1.The predictive value of serum FT3 level on clinical outcomes after PCI in patients with coronary heart disease

Aim To explore the predictive value of serum free triiodothyronine(FT3)on the long-term prognosis of patients with coronary heart disease after percutaneous coronary intervention(PCI).Methods All the subjects were from a prospective cohort study(PRACTICE study).In this study,15 250 patients with coronary heart disease after PCI in the First Affiliated Hospital of Xinjiang Medical University were selected,and the clinical data,FT3 and creatinine were collected.All the subjects were followed up regularly,and the primary follow-up endpoints were all-cause mortality and cardiogenic mortality,the secondary endpoints were major adverse cardiovascular events(MACE)and major adverse cardiovascular and cerebrovascular events(MACCE).According to the admission criteria,3 109 patients were finally in-cluded in this study.According to the baseline value of FT3,patients were divided into normal FT3 group(FT3:3.65~6.8 pmol/L,1 446 cases)and low FT3 group(FT3<3.65 pmol/L,1 663 cases).Kaplan-Meier analysis was used for survival analysis,and Log-rank test was used for survival comparison.Multivariate Cox regression analysis was used to e-valuate the risk factors of the follow-up results of the two groups.Results Compared with the normal FT3 group,all-cause mortality and cardiogenic mortality in the low FT3 group increased significantly(P<0.05).Kaplan-Meier analysis showed that the cumulative risk of all-cause mortality and cardiogenic mortality increased in the low FT3 group(P<0.05).Multivariate Cox regression analysis indicated that the risk of all-cause mortality increased by 1.639 folds in the low FT3 group(HR=2.639,95%CI:1.385~5.348,P=0.007),while no statistical difference was found in cardiogenic mortality after adjusting for multiple factors(P=0.125).Conclusion The decrease in serum FT3 levels has important predictive value for all-cause mortality after PCI in patients with coronary heart disease.

3.Relationship between psychological quality and non-suicidal self-injury behavior in middle school students: path analysis of coping style

BackgroundNon-suicidal self-injury (NSSI) behavior is highly prevalent in middle school students, which poses a significant risk to the physical and mental health of middle school students, so finding ways to improve the NSSI behavior among middle school students is of great significance for promoting their healthy growth. ObjectiveTo investigate the relationship between psychological quality and NSSI behavior among middle school students in western Anhui, and to examine the pathway of coping style in the above relationship. MethodsFrom November to December 2020, 22 872 middle school students in western Anhui were selected using stratified cluster random sampling method, and were subjected to complete the assessment of Adolescent Non-suicidal Self-injury Assessment Questionnaire (ANSAQ), Adolescent's Psychological Suzhi Scale-Simplified Version (APSS-SV) and Coping Style Scale for Middle School Students (CSSMSS). Spearman correlation analysis was adopted to examine the correlation among scores of scales and the frequency of NSSI behavior in middle school students. Amos 23.0 was utilized to test the pathway of coping style in the relationship between psychological quality and NSSI behavior. ResultsA total of 21 718 (94.95%) middle school students completed the effective questionnaire survey. NSSI behavior was detected in 7 798 middle school students (35.91%). ANSAQ total score and CSSMSS positive coping style score of middle school students were negatively correlated with the frequency of NSSI behavior (r=-0.219, -0.179, P<0.01). CSSMSS negative coping style score was positively correlated with the frequency of NSSI behavior (r=0.093, P<0.01). The direct effect value of psychological quality on the frequency of NSSI behavior was -0.136, and the indirect effect value of coping style on the relationship between psychological quality and the frequency of NSSI behavior was -0.084. Among them, the effect value of positive coping style was -0.122, accounting for 55.45% of the total effect, while the effect value of negative coping style was 0.038, accounting for 17.27% of the total effect. ConclusionCoping style may mediate the relationship between psychological quality and NSSI behavior in middle school students, and the positive coping style and negative coping style play separate roles in the pathway of psychological quality on NSSI behavior.[Funded by 2020 Natural Science Research Project of Anhui Provincial Education Department (number, KJ2020B006)]
4.Analysis of β-thalassemia gene testing results in western region of Guangxi Zhuang Autonomous Region

