2. Optimization of component compatibility of Naomaitong for anti-cerebral ischemia treatment based on artificial neural network

OBJECTIVE: To optimize the component compatibility of Naomaitong for anti-cerebral ischemia treatment based on artificial neural network. METHODS: The five effective parts in Naomaitong were divided into ten groups by uniform design. Except the rats in the sham groups, all rats were subject to right middle cerebral artery occlusion (MCAO) with the suture-occluded method by Longa. The effect of Naomaitong was evaluated based on the bulk density of necrotic zone and infarction area percentage. The artificial neural network model was established for pharmacodynamic prediction. RESULTS: The model established in this study could predict the actions of different drug combinations. The best effect was obtained by the following formula: total anthraquinones of rhubarb (TAR) 180 mg·kg-1, total saponins of Ginseng (TSG) 70 mg·kg-1, total flavonoids of Pueraria (TFP) 450 mg·kg-1, total phenolic acid of Ligusticum wallichii (TPLW) 27 mg·kg-1, rhizome oil (RO) 110 μL·kg-1. CONCLUSION: Different compatibilities of Naomaitong are all effective for the treatment of ischemia in rats. Uniform design combined with ANN can be a more efficient method to realize dose optimization for Naomaitong prescription.
3.Clinical application and assessment of the Chinese version of Fatigue Severity Scale in stroke patients

Objective To test the reliability and validity of the Chinese Version of the Fatigue severity scale (FSS)in patients with cerebral infarction.Methods The FSS was translated into Chinese language and the reverse translation was done by several experts.Validity,dimensionality,and reliability tests were implemented in 153 cases of cerebral infarction.Results One component was extracted in factor analysis,and the total cumulative contribu- tion was 64.982%.Based on the Mokken Scale analysis for Polytomous items analyses,the scale was found to be u- nidimensional and scale H is 0.6125,Cronbach?of the scale is 0.9287.Conclusion The psychometric proper- ties(reliabilities and validities)of FSS Chinese Version was satisfactory and seemed to be adaptable to Chinese cere- bral infarction patients.
4.Research and analysis on corneal endothelial cell morphology and corneal thickness in patients with diabetics

AIM: To assess the impact of diabetes on corneal endothelial cells through the quantitative analysis of corneal endothelial cell morphology for patients with diabetics.
METHODS: The corneal thickness and endothelial cell morphology of 360 eyes of 299 cases were detected using full automatic corneal endothelial cell analyzer. The normal control group included 175 eyes of 148 cases, and there were 185 eyes of 151 cases for the patients with diabetes, 110 eyes of 92 cases for the non-proliferating phase group and 75 eyes of 59 cases for the proliferating phase group. The average density of central corneal endothelial cells, proportion of hexagonal cells, coefficient of variation and corneal thickness were compared among groups, and then the statistical analysis was conducted.
RESULTS: Compared with the cornea of the normal group, in the diabetes group, the coefficient of variation of corneal endothelial cells and central corneal thickness increased, while the average density of central corneal endothelial cells and proportion of hexagonal cells decreased, showing a significant difference (P<0. 05). Compared with the cornea of non-proliferating phase group, in the proliferating phase group, the density of central corneal endothelial cells decreased, the coefficient of variation of corneal endothelial cells increased, while the proportion of hexagonal cells decreased with a significant difference (P<0. 05), and the central corneal thickness increased, showing no significant difference(P>0. 05).
CONCLUSION: Compared with the cornea of normal control group, in the diabetes group, the corneal endothelial cells show abnormal morphology, which aggravates with the severity of lesions, especially for the significant changes in the coefficient of variation and the proportion of hexagonal cells. As a result, the corneal resistance to damage in patients with diabetes will decrease.
5.Simultaneous Determination of 1-Chloroanthracene and9-Bromoanthracene by Using Matrix Isopotential SynchronousFluorescence Spectrometry Combined with Derivative Technique

Matrix isopotential synchronous fluorescence spectrometric (MISF) method combined with derivative technique has been proposed to determine two spectrally-badly-overlapped anthracene derivatives, 1-chloroanthracene and 9-bromoanthracene,simultaneously. An optimal scanning route of MISF spectra was chosen. By using the novel method, 1-chloroanthracene and 9-bromoanthracene have been determined directly and simultaneously without tedious pre-separation in its mixture.The recoveries of 1-chloroanthracene and 9-bromoanthracene for the analysis are 83.5%~94% and 90.0%~94.0% respectively and their limits of detection are 0.69μg/L and 10μg/L respectively.
7.Human bone marrow-derived mesenchymal stem cells differentiate into retinal cells in vitro

