1.Comparison of Pharmaceutical Literatures from Commonly Used Chinese Periodical Databases and Automatic Processing of the Retrieved Literatures

OBJECTIVE:To provide references for pharmaceutical workers in retrieving literatures by means of Chinese bi_omedical database(CBM),China journal full-text database(CNK) and Chinese science & technology journal database(VIP).METHDOS:With the commonly research topics of hospital pharmacy as research scope,literatures retrieved by database sampling underwent a preliminary contrast analysis and a computer-assisted automatic meta-analysis after programming.RESU_TLS:None the 3 databases indexed all the tested pharmaceutical periodicals(totaled 79 kinds).There were differences among the un-indexed periodicals, except a few are the same in kinds.The retrieved literatures of docuterm had a high proportion of repetition;however,the computer had merged the same literatures retrieved in different databases.CONCLUSIONS:The literatures indexed in the 3 databases have their own characteristics,thus literature recall ratio can only be improved by combining multiple databases.The automatic meta-analysis of the retrieval literatures can be performed by means of computer program,the repeated work can be reduced thereafter.
2.The value of magnetic resonance diffusion tensor imaging for predicting functional recovery in cerebral infarction patients

Objective To explore the changes in fractional anisotropy (FA) among cerebral infarction pa-tients using MR diffusion tensor imaging (DTI), and to verify the relationship between any FA changes and functional recovery. Methods Thirty-eight cerebral infarction patients were divided into two groups according to their recovery level using Brunnstrom's criteria. All the patients accepted routine MR and DTI examination, and FA values were measured during the acute, subacute and chronic stages of their recovery. Results Average FA values in the lesion area were significantly lower than in the corresponding contralateral area, and were highest daring the acute stage.There was no significant difference between the subacute and chronic stages. Conclusions The FA values of cere-bral infarction patients change during the different stages of recovery with a certain regularity. This may be valuable for clinical treatment and prognosis.
3.Subcutaneous injection of basic fibroblast growth factorspromotesthe proliferation of hippocampal neural stem cells in rats with vascular dementia

BACKGROUND:As a neurotrophic factor, basic fibroblast growth factors extensively distribute in the central nervoussystem,andplay an important physiological roleby combinationwith their relative receptors.
OBJECTIVE:To investigate the effect of basic fibroblast growth factors on the learning ability and proliferation of nestin-positive hippocampal neural stem cels in rats with vascular dementia.
METHODS:Totaly 30 Sprague-Dawley rats were randomly divided into vascular dementia, sham operation and treatment groups. The vascular dementia and treatment groups were for preparing vascular dementia model, and the treatment group was given subcutaneous injection of basic fibroblast growth factors. Subsequently, at 4 weeks, the learning ability of rats and the number of nestin-positive hippocampal neural stem cels was detected by the Morris water maze test and immunohistochemical staining, respectively.
RESULTS AND CONCLUSION:Compared with thevascular dementia groupthe latency period was significantlyshorterin thesham operation and treatment groups, and the number oftimes crossing the target quadrant was significantlyhigher in thesham operation and treatment groups (P< 0.05). But there was no significant difference between the sham operation and treatment groups(P> 0.05). Under fluorescence microscope, yelow-green fluorescence stained neuronspositive forbasic fibroblast growth factorcould be found in the CA1, CA2 and CA3 of the treatment group. Additionaly, the number of nestin-positive neurons in the CA1 area of the hippocampus was the most inthetreatment group, folowed by the sham operation group, andthe leastin the vascular dementia group. These results suggest that the subcutaneous injection of basic fibroblast growth factors can migrate to the hippocampus,, andimprove the learning ability of rats by inducing proliferation of nestin-positive hippocampal neural stem cels.
4.Effect of Qinggong Zhixue Granule on low immunity mice

OBJECTIVE: To probe into the pharmacological mechanism of the Chinese medicinal compound Qinggong Zhixue Granule (QGZXG), which has an effect of replenishing qi to activate blood and expelling stasis to stop bleeding, in treating irregular vaginal bleeding after medical abortion. METHODS: Healthy female KM mice and mice with immunodeficiency caused by cytoxan (CTX) were chosen as experimental subjects. The effects of QGZXG on the phagocytization of the mice's monocytes (by using the method of carbon particles expurgation), the delayed type hypersensitivity caused by 2,4-dinitrochlorobenzene (DNCB) and the content of antibody of hemolysin in the mice's serum were observed. RESULTS: (1) QGZXG improved the ability of the liver and spleen of immunodeficient mice to expurgate carbon particles. There was an significant statistical difference between the CTX treated group and the low dose group as well as the high dose group (P < 0.01) and a statistical difference between the CTX treated group and the medium dose group (P < 0.05). (2) QGZXG boosted the cellular immunity of immunodeficient mice. The ear swelling of mice in low, medium and high dose group was more obvious than that in the CTX treated group, and this difference was significant in statistics (P < 0.01). (3) QGZXG raised the content of the antibody of hemolysin in the serum of immunodeficient mice. A statistical difference occurred between the high dose group and the CTX treated group. The low and medium dose also had a tendency of such effect. CONCLUSION: QGZXG can improve both the specific and non-specific immunity of the immunodeficient mice, and therefore accelerate the recovery of the whole body and endometrium after parturition and abortion. In consequence, it leads to shorter duration and less quantity of vaginal bleeding after medical abortion.
6.Comparison of EEG complexity between rats under awaking and anesthesia

