To study the protective effects of PCF on thymocytes in the mice damaged by ultraviolet ray in vitro,The irradiation damage model of thymocytes was established.The indices of GSH-Px,SOD,MDA,T-AOC and the cell viability were determined by biochemical and MTT methods respectively.The mitochondria membrane potential was tested using flow cytometry(FCM).The results showed that PCF could enhance the activities of GSH-Px,SOD and increase the values of A-SAC and T-AOC in cells,while decrease the amounts of ROS,MDA. In addition,PCF could stabilize the mitochondria membrane potential.These results indicated that PCF had the protective effects on the thymocytes damage by UV irradiation,The mechanisms of the effects might be related to the elimination of oxygen free radical and increase of the activities of anti-oxidative enzymes in cells.

Objective To study the synergistic antitumor effect of PCF when combine used with CTX.Methods Inoculated 0.2 mL H22 tumor fluid into the right armpit of mouse,and then the animals were divided into seven groups randomly: control group;CTX group(CTX 10 mg?kg-1);three test group(PCF 1000,500 and 250 mg?kg-1) and two combined use groups (PCF 1000,500 mg?kg-1with CTX 10 mg?kg-1).PCF were given by intragastric administration for 10 days,and CTX by intraperitoneal injection for 10 days.Same volume of saline was given to the control group.The mice were killed 24 hours after the last medication and the tumor inhibition rate was calculated.Results Three PCF groups had no effect on tumor inhibition,while two combined treatusement groups showed significant effect on tumor inhibition,while at the same doses the tumor inhibition rates were raised to 55.9% and 52.9% respectively,which were higher than that of CTX when used alone.Conclusion PCF can enhance the antitumor effects of CTX.

Objective To assess whether mTOR was activated in peripheral T lymphocytes of acute coronary syndrome (ACS)patients and to investigate the relationship among mTOR,regulatory T cells and cytokins. Methods Patients with acute coronary syndrome (n=32) were selected in ACS group. Meanwhile, patients who complainted of chest pain but were proven to be normal in ECG and in coronary arteriography, were excluded as ACS patients but were diagnosed as Chest Pain Syndrome (CPS) and selected in CPS group (n=28). The expression levels of mTOR, phospho-p70S6KT389 (indicative of mTORC1 activity) and phospho-AKTS473(indicative of mTORC2 activity) were investigated in T cells, which were isolated from peripheral blood of patients in ACS group or CPS group, using Western blot. The proportion of CD4+CD25+Foxp3+Tregs over CD4+cells were evaluated by FACS. And T cell subset related cytokines such as IFN-γ, IL-4, IL-17 and TGF-β1 were exam?ined by ELISA. Then we investigated the relationship among mTOR,regulatory T cells and cytokins by Spearman analysis. Results mTOR and p-p70S6KT389 expression were significantly enhanced in ACS group as compared with those in patients from CPS group (P<0.01). Higher levels of IFN-γ, IL-17 but lower level of TGF-β1 cytokines as well as decreased propor?tion of Tregs were observed in ACS group than those in CPS group (P<0.05). There was no significant difference in the level of IL-4 and p-AKTS473 between the two groups (P>0.05). By correlation analysis, p-p70S6KT389 expression level positively correlated with IFN-γ, IL-17(rs=0.91,092,P<0.01)and negatively correlated with Tregs and TGF-β1(rs=-0.85,-0.80, re?spectively, P<0.01). Conclusion mTORC1 pathway was activated in peripheral T lymphocytes of ACS patients, and Tregs insufficiency and cytokins imbalance both contribute to the activation of mTORC1 pathway and pathological process of ACS.

Objective: To research the activity of antibacterial proteins isolated from Musca domestica during larvae-pupae metamorphosis. Methods: The 5-day-old larvae of Musca domestica were injured with a needle. 24h later, the larvae which had changed into pupae were selected. The antibacterial protein was isolated by a four-step protocol including grinding, heating, CM-sepharose F. F. cationexchange chromatography and another CM-sepharose F. F. chromatography. Antibacterial activities of samples were tested by an ultrasensitive radial diffusion assay using Staphylococcus aureus as the indicator organism. The molecular weight of the isolated protein was determined by SDS-PAGE.Results: The molecular weight of this isolated protein was 43kDa, and its antibacterial activity was strong. Conclusions: A kind of protein with strong antibacterial activity can be produced in the Musca domestica during larvae-pupae metamorphosis.

