2.The increase of micronuclei frequencies of peripheral blood lymphocyte in plywood workers exposed accumulatively to formaldehyde.

OBJECTIVETo investigate the effects of occupational exposure to formaldehyde on the micronuclei frequencies in peripheral blood lymphocytes of workers.
METHODSTwo hundred thirty six plywood workers were divided into 3 exposure groups (low, middle and high) according to internal exposure biomarker (formaldehyde human serum albumin conjugate, FA-HSA), which was detected by ELISA. The concentrations of formaldehyde (FA) in air of two workshops were measure using the high performance liquid chromatography. Cytokinesis-block micronucleus (CBMN) test was used to detect the micronuclei frequencies of peripheral blood lymphocyte in 236 workers.
RESULTSThe average concentrations of FA in the low and high exposure workshops were 0.58 +/- 0.20 and 1.48 +/- 0.61 mg/m3, respectively, there was significant difference (P < 0.01). The average concentrations of serum FA-HAS of workers in two workshops were 69.22 +/- 15.37 and 136.29 +/- 89.49 pg/ml, respectively, there was significant difference (P < 0.01). The results of CBMN test showed that the micronucleus frequencies in low, middle and high exposure groups were 1.94 +/- 1.72, 2.10 +/- 1.92 and 2.10 +/- 1.70 per thousand, respectively, there were no significant differences between groups. However, the micronucleus frequencies in accumulative low, middle and high exposure groups were 1.36 +/- 1.36, 2.31 +/- 1.81 and 2.49 +/- 1.92 per thousand, respectively, there were significant differences between different accumulative exposure groups (P < 0.01). The results of correlation analysis indicated that there was a positive correlation between accumulative exposure levels and micronucleus frequencies (r(s) = 0.321, P < 0.01). The accumulative exposure doses may be a risk factor for high micronucleus frequencies in workers exposed to FA (P(trend) = 0.002).
CONCLUSIONFA-HSA levels can serve as an internal exposure biomarker for assessing the exposure level of workers exposed to FA. Accumulative formaldehyde exposure resulted in an increase of micronuclei frequencies of peripheral blood lymphocyte in plywood workers.

3.Mechanism of coronary artery constriction induced by 5-HT

AIM:To investigate the possible mechanism of coronary artery contraction induced by 5-hydroxytryptamine(5-HT).METHODS:Isolated coronary artery rings were obtained from male Wistar rats,and the vas-cular tension meter was used to determine the tension of the coronary artery rings.The effects of inhibitors of different sig-naling pathway on vascular contraction tension induced by 5-HT were observed.RESULTS:Firstly,we found that 5-HT2A receptor antagonist sarpogrelate(1 μmol/L)completely eliminated the coronary artery contraction induced by 5-HT.Phos-pholipase Cβ(PLCβ)inhibitor U73122(10 μmol/L and 50 μmol/L), Rho-related protein kinase inhibitor Y-27632(3 μmol/L and 10 μmol/L)and protein kinase C δ subunit(PKCδ)inhibitor rottlerin(3 μmol/L and 10 μmol/L)signifi-cantly inhibited the contraction of coronary artery ring caused by 5-HT(P<0.05).In addition, compared with the un-treated group,vascular contraction tension induced by 5-HT was also decreased significantly by L-type calcium channel (Cav1.2)blocker nifedipine(1 μmol/L), store-operated Ca2+entry(SOCE)inhibitor SKF96365(10 μmol/L and 30 μmol/L)and 2-aminoethoxydiphenyl borate(2-APB,50 μmol/L and 100 μmol/L)(P<0.05).At the same time,5-HT also induced vasoconstriction after treated with nifedipine(1 μmol/L)Kerbs-Henseleit(K-H)liquid without calcium (P<0.05).CONCLUSION:5-HT activates 5-HT2Areceptor induced coronary artery contraction,possibly related to the PKC/Rho kinase signaling pathway and calcium regulation.
4.Molecular Mechanism of "Transmission Between Lung and Brain" of Influenza and Intervention Effect of Maxing Shigantang Based on JAK1/STAT1 Signaling Pathway

