1.CDK and JNK mediate FAK to enhance proliferation in human pulmonary artery smooth cells

AIM: To find out the mechanism of focal adhesion kinase(FAK) facilitating human pulmonary artery smooth muscle cells(HPASMCs) proliferation.METHODS: HPASMCs were isolated from normal part of lungs of two carcinoma patients who undergone lung partial resection.Cultured HPASMCs stimulated by fibronection(40 mg/L) were passively transfected with ODNs,sense focal adhesion kinase(FAK),mismatch sense and antisense-FAK,respectively.Expression of FAK,Jun NH2-terminal kinase(JNK) and cyclin-dependent kinase2(CDK2) proteins were detected by immunoprecipitation and Western blotting.Cell cycle and cell apoptosis were analyzed by flow cytometry.In addition,cytoplasma FAK expression was detected by immunohistochemistry staining.RESULTS: The protein expressions of FAK,JNK and CDK2 in HPASMCs decreased in FAK ASODNs group and increased in FAK SODNs group.Meanwhile,the proportion of cells at G1 phase decreased significantly in FAK SODNs group,while the cells at S phase increased significantly.In contrast,the proportion of cells at G1 phase was increased significantly in FAK ASODNs group.The level of cell apoptosis in FAK ASODNs group was higher.FAK expression in FAK SODNs group was strongly stained by immunocytochemistry,whereas that in FAK ASODNs group was weakly stained.CONCLUSION: The results suggest that FAK via JNK,CDK2 signaling pathway enhances HPASMCs proliferation.
2.Focal adhesion kinase antisense oligodeoxynucleotide promotes apoptosis in human pulmonary artery smooth muscle cells

AIM: To investigate whether focal adhesion kinase (FAK) takes part in the intracellular signaling pathway involved in apoptosis of human pulmonary artery smooth muscle cells (HPASMCs). METHODS: Cultured HPASMCs stimulated by fibronection (40 mg/L) were passively transfected with antisense-FAK oligodeoxynucleotides (ODNs). Expression of FAK and caspase-3 proteins was detected by immunoprecipitation and Western blotting. In addition, apoptosis were measured by TUNEL. RESULTS: The protein expression of FAK in HPASMCs decreased and caspase-3 expression upregulated in HPASMCs in antisense-FAK ODNs group. At the same time, antisense-FAK ODNs transfection increased the rate of cell apoptosis. CONCLUSION: These results suggest that FAK may be related with the apoptosis of HPASMCs. Antisense-FAK ODNs inhibit HPASMC proliferation and may facilitate their apoptosis via caspase-3 signaling pathway.

5.Focal adhesion kinase promotes proliferation of human pulmonary artery smooth cells

Objective To study whether focal adhesion kinase (FAK) promotes human pulmonary artery smooth cells (HPASMCs) proliferation.Methods Cultured HPASMCs stimulated by fibronectin (40 mg/L) were passively transfected with sense -FAK oligonucleotides(ODNs), FAK activity was measured by immunoprecipitation and expression of FAK protein was detected by Western blots.Meanwhile, the change of cell proliferation was measured by MTT and 3H-TdR absorbation experiment. Results The change of FAK activity and FAK protein content was dose and time dependent at diferential concentration and time passively transfected with sense-FAK ODNs in Cultured HPASMCs. At the same time, sense-FAK ODNs prompoted HPASMCs proliferation and 3H-TdR absorbation.Conclusion FAK can faciliate HPASMCs proliferation,which may play an important function in pulmonary artery hypertension development.
6.Lengthening the short nose with bilateral septal spreader graft and columellar strut

Objective To investigate a safe and effective method of lengthening the short nose.Methods Thirty-nine cases of short nose were corrected by the bilateral septal spreader graft and columellar strut using autologous cartilage,combined with expanded polytetrafluoroethylene (e-PTFE)to reconstruct the supporting structures to improve the forward and upward strength of the nasal tip in order to increase the nasal length and to correct the over rotation of nasal tip.The shield and cap grafts were also used for the patients whose nasal tip were too low with vertical dome division technique.Results All the patients showed preserved nasal length after surgery with well-proportioned nasal features and the nasal lip angles were normal.There was no evidence of postoperative nasal shortening.No operative or postoperative complications were observed by follow-up for 6 months to 1 year,except for 2 cases with graft cartilage being apparent on nasal tip.All the patients were satisfied with the surgical results achieved.Conclusions It is necessary to provide powerful forward and upward strength to lengthen nose and correct the over rotation of nasal tip effectively and safely.The application of bilateral septal spreader graft and columellar strut using autologous cartilage and combined with e-PTFE represents a valuable tool for the short nose and proper cartilage grafts of nasal tip can improve the height of the nasal tip and further correct the over rotation of the nasal tip.
7.Study on the activity of IgY against complex bacteria in pharynx and throat

