Objective:To investigate the local application of exogenous VEGF on survival of rats random flaps after radiotherapy.Methods:45 four-month old Wistar rats were taken as experimental objects and divided into 3 groups randomly to construct animal models of random flaps and simulated radiotherapy with 60Co radioactive source with 6 Gy,500 cGy/min.All the rats were killed 28 days after radiotherapy were carried out.The vitality of flaps was observed 7 days and 28 days after radiotherapy.Flaps were evaluated with cytological staining experiments,histopathological experiments and SDH measurements.Results:The group which applied VEGF was greatly distinguished from control groups(P

Interleukin-17 (IL-17) is a newly discovered proinflammatory cytokine in recent years. It has wide biological activities; therefore it may be one of the important factors in the occurrence and development of certain diseases. The article mainly reviews the current situation of the study on the related roles of IL-17 in nervous system diseases.

Objective To study the function of γδT lymphocytes in patients with lung cancer.Methods γδT lymphocytes in peripheral blood of the 124 preoperative patients with lung cancer(experiment group) and 56 healthy donors(control group) were detected by Flow Cytometry. Results The percentage of γδT cells(CD+3 γδTCR+) was (6.54±2.90) % in control group and (3.76±2.81) % in experiment group (t =3.568,P ＜0.001). The γδT cells in peripheral blood of patients with lung cancer was less than that of healthy donors (t =3.568, P ＜0.001). There was significant difference in expression of γδT cells in the peripheral blood between the all test groups and healthy control group (P =0.000). Compared with control group, γδT cells of the peripheral blood in experiment group were correlated with TNM stage, general types, pathological changes position and histology grades (P ＜0.05). The expression of γδT cells in the peripheral blood were not associated with age, gender (P ＞0.05). Conclusion The results suggested that the dysfunction of γδT had important function on the development of lung cancer.

Objective To evaluate the changes and clinical significance ot the serum levels of homocysteine (Hcy) in gastric cardia cancer patients before and after surgery.Methods Serum Hcy concentrations of 102 patients with gastric cardia cancer (including 69 case of males) and 50 healthy human were measured by enzymatic cycling assay.Results Total Hcy levels were significantly increased in male patient group compared with the levels in control group (t =5.143,P =0.001).Hcy levels were significantly lower in postoperative group compared with preoperative group [(17.08±5.31) μmol/L vs (20.34±9.26) μmol/L,(14.07±4.87) μmol/L vs (20.34±9.26) μmol/L,P ＜ 0.05].Compared with Ⅳ stage group and other TNM stage groups,significantly lower levels of Hcy were detected in patients with gastric cardia cancer (t =2.306,3.285,P =0.030,0.002).Hcy levels in patients with gastric cardia cancer were also significantly higher than those in the tumor length ＜ 3 cm,3-5 cm and ＞ 5 cm groups (t =2.461,2.147,P =0.017,0.038).Multiple logistic regression analysis indicated a statistically significant association between serum Hcy concentration and gastric cardia cancer incidence (OR =1.136,95 ％ CI 1.010-1.278,P =0.033).Increasing serum Hcy levels were significantly associated with a decreasing risk of metastatic lymph node (OR =0.865,P =0.010).Conclusion Serum Hcy levels are directly associated with risk of male patients with gastric cardia cancer,and play important roles in the development of gastric cardia cancer.

Objective To determine the relationship between zinc and nutritional status or immunity in patients with end-stage renal disease undergoing continuous ambulatory peritoneal dialysis (CAPD). Methods Forty-five stable CAPD patients undergoing peritoneal dialysis were enrolled in this study. The dietary zinc intake and serum zinc levels were investigated, and the results were compared with the gender- and age-matched healthy populations (n = 45). The relationship between dietary zinc intake and serum zinc levels and subjective global assessment (SGA) score, blood cell counts, serum albumin (ALB), prealbumin (PA), C-reactive protein (CRP),and lymphocyte subsets were analyzed. Results The percentages of inadequate dietary zinc intake (P= 0. 007)and low blood zinc (P = 0. 036) were significantly higher in CAPD patients than in healthy group. CD8 levels were significantly lower (P = 0. 000) while CD4/CD8 ratio was significantly higher (P = 0. 033) in CAPD patients than in healthy group. In CAPD patients, correlation analysis showed that dietary zinc intake was significantly correlated with serum prealbumin levels (r = 0. 577, P = 0. 000), but was negatively correlated with SGA score (r = - 0.354,P = 0. 015) and CRP (r = - 0.354, P = 0. 015); however, it was not significantly correlated with lymphocyte subsets and CD4/CD8 ratio. Serum zinc level was significantly positively correlated with hemoglobin (r= 0. 411, P=0.005), hematocrit (r=0.345, P=0.023), WBC (r=0.318, P=0.035), SGA score (r=0.417, P=0. 005), and CRP (r=0.342, P = 0. 027), but was not significantly correlated with lymphocyte subsets and CD4/CD8 ratio. Conclusions Zinc deficiency is common among patients with CAPD. Adequate dietary zinc intake can facilitate protein synthesis and improve the overall nutritional status. High serum zinc concentrations are beneficial for the synthesis of hemoglobin and the improvement of anemia.

