1.Application of Mutislice CT angiography in cervical vascular diseases

Objective To discuss the clinical value of Mutislice CT angiography (MSCTA)in cervical vascular diseases. Methods 60 patients highly suspected suffering cervical vascular diseases,were implemented 16-slices spiral CT angiography of cervical vascular,and blood vessel reconstruction and analysis were done subsequently. Results 43 cases with cervical vascular abnormality were presented in the 60 patients,who were implemented MSCTA. Positive ratio was 71.7%. 171 pieces vessel stenosis were detected among 240 pieces carotid artery and vertebra artery, and 4 cases carotid artery aneurysm and 1 case AVM were found also. Conclusion MSCTA is a convenient and shortcut modality to inspect cervical vascular diseases, and the blood vessel can be observed 3-dimensionally and in any direction in high quality reconstruction images.
2.Analysis of microvessel density in pterygium tissue with corneal laser confocal microscopyc in vivo and immunohistochemistry in vitro

BackgroundPterygium is an ocular surface disease of abnormal cell proliferative kind and angiogenesis plays an important role in its development and recurrence.Several anti-angiogenic therapies have been used to treat pterygium,but there very few studtes for the in vivo observation of the microvessles in pterygium.ObjectiveThis study was to observe angiogenesis in pterygium with a high-resolution confocal microscope in vivo and to perform immunohistochemical study in vitro.MethodsA prospective case-controlled study was designed.Twenty eyes of 20 consecutive patients with primary pterygia and 20 age- and sex-matched patients with inner eye diseases and strabismus with normal conjunctiva were enrolled in this study.An in vivo confocal microscopy imaging system (Heidelberg Retina Tomograph Ⅱ Rostock Cornea Module) was used to collect microvascular pictures from the anterior part of pterygia and normal nasal conjunctiva of controls,and then immunochemistry was performed to examine the expression of CD31 in microvessel in vitro.The vascular density values were compared between these two groups.The correlation of vascular density values between in vivo Heidelberg Retina Tomograph and in vitro immunohistochemistry was calculated.Written informed consent was obtained from pationts before any examination and surgery.ResultsUnder the in vivo confocal microscope,the microvessel density was (8929±2993) μm/mm2 and (4202 ±692)μm/mm2,respectively,in pterygium and the normal conjunctiva group with a statistically significant difference between them (t =6.881,P<0.01 ).Immunochemistry revealed that the expression of CD31 to measure vascular density was ( 21.00 ± 4.06/400 × field ) and ( 6.07 ± 1.75/400 × field ) in pterygium and the normal conjunctiva group,showing significant difference (t =12.312,P<0.01 ).Positive correlations were found in the vascular density values between in vivo corneal laser confocal microscopy examination and in vitro immunochemistry examination in both the pterygium group and normal conjunctiva group (pterygium group:r=0.649,P<0.01 ;normal conjunctiva group: r=0.572,P<0.01 ) ConclusionsIn vivo confocal microscopy imaging is superior to in vitro immunochemistry in evaluating the microvessel of pterygium.The results of this study offer a new way index for further investigation of the biological behavior of pterygium and its mechanism.

Objective To know the antitumor effect and value of vinorelbine in chemotherapy with relapsed breast cancer.Methods To receive breast cancer tissue from primary and relapsed patients by surgery,then to obtain mammary cancer cells by collagenase.The antitumor effect of 5 chemotherapy drugs with breast cancer cells was measured by culture in vitro and MTT.Results With primary cancer cells from primary breast cancer,the antitumor effect of paclitaxel and docetaxel(sensitivity is 91.04%,92.54% respec- tively) is better than adriamycin,epirubicin and gemcitabine(sensitivity is 73.13%,74.63%,71.64% re- spectively;P
4.Effect of Neoadjuvant Chemotherapy Treatment on Prognosis of Patients with Advanced Gastric Cancer: a Retrospective Study.

OBJECTIVETo evaluate the prognostic effects of neoadjuvant chemotherapy (NAC) in patients with local advanced gastric cancer.
METHODSWe retrospectively analyzed prognosis in 191 patients with advanced gastric cancer, of whom 71 were treated with NAC and 120 received surgery only between February 2007 and July 2013. Postoperative complication rate was recorded. Survival by clinicopathological features, pathological T and N stages, and histopathological tumor regression was retrospectively compared between the two groups.
RESULTSAccording to Response Evaluation Criteria in Solid Tumors, none of the 71 patients in the NAC followed by surgery group showed complete response, 36 showed partial response, 25 had stable disease, and 10 had progressive disease. The chemotherapy response rate was 50.7%; the disease control rate was 85.9%. Grade 3/4 adverse events were seen in less than 20% patients, with acceptable toxicities. No difference was found in the overall postoperative complication rates between the two groups (7 versus 22 cases, P=0.18). Median survival time was significantly different, at 54 months in the NAC combined with surgery group and 25 months in the surgery-only group (P=0.025).
CONCLUSIONIn patients with operable gastric adenocarcinomas, NAC can significantly improve overall survival without increasing surgical complications.

5.Research of autophagy activity between rat bone marrow mesenchymal stem neural differentiation.

OBJECTIVETo study the autophagy activity between rat bone marrow stem cells (BMSCs) neural differentiation in order to explore the mechanism involve in this process.
METHODSBMSCs were passed by 3 generation, then was induced with the revulsant 2% (DMSO) + 200 µmol/L (BHA), NSE expression was detected by immunocytochemical stain, the mRNA expression of autophagy associated genes L3B, Beclinl, Atg5, Atg7, Atg10 were detected by RT-PCR, the autophagy protein LC3B was examined by Western blot and flow cytometry analysis.
RESULTSBMSCs were passed by 3 generation, the purity of BMSCs could reach more than 90%, the morphology of cells were like fibroblasts, after the revulsant 2% DMSO + 200 µmol/L BRA induced, cells were extended long neurites, like nerve cells, positive rate of NSE staining was (83±5) %, RT-PCR results showed that the expression of autophagy associated genes LC3B, Beclinl, Atg5, Atg7 Atg0 were rised after BMSCs neural differentiation, Western blot analysis showed that the LC3B-II protein expression was increased after neural differentiation and the MFI of L3B was highten by flow cytometry.
CONCLUSIONAutophagy is increased after rat BMSC neural differentiation.

