1.Clinical significance of combined measurement of PreS1 antigen and HBV serological markers in patients with hepatitis B

Objective To investigate the relationship between PreS1 antigen and five items of HBV serological markers(HBV M), as well as the clinical significance of combined measurement of PreS1 antigen and HBV M in patients with hepatitis B. Methods Enzyme linked immunosorbent assay (ELISA) was applied to testing serum in 268 patients with hepatitis B. Results The detection rate of PreS1 antigen was 85.88% in HBeAg'positive samples,significantly higher than that in HBeAg nega-tive samples(39.34%), P<0.01. Conclusion PreS1 antigen may effectively reflect the situation of HBV amplification. Combined measurement of PreS1 antigen and HBV serological markers in clinical examination contributes to clinical diagnosis,therapeutic effect monitoring and prognosis evaluation.
2.Clinical significance in detecting myocardial markers in acute myocardial infarction

Objective To explore the Troponin I(cTnI) ,myoglobin(MYO) and creatine kinase isoenzyme(CK‐MB) in early di‐agnosis of acute myocardial infarction(AMI) ,and compare their significance in the diagnosis of AMI .Methods used chemilumines‐cence method in 126 patients with AMI and 82 cases of myocardial infarction group to test cTnI ,MYO and CK‐MB ,the results were compared .Results The cTnI ,MYO ,CK‐MB quality appear time and peak time was different ,in patients with acute myocardial in‐farction(cTnI ,MYO and CK‐MB joint detection sensitivity ,specificity ,negative predictive value ,positive predictive value was higher than single test .Conclusion cTnI ,MYO and CK‐MB joint detection is helpful to early diagnosis of AMI .
3.In vitro Drug Resistance of 318 Gram-positive Strains in Nosocomial Infection

OBJECTIVE To investigate the drug resistance of Gram-positive organisms infection in our hospital and offer scientific basis for reasonable usage of antibiotics.METHODS The 318 strain organisms were isolated from the specimens of Aikang Hospital since Jan 2001 to Dec 2005.Gram-positive organisms were isolated from the specimens by the routine methods.The drug susceptivity was tested by K-B method.We got the results by 1999 standard of NCCLS.RESULTS Among the 318 strains,Staphylococcus aureus were 128 strains,40.3% of the total organisms;coagulase-negative S.aureus were 69 strains,21.7% of the total organisms;Enterococcus faecalis were 67 strains,21.1% of the total organisms;meticillin-resistant S.aureus(MRSA) were 32.8%;Meticillin-resistant coagulase-negative S.aureus(MRCNS) were 29.0%.The results of drug sensitivity test showed the drug resistant rate of Gram-positive strains for commonly used antibacterials was lower than in Shenzhen,Beijing,Shanghai,Wuhan,Guangzhou and other cities.CONCLUSIONS We should attach in our city importance to supervise the drug-resistance of Gram-positive organism,and preclude dissemination and epidemic of drug resistant strains.
4.Extended-spectrum ?-Lactamases-producing Klebsiella pneumoniae and Escherichia coli:Distribution and Drug-resistance Analysis

OBJECTIVE To study the distribution and characteristic about drug resistance of extended-spectrum ?-lactamases(ESBLs) producing Klebsiella pneumoniae and Escherichia coli for guiding the clinical drug using. METHODS Using K-B agar diffusion method and confirmed test to detect the drug resistance and distributions of ESBLs-producing K.pneumoniae and E.coli. RESULTS Among 582 K.pneumoniae and E.coli isolates,128(22%) were ESBLs-producing strains.For E.coli,the ratio was 17.5%,while K.pneumoniae was 28.3%.The sensitive rate of K.pneumoniae to imipenem was 100%. CONCLUSIONS ESBLs-producing K.pneumoniae and E.coli are the most common species causing infections,and imipenem is the best choice.
5.Expression and diagnostic value of miRNA-15 a in prostate cancer