Objective To observe the effect of nicotine(NIC) on the activation and resultant death of microglia induced by LPS. Methods The animal model that exposed to chronic nicotine treatment was established and LPS was injected intraperitoneally to induce the activation of microglia. Furthermore, the CD11b-positive microglia in cerebral cortex, hippocampal and substantia ngra were observed through immunohistochemical staining. BV2 cells(Microglial cell line of mouse) were subcultured, simultaneously the following kits were used including CCK-8 kit assay for cell activity, Nitric oxide assay kit assay for NO release, RT-PCR assay for the iNOS,TNF-α,IL-1β,IL-6,COX-2,IRF-1,Caspase-11 mRNA expression, Western blotting assay for the protein expression of P-I-κB and Caspase-3. Results Nicotine suppressed the CD11b-positive microglia expression in cerebral cortex,hippocampal and substantia ngra induced by LPS;Nicotine inhibited the activation-induced cell death (AICD), attenuated NO release, reduced iNOS,TNF-α,IL-1β,IL-6,COX-2,IRF-1,Caspase-11 mRNA expression, decreased the protein expression including P-I-κB and Caspase-3 of BV2 cells. Conclusion Nicotine pretreatment can suppress the activation and resultant death of microglial cells induced by LPS, which suggests that nicotine may play a neuroprotective role on inflammatory reaction of brain.;

Objective To investigate the clinical features and the gene mutation of patients with spinocerebellar ataxia type3 and type7.Methods The trinucleotide repeat mutations were detected by polymerase chain reaction (PCR),agarose gel electrophoresis method,and DNA sequencing in 13 patients,4 related members and 4 common members from 3 spinocerebellar ataxia families.Results Among the 13 patients,four patients had SCA3/MJD(CAG) n expansion mutation(n = 65 ～ 74),nine patients had SCA7 allele expansion for 40 ～ 52 times.Patients with type 3 or 7 showed significant difference in nervous system injury.Conclusion The difference of clinical feature might be used in diagnosis of SCA3/MJD and SCA7,but genotype determination would be the only method of definite diagnosis.

Objective To observe the adipocytic differentiation potential of bone marrow stromal cells (BMS), and the effect of simvastatin on adipocytic differentiation of bone marrow stromal cells in vitro, and to elucidate the mechanisms of anabolic effect of simvastatin on bone formation. Methods BMS from femur and tibia of adult female BALB C mice were cultured in vitro. Changes of alkaline phosphatase (ALP) activity were determined after treatment with adipogenetic agonist (hydrocortisone 0 5 ?mol/L and indomethacin 60 ?mol/L, HI) for 6 days. Thenexpression of lipoprotein lipase (LPL) mRNA was detected by RT PCR after treatment with HI and different concentration of simvastatin for 72 h. Adipogenetic differentiation were also observed with Oil Red O staining and fluorescence activated cell sorting (FACS) after treatment with HI and different concentration of simvastatin or 100 ?g/L rhBMP 2 for 12 days. Objective: To observe the effect of simvastatin on osteoblastic cell differentiation of bone marrow stromal cells in vitro, and to elucidate the mechanisms of anabolic effect of simvastatin on bone formation. Methods: Bone marrow stromal cells from femur and tibia of adult female BALB C mice were cultured in vitro , after being treated with different concentrations of simvastatin for 72 h, changes of mRNA level of osteocalcin (OCN) were detected by RT PCR, change of OCN, and osteopontin (OPN) expression were examined by Western blot, and the changes of cellular alkaline phosphatase activity (ALP) were examined by histochemistry and enzymologic measurement. Results: After bone marrow stromal cells were treated with different concentration of simvastatin for 72 h, level of OCN mRNA increased, and expression of OCN and OPN also increased in a concentration dependent manner, and cellular ALP activity significantly increased in a concentration dependent manner. Conclusion: Simvastatin can stimulate osteoblastic differentiation,and improve cellular ALPase activity with high expression of osteocalcin and osteopontin in vitro. These may be parts of the mechanism of anabolic effect of simvastatin on bone formation.

Objective:To find an applicable condition of the single-plasma-sample method (SPSM) to measure the glomerular filtration(GFR) with 99m Tc-Diethylene Triamine Pentaacetic Acid ( 99m Tc- DTPA) , and predict the value of 99m Tc-DTPA plasma clearance by dual plasma sample method (DPSM ) from that by SPSM.Methods:Three hundred and thirty five patients with chronic kidney disease (CKD) were selected (192 males and 143 females). The average age was 51.91?14.76 years. The GFR was determined simultaneously by 2 methods: (1) SPSM (sGFR); (2) DPSM (tGFR), using DPSM as reference standard, sGFR calculated from the different SPSM was compared with tGFR. An equation was developed to predict tGFR from sGFR. GFR estimated by abbreviated Modification of Diet in Renal Disease (MDRD) equation (aGFR) was evaluated as the criterion in selection of DPSM and SPSM. The condition that DPSM could be substituted by SPSM in GFR measurement was given. Results: When tGFR ≥ 30 mL/ (min?1.73 m2), all of the sGFR were significantly correlated with tGFR.