ObjectiveTo explore the molecular mechanism of "transmission between the lung and brain" of influenza based on Janus kinase 1/signal transducer and activator of transcription 1(JAK1/STAT1) signaling pathway and further investigate the intervention effect of Maxing Shigantang (MXSGT). MethodA total of 100 SPF BALB/c mice were randomly divided into a normal group,a model group,an oseltamivir group (21.63 mg·kg-1·d-1),an antiviral granules group(3.9 g·kg-1·d-1), and an MXSGT group(6.05 g·kg-1·d-1), with 20 mice in each group. The pneumonia model was induced in mice except for those in the normal group by intranasal infection of influenza A virus(IAV). Twenty-four hours after modeling,mice were treated with corresponding drugs, while those in the normal group and the model group received the same amount of normal saline by gavage, once a day for 3 and 7 days. The pathological changes in the lung and brain were observed by hematoxylin-eosin(HE)staining. The mRNA expression of IAV nucleoprotein(NP),JAK1, and STAT1 in the lung and brain was detected by real-time quantitative polymerase chain reaction(Real-time PCR), and the protein expression of JAK1 and STAT1 in the lung and brain was detected by Western blot. Immunohistochemical method was used to detect the expression of phosphorylated(p)-STAT1 in the lung and brain tissues, and enzyme-linked immunosorbent assay(ELISA) was used to detect the serum levels of interleukin-1β(IL-1β) and interleukin-10(IL-10). ResultCompared with the normal group, the model group showed obvious pathological changes in the lung tissues and cerebral cortex, increased relative mRNA expression of IAV NP in the lung (P<0.01), elevated mRNA and protein expression of JAK1 and STAT1 in the lung and brain tissues (P<0.05,P<0.01),up-regulated expression level of p-STAT1 in lung tissues and cerebral cortex (P<0.05,P<0.01), and increased serum level of IL-1β (P<0.05). Compared with the model group, the MXSGT group showed alleviated pathological damage to lung tissues and cerebral cortex, decreased relative mRNA expression of IAV NP in lung tissues(P<0.01),reduced mRNA and protein expression levels of JAK1 and STAT1 in lung tissues and brain tissues(P<0.05,P<0.01), and increased serum level of IL-10(P<0.01). ConclusionThe abnormal activation of the JAK1-STAT1 signaling pathway may be one of the molecular mechanisms of "transmission between the lung and brain" of influenza. As an effective compound prescription against the influenza virus,MXSGT can alleviate the pathological damage of brain tissues in mice infected with IAV by regulating the level of cytokines mediated by this pathway.

With the development of small-molecule immunotherapy drugs, its combination with the programmed cell death ligand 1/programmed cell death protein 1 (PD-L1/PD-1) antibodies would provide a new opportunity for cancer treatment. Therefore, targeting PD-L1/PD-1 axis by small-molecule drug is an attractive approach to enhance antitumor immunity and considered as the next generation of tumor immunotherapy. Acetyl-CoA carboxylase (ACC) is the rate limiting enzyme of fatty acid synthesis pathway. Studies have shown that ACC1 is implicated in a variety of metabolic diseases and cancer. However, the role and mechanism of action of ACC1 in clear cell renal cell carcinoma (ccRCC) have not been reported. In this study, 786-O and Caki-1 clear cell renal carcinoma cells were used as research objects to investigate the effect of abnormal expression of ACC1 on their proliferation and unravel the underlying mechanism. Red oil-O-staining results showed that the lipid content of 786-O and Caki-1 cells was significantly higher than that of human kidney 2 (HK2) cells. By searching TCGA database, we found that the expression of ACC1 proteins in ccRCC was significantly higher than that in normal renal tissues (P < 0.001). Plus, ACC1 protein expression in all clinical TNM stages was significantly higher than that in normal tissues, and the higher the expression of ACC1, the higher the pathological grade. Furthermore, high expression of ACC1 mRNA is positively correlated with poor prognosis in ccRCC patients. Western blotting analysis showed that the expression of ACC1 in 786-O and Caki-1 cells was significantly higher than that in HK2 cells. The results of red oil-O-staining showed that knocking down ACC1 could significantly reduce the lipid content of 786-O and Caki-1 cells. The results of CCK-8 assays and clonogenicity analysis showed that knocking down ACC1 could significantly reduce the proliferation and colony forming ability of 786-O and Caki-1 cells. In this paper, the key technical problems in the research and development of famous classical formulas are analyzed. Firstly, the puzzled problem for a long-time, which is conversion relationship from medicinal metrology of Han dynasty (HD) to that of modern (gram,g), is comprehensively expounded that one Liang (两) of HD=3 g is more appropriate. Secondly, the model and principles of quality consistency evaluation are given for the transformation from the quality of authoritative basic sample prepared by casserole (ABS-C) to the quality consistency in Laboratory process, pilot-scale process and industrial production. The consistency evaluation model is ξABS-X=K1(Q1ABS-X/Q1ABS-C)+K2(Q2ABS-X/Q2ABS-C)+……+Ki(QiABS-X/QiABS-C)=∑Ki(QiABS-X/QiABS-C)(i=1,2,3……n). In the formula, ABS-X means laboratory reference sample ABS-C (ABS-L), pilot-scale ABS-C (ABS-mP) or industrial production ABS-C (ABS-P), ξABS-X means the quality consistency rate or similarity degree of ABS-L, ABS-mP and ABS-P processes with ABS-C, Ki means the weight of each quality evaluation index (i), QiABS-X is the data of i in ABS-L, ABS-mP, ABS-P samples, and QiABS-C is the data (or mean) of i in ABS-C sample. Thirdly, in order to control the quality of the herbal medicines whose active ingredients were unknown, their chemical constituents should be studied deeply, and if necessary, the bioassay research should be carried out according to the main efficacy or indication of famous classical formulas. Finally, for the special processing of some herbal medicines, it is difficult to formulate the processing method, technology and standard of prepared slices. It is suggested that the scientific connotation and historical evolution of the special processing method should be thoroughly sorted out, and its technological characteristics are summarized, the modern processing technology and production processes are simulated, and then the corresponding processing methods and quality standards are formulated.