Objective:To explore the diagnostic value of miRNA-15a in patients with prostate cancer.Methods:From January 2014 to January 2015,Gene expression of MiRNA-15a were examined in 36 serum and tumor tissue samples from prostate cancer ,40 benign prostatic hyperplasia ( BPH) and 40 health volunteers ,which examined by quantitative Real-time reverse transcription polymerase chain reaction(RT-qPCR).Results: The serum expression of miRNA-15a were (0.193±0.081),(0.359±0.04) and (0.376± 0.037 ) in patients with prostate cancer ,benign prostatic hyperplasia and healthy controls respectively .miRNA-15 a expression was sig-nificantly lower in BPH and healthy controls than patients with prostate cancer (P<0.05).There were no significant difference between BPH and control group(P>0.05).Serum miRNA-15a expression in patients with prostate cancer at different expression levels PSA , Gleason score,clinical stage,the difference between the presence of distant metastasis was statistically significant (P<0.05),and other pathological factors unrelated (P>0.05);Prostate cancer tumor tissue expression and PSA expression levels of miRNA-15a,Gleason score,clinical stage,presence of distant metastasis was statistically significant difference between (P<0.05),and other pathological factors unrelated(P>0.05).Conclusion:Low expression of miRNA-15a may be contributed in the development of prostate cancer ,and the expression levels miRNA-15a may be considered potential novel biomarkers for the diagnosis of prostate cancer .
6.Constructing the dual luciferase reporter vector containing human DRD1 promoter region

BACKGROUND:The polymorphisms of dopamine receptor in promoter region wil affect the expression of the receptor, thereby affecting the dopaminergic neurotransmitter, final y lead to related diseases.
OBJECTIVE:To construct the dual luciferase reporter vector containing human DRD1 promoter region and determine its activity, which could provide the basic tool for studying the transcriptional regulation of DRD1 gene.
METHODS:DRD1 promoter sequence was amplified by PCR using the human blood genomic DNA and cloned into pGM-T vector. After sequencing, the correctly constructed vectors were ligated to the firefly luciferase reporter plasmid pGL3-Basic. The cloned pGL3-Basic vectors were transfected into HEK293 using cationic liposome method. In the meanwhile, PGL3-Basic vector with no promoter was co-transfected with pGL3-TK plasmid as negative control group. The relative fluorescence intensity was measured by chemiluminescence.
RESULTS AND CONCLUSION:(1) Recombinant luciferase reporter gene vectors were confirmed by restriction analysis and sequencing. (2) Compared with the negative control group, the HEK293 cel s transfected by recombinant vectors presented transcriptional activity. (3) In conclusion, luciferase reporter gene vectors containing DRD1 promoter region are successful y constructed and can provide the basic tool for further study on the transcriptional regulation of DRD1.
7.Clinical analysis on thyroid hormone and autoantibodies in patients with type 2 diabetes mellitus

Objective To explore the relationship between type 2 diabetes mellitus(T2DM)and thyroid diseases by analyzing the abnormal status of thyroid hormone and antibodies in T2DM patients and the healthy control group.Methods The serum thyroid stimulating hormone (TSH),free triiodothyronine (FT3),free thyroxine (FT4),anti-thyroglobulin antibody (TGAb)and anti -thyroid peroxidase antibody (TPOAb)were measured in 396 patients with T2DM and 411 healthy persons.Results In the healthy group,the incidence rate of abnormal thyroid hormone was 7.5%,in which the male was 5.5% and female was 9.4%,the incidence rate of subclinical hypothyroidism in females was 4.7%,which was higher than 1.5% in males,there was statistically significant difference between them(P <0.05);the positive rate of TPOAb in the healthy control group was 10.2% and the positive rate of TGAb was 6.6%;in T2DM patients,the incidence rate of thyroid hormone abnormalities was 16.2%,which in males and females were 12.3% and 20.5% respectively,among them the incidence rate of subclinical hypothyroidism in females was 9.2%,which was significantly higher than 4.3% in males,the difference had statistical significance (P <0.01),T2DM patients with subclinical hypo-thyroidism accounted for 40.6% of thyroid dysfunction incidence rate;the positive rate of TPOAb was 15.2% and the positive rate of TGAb was 7.1 %.Conclusion The incidence rate of thyroid diseases in T2DM patients is significantly increased compared with the healthy control group,subclinical hypothyroidism in females is predominant;screening thyroid autoantibodies and monitoring FT3,FT4 and TSH at regular intervals in T2DM have the important clinical significance for evaluating the disease condition,jud-ging prognosis and guiding treatment.
8.EFFECTS OF THE ESTROGENIC ACTIVITIES OF GENISTEIN ON IMMUNE RESPONSE IN IRRADIATED MICE

