The nuclear factor erythroid 2 related factor 2 (Nrf2) is a key protein of endogenous antioxidant defense systems in the body. In response to oxidative stress, Nrf2 translocates to nucleus and binds to antioxidant response elements (ARE), regulating the expression of a large amounts of antioxidant genes and maintaining a proper redox balance. The pathological processes of neurodegenerative diseases are associated with generation of reactive oxygen species, which cause oxidative stress. Oxidative stress plays a cardinal role in the onset and progression of neurodegenerative diseases. Nrf2-inducer compounds can reduce oxidant stress and have shown therapeutic efficacy in many neurodegenerative disease models. How to activate the Nrf2 signaling pathway effectively has been received much attention. Here we provided an overview of specific mechanism of Nrf2-ARE pathway and the protective effects of Nrf2 in different neurodegenerative diseases, and summarized the Nrf2 activators recently in preclinical study.

Acta Anatomica Sinica 2019;50(4):431-437
Objective: To investigate the effect of scutellarin after cerebral ischemia-reperfusion injury in rats with hyperlipemia, by observing the expression of B-cell lymphoma-2 (Bcl-2) and Bcl-2 assaciated X protein (Bax) in cerebral cortex with hyperlipemia. Methods: After the rat model of hyperlipidemia was established, the focal cerebral ischemiareperfusion model in hyperlipemia rats was established with thread embolism of the middle cerebral artery. The neurobehavioral score was used to observe the symptoms of neurobehavioral injury. 2,3,5 -triphenyl tetrazolium chloride (TTC) staining was used to observe the cerebral infarct volume. HE staining was used to observe the pathological changes of brain tissue. Immunohistochemistry and Western blotting were used to observe the expressions of Bcl-2 and Bax. Results: Compared with the sham-operation group, the neurobehavioral scores, cerebral infarction volum and pathologic damage were significantly increased, the expressions of Bcl-2 was significantly lower, the expressions of Bax was significantly increased in ischemia-reperfusion group(P<0. 05). Compared with the saline group, the neurobehavioral scores, cerebral infarction volum and pathologic damage were significantly decreased, the expression of Bcl-2 was significantly increased, the expression of Bax was significantly decreased in scutellarin treatment group (P<0. 05). Conclusion: In cerebral ischemiareperfusion injury rats with hyperlipemia, scutellarin can alleviate cerebral ischemia-reperfusion injury by promoting the expression of Bcl-2 and inhibiting the expression of Bax.

Mitochondrial dysfunction has been implicated in the aetiology of sporadic Parkinson's disease but its role in the disease mechanism remains unclear. To investigate the effect of synuclein on mitochondrial dysfunction induced by rotenone. The human dopaminergic SH-SY5Y cells were used as a cell model. The cells over-expressed the wild-type ?-synuclein were treated with complex I inhibitor rotenone. The cell viability, complex I activity,Mitochondrial swelling and O2-content were tested at different time point-1w,2w,4w after rotenone treated.CCK-8 test results showed that the cell viability of overexpressed ?-synuclein(SH-SY5Y-Syn)was much lower than the control group(SH-SY5Y-Ctr).After administrating with rotenone about 1w or 2w the cell viability of SH-SY5Y-Syn became higher than that of SH-SY5Y-Ctr.On the 4th week the results were contrary to the first 2 weeks.Similar results were got when test the mitochondrial function. In the first 2 weeks after roteoone administrating, the mitochondrial function of SH-SY5Y-Syn was better than that of SH-SY5Y-Ctr.This suggest that the ?-synuclein could protect the mitochondrial against the injury induced by rotenone in the early stage-1w,2w,while this effect disappeared in the final stage-4w.
