2.Screening serum biomarkers in gastric cancer by iTRAQ-Coupled 2D-LC-MS/MS

Objective To apply the Isobaric tags for relative and absolute quantitation (iTRAQ) to identify proteins differentially in serums of gastric cancer (GC) patients, gastric mucosal intestinal metaplasia (IM) patients and normal control population. Methods The study included serum samples from GC group, IM group and normal control group with 45 cases each group,Proteins differentially regulated in serums were identified by the iTRAQ coupled with two-dimensional liquid chromatography/tandem mass spectrometry (2D-LC-MS/MS) technology. Results 10 540 unique peptides , of which correspond to a set of 199 proteins were identified , 23 proteins were over expressed and 14 proteins were underexpressed in GC and IM.Nine immune response proteins such as Lipopolysaccharide-binding protein,C-reactive protein and Ficolin-3 were found in the differential proteins, implying a close linkage between immune response and GC. Conclusions iTRAQ technology may help to identify novel serum markers for early diagnosis of GC.
3.Time difference attack therapy in multiple Acinetobacter bauamnnii.

Objective To explore time difference attack therapy in Acinetobacter bauamnnii infection. Methods 67 patients with Acinetobacter bauamnnii were divided into two groups. The experimental group were first given Fosfomycin 4 g 5% GS 100 ml iv girt to finish within 60 min and then given Cefperazone/Sulbactam 4 g 0.9% NS 250 ml iv gitt immediately bid. The control group were given: Ampicillin/Sulbactam 3 g 0.9% NS 250 ml iv gitt (tid) + Ciprofloxacin 0.2 g iv girt (bid). The treatment course all was 11 days. Results The overall effective rate of experimental group methods was superior to that of control group(X2 =9.56 ,P =0. 023). Conclusion The Fos-fomycin Cefperazone/Sulbactam time difference attack therapy for the treatment of Acinetobacter the bauamnnii in-fection is a new way.
4.Advances in Isolation and Synthesis of Xanthone Derivatives

Xanthone and its derivatives occupy a large part of the family of natural polyphenolic compounds with various biological and pharmacological activities.In recent years (from 2006 to 2011),it was reported that 127 xanthones were discovered from plants and fungi using various modem separation methods including silica gel/polyamide column chromatography,HPLC,high-speed counter-current chromatography,high-performance centrifugal partition chromatography,etc.Since total synthesis and structure modification for xanthone and its derivatives have been given attention worldwide,we introduced the synthetic methods of xanthone skeletons as well.Unfortunately,to date,there are still weaknesses in current methods of separation and synthesis,which need to be improved.This review,to a certain extent,provides necessary foundation for the further research and development of medicines containing xanthone and its derivatives.
5.Stromal cell derived factor-1 promotes the proliferation of bone marrow stem cells:the optimal concentration is 200 μg/L

BACKGROUND:Stromal cel derived factor-1 is a smal molecular protein with a wide range of biological activity that can cause immune cel chemotaxis, and it also has a chemotactic effect on bone marrow stem cels and periodontal ligament cels.
OBJECTIVE:To investigate the effect of stromal cel derived factor-1 with different concentrations on the proliferation of bone marrow stem cels and to probe the best concentration.
METHODS:Bone marrow stem cels from beagle dogs were culturedin vitro and stimulated by different concentrations of stromal cel derived factor-1 (100, 200, 300 μg/L). MTT was used to detect the influence of stromal cel derived factor-1 on the proliferation of bone marrow stem cels so as to screen the best concentration of stromal cel derived factor-1. Then, stromal cel derived factor-1 at the best concentrations was used to intervene the bone marrow stem cels, and MTT was used again to detect the proliferation of bone marrow stem cels.
RESULTS AND CONCLUSION:Stromal cel derived factor-1 at concentrations of 100, 200, 300 μg/L could promote the proliferation of bone marrow stem cels, and the effect was more notable at 200 and 300 μg/Lbut withno significant difference. Therefore, 200 μg/L was considered to be the best concentration of stromal cel derived factor-1 for intervention of bone marrow stem cels. Compared with the blank control group, 200 μg/L stromal cel derived factor-1 could significantly promote the proliferation of bone marrow stem cels. Taken together, stromal cel derived factor-1 can promote the proliferation of bone marrow stem cels, and its best concentration is 200 μg/L.
6.Determination of eugenol in Bawei Tanxiang Pills by Capillary Gas Chromatography

