1.Therapeutic effect of telmisartan combined nifedipine in patients with obese and hypertension

Objective:To explore the therapeutic effect of telmisartan combined nifedipine in patients with obese and hypertension and its influence on patients′blood lipid level.Methods:A total of 180 patients with obese and hyper-tension,who hospitalized in our hospital from Jan 2011 to Dec 2012,were enrolled as subjects.According to random number table method,they were equally divided into nifedipine group and nifedipine combined telmisartan group (combined medication group).Therapeutic effects,changes of levels of blood lipids etc.were observed in two groups.Results:After six-week treatment,compared with nifedipine group,total effective rate of combined medi-cation group significantly rose (75.56%vs.93.33%,P <0.05);the levels of total cholesterol [TC,(5.20±0.59) mmol/L vs.(4.75±0.73)mmol/L],triglyceride [TG,(1.87±0.49)mmol/L vs.(1.67±0.48)mmol/L]and blood pressure [(138±9)/ (90±6)mmHg vs.(125±9)/ (81 ±5)mmHg]significantly reduced in combined medication group,P <0.05 all.Conclusion:Telmisartan combined nifedipine possess significant therapeutic effect on obese and hypertension,it also can lower blood lipid levels.
2.Effect of Drinking Water Contained Low Level Calcium and High Level Magnesium on Bone Metabolism of Ovariectomized Rats

Objective To study the influence of excessive magnesium and/or deficient calcium intake on the bone mass of ovariectomized rats. Methods Three-month-old ovariectomized rats were fed with different types of drinking water: Interaction group was fed with deficient-Ca and excessive-Mg water: Ca 5 mg/L, Mg 60 mg/L; Excessive-Mg group was Ca 60 mg/L, Mg 60 mg/L; Deficient-Ca group was Ca 5 mg/L, Mg 30 mg/L; Normal group was Ca 60 mg/L, Mg 30 mg/L. After 3 months of treatment, the left femur was measured with the length and weight, then the content of Ca and Mg were determined, right femur was X-rayed, and right tibia was taken for the pathological examination. Results Calcium deficiency in water caused significant changes in all of the bone mass indexes, magnesium excess in water showed a tendency of Ca decreasing and Mg increasing in the bone. It was easily distinguish the difference in bone mineral and bone microstructure by X-ray and pathological examination: control group was the best, then was Mg excess group, Ca deficiency group was inferior, and the interaction group was the worst. Conclusion Calcium deficiency is one of the major reasons of osteoporosis, while the excessive intake of magnesium may not only disturb the bone metabolism but also aggravate the adverse effect of Ca deficiency on bone mineral and bone health.
3.Effects of estrogen on the cognitive function,the expression of Bcl-2 and Bax proteins and the neuroal apoptosis in hippocampus in rats with vascular dementia

Objective To explore the effects of estrogen on the cognitive function,the expression of Bcl-2 and Bax proteins and the neuroal apoptosis in hippocampus in rats with vascular dementia(VD).Methods 60 male Wistar rats were randomly divided into sham-operated group,VD group and estrogen group,20 rats in each group.VD models were made with permanent bilateral common carotid arteries occlusion.The 17-?estrogen (200 ?g/kg,dissolution in peanut oil ) was intraperitoneal injected in estrogen group,once in other day,for 30 times.While,same-dose peanut oil was intraperitoneal injected in the sham-operated group and VD group once in other day,total 30 times.After 60 d the capability of learning and memory were observed by Y-maze test.Neuroal morphology and the expression of Bcl-2 and Bax proteins in hippocampal CA1 subregion were detected by HE and immunohistochemistry stain respectively;neuroal apoptosis was measured by dUTP nick-end labeling(TUNEL).Results Comparing to sham-operated group,cognitive ability in the VD group and estrogen group were significently decreased;both the expressions of Bcl-2 and Bax proteins were increased in hippocampal CA1 subregion;the number of TUNEL positive cells was obviously increased (allP

5.Expression of Thymus and Activation-regulated Chemokine in die Kidney of MRL/lpr Mice

Objective To study the expression of thymus and activation-regulated chemokine(TARC)in the kidney of MRL/lpr mice and to elucidate the molecular pathogenesis of lupus nephritis.Methods The 24-hour urine protein of 4 MRL/lpr mice of 16 weeks of age and 4 age-and sex-matched BALB/c mice was measured.The expressions of TARC mRNA and protein in the kidneys of MRL/lpr mice and BALB/c mice were detected by reverse transcription polymerase chain reaction and Western blot,respectively.Results The 24-hour urine protein and the expression level of TARC mRNA were significantly higher in MRL/lpr mice than in BALB/c mice(P<0.05).The expression of TARC protein was detectable only in MRL/lpr mice.Conclusion The expression of TARC increases in the kidney of mice with systemic lupus erythematosus,suggesting that TARC may be involved in lupus nephritis in mice.

