancerous tissues were collected, hepatic sinusoidal capillarization was detected by immunohistochemistry, mRNA expression and protein expression of CD34, VEGF and endothelin-1 genes were detected by QPCR, Western blot, and the relationship between gene expression and metastasis of HCC was observed.
Results:
Hepatic sinusoid capillarization was observed in carcinoma tissues, not in the adjacent tissues, whereas mRNAs and proteins of CD34, VEGF and endothelin-1 were higher in liver cancer tissues than that in the adjacent tissuess. Traumatic cerebrospinal fluid leakage commonly presents in traumatic brain injury patients, and it may lead to complications such as meningitis, ventriculitis, brain abscess, subdural hematoma or tension pneumocephalus. When misdiagnosed or inappropriately treated, traumatic cerebrospinal fluid leakage may result in severe complications and may be life-threatening. Some traumatic cerebrospinal fluid leakage has concealed manifestations and is prone to misdiagnosis. Due to different sites and mechanisms of trauma and degree of cerebrospinal fluid leak, treatments for traumatic cerebrospinal fluid leakage varies greatly. Hence, the Craniocerebral Trauma Professional Group of Neurosurgery Branch of Chinese Medical Association and the Neurological Injury Professional Group of Trauma Branch of Chinese Medical Association organized relevant experts to formulate the " Chinese expert consensus on the diagnosis and treatment of traumatic cerebrospinal fluid leakage in adults ( version 2023)" based on existing clinical evidence and experience. The consensus consisted of 16 recommendations, covering the leakage diagnosis, localization, treatments, and intracranial infection prevention, so as to standardize the diagnosis and treatment of traumatic cerebrospinal fluid leakage and improve the overall prognosis of the patients.
9.Teaching reform and practice of 'Production Internship' course for biotechnology specialty from the perspective of training application-oriented talents.

Production internship is an important teaching tache for undergraduate students to carry out engineering training by using professional skills, and it is a key starting point for fostering application-oriented talents in biotechnology. The Course Group of 'production internship of biotechnology majors' of Binzhou University is investigating application-oriented transformation for local regular colleges and universities, as well as fostering high-level application-oriented talents. By taking green fluorescent protein (GFP) polyclonal antibody as an example, the reform and practice on teaching content, teaching mode, assessment method, continuous improvement of curriculum were carried out. Moreover, the characteristics of the Yellow River Delta-Binzhou Biotechnology & Pharmaceutical Industrial Cluster were taken into account to intensify academic-enterprise cooperation. On one hand, this Course Group designed and rearranged the course contents, carried out essential training through online resources and platforms such as virtual simulation, and recorded, tracked and monitored the progress of production internship through practical testing and software platforms like 'Alumni State'. On the other hand, this Course Group established a practice-and application-oriented assessment method in the process of production internship and a dual evaluation model for continuous improvement. These reform and practices have promoted the training of application-oriented talents in biotechnology, and may serve as a reference for similar courses.

Bioengineering majors require students to acquire excellent abilities of thinking and analyzing complex problems and have high requirements for students' comprehensive practical skills. Because of the professional characteristics, it is necessary to develop students' abilities to solve complex problems via the teaching of a series of experiments. Therefore, it is particularly important to reform the traditional experiment teaching for students majoring in bioengineering to improve the teaching quality, which have great significance for the cultivation of comprehensive talents. In this study, with the advantages of geographical location and resources to cultivate application-oriented innovative talents, the course group of Comprehensive Experiment of Bioengineering has designed the course based on virtual simulation technology in Binzhou University. Taking the experiment of extraction and bioactivity analysis of Suaeda salsa (growing in the Yellow River Delta) polysaccharide in fermentation as a case, we studied the course design idea, experimental process, teaching method and result analysis, and have improved the teaching performance. This case analysis provides new ideas and content reference for the teaching reform of similar courses.