Aim To study the differentiation of human bone marrow-derived mesenchymal stem cells ( HM-SCs) into retinal cells in vitro. Methods HMSCs were isolated from human bone marrow after Ficoll den-sity gradient centrifugation. The adherent cells after at least 5 passages were used for study. Immunopheno-type of the cells was analysed by flow cytometer, and cellular differentiation was identified by immunofluores-cence labeling technique. Results The target cells derived from human bone marrow adhered to the plate with fibroblastic-like morphology, whose surface mark-ers were similar to mesenchymal stem cells. Major cells
were positive for CD90 , CD44 , CD147 , while they were all negative for CD34, CD45, HLA-DR. In the differentiation study, HMSCs cultured in induced me-dium can differentiate into nestin ( neural stem cell ) -positive cell, GFAP ( glial fibrillary acidic protein ) -positive glial cells and retina-specific neurons express-ing Rhodopsin with CD90 ( mesenchymal stem cells )-negative. Conclusion HMSCs have the ability to dif-ferentiate into retinal neural cells in vitro.
8. Compatibility study of five rhubarb aglycones in treatment of cerebral ischemia based on support vector machine

Objective: To optimize the compatibility of five rhubarb aglycones (aloe-emodin, rhein, emodin, rhubarb, and physcion) in treatment of cerebral ischemia. Methods: The rats were divided into 14 groups based on uniform design. The rats were subjected to right middle cerebral artery occlusion (MCAO) with the suture-occluded method by Longa. The effect of rhubarb aglycone was evaluated using neurological symptoms and infarction area percentage as indexes. The support vector (SVM) machine model was established for pharmacodynamics prediction of rhubarb aglycone. Results: The model established in this study could predict the drug actions of different combinations. The best compatibility was aloe-emodin 6.6534 mg/kg, rhein 26.0008 mg/kg, emodin 11.0042 mg/kg, chrysophanol 3.8414 mg/kg, and physcion 3.8620 mg/kg. Conclusion: Different compatibilities of rhubarb aglycone are effective in the treatment of ischemia to rats. Uniform design combined with SVM method can be more efficient to realize the dose optimization for rhubarb aglycone prescription.
9.Isolation,Identification and Bioactivity Screening of Streptomyces pseudogriseolus Associated with Marine Sponge Hymeniacidon perleve

An actinomycete B37 was isolated from an intertidal marine sponge Hymeniacidon perleve, which has strong activity against Gram positive bacteria and moderate activity against tumor cells. The mycelium and spore morphology, physiological properties and 16SrDNA sequence suggested that B37 is Streptomyces pseudogriseolus. The fermentation conditions of this strain were investigated for the biosynthesis of bioactive metabolites.
10.Intervention of empowerment with multi-stage change theory on diet behavior for the influence of metabolic indices in patients with diabetes

Objective To investigate the influence of different educational methods on the metabolic indices in patients with diabetes .Methods A total of 218 patients with diabetes in the First Affiliated Hospital of Guangxi Medical University were ran-domized divided into two groups .Different intervention ways were used :patients in the control group received conventional educa-tional methods ,while patients in experiment group received the educational methods combined with empowerment and multi-stage theory of change .The C-DES-SF scale scores and the HbA1c ,FPG ,2 h PG ,TC ,TG ,LDL ,HDL indices was compared between the two groups .Results Indices and empowered ability of HbA1c ,FPG ,2 h PG ,TC ,LDL were changed in accordance with time .As compared with control group ,the educational method in intervention group had better change of FPG ,2 h PG ,TC ,LDL(P< 0 .05) . Indices and empowered ability of HbA1c ,FPG ,2 h PG ,TC ,LDL had inter-action with time ,and indexes in intervention group were better than those in the control group(P< 0 .05) .At the 3rd and 6th month after invention in the intervention group ,patients in movement stage and movement maintaining stage were higher than those in control group(P< 0 .05) .Conclusion Well correction of bad diet behaviors by application of educational methods of combined with empowerment and multi-stage theory of change for the type 2 diabetes patients would help the patients to improve the ability of self-care and control the level of metabolic indices .