Objective To study the radiation-protective effects produced by the estrogenic activities of genistein(Gen) for providing experimental basis to develop the food for radiation protection. Method After blocking estrogen receptor (ER) activities by Tamoxifen and Faslodex,the effects of Gen on peripheral leucocytes and lymphocyte counts,serum haemolysin and delayed type of hypersensitivity of the mice were examined after exposure to 4 Gy ? radiation. Results Gen was shown to inhibit the decrease of peripheral leucocytes and lymphocyte counts and the decreased immune response of the ?-irradiated mice. Faslodex (ER? blocker) + Gen eliminated the protective effect of Gen and still caused the similar effects in hematological response and immune response after irradiation. Tamoxifen (ER? blocker) did not have these effects. Conclusion Gen can enhance the immune function in irradiated mice through activation of estrogen receptor ? pathway.
9.An investigation on binocular summation response of visual evoked potential in children with hyperopia amblyopia

Objective Conventional examination for vision function adopts subjective psychophysics methods.Pattern visual evoked potential (P-VEP) binocular summation response is a new objective way for the test of vision function.But its clinical value in evaluating vision function of amblyopia children is still in controversy.This study was to explore the binocular vision and the feature of P-VEP binocular summation in children with amblyopia and evaluate the significance of P-VEP binocular summation in binocular vision.MethodsThis is a case-controlled study.P-VEP binocular summation response and single ocular P-VEP response was respectively recorded in 151 hyperopia amblyopia children and 80 age- and gender-matched normal children.P-VEP response from children with hyperopia amblyopia was recorded under the corrected vision.The hyperopia amblyopia was diagnosed based on the standard of National Children Amblyopia and Strabismus Prevention and Treatment Working Group.The feature of P-VEP binocular summation was further analyzed and compared with monocular VEP response.Informed consent was obtained from each subject or custodian prior to the trail.ResultsNo significant difference was found in latency between P-VEP binocular summation response and monocular P-VEP response whether amblyopia children or normal children(P>0.05).Binocular response/monocular response value in amblyopia children was lower than that in normal children(P<0.05).Binocular response/monocular response value in mild or moderate amblyopia children was higher than that in severe amblyopia children(P<0.05).Binocular response/monocular response value was significantly reduced in the children with binocular vision than in the children without binocular vision (P<0.05).ConclusionP-VEP binocular vision,acting as an evaluation factor,could impartially reflect the function of binocular vision of amblyopia children.
10.The relationship between behavioral problems and family environment in preschool-age children

Objective To study the relationship between behavioral problems and family environment in preschool-age children. Methods Children between 4-6 years of age in the kindergartens of Zhongshan city were randomly selected. Child Behavior Checklist (CBCL) was used to assess the behavioral problems in children, and self-designed general situation question-naire and Family Environment Scale-Chinese Version (FES-CV) were applied to evaluate the family environment. Results A total of 3445 children were enrolled and 29.87%children showed behavioral problems. There were signiifcant differences in the scores of feeling expression, family conlfict, intelligence, recreation, organization and control between children with and without behavior problems (P<0.001). The scores of feeling expression, intelligence, recreation, organization and control were signiif-cantly higher and the score of family conlfict was lower in healthy children in comparison with children with behavior problems. Through multivariate logistic regression, it was suggested that female (OR=2.04), non-urban population (township OR=1.45, rural OR=1.51), non-local Zhongshan resident (OR=1.39), father’s low educational level (technical secondary school OR=1.77, junior high school OR=2.17, primary school and below OR=3.24), discipline styles (indulgent OR=1.47, mixed OR=1.37), family conlfict (OR=1.13) were independent risk factors for behavioral problems (P<0.05). Mother’s age over 30 (OR=0.74) or over 40 (OR=0.46), feeling expression (OR=0.94) and intelligence (OR=0.87) were independent protective factors for behavioral prob-lems (P<0.05). Conclusions Behavioral problems in preschool-age children are closely related to family environment, which needs to be paid more attention.