7.The relationship between cell apoptosis and Phospho-Ser~(727) Stat1 expression in the rat with experimental diffuse axonal injury

Objective To investigate the role of Stat1 in pathological process of nerve cells apoptosis induced by diffuse axonal injury (DAI) on rats. MethodsThe DAI model was established by using an injury model adapted from Marmarou et al. in 1994. All animals were divided into three groups, including control group, mock group and test group sacrificed on 6, 12, 24, 48, 72, 120 and 240 hours post injury (hpi). The paraffin-embedded sections of brain tissue were processed for HE staining and Bielschowsky’s silver method. Cell apoptosis was examined by flow cytometry and the expression of bax and bcl-2 were analyzed by RT-PCR. And Phospho-Ser727 Stat1 expression was examined by immunohistochemistry in different brain regions. ResultsThere was no brain contusion within HE staining, however, waving and enlargement of axons were observed within Bielschowsky’s silver method. The apoptotic rate of brain cells as well as PCR products ratio of bax to bcl-2 was highest at 24 hpi and decreased with time. An up-regulation of Phospho-Ser727 Stat1 at 6 hpi was discernible, and then reached the top at 24 hpi in cortex, cerebellum, brain stem and corpus callosum, and at 12 hpi in hippocampus. This increase was associated with the nerve cells apoptosis, r=0.921. In addition, the Phospho-Ser727 Stat1 positive cells were neurons and glial cells assessed from morphous. ConclusionsOur data indicate that Stat1 may contribute to the apoptosis of DAI on rats. In addition, the expression of Phospho-Ser727 Stat1 in glial cells suggested that glial cells may play an important role in the pathogenic mechanism of DAI.
9.Expression of lymphokine-1 and clinical significance of p53 protein mutation in hilar cholangiocarcinoma

Objective:To analyze the expression and clinical significance of lymphoid enhancer factor-1 (LEF-1) and P53 protein in hilar cholangiocarcinoma.Methods:A total of 50 cases with hilarcholangiocarcinoma in the Tangshan People′s Hospital from March 2010 to December 2017 were retrospectively analyzed.P53 protein mutation was detected by immunohistochemistry.At the same time, the expression of LEF1 protein in cholangiocarcinoma and adjacent normal tissues was detected.The expression of LEF1 and its correlation with clinicopathological parameters were analyzed.At the same time, the relationship between p53 protein mutation and LEF1 protein expression and the prognosis of patients were discussed.Results:The positive expression rate of LEF1 in tumor tissues were 62.00%(31/50), which was higher than in adjacent tissues 36.00% (18/50). The difference was statistically significant (χ 2=6.763, P=0.016). There were 28 patients with TP53 protein mutation and 22 patients with wild type.The positive rate of LEF1 in TP53 mutant group was 75.00% (21/28), which was significantly higher than that in TP53 wild type group(45.45%, 10/22), and the difference was statistically significant(χ 2=4.565, P=0.03). The LEF1 expression was associated with tumor differentiation, TNM stage, lymph node metastasis and TP53 protein mutation (all P<0.05). LEF-1 expression was positively correlated with TP53 protein mutation ( r=0.294, P=0.04). The 1-year cumulative survival rate of patients with TP53 protein mutation was significantly lower than that of patients with TP53 protein wild type (60% vs.81%, P=0.0416). The 1-year cumulative survival rate of patients with LEF1 positive expression was also significantly lower than that of patients with negative expression (53% vs.82%, P=0.0180). Conclusion:LEF-1 is highly expressed in hilar cholangiocarcinoma, and patients with high expression have poor prognosis.The positive expression of LEF-1 in hilar cholangiocarcinoma patients with TP53 protein mutation was increased, and the prognosis of patients with TP53 protein mutation was poor.
10.Effect of combination nursing intervention on quality of life in cancer patients undergoing chemotherapy

Objective To explore the effect of health education and comprehensive nursing intervention on quality of patients with lung cancer.Methods One hundred lung cancer patients who were treated with chemotherapy from February 2010 to February 2013 in our hospital were selected and assigned to the intervention group (n=50) and the control group (n=50).The participants in the control group received conventional anti-cancer drug and routine primary care,however those of the intervention group additionally obtained health education and comprehensive nursing intervention.The Zung Anxiety Scale (SAS),Depression Rating Scale (SDS) and the core quality of life questionnaire (QLQ-C30) were used to evaluate the quality of life of the patients.Paired samples t test and ANOVA were used for data analysis.Results Differences of age,gender,disease duration,SAS,SDS score and EORTC QLQ-C30 score were not statistically significant between the two groups (all P>0.05).After the intervention,SAS and SDS scores of the control subjects were 58.8±7.6 and 49.5±10.1 respectively,and SAS and SDS scores of the intervention group were 46.5±7.8 and 37.4±9.2 respectively (t velues were 5.347 and 6.215,respectively; both P<0.05).Quality of life scores in the intervention group were as followed:general condition 57.2± 18.3,physical fitness 63.0±25.9,mutual affection 67.3±23.3,cognitive score 75.4±28.2,social role 67.7±24.8 and function scores 58.5±22.3.Quality of life scores in the control group were as followed:general condition 47.3±23.0,physical fitness 54.9±26.6,mutual affection 60.4±19.1,cognitive score 66.2±25.8,social role 54.1±26.2 and function scores 49.4±21.5 (t values were 5.725,6.144,5.021,5.702,7.137 and 6.357,respectively; all P< 0.05).Conclusion Health education and comprehensive nursing intervention for lung cancer patients may reduce their anxiety and depression symptoms and improve the quality of life.
