1.Effect of sodium selenite on Treg cells in peripheral blood mononuclear cells in patients with Graves disease

Objective:Recently,selenium was used for the treatment of Graves disease. However,there has been no reports that selenium affected the immune function of Graves disease at the cellular level so far. It is reported the effect of sodium selenite on Treg cells in peripheral blood mononuclear cells in patients with Graves disease in the present study. Methods:AFS were used to detect the level of serum selenium;PBMC( Peripheral blood mononuclear cells) were extracted,then,cultured with sodium selenite. Foxp3 mRNA expression was tested by Real-time fluorescence PCR,and ELISA was used to detect the secretion of IL-35. Results:The serum sodium selenite levels of GD group are significantly lower than the HC group;The expression of Foxp3 mRNA and secretion of IL-35 are both increased. Conclusion:It shows sodium selenite could improve the state of immune disorders by recovering the function of Treg cells.

As a member of tyrosine kinases(TK) family,Epidermal growth factor receptor(EGFR) has an activity of intrinsic protein tyrosine kinase,which plays an essential role in the regulation of signal transduction in the cells.Due to the abnormal expression of EGFR-TK,the certain type cancers may developed and progressed.Based on that,the inhibitors of EGFR-TK could be effective medicines for the treatment of cancer.The EGFR-TK domain was amplified by RT-PCR with RNA of HUVCEs cells as the template and expressed in E.coliBL21(DE3) using plasmid pET30a as vector.The recombinant protein was purified with the affinity chromatography(Ni-NTA),which was identified to have kinase activity catalyzing the substrate phosphorylated with ATP in the enzymatic reaction.Using the recombinant EGFR-TK as target,the screening model for enzymatic inhibitors was constructed.

Objective To study the relationship between type 1 and type 2 cytokines and human acute graft-versus-host disease (aGVHD).Methods In 20 patients undergoing allogeneic peripheral blood stem cell transplantation (allo-PBSCT), plasma concentrations of interleukin-18 (IL-18), interleukin-2R (IL-2R), tumor necrosis factor-a (TNF-?) and interleukin-10 (IL-10) were measured by using enzyme-linked immunosorbent assay (ELISA).Results All the patients achieved engraftment. Eight cases deve- loped grade Ⅰ-Ⅱ aGVHD and 3 cases developed Ⅲ-Ⅳ aGVHD. The concentrations of IL-18 and IL-2R in the patients with aGVHD were significantly higher than those without aGVHD. The levels of IL-18 were correlated with the severity of aGVHD; The levels of TNF-? showed no difference between the patients with or without aGVHD; The concentrations of IL-10 in the patients with aGVHD were significantly declined but in those without aGVHD were significantly increased.Conclusion Type 1 and type 2 cytokines play an important opposite role in development of aGVHD.
6.Study on Mechanism of HL-60 Cell Apoptosis Induced by DMDAI-L

Objective: To explore the effect and mechanism of DMDAI-L in inducing the HL-60 cells apoptosis. Methods:Caspase-3 activity in HL-60 cells was measured with the enzymatic visible substrate DEVD-pNA. The fluorescence changes of mito-chondrial membrane potential (△Ψm) in HL-60 cells were investigated with the fluorescent probe JC-1. Results:The caspase-3 activ-ity was significantly increased in HL-60 cells after the DMDAI-L treatment at the concentration of 1. 25, 2. 5, 5, 10 and 20μg·ml-1 for 24h(P<0. 05). DMDAI-L could significantly reduce the mitochondrial membrane potential in HL-60 cells. Conclusion: The mechanism of DMDAI-L in inducing HL-60 cells apoptosis may involve the activation or regulation of caspase-3 activity as well as the reduction of mitochondrial membrane potential in HL-60 cells within certain concentration and time range.
7.Temperament and Self-esteem of Children Aged 8~12

ObjectiveTo study the level of temperament and self-esteem of children aged 8~12 in Beijing city,as well as their interaction and influenced factor.Methods203 school children aged 8~12 were assessed with Carey's temperament questionnaire(8~12:MCTQ) and The Self-Esteem Scale(SES).ResultsTemperament and self-esteem of school children aged 8~12 were associated with the parental behavior,especially with work style of their fathers.The rhythmicity was found to be negative correlated to the self-esteem scores,and the intensity of reaction was found to be positive correlated to the self-esteem scores.ConclusionParents' behavior and family environment were important to the temperament and self-esteem of school children.
8.Impact and quality of life of minimally invasive surgery combined with catheter attract for patients with cere-bral hemorrhage

