Objective To explore the evaluation value of forkhead box M1(FOXM1)and CC chemokine receptor 5(CCR5)on lung function and prognosis in elderly patients with acute exacerbation of chronic obstructive pulmonary disease(AECOPD).Methods A total of 128 AECOPD patients admitted to Yueyang People's Hospital from January 2022 to January 2023 were collected as the acute exacerbation group,135 stable COPD patients admitted at the same time were regarded as the stable phase group,and 120 health examination volunteers of similar age and gender were regarded as the control group.Enzyme-linked immunosorbent assay(ELISA)was applied to detect the expression levels of FOXM1 and CCR5 in serum,and the vital capacity meter was applied to measure pulmonary function.Pearson method was applied to analyze the correlation between serum FOXM1,CCR5 and lung function indicators in AECOPD patients.Multivariate logistic regression analysis was applied to screen prognostic factors for AECOPD patients.The evaluation value of FOXM1 and CCR5 levels in evaluating poor prognosis of AECOPD patients was analyzed by receiver operating characteristic(ROC)curve.Results In the serum of patients in the AE COPD group and stable stage group,the levels of CCR5(49.36±12.31 ng/ml,34.25±8.87 ng/ml)and FOXM1(40.21±10.74 pg/ml,23.38±5.77 pg/ml)were significantly higher than those in the control group(14.55±4.58 ng/ml,15.06±3.55 pg/ml),while FEV1(1.15±0.13 L,1.67±0.19 L),FVC(2.93±0.30 L,3.28±0.36 L)and FEV1/FVC(39.25％±3.97％,50.91％±5.01％)were lower than those in the control group(1.95±0.26 L,3.57±0.44 L,54.62％±5.20％),with significant differences(F=11 1.034～641.907,all P＜0.05).The levels of serum FOXM1 and CCR5 were gradually increased with the aggravation of lung function grading,while the levels of lung function indicators FEV1 and FVC were gradually decreased with the aggravation of lung function grading,and the differences were statistically significant(F=31.27,49.37;42.72,29.48,all P＜0.05).The serum levels of FOXM1 and CCR5 were negatively correlated with FEV1,FVC and FEV1/FVC(r=-0.639～-0.479,all P＜0.05).Logistic regression analysis found that CCR5[OR(95％CI):3.380(1.944～5.876)],FOXM1[OR(95％CI):5.711(3.175～10.273)],APACHE Ⅱ score[OR(95％CI):2.132(1.243～3.660)],and lung function grading[OR(95CI):2.017(1.007～4.037)]were all risk factors for poor prognosis(all P＜0.05),while FEV1[OR(95％CI):0.649(0.441～0.955)]and FVC[OR(95％CI):0.120(0.073～0.198)]were protective factors for poor prognosis(all P＜0.05).ROC curve results showed that the areas under the curve(AUC)of serum FOXM1 and CCR5 levels in predicting poor prognosis in AECOPD patients were 0.821 and 0.831,respectively.The AUC predicted by the combination of the two was 0.895,which was higher than that detected by a single indicator(Z=2.800,2.654,all P＜0.05).Conclusion FOXM1 and CCR5 were both high levels in the serum of AECOPD patients.Early detection of them can serve as serum markers for evaluating lung function and poor prognosis in AECOPD patients.

