1.SEN-V DNA found in liver tissue from patients with non A to E hepatitis in Guangzhou area

Objective To study the SEN-V DNA in liver tissue from patients with non A to E hepatitis in Guangzhou area. Methods A set of primers according to the original sequence from GenBank were designed , and SEN-V DNA sequence was amplified by in situ-PCR in liver tissue. Results Two of 6 patients with non A to E hepatitis were found to be SEN-V-DNA positive in liver tissue. While 8 patients with HBV infection and 6 of other liver diseases including 2 of autoimmune liver disease, 1 of Dubin Johnson syndrome, 1 of fatty liver, 1 of the primary biliary cirrhosis and 1 of drug induced hepatitis were all SEN-V DNA negative. SEN-V DNA were mainly distributed in liver cell cytoplasm, partially in nucleus. There presented the phenomenon of cell fuse. SEN-V positive cells accounted for about 3%~10% of total cells.Conclusions SEN-V can be found in liver tissue of patients with non A to E hepatitis in south China. Maybe it can cause liver damage.
2.Application value of adaptive iterative reconstruction algorithm in low dose pediatric cervical CT

Objective To explore the application of adaptive statistical iterative reconstruction (ASIR)scanning technique in low dose pediatric cervical CT, especially in reducing the radiation dose for thyroid. Methods We retrospectivly analyzed 140 pediatric cervical CT. 70 cases of group A scaned with ASIR technology and preseting noise index 12 HU in Discovery CT 750 HD (GE Healthcare), and 70 cases of group B scaned with filtered back projection (FBP) technology and preseting noise index 14 HU in VCT 64 (64 Lightspeed VCT, GE Healthcare). The other scan parameters were same in 2 groups. The distribution characteristics of gender and age were completely consistent between groups. The minimum current, the maximum current, the current of thyroid on CT images and CT dose volume index (CTDIvol), scanning length and dose length product (DLP) on dose reports were all recorded for each child. The noise values of CT images were measured. 4 scores standard was used to evaluate the subjective image quality. The data were analyzed with SPSS 13, Kappa method for the test of consistency and t test for the differences of the measured data. Excel was used for mapping. Results The minimum current, the maximum current, the average current, the current for thyroid of the two groups were respectively (26.8 ± 4.2), (43.2 ± 15.4)mA;(35.8±9.6),(41.8±13.6)mA;(34.6±8.6), (41.0±13.2)mA;(36.8±12.3), (44.7±21.0)mA. The CTDIvol, DLP and the scanning length of the two groups were respectively(1.3 ± 0.3), (1.8 ± 0.6)mGy;(29.0 ± 8.3), (40.3 ± 15.6) mGy·cm and(157.5±20.2), (157.6±21.8)cm. The average current, the thyroid current, CTDI and DLP in group A were 15.8%, 17.9%, 27.3%and 28.4%lower than those in group B (t=-5.50—-0.02, P<0.01). The subjective image quality scores of the two doctors were respectively 3.4 ± 0.9, 3.3 ± 1.0, they were highly consistent (Kappa=0.834, P<0.01). The objective image noise of cancellous bone and neck muscles were respectively (32.5 ± 5.3),(32.9 ± 5.1)HU and (9.2 ± 2.3), (9.1 ± 2.0)HU, there were not statistically difference between the two groups (t=-0.47—0.24, P<0.05). Conclusion Compared with FBP, low dose pediatric cervical CT with 30%ASIR could provide dose reduction for thyroid and the whole neck while maintaining diagnostically acceptable images .
3.Voxel based morphometric study of brain structure in patients with posttraumatic stress disorder.