Objective To analyze the impact and quality of life of minimally invasive surgery combined with catheter attract surgery on patients with cerebral hemorrhage.Methods A total of 90 patients with cerebral hemorrhage in our department from February 2013 to De-cember 2014 were randomly divided into invasive group and the control group.Minimally invasive surgery group were given to cerebral hemor-rhage minimally invasive surgery combined with catheter attract surgery,the control group received traditional craniotomy.The postoperative recovery,nerve function scores,quality of life and morbidity between two groups were compared.Results The operation time and edema vol-ume of tissue postoperative 7 days in minimally invasive surgery group was significantly lower than those of the control group.The hematoma clearance rate 1 day after surgery was higher than that of control group,with statistically significant difference(P <0.05).NIHSS scores of two groups were gradually decreased 7 d,15 d,30 d after surgery(P <0.05).NIHSS scores of the minimally invasive surgery group 7 d,15 d, 30 d after surgery were lower than that of control group,with statistically significant difference(P <0.05).The scores of vitality,physiological function,physical function,general health,bodily pain,emotional function,social function,mental health after 6 months of the surgery in the min-imally invasive group was higher than those of control group,with statistically significant difference(P <0.05).The rate of postoperative pulmo-nary infection,electrolyte imbalance,gastrointestinal bleeding in MIS group were lower than those in the control group,with statistically signifi-cant difference(χ2 =5.400 0,4.405 7,4.615 4,P <0.05).Conclusion Minimally invasive surgery combined with catheter attract surgery can improve neurological function and quality of life for patients with intracerebral hemorrhage, and reduce the incidence of complications.
9.Application of role-play test in caring nursing service

Objective To make nurses understand the essential factors of caring nursing service and satisfy the requirement of in-hospital patients. Methods To establish the plan of role-play test(RPT).The roles of patients were played by nurses. They experienced the caring requirement of patients, true feeling, the state of physiology and psychology, etc. Then we discuss and analyze the collected information to consummate measures of caring nufing service. Results The RPT deeped the nurses' understanding about essential factors of caring nursing service. The nursing quality and patients' satisfaction degree were improved compared with those of before(P<0.05).The nursing complaints and service quality were also increased. Conclusion Application of RPT proved to be an effective way to deepen the caring nursing service.
10.Primary methods and applications of image texture research in computer-aided diagnosis

OBJECTIVE: To analyze recent developments in image texture research both from methodology and from medical image analysis. DATA SOURCE: With the key words of "medical image, texture research, image analysis, application", we retrieved PubMed database (http://www.ncbi.nlm.nih.gov/sites/entrez/), ScienceDirecr database (http://www.sciencedirect.com/) from 1983 to 2009 and CNKI database (http://www.cnki.net/) from 2004 to 2009. DATA SELECTION: Original research thesis, and reviews with clear opinion, sufficient data and reliable conclusion were included. Repetitive studies and studies concerning unrelated to the objective were excluded. MAIN OUTCOME MEASURES: A total 104 literatures were selected, including 10 Chinese literatures and 94 English literatures. These literatures were primarily collected by reading titles and abstracts. A total of 33 literatures with unrelated objective, 18 literatures with repetitive studies were excluded. Finally, 53 Chinese and English literatures were included for further analysis. RESULTS: Primary methods used in texture analysis are structural, statistical, model-based and transform-based-method. When we are interested in identifying texture primitives and their distribution to analyze regular texture, structural approaches are suited. Characteristics of texture like smoothness and coarseness are well analyzed by statistical approaches. Model-based-method is based on the construction of an image model that can be used not only to describe texture, but also to synthesize it. Digital features of texture are got by using some signal processing theories in transformation domain. Texture applications have been widely used in medical imaging domain. CONCLUSION: Because of the specific and complication of medical image and texture, not all texture measure can be used for medical image analysis. One of the development directions of medical image texture research is how to integrate and educe advantages of different methods to fully extract texture features and exactly attribute medical image texture and the relation between its changes and pathological state, resulting in an important component of computer-aided diagnosis.
