1.PLOD2 is involved in human skin fibrosis

Objective To discuss the relationship between the expression of PLOD2 and the formation of collagen I in the scar. Methods To detect the mRNA expression of PLOD2 and COLIα1 in the scar and normal skin tis-sue, as well as the content of COLIα1 by RT-PCR and Western blot; design and synthesize three kinds of PL0D2 ASODN targeted at the translational initiation, the joint of the first and the second exons and termination codon re-spectively , transfected into NIH/3T3 mouse fibroblast cell and detect the mRNA expression of COLIα1 and forma-tion of collagen I by RT-PCR and Western blot. Results The expression of PLOD2 in the scar tissue was signifi-cantly higher than that in the normal tissue ( P < 0. 05 ) , three kinds of ASODNs showed inhibitory effects on the mRNA expression of COLIα1 and formation of collagen I in fibroblast (P <0. 05) , during which the repressive ef-ficiency of ASODN targeted against the translational initiation was the highest (P <0. 001) ; while the mRNA ex-pression level of COLIα1 did not change in the scar tissue, normal tissue and the groups treated with three kinds of ASODN. Conclusion The high expression of PLOD2 is tightly related to the formation of collagen I , PLOD2 ASODN can suppress the formation of collagen I post-transcriptionally.
2.Physicochemical Properties and Gastric Mucosa Irritation of Cantharidin-hydroxypropyl-β-cyclodextrin Inclusion Complex

Objective To increase the solubility and relieve the mucous irritation of cantharidin (CA) by preparing cantharidin-hydroxypropyl-β-cyclodextrin (CA/HP-β-CD) inclusion complex.Methods The inclusion complex was prepared by co-evaporation method and characterized by differential scanning calorimetry (DSC),X-ray diffractometry (XRD),and nuclear magnetic resonance (NMR).Results The disappearance of CA as well as the shift of exothermic peaks shown in DSC results indicated the complexation phenomenon.XRD results showed that the crystalline CA pattern had disappeared,and in NMR results,H-5 shifted from δ 3.731to 3.695 after complexation and H-2 shifted from δ 3.626 to 3.598,which suggested that part of the drug had entered the HP-β-CD cavity to form an inclusion complex.The solubility increased 10.3 times after complexation and the mucous irritation of CA was relieved remarkably.Conclusion Through complexation with HP-β-CD,the solubility and dissolution rate of CA are improved significantly,and the irritation of musous is relieved.
3.Multivariate analysis of risk factors for clinically overt hemorrhagic cystitis after allogeneic hematopoietic stem cell transplantation

Objective To analyze the risk factors of clinically overt hemorrhagic cystitis(HC)(grade ≥Ⅱ) in 114 patients undergoing allogeneic hematopoietic stem cell transplantation(allo-HSCT) to predict the occurrence of HC.Methods We retrospectively analyzed 29 cases of clinically overt HC from a series of 114 patients subject to allo-HSCT from April 1997 to December 2004.The time of follow-up began from the day of initiating conditioning to day 180 post-transplant.The 11 clinical parameters were selected for univariate analysis using a Cox regression: age,sex,underlying disease,conditioning regimen,disease status at transplant,aGVHD,donor type,use of ATG,GVHD prophylaxis,platelet and neutrophil engraftment.Factors that were significant at the 0.1 level on univariate analysis were evaluated by multivariate analysis using a Cox regression.The cumulative incidence of grade ≥Ⅱ HC within the day 180 after transplantation was calculated by the method of Kaplan and Meier.Results Twenty-nine out of 114 patients(26 %) developed HC with grade Ⅱ in 12/29 cases(41.4 %),grade Ⅲ in 11/29 cases(37.9 %) and grade Ⅳ in 6/29 cases(20.7 %).The following factors were associated with an increased risk of HC by univariate analysis: male gender(RR=2.885,P=0.021),younger than 26 years(RR=3.265,P=0.002),gradeⅢ~Ⅳ aGVHD(RR=4.039,P=0.002),unrelated donor(RR=4.347,P=0.000),intense GVHD prophylaxis(RR=2.218,P=0.045),advanced disease(RR=2.668,P=0.009).These risk factors were entered into a multivariate model.Only male gender(RR=2.993,95 % CI 1.218-7.358;P=0.017) and unrelated donor(RR=4.478,95 % CI 2.049-9.786;P=0.000) were identified as being significantly associated with the occurrence of HC.Conclusion In multivariate analysis,patients were at increased risk of HC if they were male or had received grafts from unrelated donors.
4.Chtonic diseases and the MOS 36 item short from health survey score in community residents older than 60

This study was to investigate the relationship between chronic diseases and quality of life among community residents beyond 60.From September 2005 to November 2005,1731 residents aged 60-89 were surveyed with a general information form and the the MOS 36 item short from health survey (SF-36).The results showed that the subjects with no chronic diseases seemed to have higher scores,while SF-36 values dropped to some extent when the number of diseases increased.In comparison with diabetic or hypertensive subjects,those with heart disease had lower SF-36 scores for all items,except for general conditions and physical capability.
5.Progress in studies of viperin, an important cellular antiviral factor.

Viperin is an endoplasmic reticulum (ER)-associated protein that has been identified as an innate antiviral protein. Viperin expression can be largely upregulated by viruses, interferons, and oligonucleotides such as poly I:C and lipopolysaccharides. Viperin inhibits viral replication by interactiing with host cell proteins and several viral proteins, and disrupting the cell membrane system. It shows a broad-spectrum of antiviral activity. Some viruses have developed activities that counteract the action of viperin during a long- term period of evolution with hosts by impairing viperin expression. In addition to its antiviral effects, viperin has several other biological functions. This article review the basic characteristics of viperin and the state of current research into its antiviral effects, demonstrating the rapid progress that has been made in this field.