Voxel based morphometry (VBM) methods are used to detect the difference in brain structures between the posttraumatic stress disorder (PTSD) sufferers and the normal controls. Standard VBM method can detect the difference of the gray matter or white matter densities while the optimized VBM method can detect the difference of gray matter or white matter volumes in the whole brain. The experiments showed that for the patient group, gray matter density or volumes significantly increased in the right frontal lobe, middle frontal gyrus, vermis, left caudate and parietal lobe, compared with the normal controls. However, in the left frontal lobe and middle frontal gyrus, gray matter density significantly decreased. There is no significant difference in white matter between the two groups. These results are consistent with those of the fMRI, which not only provide the evidence for further study of the pathogeny in PTSD but also validate the efficiency of the VBM methods for detecting the difference in the whole brain structure.

4.Experiences of students in the MOOC-based flipped classroom of Basic Nursing:a qualitative study

Objective To investigate students' experiences of learning achieved through the flipped classroom based on Massive Open Online Course(MOOC). Methods Qualitative study in which data were gathered from semi-structured and in-depth interviews conducted with 8 key students from the flipped classroom. The interviews were analyzed using Nancy′s phenomenology 7-step analysis method. Results Four themes emerged from the interviews: personalized learning, active learning, cooperative learning, and the critical role of teacher. Conclusions Overall, our research highlights the efficacy of MOOC-based flipped classroom. The implementation of the flipped classroom in this study improved personalized, active, and cooperative learning, and promoted the positive teacher-student relationship. Further research is needed to continue the investigation into maintaining students′ learning initiatives, controlling teach-learning progression,and strengthening formative evaluation.
5.Prediction of myometrial invasion of bladder cancer based on texture analysis of the bladder wall at tumor base

Objective:To identify the value of ultrasound radiomic features extracted from the bladder wall at tumor base in predicting myometrial invasion of bladder cancer.Methods:A total of 175 cases with bladder cancer confirmed by pathology from January 2017 to February 2022 in the First Affiliated Hospital of Guangxi Medical University were retrospectively analyzed. They were divided into training set and testing set in a ratio of 7∶3. The MaZda texture analysis software was used to draw the region of interest (ROI) of the bladder wall and the tumor region for extracting texture features. The minimum absolute reduction and variable selection operator (LASSO) regression and 10-fold cross-validation were used to screen the features of training set for establishing the models. And the ROC curve was used to evaluate the efficiency of the models.Results:A total of 279 texture features were extracted from the ROI of the bladder wall and the tumor region, and 5 texture features were screened out for constructing omics scoring models by LASSO regression and 10-fold cross-test. The area under ROC curve (AUC)s used in training set and testing set of the bladder wall were 0.921 and 0.856, while the AUCs applied in training set and testing set of the tumor region were 0.849 and 0.704. Both in the training set and test set, the AUCs of the model of the bladder wall were higher than those of the model of the tumor region (all P<0.05). Conclusions:The omics scoring model based on the texture features of the bladder wall at tumor base can effectively identify muscle-invasive bladder cancer(MIBC) and non-muscle-invasive bladder cancer(NMIBC), and has better performance than the model based on the texture feature of the tumor region.
6.Study on Relationship of CYP3A4,CYP2C8 and CYP3A5 Gene Polymorphism with ADR/Blood Concentration of Hydroxy- chloroquine in Patients with Autoimmune Disease