Objective:To analyze the positive detection rate, main genotypes of β-thalassemia in western region of Guangxi Zhuang Autonomous Region (referred to as Guangxi).Methods:Retrospective analysis of 26 189 individuals who underwent gene testing for thalassemia at the Affiliated Hospital of Youjiang Medical University for Nationalities from January 2013 to December 2019. Using the crossing breakpoint PCR (Gap-PCR) and reverse dot blot (RDB) techniques to detect Chinese common type of 7 kinds of α-thalassemia and 17 kinds of β-thalassemia genotypes, high-throughput sequencing(Sanger) was performed for suspected rare β-thalassemia. Gap-PCR was used for suspected deletion β-thalassemia types.Results:β-thalassemia was diagnosed in 4 495 (17.16%) of 26 189 samples. A total of 6 177 alleles of 20 types of β-thalassemia were detected, mainly CD17 (2 712 cases, 43.90%) and CD41-42 (2 240 cases, 36.26%), including 7 rare alleles: Gγ +( Aγδβ) 0, SEA-HPFH, Hb New York, Hb G-Taipei, Hb Hezhou, Hb G-Coushatta and IVS-Ⅱ-81. There were 3 903 case (86.83%) heterozygous, 273 case (6.07%) double heterozygous, and 319 case (7.10%) homozygous among 4 495 β-thalassaemia subjects. A total of 48 genotypes were detected. The two most common genotypes were CD17/β N (1 890 cases, 42.05%) and CD41-42/β N (1 212 cases, 26.96%), accounted for 69.01% (3 102/4 495). Seven rare genotypes were detected: Gγ +( Aγδβ) 0/β N in 3 cases, Hb New York/β N in 3 cases, Hb G-Taipei/β N in 2 cases, SEA-HPFH/β N, Hb Hezhou/β N, Hb G-Coushatta/β N and IVS-Ⅱ-81/β N in 1 case each. A total of 1 041 cases (3.97%, 1 041/26 189) of 116 types of αβ-thalassemia were detected, mainly -- SEA/αα composite CD17/β N (144 cases, 13.83%), followed by -α 3.7/αα composite CD17/β N (112 cases, 10.76%). Conclusions:Western region of Guangxi is a high prevalence area of β-thalassemia, CD17/β N and CD41-42/β N are the main genotypes. The variation spectrum of β-thalassemia is complex and diverse, with rich genotype.

Many individuals experience all three conditions concurrently, significantly heightening the risk of cardiovascular disease and mortality. Although the National Metabolic Management Center(MMC) has been established for over eight years and has its unique features, the awareness, treatment, and control rates of these diseases in China remain low, and the efficiency of community management is insufficient. According to the previous two editions of management guidelines and the most recent domestic and international diagnostic and treatment guidelines, this paper conducts an in-depth analysis of the operational experience and management strategies of the MMC. Its aim is to improve the efficiency of grassroots MMC mode management for " Three Highs" patients and ensure that patients receive more standardized management.

7.Improvement effects and mechanism of total flavonoids of Bidens pilosa on lipopolysaccharide-induced neuroin-flammation in mice