Objective:To detect the activity of IgY against complex bacteria in pharynx and throat.Methods:Purified antigens against bacteria in pharynx and throat was used to immunize egglaid hens.The eggs from immunized hens were collected and abstract IgY from the yolks.The antibody activity of IgY was detected by SDS-PAGE electrophoresis and ELISA.Results:SDS-PAGE electrophoresis represented at least twelve ladders,and the titer of ELISA was 1∶512.Conclusion:IgY antibody was obtained in egg yolk after immunized hens with complex bacteria.The activity of IgY was detected.IgY showed stable to heat.
8.Role of adenosine A1 receptors in hippocampal neurons in cognitive dysfunction caused by isoflurane anesthesia in aged mice

Objective To evaluate the role of adenosine A1 receptors in hippocampal neurons in the cognitive dysfunction caused by isoflurane anesthesia in aged mice.Methods Sixteen male adenosine A1 receptor gene knockout homozygote mice (gene knockout mice) and 16 male wild-type mice,aged 18-22 months,weighing 27-32 g,were studied.Each type of mice was randomly divided into 2 groups (n=8 each) using a random number table:control group (group C) and isoflurane anesthesia group (group Ⅰ).Mice inhaled 1.4% isoflurane in 100% O2 for 2 h in group Ⅰ,and 100% O2 for 2 h in group C.All the mice underwent Morris water maze test at 24 h after isoflurane or O2 inhalation.After the test,the mice were sacrificed and the hippocampal tissues were harvested to determine the number of β-amyloid1-42 (Aβ1-42) plaques (using immunohistochemistry) and expression of phosphorylated tau (p-tau) protein,and 2B subunit-containing N-methyl-D-aspartate receptors (NR2B) (by Western blot analysis).Results Compared with group C of wild type mice,the escape latency was significantly prolonged,the number of Aβ1-42 plaques was enlarged,the expression of p-tau protein was up-regulated,and the expression of N R2B was down-regulated in group Ⅰ of wild type mice.Compared with group Ⅰ of wild type mice,the escape latency was significantly shortened,the number of Aβ1-42 plaques was decreased,the expression of p-tau protein was down-regulated,and the expression of NR2B was up-regulated in group Ⅰ of gene knockout mice.There was no significant difference in the parameters mentioned above between group Ⅰ and group C of gene knockout mice.Conclusion Adenosine A1 receptors in hippocampal neurons mediate isoflurane anesthesia-induced cognitive dysfunction in aged mice,and the mechanism may be related to promotion of deposition of Aβ,phosphorylation of tau protein and inhibition of activities of NR2B.
9.The detection and analysis of serum IL-8 and IgA/C3 in henoch-schonlein purpura

Objective To investigate the correlation between serum interleukin-8 (IL-8 )and immunoglobulin/complement 3A (IgA/C3)and immune imbalance in Henoch-Schonlein Purpura(HSP)in children,and provide evidence and guidance for clinical therapy of HSP.Methods Serum IL-8 was measured by Enzyme-linked immunosorbent assay(ELISA),and IgA and C3 were de-tected by automatic biochemistry analyzer,and calculate the ratio of IgA/C3.Results Serum IL-8 levels were (389±5 1.68)ng/L, significantly higher than that in healthy children (P < 0.05 ).Serum IgA levels were(306.2 ± 32.21 )mg/L,significantly higher (P <0.05).Serum C3 levels were (1 14.5 ±20.7)mg/L,of no statistical difference.IgA/C3 were (3.041 ±0.508 5),significantly higher(P <0.05).There was a positive correlation between IL-8 and IgA/C3 (r =0.534,P <0.05 ).Conclusion The children of HSP has cytokine and immunoglobulin disturbance,and presents a hyperreactivity in both humoral immunity and cellular immunity. Positive correlation between IL-8 and IgA/C3 was observed.
10.Clinical application and Fuzzy evaluation of autologous platelet-rich plasma mixed with fat graft in facial tissue depression

Objective To evaluate the clinical effect of the facial soft tissue defect treated with platelet-rich plasma (PRP) mixed with purified fat.Methods 35 patients with facial soft-tissue defects were treated by PRP mixed with centrifuged fat tissue.Using Fuzzy judgment method the clinical effectiveness of therapeutic outcomes were evaluated on fullness,injection of frequency and satisfaction during 3 to 6 months of follow-up.Results The therapeutic outcomes showed that the reconstructed face became a relative symmetrical and natural appearance with not evidently fat resorption and stable after operation.Both patients and surgeons were satisfied with the results.The score of effectiveness was highest in Fuzzy judgment.Conclusions The efficacy of PRP is significant in enhancing fat graft,reducing fat necrosis and resorption.The fat tissue mixed with PRP is a reliable reconstruction option for the facial soft tissue defect.
