1.Correlation between Fibrinogen in Pleural Effusion and Pleural Adhesion in Tuberculosis Pleurisy

ObjectiveTo investigate the corelation between fibrinogen in pleural effusion and pleural adhesion in tuberculosis exudative pleurisy.Methods234 cases of primary tuberculosis pleurisy were divided into 3 groups (A、B、C) according to their level of fibrinogen in pleural effusion from low to high. The incidence rates of pleural adhesion were assessed during the course of treatment and after treatment.ResultsThe incidence rate of pleural adhesion in the course of treatment were as those: group A 10.5%, group B 32.3%, group C 54.5%. After treatment, it was as those: group A 10.5%, group B 16.9%, group C 42.4%. Whenever, there was significant difference between group C and group A or B (P<0.05).ConclusionThe level of fibrinogen in pleural effusion may be associated with pleural adhesion, which hinder the recovery of patients.

3.Effect of Specific Growth Rate and Nitrogen Source on the Production of Recombinant Lateolabrax japonicus Growth Hormone by Pichia pastoris

The bioreactor production of recombinant Lateolabrax japonicus growth hormone (rljGH) expressed intracellularly by Pichia pastoris was investigated. A strategy of feeding methanol at the exponential rate was established and the effect of specific growth rate on the rljGH production was examined. The results indicated that the average specific production rate increased and the rljGH production duration decreased as the specific growth rate increased. The maximum specific rljGH production (0.58 mg/g WCW) was achieved at a specific growth rate of 0.029/h. The effect of supplementing ammonium sulfate, peptone and yeast ex- tract on the rljGH production was further investigated. The results indicated that the effects of ammonium sulfate and peptone were not significant. Supplementing yeast extract of 2.5 g/L was advantageous for the rljGH production. The duration of the rljGH production was increased to 23 h from 17 h and the fermenta- tion stability of run-to-run could be improved.
4.Progress in the biomarker discovery for drug-induced liver injury.

The leading cause of drug withdrawal from market and clinical trials failure is drug-induced liver injury (DILI). Varying clinical, histological and laboratory features of DILI, as well as undefined underlying mechanisms, hinder patients to be diagnosed in the early-stage of the disease and receive effective treatments. Conventional indicators, like serum transaminases and bilirubin, have inevitable limitations referring to sensitive prediction and specific detection of DILI. In order to reduce the occurrence of DILI, researchers have attempted to discover potential biomarkers with higher specificity and sensitivity from blood and urine in recent years. This article aims to review recent advances in biomarkers of DILI.

6.Q-analysis real-time ultrasonic elastography quantitative analysis in the diagnosis of breast benign and malignant lesions

Objective To evaluate the value of GE′s Q-analysis real-time ultrasonic elastography quantitative analysis software in the diagnosis of breast diseases and explore the best cutoff point of the breast diseases diagnosis.Methods The elastograms of 71 breast lesions(66 patients)were evaluated by using the GE LOGIQ E9 with the Q-analysis software.All the 71 lesions were diagnosed as malignant and benign lesions by surgery or biopsy.The receiver operating characteristic (ROC)curves of the entirety elasticity rate and local elasticity rate was performed.We compared the areas under ROC curves ( AUC) of the two modalities and confirmed the best cutoff point of the breast diseases diagnosis .We also assessed the sensitivity and specificity of the two modalities .Lastly we assessed the ROC curves with Z test to determine whether the difference between the two modalities was significant .Results Among the 71 leisions (66 patients),there were 37 benign lesions and 34 malignant ones.The elastograms of the leisions showed that the elasticity of 37 benign leisions was good ,the hardness value was low and the texture was even .As for the 34 malignant leisions,the elasticity was bad,the hardness value was high and the texture was uneven .The AUC of the local modality was 0.899 ( >0.5).The AUC of the entire modality,was 0.985 ( >0.5).Both modalities were accurate for the diagnosis of breast diseases .When the cutoff point of entirety elasticity rate was set as 3.60,the sensitiveness was 82.4%and the specificity was 86.5%.When the cutoff point of local elasticity rate was set as 2.60,the sensitiveness was 97.1% and the specificity was 91.9%.The difference between the two modalities was significant (Z =2.621,P=0.0088).Conclusions Q-analysis real-time ultrasonic elastography quantitative analysis is able to evaluate the rigidity and texture of tissue and is valuable to distinguish benign breast lesions between malignant breast lesions .The sensitiveness and specificity of local modality are higher than that of entirety modality .
7.A qualitative research on the behaviors caused by fear of hypoglycemia in type 2 diabetic patients