Objective To compare the EEG complexity between rats under awaking and differ-ent depth of anesthesia via analyzing sample entropy and fractal dimension.Methods Sixteen SD rats were intraperitoneally injected with urethane twice,first with 500 mg/kg and second with 800 mg/kg one hour later.The scalp EEG was collected in stage of awaking (W),light anesthesia (LA)and heavy anesthesia (HA).The sample entropy (SampEn)and fractal dimension (FD)were computed by MATLAB.The characteristic values were denoised by linear dynamic system method during the whole process.Results The value of SampEn and FD gradually dropped from awaking to heavy anes-thesia.The SampEn and FD in W was significantly higher than the value in LA or in HA (P <0.05). The SampEn and FD in HA was significantly lower than that in LA (P < 0.05 ).Conclusion The SampEn and FD of EEG could be used to monitor the depth of anesthesia.
7.Clinical Observation of Mailuoning Injection Combined with Benazepril in the Treatment of Chronic Glo-merulonephritis

OBJECTIVE:To observe clinical efficacy and safety of Mailuoning injection combined with benazepril in the treat-ment of chronic glomerulonephritis. METHODS:130 patients with chronic glomerulonephritis were randomly divided into observa-tion group and control group,with 65 cases in each group. Both groups received rountine treatment as low salt and fat diet. Control group was additionally given Benazepril tablet 10 mg,qd;observation group was additionally given Mailuoning injection 20 ml added into 5% Sodium chloride injection 250 ml,ivgtt,qd,on the basis of control group,20 days of drug withdrawal every 10 days. Both group received 4 months of treatment. Clinical efficacy of 2 groups were observed as well as the levels of BUN,UCr, SCr and 24 h urine protein quantitative (UPE) before and after the treatment. The incidence of ADR was compared between 2 groups. RESULTS:Total effective rate of observation group was 83.1%,which was significantly higher than 67.7% of control group,with statistical significance (P<0.05). There was no statistical significance in BUN,UCr,SCr and 24 h UPE between 2 groups before treatment (P>0.05);those indexes of 2 groups decreased significantly after treatment,and the observation group were lower than the control group,with statistical significance(P<0.05). There was no statistical significance in the incidence of ADR between 2 groups (P>0.05). CONCLUSIONS:Mailuoning injection combined with benazepril is effective for chronic glo-merulonephritis,and can effectively improve renal function with good safety.
8.Development of an immunomagnetic bead assay for quantitative detection enolase of Candida albicans

Objective To develop an immunomagnetic bead ( IMB) assay for quantitative detection enolase ( Eno) of Candi-da albicans, and to improve the diagonosis of invasive candidiasis . Methods The immunomagnetic bead was prepared by conjuga-ting with Anti Eno of Candida albicans monoclonal antibody .HRP-conjugated goat polyclonal antibody against Candida albicans Eno was employed as detecting antibody .The performance parameters of the IMB assay including precision , specificity, linear range and limit of detection were verified by using recombinant Candida albicans Eno.Then the developed assay was applied to determine Eno levels in supernatant of pathogenic fungi cultures . Results The intra and inter-coefficient of variation was 4.54%, 5.87% and 5.26%, 8.82%at the concentration of 25 ng/mL and 5 ng/mL, respectively.The limit of detection was 0.5 ng/mL.The linear range was(0.5-50) ng/mL.The level of Eno in Candida albicans culture after incubated in 37℃for 24 h was 3.89 ng/mL and gradually in-creased to 37.89 ng/mL at 120 h.There was a positive correlation between the level of Eno and growth hyphae of Candiad albicans. There was weak cross reaction with Candida parapsilosis and no cross reaction with Candida tropicalis, Candida guilliermondii, Candida glabrata, Cryptococcus neoformans and Saccharomyces cerevisiae. Conclusion An IMB assay for quantitative detection Eno of Candida albicans was developed , which was more sensitive , rapid and reliable than previous qualitative ELISA .The IMB assay has the potential to be applied to the research in invasive candidasis .

10.Determination of AKAP12 methylation levels in peripheral blood using methylation-sensitive high resolution melting analysis in colorectal cancer

Objective To detect quantitatively AKAP12 methylation and evaluate its clinical significance in peripheral blood in colorectal cancer. Methods MS-HRM technology was used to detect quantitatively AKAP12 methylation in peripheral blood from 80 colorectal cancer patients and 20 healthy volunteers. They also validated the reproducibility and compared with MSP. Results Thirty-eight of the 80 colorectal cancer samples (47. 5% ) were found to be methylated at the AKAP12 promoter region by MS-HRM (the methylation levels of 24 cancer samples ranged between 1 % and 20% , the methylation levels of 12 cancer samples ranged between 20% and 60% , the methylation levels of 2 cancer samples ranged between 60% and 100% ). The methylation levels of 2 health samples were less than 10% . They also compared the results generated by MS-HRM with a traditional MSP assay. The AKAP12 MS-HRM assay was able to reproducibly detect 1% AKAP12 methylated DNA, whereas the MSP method was unable to detect less than 10% methylation. No significant correlation was observed between the AKAP12 methylation levels and patients' age and gender. However, AKAP12 methylation was significantly higher in DNA from colorectal cancer patients with high Dukes stage and differentiation (x2 =5. 93 or 8. 41, P = 0.01). Conclusions The authors demonstrate here for the first time, the utility of quantitative AKAP12 MS-HRM analysis of promoter methylation in peripheral blood samples. AKAP12 MS-HRM quantitative methods have many promising applications in the detection of colorectal cancer.
