2.The expression and significance of human macrophage metalloelastase in both gastric cancer cell lines and gastric cancer tissues

Objective To investigate the expression of human macrophage metalloelastase(HME) both in gastric cancer cell lines and gastric cancer tissues,and to find the role of HME in gastric carcino genesis.Methods Fifty eight patients who were operated in our hospital during April to Aug.2003 were enrolled.The samples taken from cancer,paracancer or normal tissues of these patients and cancer cell lines(MGC-803,SGC-7901,AGS)were detected for HME protein and HME mRNA expressions by Western blot and immunohistochemistry,RT-PCR and fluorescence quantitative PCR,respectively. Results Both HME mRNA and HME protein expressions were found in all three gastric cancer cell lines.The expressions of HME mRNA and HME protein in gastric cancer tissues was increased signifi- cantly compared with that in normal tissues(P0.05).Conclusions The increased HME expression in gastric cancer tissures compared with normal tissue indicate that HME may be a potential tumor marker for gastric cancer.
5.Experimental study on the theraputic effect of crescent Euphorbia on lewis lung cancer in mice.

OBJECTIVETo observe the inhibitory effects of crescent euphorbia on tumor growth, immunoregulation, the side effect on the mice blood system, cell cycle and to investigate its effect on apoptosis of tumor cells.
METHODCrescent euphorbia was administered gastrally to C57BL/6J mouse implanted with Lewis lung cancer for 12 days. Inhibition on tumor growth, immunoregulation,the side effect on the mice blood system and cell cycle were observed and its effect on apoptosis of lung cancer cells were investigated.
RESULT(1) Inhibitory rates of crescent euphorbia at doses of 7.5, 15, 30 g x kg(-1) and 60 g x kg(-1) were 0.61%, 16.93%, 32.81% and 58.26% respectively, significant differenel with those of controls at doses of 30 g x kg(-1) and 60 g x kg(-1)(P < 0.05). (2) Although no effect on thymus index in normal mouse at all dose of crescent euphorbia was found, spleen index in tumor-bear mice was reduced significantly at dose of 60 g x kg(-1)(P < 0.05). (3) No side effect on the miceblood system was found. (4) The percentage of S phase of cell cycle was increased in the group of crescent euphorbia at dose of 30 g x kg(-1), 60 g x kg(-1) and the apoptotic rate was (16.43 +/- 18.69)% and (24.37 +/- 15.48)% by flow eytometry (P < 0.05). The apoptotic index (AI)of these two groups were (4.00 +/- 7.50)% and (5.93 +/- 5.96)% by TUNEL, however apoptosis was found in the group at doses of 60 g kg(-1) (P < 0.01).
CONCLUSIONCrescent euphorbia can inhibit the growth of Lewis lung cancer in mice, no side effection for the mice blood system and the effect might be associated with induction of apoptosis on cancer cells.

7.The clinical significance of CT angiography in the diagnosis of arterioportal shunts associated with hepatocellular carcinoma

Objective To evaluate the clinical significance of CT angiography(CTA)in the diagnosis of arterioportal shunts(APS)associated with hepatocellular carcinoma(HCC).Methods One hundred and twenty-seven consecutive HCC patients accepted both dynamic enhancement CT and DSA examinations.The interval between CT and DSA exam was from 3 to 15 days.Based on transverse CT images in hepatic artery phase,CTA was performed for all the patients.By contrast with DSA results,the capabilities of transverse CT and transverse images combined with CTA in APS diagnosis were analyzed. Results In all 127 HCC cases,52 cases with APS were confirmed by DSA(40.94%),33 with central type of APS and 19 with peripheral type.Diagnostic sensitivity of APS based on transverse CT and combined CTA with transverse CT images were both 94.23%(49/52).However,specificity was 84.00%(63/75) and 97.33%(73/75),respectively,accuracy was 88.19%(112/127)and 96.06%(122/127),the predictive value of positive cases was 80.33%(49/61)and 96.08%(49/51),and the predictive value of negative cases was 95.45%(63/66)and 96.05%(73/76).Combined with CTA,false positive cases of 4 central type of APS and 6 peripheral type of APS were excluded which were demonstrated by transverse CT images.By contrast with DSA,the coincidence rate of the type of APS diagnosed by transverse images combined with CTA was 88.46%(46/52),including 90.91%(30/33)of central type of APS and 84.21%(16/19)of peripheral type.The supplying arteries of central type of APS were intuitively displayed by CTA in 23 cases,19 from proper hepatic artery and 4 from gastro-duodenal artery.Conclusion CTA techniques based on the dynamic enhancement CT exams could effectively promote the specificity and the accuracy of APS diagnosis.
8.Antitumor components screening of Stellera chamaejasme L. under the case of discrete distribution of active data.

This is to report the screening, extracting and validating antitumor components and compounds from Stellera chamaejasme L. under the case of discrete distribution of active data. In this work, different components from Stellera chamaejasme L. were collected by HPD macroporous resin and polyamide resin column, and their antitumor activity on A549 were tested by MTT assay. Activity results indicate that activity of components at 30-39 min is more potent than that of Stellera chamaejasme L. extract, and the activity of components at 33.97 min is equivalent to positive drug, cis-platinum at 100 microg x mL(-1), but with totally different mode of action. Under the case of discrete activity, the weight analysis is capable of screening active components and compounds from natural products.