AIM: To establish a capillary GC-FID programmed temperature method for determining eugenol in Bawei Tanxiang Pills(Lignum Santali Albi, Flos Caryophylii, etc.). METHODS: Chromatography was performed on a 25m?0.25mm(0.25?m film thickness), SE-54 fused-silica capillary column coupled with a FID-detector. The internal standard was cyclohexanone. RESULTS: A good linearity was obtained in the range of 0.25?g~8?g of Eugenol with a correlation coefficient of 0.9993. The average recovery was 96.5%, the RSD was 2.4%. CONCLUSION: The established method was provided to be a reliable quantitative method for quality control of Bawei Tanxiang Pills.
7.Effect of matrine compound injection on interleukin-1? level in serum of rat with liver fibrosis

Objective:To study the anti-fibrogenetic effect of matrine compound injection and its mechanism. Methods: Experimental liver fibrosis induced by tetrachloride in rats was treated by matrine compound injection. Liver function index, interleukin-1? (IL-1?) and pathological changes in liver tissue were observed in model and therapy groups during the 4th,6th and 9th week. Results:The serum IL-1? levels of model and therapy groups were (25. 51?14. 31) and (10. 49?7. 49) pg/ml in the 4th week, (109. 67?20. 87) and (15. 06?2. 65) pg/ml in the 6th week, and (40. 26?10. 63) and (23. 27?7. 56) pg/ml in the 9th week. The liver function (ALT, AST, TBil) of therapy group was better than that of model group at each stage. Pathological changes indicated that the infiltration of inflammatory cells,the degree of liver cell necrosis and the degree of fi-broplastic proliferation in the therapy group were obviously better than those of the model group. Conclusion : Matrine compound injection has an anti-fibrogenetic action on the liver fibrosis induced by tetrachloride.
8.Correlations of neurophysiological features with clinical manifestation in patients with diabetic peripheral neuropathy

Objective To explore the correlation of electrophysiological features with clinical manifestation in patients with diabetic peripheral neuropathy (DPN).Methods The clinical data of 306 patients with DPN undergoing nerve conduction study (NCS) during May 2013 to June 2015 were retrospectively analyzed.Among 306 patients,304 were type 2,and 2 were type 1 diabetes with a mean age of 64.1 years and disease duration of 1 week-31 years.The relationship between the electrophysiological features and clinical manifestations was evaluated.Nerve conduction studies and F-wave and in sympathetic skin response were analyzed.Results ① NCS in lower limbs presented more abnormalities than that in upper limbs.There was the highest prevalence of not elicited sensory potential in Tibial nerve [56.9%(174/306),x2 =175.9] and the highest prevalence of not elicited motor potential in peroneal nerve [7.8%(24/306),x2 =1 103.1].Amplitude abnormality was more severe in sensory nerve than that in motor nerve[41.4% (507/1 224) vs.10.8% (132/1 224),x2 =559.2].The ratio of abnormal motor conduction velocity or distal latency was higher than that of amplitude decreased [22.9% (280/1 224) vs.10.8%(132/1 224),x2 =1 077.4].② The abnormal ratios of the shortest F-wave latency was lower than that of the prolonged distal latency in median and tibial nerve [5.4% (33/612) vs.20.6% (126/612),x2 =607.6].③ The abnormal ratio of sympathetic skin response (SSR) in lower limbs were higher than that of upper limbs [40.2% (246/612) vs.27.3% (167/612),x2 =129.4,P < 0.001],abnormal ratio of amplitude was higher than that of the prolonged latency [30.1% (368/1 224) vs.4.7% (58/1224),x2 =1040.5,P <0.001].Among the 99 cases presenting abnormal SSR in upper limbs,32 cases were normal in conventional NCS.Among the 127 cases presenting abnormal SSR in lower limbs,7 cases were normal in conventional NCS.④Disease course and glycosylated hemoglobin values were positively correlated with the distal motor latency,and negatively correlated with the nerve conduction velocity and amplitude.Conclusion Electrophysiological features of DPN is mainly shown in axon of the distal sensory nerve.Long duration and hyperglycemia may lead to more abnormalities in electrophysiological tests.
9.Arthroscopic reconstruction of anterior cruciate ligament with hamstring tendon graft using bioabsorbatble rigidfix cross pins