Natural products are important sources for the discovery of anti-tumor drugs. Evodiamine is the main alkaloid component of the traditional Chinese herb Wu-Chu-Yu, and it has weak antitumor activity. In recent years, a number of highly active antitumor candidates have been discovered with a significant progress. This article reviews the research progress of evodiamine-based antitumor drug design strategies, in order to provide reference for the development of new drugs with natural products as leads.
7.A two-year animal experimental study on the pathological effects of Helicobacter pylori on liver tissues.

OBJECTIVETo observe whether H. pylori administered orally in mice could arrive in their livers after a long-term infection, leading to active inflammation and even causing HCC as an independent etiological factor.
METHODSTwenty C57BL/6 mice were orally administered H. pylori SS1 and kept for 24 months (experimental group) along with 13 mice which served as blank controls (control group). H. pylori colonization and pathologic consequences were studied in the livers and gastric tissues of the mice. The bacterial DNA extracted from liver tissues was examined by nested PCR for H. pylori 16S rRNA genes. 16S rRNA PCR amplicons were sequenced and compared with sequencing results of 16S rRNA PCR amplicons of the bacteria cultured from gastric mucosa and compared with that of the inoculated H. pylori SS1.
RESULTSOf the 20 mice in the experimental group, H. pylori was found in the gastric mucosa of 12, and in 11 of them pathological gastric lesions were found, including one with gastric lymphoma. H. pylori were found in the livers of 7 mice. Liver lesions, one with mild inflammation, 3 with inflammation and fibrosis, 2 with inflammation, fibrosis and hepatocyte hyperplasia with atypia were found in 6 of them. No liver lesions were found in the mice of the control group. In the mice of the experimental group no liver lesions were found in those mice with no H. pylori in their gastric mucosae. Sequencing results of 16S rRNA PCR products of the liver showed 100% homogeneity with the cultured H. pylori from gastric mucosa and the administered H. pylori SS1.
CONCLUSIONTwo years after oral administration of H. pylori to C57BL/6 mice, gastric mucosal lesions and liver lesions, including inflammation, cirrhosis and hepatocyte hyperplasia with atypia were found in those animals.

8.Effect of DNA methyltransferases 1 inhibition on proliferation, invasion, and metastasis in ACC-M line.

OBJECTIVETo investigate the effect of DNA methyltransferases 1 (DNMT-1) inhibition on the ACC-M cells in vitro and in vivo and discuss the role of DNMT-1 in the development, invasion and metastasis of salivary adenoid cystic carcinoma (SACC).
METHODSACC-M cells of stable DNMT-1 inhibition were established in a previous research. In vitro, the growth and invasion of ACC-M cells which stably inhibited DNMT-1 were detected and analyzed by methyl thiazolyl tetrazolium (MTT) growth curve, flow cytometry, plating efficiency and invasion assay. In vivo, the growth and metastasis of ACC-M cells which persistently inhibited DNMT-1 were observed and analyzed by subcutaneous injection and tail vein injection into the nude mice.
RESULTSIn vitro, the doubling time [(34.7 +/- 2.1) h], S phase fraction [(17.4 +/- 1.7)%], plating efficiency [(43.0 +/- 1.3)%] of ACC-M cells was significantly different from those of blank [(26.2 +/- 3.1) h, (31.5 +/- 2.0)%, (71.0 +/- 4.7)%], empty load control [(28.4 +/- 3.9) h, (39.0 +/- 2.0)%, (66.0 +/- 5.2)%], P < 0.05, and the invasion ability was not significantly different among these groups (P > 0.05). In vivo, the subcutaneous tumor forming rate (6/10), volume [(2.18 +/- 0.83) mm(3)], weight [(0.0156 +/- 0.0046) g] of ACC-M cells was also significantly lower than that of blank [10/10, (155.44 +/- 1.67) mm(3), (0.0724 +/- 0.0157) g], empty load control [10/10, (147.46 +/- 1.73) mm(3), (0.0729 +/- 0.0177) g], P < 0.05, but the rate of lung metastasis was not significantly different among these groups (P > 0.05), and the masses (2.0 +/- 0.5), diameter (70.0 +/- 20.3) microm of ACC-M cells was significantly lower than that of blank [(28.0 +/- 5.5), (195 +/- 25.4) microm], empty load control [(27.0 +/- 4.5), (190.0 +/- 19.9) microm], P < 0.05.
CONCLUSIONSInhibition of DNMT-1 is able to inhibit the proliferation and metastasis of ACC-M cells in vitro and in vivo.

9.Diverse components of voltage-gated calcium channel currents in primary cultured hippocampal neurons.

The present study was to record and separate the voltage-gated calcium channel currents (VGCCs) in primary cultured hippocampal neurons. An improved method is described for efficient and stable recording of VGCCs from primary cultured hippocampal neurons. The procedure allows the obtained hippocampal neurons overcome the defects from acutely isolated hippocampal neurons and currents do not rundown apparently after 20 min of membrane rupture. Using whole-cell configurations of patch clamp technique, it is shown that the method does not damage the neuron membrane electrical properties, and successfully records a variety of VGCCs components in hippocampal neurons (such as L, N, P/Q, T, etc.).