OBJECTIVE To investigate the effect and mechanism of total flavonoids of Bidens pilosa L. (TFB) on lipopolysaccharide (LPS)-induced neuroinflammation in mice. METHODS Fifty C57BL/6 mice were randomly divided into normal control group, LPS group and TFB low-dose, medium-dose and high-dose groups, with 10 mice in each group. TFB low-dose, medium-dose and high-dose groups were given TFB solution intragastrically at 60, 120 and 240 mg/kg, and the normal control group and LPS group were given corresponding volume of normal saline, once a day, for consecutive 21 d. From the 15th day of administration, except for the normal control group, other groups were given LPS (400 μg/kg) intraperitoneally for 7 consecutive days to establish neuroinflammatory model. Brain tissues were taken under anesthesia 4 h after the final administration. The morphological changes of neuronal cells in mice were observed; the contents of nitric oxide (NO), tumor necrosis factor α (TNF- α), interleukin-1β (IL-1β), IL-6 and IL-10 were measured, and the expressions of inflammatory pathway-related proteins [inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2), myeloid differentiation factor 88 (Myd88) and protein kinase C (PKC)] were measured in the brain tissues of mice. RESULTS Compared with the normal control group, the neuronal arrangement in the hippocampal region of the brain tissue of mice in the LPS group was sparsely disorganized, with a large number of neuronal fixations and shrunken nuclei; the contents of TNF-α, IL-1β, IL-6 and NO in the brain tissue were significantly increased, the contents of IL-10 were significantly decreased, and the relative expressions of iNOS, COX-2, Myd88 and PKC proteins were significantly increased (P<0.05). Compared with the LPS group, the neuronal pathological changes in the brain tissue of mice in the TFB low-dose, medium-dose and high-dose groups were 202014810) significantly improved, and the changes of the above indices in the brain tissue were significantly reversed (P<0.05) CONCLUSIONS TFB has an inhibitory effect on E-mail:pangxjun@163.com neuroinflammation, and its mechanism of action may be related to down-regulation of the expressions of inflammatory pathway-related proteins iNOS, COX-2, Myd88 and PKC, and reduction of inflammatory factors release.

Porcine epidemic diarrhea (PED) is a major disease of pigs that inflicts heavy losses on the global pig industry. The etiologic agent is the porcine epidemic diarrhea virus (PEDV), which is assigned to the genus Alphacoronavirus in the family Coronaviridae. This review consists of five parts, the first of which provides a brief introduction to PEDV and its epidemiology. Part two outlines the passive immunity in new born piglets and the important role of colostrum, while the third part summarizes the characteristics of the immune systems of pregnant sows, discusses the concept of the "gut-mammary gland-secretory IgA(sIgA) axis" and the possible underpinning mechanisms, and proposes issues to be addressed when designing a PEDV live vaccine. The final two parts summarizes the advances in the R&D of PEDV vaccines and prospects future perspectives on prevention and control of PEDV, respectively.

9.Production of high-purity recombinant human vascular endothelial growth factor (rhVEGF165) by Pichia pastoris.

Vascular endothelial growth factor (VEGF165) is a highly specific vascular endothelial growth factor that can be used to treat many cardiovascular diseases. The development of anti-tumor drugs and disease detection reagents requires highly pure VEGF165 (at least 95% purity). To date, the methods for heterologous expression and purification of VEGF165 require multiple purification steps, but the product purity remains to be low. In this study, we optimized the codons of the human VEGF165 gene (vegf165) according to the yeast codon preference. Based on the Pichia pastoris BBPB vector, we used the Biobrick method to construct a five-copy rhVEGF165 recombinant expression vector using Pgap as the promoter. In addition, a histidine tag was added to the vector. Facilitated by the His tag and the heparin-binding domain of VEGF165, we were able to obtain highly pure rhVEGF165 (purity > 98%) protein using two-step affinity chromatography. The purified rhVEGF165 was biologically active, and reached a concentration of 0.45 mg/mL. The new design of the expression vector enables production of active and highly pure rhVEGF165 ) in a simplified purification process, the purity of the biologically active natural VEGF165 reached the highest reported to date.

ORF3 protein, the single accessory protein encoded by porcine epidemic diarrhea virus (PEDV), is related to viral pathogenicity. In order to determine the cytoplasmic location signal of PEDV ORF3, we constructed a series of recombinant plasmids carrying full-length or truncated segments of PEDV DR13 ORF3 protein. When the acquired plasmids were transfected into Vero cells, expression and distribution of the EGFP-fused full-length ORF3 protein and its truncated forms in the cells were observed by laser confocal microscopy. The results showed that ORF3 protein or their truncated forms containing 40-91 aa segment including two transmembrane domains were localized in the cytoplasm, whereas ORF3 truncated peptides without the 40-91 aa segment were distributed in the whole cell (in both cytoplasm and nucleus). This suggests that the 40-91 aa is the key structural domain determining cytoplasmic location of PEDV ORF3 protein. The discovery provides reference for further clarifying intracellular transport and biological function of PEDV ORF3 protein.
