Journal of Chongqing Medical University 2007;0(10):-
Objective:To investigate whether PLC? gene down regulation by RNA interference(RNAi) leads to inhibition of proliferation in human bladder carcinoma T24 cell. Methods: The shRNA recombinant plasmids targeting to PLC? gene was constructed and transfected into bladder carcinoma T24 cell with Lipofectamine 2000.RT-PCR was used to monitor the validity of specific s h R N A in down regulation of PLC?.Then MTT assay was performed for detecting cell proliferation, the changes of PCNA were analyzed by immunocytochemical method,Electron microscope was used to observe the morphological changes. Results: The specific PLC? shRNA was confirmed to be efficient in silencing PLC? expression.PLC? gene down regulation by the shRNA recombinant plasmids inhibition cell proliferation rate about 78.01%,They were significantly different from that of control group(P
2.Proliferation inhibitory effect of crocin on transitional cell carcinoma of urinary bladder
Journal of Chongqing Medical University 2007;0(11):-
Objective:To investigate the proliferation inhibitory effect of crocin on transitional cell carcinoma of urinary bladde(rTCCB). Methods:MTT assay was used to evaluate the inhibitory effect of Crocin on the proliferation of T24 cells. Microscope was used to observe the morphological changes of the T24 cells. Flow cytometry was used to measure the cell cycle. T24 cells were inoculated in to BALB/c nude mice to establish bladder cancer model. After treatment with Crocin, the inhibitory effects were observed on the growth of tumors. Results:The proliferation of T24 cells were inhibited remarkably. Morphological changes of cells were observed. Flow cytometric profiles revealed that crocin led to the increase of the cells in G0/G1 phase and the decrease of cells in S phase and G2/M(P

4.Expression of cytokeratin 20 in exfoliated urothelial cells of bladder cancer
Chinese Journal of Urology 2001;0(04):-
Objective To study the expression of cytokeratin(CK)20 in exfoliated urothelial cells and to evaluate its clinical significance. Methods The expression of CK20 in exfoliated urothelial cells in 40 patients with bladder cancer and 20 subjects with normal urothelium was observed with immunohistochemical methods. Results The positive staining of CK20 in patients with bladder cancer was 52.5%, the expression of CK20 was negative in all controls, the positive rate of expression of CK20 combining with HE staining in patients with bladder cancer was 90%. The expression of CK20 was significantly higher in recurrent cases(80%) than in primary cases(43.3%). CK20 was not related to the tumor grade or stage. Conclusions The results suggested that the method had a higher specificity and therefore CK20 would be a good diagnostic marker.

Objective To evaluate urinary vascular endothelial growth factor (VEGF) in the diagnosis of bladder cancer. Methods VEGF in urine was measured by enzyme linked immunosorbent assay in 33 cases of transitional cell bladder carcinoma,10 benign urological conditions and in 10 normal individuals. Results The mean urinary VEGF level in 33 patients with bladder cancer was ( 309.8 ? 86.6 )ng/g Cr, significantly higher than in the 20 controls which was (107.6?35.4)ng/g Cr ( P

Objective To investigate the role of the cough reflex sensitivity and airway neurogenic inflammation in the pathogenesis of cough post infectious(CPI).Methods Capsaicin provocation test and sputum inducing were performed in 10 patients with CPI(CPI group)and 10 healthy subjects(normal control group)to detect the cough reflex sensitivity and neurokinin of sputum.Total and differential cells count of induced sputum was performed.Substance P(SP),neurokinin A(NKA),neurokinin B(NKB)and calcitonin gene-related peptide(CGRP)in the supernatant of induced sputum were measured. The expression of SP,neurokinin 1 receptor and NKA in sputum cells was detected. Results The cough threshold(lg C5)of CPI was significantly lower than normal(1.16?0.61 vs 2.64?0.28,P

Objective BOLD-fMRI was used to observe the activated auditory cortex evoked by pure tone stimuli in patients with sensorineural hearing loss and this paper is to discuss the objective measure for patient with sensorineural hearing toss. Methods BOLD-fMRI examinations were taken in 22 patients with unilateral moderate to severe sensorneural hearing loss and 15 control subjects. The volumes and intensities of the two hemispheres of the activated auditory cortex were analyzed quantitatively. Results Significant activation was found in the temporal lobe in control subjects, and significant differences in the volume and intensity were noted between the contralateral and ipsilateral activated auditory cortexes in them (P<0.01), exhibiting clearly eontralateral predominance. When the normal ear of patients with sensorineural hearing loss received signals, there was no significant difference be-tween contralateral and ipsilateral activated auditory cortexes (P>0.05). Conclusion When the normal ear of pa-tients with sensorneural hearing loss was stimulated by pure tone, the contralateral hemisphere predominance disap-peared. This result seems to show the plasticity of auditory cortex of patients with unilateral hearing loss.

Objective To investigate the severity of cough in patients with different etiology of chronic cough and its related factors. Methods One hundred and fifty patients with chronic cough were recruited. The diagnosis of the patients was as follows: 24 cases with post nasal drip syndrome (PNDs), 26 cough variant asthma (CVA), 20 gastroesophageal reflux-induced cough (GERC), 31 eosinophilic bronchitis (EB), 30 atopic cough (AC) and 19 cough post infection (CPI). The severity of cough at daytime and night-time were scored by the doctor and the patient at the first visit. The correlation between cough score and age, gender and course duration of patients was analyzed. Results There was no significantly difference of cough score at daytime among different groups. Cough score at night in CVA group was significantly higher than that of other five groups (all P <0.01). Regression analysis showed that cough score at daytime was significantly correlated with age of patients (P < 0.05), while cough score at night was correlated significantly with gender (P < 0.01). Conclusion Cough severity at daytime in different etiology of chronic cough is similar, while that at night in patient with CVA is higher than that with other etiology.