OBJECTIVE: To study the relationship of CYP3A4, CYP2C8 and CYP3A5 gene polymorphism with ADR/blood concentration of hydroxychloroquine in patients with autoimmune disease (AID), and to provide reference for individual medication of hydroxychloroquine. METHODS: Totally 77 AID patients,who were treated with hydroxychloroquine (daily dose of 200 mg to 400 mg) for a long-term (>6 months), were selected from the department of rheumatology and immunology, the First Affiliated Hospital of Anhui Medical University during Jul. 2017 to Aug. 2018. The information, blood sample and ADR of them were collected. Those patients were divided into normal liver function group, abnormal liver function group, normal renal function group, abnormal renal function group, normal eye group and abnormal eye group according to the site of ADR. The concentration of hydroxychloroquine was determined by HPLC. Genotype of CYP3A4, CYP2C8 and CYP3A5 were detected by MassARRAY microarray system. The differences of hydroxychloroquine-induced ADR in different genotypes were analyzed by χ2 test. The blood concentration difference of hydroxychloroquine in different genotypes were analyzed by independent sample t-test and one-way ANOVA. RESULTS: There was statistical significance in the distribution of CYP3A5 rs4646453 locus between normal renal function group and abnormal renal function group(P<0.05). The incidence of CC genotype was higher than that of AA+AC genotype in abnormal renal function group. There was statistical significance in the distribution of CYP2C8 rs10882526 locus between normal liver function group and abnormal liver function group(P<0.05). The incidence of allele G was higher than that of allele A in abnormal liver function group, and the incidence of AG genotype was higher than that of AA genotype. There was no significant correlation of the gene polymorphisms of CYP3A4, CYP2C8 and CYP3A5 with blood concentration among 77 AID patients. In subgroup analysis, blood concentration of GT, GG and TT genotypes of CYP2C8 rs10882521 in 58 patients with systemic lupus erythematosus (SLE) were 514.1,735.3 and 785.9 ng/mL, respectively; GG and TT genotypes were significantly higher than GT genotype (P<0.05). CONCLUSIONS: AID patients with CYP3A5 rs4646453 CC genotype have a higher incidence of renal dysfunction due to taking hydroxychloroquine; patients with CYP2C8 rs10882526 locus allele G and AG have a relatively high incidence of renal dysfunction due to taking hydroxychloroquine. When SLE patients taking the same dose of hydroxychloroquine, the blood concentration of hydroxychloroquine in patients carrying CYP2C8 rs10882521 GT genotype is lower than other genotypes.
7.Role of METTL3 in homocysteine-induced autophagy in mouse islet beta cells

BACKGROUND:Hyperhomocysteinemia is closely related to the function of islet β cells,but its specific molecular mechanism is not fully understood. OBJECTIVE:To investigate the role of N6 methyltransferase-like 3(METTL3)in homocysteine(Hcy)-induced autophagy of mouse islet β cells. METHODS:The 3rd and 4th generation mouse islet β cells were taken for the experiment.(1)Cell modeling and grouping:cells in control group were not treated with Hcy,while those in homocysteine group were treated with 100 μmol/L Hcy for 48 hours.(2)The mouse islet β-cells were transfected with the plasmids overexpressing Ad-METTL3 and si-METTL3 according to the instructions of LipofectamineTM 2000.Three different interfering fragments were designed,and the one with the best interfering efficiency was verified and screened by PCR.(3)After transfection,the cells were divided into control group,Hcy group,Ad-NC(negative control)+Hcy group,Ad-METTL3+Hcy group,si-NC(negative control)+Hcy group and si-METTL3+Hcy group.(4)qRT-PCR and western blot were used to detect the expression levels of METTL3 and autophagy-related proteins LC3Ⅱ/Ⅰ and p62 in cells.Insulin level was determined by ELISA to evaluate insulin secretion capacity of islet cells.Autophagy-related proteins and insulin level were detected after overexpression and interference with METTL3. RESULTS AND CONCLUSION:Compared with the control group,the expression level of LC3Ⅱ/Ⅰ was increased(P<0.05),the expression of p62 was significantly reduced(P<0.05),and the insulin secretion capacity was significantly decreased(P<0.05)in the Hcy group.Compared with the control group,the protein and mRNA levels of METTL3 were reduced in the Hcy group(P<0.05).After METTL3 silencing in islet β cells,Hcy further upregulated the expression of LC3Ⅱ/Ⅰ(P<0.05),significantly dowregulated the expression of p62(P<0.05),and increased the insulin level(P<0.05).After overexpression of METTL3,Hcy significantly decreased the LC3Ⅱ/Ⅰ expression and increased the p62 expression in islet β cells(P<0.05).To conclude,METTL3 is involved in the Hcy-induced autophagy regulation of islet β cells and plays a role in the regulation of insulin secretion.