6.Serum IGF1's variation and VEGF's expression in hyperglycemia mice with colorectal tumor

Objective To observe colorectal tumor's growth in hyperglycemia mice and its vascular endothelial growth factor (VEGF)'s expression, insulin-like growth factor-1 (IGF-1)'s variation of blood through the experiment, then to ascertain whether type 2 diabetes mellitus (T2DM) danger factors to promote colorectal cancer happen and progress or not. Methods The mouse model of colorectal cancer combined T2DM was established. The volume of tumor was observed. After 5 weeks, all mice were executed and IGF-1 in the blood and the expression of VEGF in the tumor tissue was examined. Results The average tumor volume of colorectal tumor-diabetes group [(1628.5±882) mm3] were larger than that of colorectal tumor group [(1950.2±726) mm3] (P <0.05), and its expression IGF-1 of blood [(105.33±32.32) ng/ml] were higher than that of the control group [(69.83±25.57) ng/ml] and colorectal tumor group [(70.17±25.27) ng/ml] (P <0.05). The expression of VEGF [(70.0±11.5)%] in colorectal tumor-diabetes group were significantly higher than that of colorectal tumor group [(42.9±7.5)%] (P <0.05), too. Conclusion The model of T2DM and transplanted colorectal tumor can be duplicated successfully in ICR mice. Diabetes mellitus may be one reason of promoting colorectal cancer progress. Besides high blood glucose, its mechanism is the high level of IGF-1 which can inhibit apoptosis, promote cell differentiation and hyperplasia, and through inducing VEGF duplicating, heighten its expression, promote the tumor vessel growth, lead to tumor happen and metastasis.
7.Diagnostic value of AFU, TGF-α and Fer in the AFP lowly expression primary hepatic carcinoma

Objective To study the diagnostic value of serum α-L-fueosidase (AFU), transforming growth factor alpha(TGF-α)and ferritin (Fer) in primary hepatic carcinoma (PHC). Methods AFU and Fer in the serum were studied in 36 patients with benign hepatism and 56 patients with low concentration AFP of PHC hepatic carcinoma by automatic biochemistry analyzer Roche Modular. ELISA was used to assay the degree of TGF-α The sensitivity and specificity of AFU, TGF-α and Fer for low expression AFP of PHC were evaluated by ROC curve. Results The serum AFU, Fer and TGF-α levels were all significant difference in the patients than those in controls (P <0.05). The area under ROC curve of these indexes in descending order was AFU (0.707), TGF-α (0.677) and Fer (0.592). The diagnostic sensitivity of Fer was lower than AFU and TGF-α in diagnosis of AFP lowly expression PHC. The ratio showed AFU better than TGF-α The diagnostic specificity of AFU (64 %) was higher than that of TGF-α (61%) when TGF-α and AFU was at the same diagnostic sensitivity (64 %). Conclusion AFU is more valuable than Fer and TGF-α for negative or AFP lowly expression PHC, meanwhile it has much more accuracy.
8.Survey on depression status of elderly residents in a Shanghai community

Surveys on depression status of elderly population in a community of Shanghai Jing'an District were carried out in 2005 and 2008 respectively. The cluster sampling method was used in the study,1731 and 2315 community residents aged 60 -89 were selected in two surveys. The depression status was evaluated with geriatric depression scale (GDS). During the first survey 297 out of 1731 people were identified as depression with a prevalence rate of 17. 2%; during the second survey 374 out of 2315 people were found to have depression with a prevalence rate of 16. 2%. The factors related to depression status included family support, hobby, ability of self-taking care, family finance and chronic diseases.
9.Isolation of Brucella melitensis from the blood of infective endocarditis infected patient

The aim of the present study was to explore the prevalence characteristics of brucellosis in Hainan by investigating the biological characteristics and clinical type of pathogenic bacteria isolated from the blood of a patient infected by infective endocarditis.Bacteriological experiments were conducted according to the standard identification methods of Brucella including morphology,cultural characters,biochemistry characters,serological test and phage test,etc.At the same time,systematic analysis on the information about epidemiology,clinical manifestation and laboratory data of the patient was carried on.The Brucella melitensis was identified as Brucella melitensis biotype 2 and was significantly different from the Brucella melitensis biotypes 1 and 3 isolated in China reported in recent years.The urease test result of Brucella melitensis was variable and there is no strong positive result reported except the isolates in Hainan.As Brucellosis cause by Brucella melitensis biotype 2 was firstly reported in Hainan Province,great importance should be attached to its point-like prevalence.
10.Effect of matrine compound injection on interleukin-1? level in serum of rat with liver fibrosis

Objective:To study the anti-fibrogenetic effect of matrine compound injection and its mechanism. Methods: Experimental liver fibrosis induced by tetrachloride in rats was treated by matrine compound injection. Liver function index, interleukin-1? (IL-1?) and pathological changes in liver tissue were observed in model and therapy groups during the 4th,6th and 9th week. Results:The serum IL-1? levels of model and therapy groups were (25. 51?14. 31) and (10. 49?7. 49) pg/ml in the 4th week, (109. 67?20. 87) and (15. 06?2. 65) pg/ml in the 6th week, and (40. 26?10. 63) and (23. 27?7. 56) pg/ml in the 9th week. The liver function (ALT, AST, TBil) of therapy group was better than that of model group at each stage. Pathological changes indicated that the infiltration of inflammatory cells,the degree of liver cell necrosis and the degree of fi-broplastic proliferation in the therapy group were obviously better than those of the model group. Conclusion : Matrine compound injection has an anti-fibrogenetic action on the liver fibrosis induced by tetrachloride.