Objective To describe the surgical technique in reconstructing anterior cruciate ligament (ACL) with six strands harmstring tendon graft fixed by bioabsurbable rigidfix cross pins under arthroseopy and to e-valuate its efficacy. Methods From March 2005 to June 2008,39 patients with ACL injury were treated with ACL reconstruction by transplantation of six strands autogenous harmstring tendon , fixed by bioabsorbable rigidfix cross pins in femoral side. There are 22 male and 17 female,ages from 22 to 55 (the average age is 37). 19 cases were hurt in traffic accident, and another 20 cases in accidental injury. The state of illness is 7 days to 38 months. 13 cases merge the meniscus rupture, and 4 cases of meniscus suture,8 partial meniscectomy, 1 meniscectomy were performed simultaneously ;4 cases associating with the medial collateral ligaments Ⅲ degree injure underwent medial collateral ligament neo-plasty or reconstruction ;no cases merge posterior cruciate ligament injury, the patients were followed up 12 to 51 months , Pre-and post-operative knee joint function and stability were evaluated according to the Lysholm scoring scale system and the results of KT-2000 arthrometer , the clinical results and the reliability of the fixation were analyzed. Results 32 patients were followed-up and there is no limitation of the extention in the knee joints. The flexation of the knee joint is greater than 120°,and the anterior drawer test in 90° of flexation were negative in all patients. The postoperative Lachman test was strong positive in 1 case, negative in 26 cases and positive in 5 cases. The Lysholm scores was (92.6±4.2) points. The results of KT-2000 arthrometer: 31 cases 0-4.5 nun, average 3.2 mm;1 case 6. 5 mm. Conclusions It is a safe and reliable method to reconstruct ACL with six strands harmstring tendon graft fixing by bioabsorbable rigidfix cross pins under arthroscopy, and this procedure can obtain primary stabilization and long term stabilization of the autografts.
10.The effects of an enriched environment on psychological and behavioral symptoms of senescence

Objective To investigate the effects of an enriched environment (EE) on the psychological and behavioral symptoms of senescence and on the level of plasmic adrenocorticotropic hormone (ACTH) and serum corticosteroid (Cor). MethodsTwenty male senescence-prone SAMP8 mice of 3 months old were divided randomly into an enriched environment (EE) group and a standard environment (SE) group.Five male senescence-resistant SAMR1 mice of the same age served as the control group.Behavioral symptoms were assessed after 2 months using autonomic activity and elevated-plus maze (EPM) test performance.Plasma ACTH and serum Cor were detected using radio-immunologic methods. ResultsAverage autonomic activity frequency and EPM open-arm times were significantly lower with the SAMP8 mice than among the control group,and the autonomic activity frequency was significantly higher in the EE group than in the SE group of SAMP8 mice.The average EPM open-arm times of the EE group and the SE were not significantly different.Average ACTH and Cor levels were both significantly lower in the SAMP8 mice,and the level of ACTH was significantly higher in the EE group than in the SE group.There was no significant difference in these groups' average Cor levels. ConclusionsThe SAMP8 mice of 5 months demonstrated significant mental and behavioral abnormalities,as well as down-regulated plasma levels of ACTH and Cor.EE An enhanced environment can reduce behavioral disturbance through up-regulating ACTH.