Objective To explore the behaviors caused by fear of hypoglycemia in type 2 diabetic patients, and deeply analyze the causes of these behaviors, providing help for medical staff to fully understand the inner demands of diabetic patients with hypoglycemia, so as to manage the blood glucose effectively, laying the foundation of Hypoglycemia Fear Survey-behavior scale suitable for domestic type 2 diabetic patients. Methods The interpretative phenomenological analysis method was used guided by the knowledge-attitude-belief-practice and social cognitive theory, 33 patients with hypoglycemia from 5 hospitals with different levels were interviewed by semi-structured interview. Results Four themes were extracted, including excess sensitivity, caution, avoidance and compromise. The theme excess sensitivity contained three kinds of behaviors, that were meal with sensitive alertness, drug sensitivity and excessive response to the hypoglycemia, while the theme caution contained two kinds of behaviors, that were strive to blood sugar stable and must prepare food. Conclusions The results show that the fear of hypoglycemia is serious in type 2 diabetic patients, and they make many behavior change to avoid it, affecting their quality of life and glycemic management. Medical staff should attach importance to the inner feelings and behaviors of diabetic patients with hypoglycemia, and provide guidance and support after comprehensive assessment, working together with the patient to realize the effective management of blood glucose.
8.Effect of Melatonin on CD4~+CD_(25)~+ Regulatory T Cell and Airway Inflammation in Asthmatic Rat

Objective To explore the effect of Melatonin(MT) on CD4+CD25+ regulatory T cell (CD4+CD25+Tr)and airway inflammation in asthmatic rat.Methods Thirty-two SD rats were randomly divided into 4 groups,8 rats in each group.Asthmatic group:rats were immunized on day 1 and 7 by intraperitoneal inject of mixture of ovalbumin(OVA) and aluminumhydroxide.From day 14,the animals were allenged with aerosolized OVA for 20 min per day for 7 consecutive days.MT group:OVA-sensitized rats were injected intraperitoneally with 0.1 mg/kg MT 30 min before each OVA challenge.Dexamethasone group:OVA-sensitized rats were injected intraperitoneally with 0.5 mg/kg Dexamethasone 30 min before each OVA challenge.Control group:OVA for inhalation and MT for intraperitoneal injection was replaced with saline.After the last challenge,peripheral blood was stained to count the percentage of eosinophil(EOS).Then the rats were lavaged and total leukocytes counts in bronchoalveolar lavage fluid(BALF) were performed after staining with Wright-Giemsa staining.The EOS counts around the airway was counted after the histological section of lung staining with hematoxylin and eosin staining.The serum level of immunoglobulin E(IgE) was detected by immunoenhancement.The change of CD4+CD25+Tr was assessed with flow cytometry.SPSS 10.0 software was applied to analyze data. Results In asthmatic rats,the CD4+CD25+ Tr/ CD4+T cells ratio had significant negative relationship with the EOS counts around the airway and the total leukocytes counts in BALF (r=-0.73 P0.05).There was a significant decrease in the percentage of the eosinophils in peripheral blood,the eosinophil counts around the airway,the total leukocytes counts in BALF and the serum level of IgE in MT group compared with asthmatic group (Pa

10.A Modified Method for Isolating DNA from Fungus

Genomic DNA of two fungi Thamnidium elegans and Umbelopsis isabellina were extracted with an amended Cetyltrimethyl Ammonium Bromide (CTAB) method. This modified method uses repeated freezing in liquid nitrogen and thawing with combination of shocking with glass beads to replace of the tra- ditional method. Quality and concentration of DNA extracted by the modified methodwere tested. Compared with the traditional method, higher yield and purity of genomic DNA were obtained with less amount ofmy- celium. The result indicted that this is a simple and highly efficient method, which is suitable to treat many samples at one time and for basic molecular experiments, such as restriction endonuclease reaction and PCR.