Objective:To establish a nomogram model for predicting positive resection margins after prostate cancer surgery, and to perform the corresponding verification, in order to predict the risk of positive resection margins after surgery.Methods:A total of 2 215 prostate cancer patients from The First Affiliated Hospital of Naval Medical University, Hospital, Peking University First Hospital, Peking University Third Hospital, Peking University, and First Affiliated Hospital of Xi′an Jiaotong University were included in the PC-follow database from 2015 to 2018, and a simple random sampling method was used. They were divided into 1 770 patients in the modeling group and 445 patients in the verification group. In the modeling group, the age (<60 years, 60 to 70 years, >70 years), PSA (<4 ng/ml, 4-10 ng/ml, 11-20 ng/ml, >20 ng/ml), pelvic MRI (negative, suspicious, positive), clinical stage of the tumor (T 1-T 2, ≥T 3), percentage of positive needles (≤33%, 34%-66%, >66%), Gleason score of biopsy pathology (≤6 points, 7 points, ≥8 points). Univariate and multivariate logistic analysis were performed to screen meaningful indicators to construct a nomogram model. The model was used for validation in the validation group. Results:The results of multivariate analysis showed that preoperative PSA level ( OR=2.046, 95% CI 1.022 to 4.251, P=0.009), percentage of puncture positive needles ( OR=1.502, 95% CI 1.136 to 1.978, P=0.002), Gleason score of puncture pathology ( OR=1.568, 95% CI 1.063 to 2.313, P=0.028), pelvic MRI were correlated ( OR=1.525, 95% CI 1.160 to 2.005, P=0.033). Establish a nomogram model for independent predictors of positive margin of prostate cancer. The area under the receiver operating characteristic (ROC) curve of the validation group is 0.776. The area under the ROC curve of the preoperative PSA level, percentage of puncture positive needles, puncture pathology Gleason score, pelvic MRI, postoperative pathology Gleason score were 0.554, 0.615, 0.556, 0.522, and 0.560, respectively. The difference between the nomogram model and other indicators was statistically significant ( P<0.05). Conclusions:The constructed nomogram model has higher diagnostic value than the preoperative PSA level, percentage of puncture positive needles, Gleason score of puncturing pathology, pelvic MRI, and postoperative pathological Gleason score in predicting positive margin.

Objective:To investigate the risk factors for Gleason score upgrading after radical prostatectomy in clinical low-risk prostate cancer patients aged≥65 years.Methods:A total of 485 clinical low-risk prostate cancer patients aged≥65 years at five centers of the national multi-center PC-follow database from January 2015 to March 2019 were retrospectively analyzed.Data including age at diagnosis, prostate-specific antigen(PSA), MRI prostate imaging, puncture Gleason score, operation method, puncture method, positive incision margin and capsule penetration were collected.Differences in Gleason scores before and after operation were compared, and the risk factors for Gleason score upgrading after radical resection were evaluated by univariate and multivariate Logistic regression analysis.Results:Of 485 patients with a puncture Gleason score of 3+ 3=6, 261(53.8%)cases had postoperative pathological upgrading, in whom 228(87.4%)cases had Gleason score upgrading of 7, 22(8.4%)had Gleason score upgrading of 8, and 11(4.2%)had Gleason score upgrading of 9 or more.The rate of Gleason score upgrading was elevated with increased preoperative PSA levels, positive pelvic MRI, and higher positive rates of puncture biopsy.The incidences of postoperative capsule penetration(27.2% vs.12.5%, P<0.001)and positive incision margin(25.2% vs.17.4%, P=0.036)had statistically significant differences between the pathologically upgraded group and the pathologically non-upgraded group.Multivariate analysis showed that preoperative PSA level, percentage of positive puncture biopsies, biopsy Gleason score and pelvic MRI were independent predictors of prostate cancer. Conclusions:For clinical low-risk prostate cancer patients aged≥65 years with high risk factors for Gleason score upgrading, repeated biopsies should be carried out when necessary and the treatment plan should be adjusted accordingly.

Production internship is an important teaching tache for undergraduate students to carry out engineering training by using professional skills, and it is a key starting point for fostering application-oriented talents in biotechnology. The Course Group of 'production internship of biotechnology majors' of Binzhou University is investigating application-oriented transformation for local regular colleges and universities, as well as fostering high-level application-oriented talents. By taking green fluorescent protein (GFP) polyclonal antibody as an example, the reform and practice on teaching content, teaching mode, assessment method, continuous improvement of curriculum were carried out. Moreover, the characteristics of the Yellow River Delta-Binzhou Biotechnology & Pharmaceutical Industrial Cluster were taken into account to intensify academic-enterprise cooperation. On one hand, this Course Group designed and rearranged the course contents, carried out essential training through online resources and platforms such as virtual simulation, and recorded, tracked and monitored the progress of production internship through practical testing and software platforms like 'Alumni State'. On the other hand, this Course Group established a practice-and application-oriented assessment method in the process of production internship and a dual evaluation model for continuous improvement. These reform and practices have promoted the training of application-oriented talents in biotechnology, and may serve as a reference for similar courses.