Objective To obtain two-dimensional gel electrophoresis maps of the renal tissue proteins of normal and chronic intermittent hypoxia(CIH) rats for identifying the diferentially expressed proteins in the CIH rats.MethodsCIH rat models were established, and the proteins in the renal tissue underwent two-dimensional gel electrophoresis with immobiline pH gradientisoeleclricfocusingasthefirstdimensionandverticalSDS-PAGEasthesecond dimension.Analysis of 2-DE maps was used to determine differential expression of proteins between the two groups by ImageMaster 2D Platinum 5.0, and four protein spots expressed differently were picked up for further identification by MALDI-TOF-MS.Results Matched and compared with those of control group, 112 protein spots were determined with differently expressive levels in CIH group.By MALDI-TOF-MS, three protein spots of them were identified as ATP synthase delta subunit of mitochondrial precursor, catechol O-methyltransferase, apurinic/apyrimidinic endonudease.Conclusions There is obvious difference in expressive proteomes in renal tissue between normal and chronic intermittent hypoxia rats.The functions of those identified proteins are involved in cellular energy metabolism, apoptosis, signal transduction, anti-cell injury and hormone metabolism,so that proteomics can serve as a new approach in the study of obstructive sleep apnea-hypopnea syndrome to discover new therapeutic targets.

Objective To investigate whether mTOR/p70S6K signal was activated in peripheral T lymphocytes of the coronary heart disease (CHD) patients and to investigate its possible role in the pathogenesis of coronary atherosclerosis. Methods The mRNA expressions of mTOR andp70S6K were detected by RT-PCR and protein expressions of mTOR, p70S6K and phosphorylated-p70S6K (p-p70S6K) were detected by Western blot assay in T cells isolated from peripheral blood of the CHD patients (including AMI, UAP, SAP) and the CPS patients. Results Levels of mTOR and p-p70S6K were significantly enhanced in the AMI and UAP groups as compared with those in the SAP and CPS groups (P < 0.05, respectively). The mRNA expressions of mTOR and p70S6K were significantly increased in the AMI and UAP groups as compared with those in the SAP and CPS groups (P < 0.05). Conclusion The mTOR/p70S6K pathway is activated in the peripheral T lymphocytes of CHD patients with AMI and UAP , which may participate in the pathological process of coronary atherosclerosis.

Objective To explore an efficient way to modulate the expression of estrogen receptor (ER) ? and ?, and to build up a model of endometrial cancer cell expressing predominantly one isoform of ER and to verify the roles of ER ? and ? in the tumorigenesis of endometrial cancer associated with estrogen and tamoxifen (TAM). Methods A series of oligodeoxyribonucleotides (ODN) against ? or ? regions of ER ? or ? were synthesized and tested in human endometrial cancer cell lines (Ishikawa) that express functional ER ? and ?. The expressions of two ER isoforms were detected by western blot using specific antibodies. Then we studied the change of Ishikawa proliferation in response to 17?-estradiol and TAM under the influence of antisense ODN. Results (1) Transfection with antisense ODN directed against the ER? and ER? could significantly inhibit target protein production. (2) 17?-estradiol could increase the proliferation of Ishikawa cells, but they lost the ability to proliferate in response to 17?-estradiol after transfected with ER? antisense ODN especially at hours 24, 48 and 72 ( P

Objective In order to offer a noninvasive and objective examination method for assessing the clinical diagnosis and eurative criterion of concussion patients, the patients color three - dimensional transcranial doppler( 3D - TCD) characteristic at deferent phases, parameter and clincal manifestation were studied Method Parameter of Median Cerebral Artery(MCA), Base Artery(BA), the average peak forwand velocity of cerebral blood flow, train figure were tested by 3D - TCD within 24 hours, 3 - 6 days and at the end of treatment, respectively. The results were compared with clinical diagnoses and curative criterion. Result Among 135 patients who had clinically diagnosed concussion, agreeable diagnoses 101 cases, basically agreeable diagnooses 12 cases, disagreeable diagnosis 22 cases(16. 2%) .At the end of treatment, according to clinical determination 96 cases(71.1%) were cured, 39 cases(28.9%) were improved. Wave train character, figure and diagnoses prameter of cerebral spasm caused by concussion were suggested Conclusion To be one of the diagnostic bases and curative criterion, 3D- TC D technology is performable. Objective To investigate the early nursing intervention on recovery of upper limb edema after radical mastectomy, and subjective comfort of patients and satisfaction degree with the service quality of nursing staff. Methods 70 patients according to different methods of nursing intervention were randomly divided into the control group and the observation group with 35 patients in each group. The observation group was given comprehensive care, including psychological care, raising affected limb, strengthen the training arm function, conducting various forms of aerobic motion, pressure gloves and massage, hot packs, physical therapy, the use of elastic bandages, guidance and other aspects of hospital care. The control group only received routine care. The nursing effect, subjective comfort degree, satisfaction degree with the service quality, time of edema alleviation and disappearance and hospitalization time of two groups were compared. Results Compared with the control group, nursing effect of the observation group was better,subjective comfort of patients and the satisfaction degree with the quality of service significantly improved,and the time of edema alleviation and hospitalization time was significantly shorter, the difference was statistically significant. Conclusions Early implementation of nursing interventions for patients with limb swelling after radical operation of breast cancer can improve patients satisfaction degree, promote the recovery of upper limb edema, reduce hospital stay.
