2.Effect of Perforin Antibody on the Expression of Granzyme B in Viral Myocarditis

Objective To explore the effect of perforin(PFP) antibody on the expression of Granzyme B (GzmB) in viral myocarditis.Methods Forty-five 4-week-old BALB/c male mice were randomly divided into 3 groups,including normal control group (n=15),viral control group (n=15) and PFP antibody therapy group (n=15).The mice in normal control group were inoculated with 0.15 mL Eagle reagent,and the mice in viral control group and PFP antibody therapy group were moculated with 0.15 mL TCID501012 L-1 coxsackievirus B3.The mice in PFP antibody therapy group were inoculated with PFP antibody (0.1 mg/kg) on 6 h and 3 d post inoculation.The mice in 3 groups were sacrificed on day 10 post inoculation.Extracted their hearts,then fixed,dehydrated,embeded and sliced the myocardial tissues.The expressions of GzmB proteins in myocardium were determined by immunohistochemistry.The areas of GzmB were measured by the Leica Qwin V3 system.SPSS 11.5 software was used to analyze the data.Results There were 4 mouse dead in viral control group,and 2 mouse dead in PFP autibody therapy group,no mice dead in normal contol group.The expressions of GzmB were not found in myocardial tissues of normal control group,but there were lots of expressions of GzmB in myocardial tissues of viral control group[(67.13?1.82)%].Athough the expressions of GzmB in PFP antibody therapy group[(6.98?1.34)%] were significantly decreased compared with those in myocardial tissues of viral control group(q=66.180 P
3.Effect of Different Types of Tube Feeding on Swallowing Function for Stroke Patients

Objective To evaluate the effect of different types of tube feeding on swallowing function of stroke patients. Methods 60 patients with dysphagia post stroke were divided into observation group and control group with 30 cases in each group. The control group received indwelling nasogastric tube feeding, and the observation group received intermittent nasogastric tube feeding. They were assessed with Kubota Water Swallow Test (WST) and VGF on the 1st day and 30th day after admission. Results The scores of drinking test and VGF were better in the observation group than in the control group (P<0.05) on the 30th day. Conclusion Long-term nasogatric tube feeding is effective on dysphagia in stroke patients.

6.Comparative study on effects between electroacupuncture and auricular acupuncture for methamphetamine withdrawal syndrome.

OBJECTIVETo observe the efficacy difference of electroacupuncture and auricular acupuncture in the treatment of methamphetamine withdrawal syndrome.
METHODSNinety male patients of methamphetamine addiction were randomized into an electroacupuncture group, an auricular acupuncture group and a control group, 30 cases in each one. In the electroacupuncture group, Neiguan (PC 6), Shenmen (HT 7), Zusanli (ST 36), Sanyinjiao (SP 6), Jiaji (EX-B 2) at T5 and L2 were selected bilaterally. In the auricular acupuncture group, jiaogan (AH(6a)), shenmen (TF4), fei (CO14) and gan (CO12) were selected unilaterally. The treatment was given 3 times a week, totally 12 treatments were required. In the control group, no any intervention was applied. Separately, before treatment and after 1, 2, 3 and 4 weeks treatment, the scores of methamphetamine withdrawal syndrome, Hamilton anxiety scale and Hamilton depression scale were observed in each group.
RESULTSThe total score of methamphetamine withdrawal syndrome, anxiety score and depression score were obviously reduced in 2, 3 and 4 weeks of treatment as compared with those before treatment in the electroacupuncture group and the auricular acupuncture group (all P < 0.05), and showed a trend of gradual decline as the extension of treatment. In 1,2,3,4 weeks of treatment, the total score of withdrawal syndrome, anxiety score and depression score in the electroacupuncture group and auricular acupuncture group were lower significantly than those in the control group (all P < 0.05), in which, the total score of withdrawal syndrome in the electroacupuncture group was lower significantly than that in the auricular acupuncture group in the 4th week of treatment (3.69 +/- 2.446 vs 5.73 +/- 3.169, P < 0.05); the anxiety scores were lower significantly than those in the auricular acupuncture group in 3 and 4 weeks of treatment (8.19 +/- 4.57 vs 9.65 +/- 4.24, 5.27 +/- 2.89 vs 7.38 +/- 3.10, both P < 0.05); the depression scores were lower significantly than those in the auricular acupuncture group in 2, 3 and 4 weeks of treatment (15.35 +/- 5.64 vs 19.81 +/- 5.37, 10.96 +/- 4.52 vs 15.00 +/- 4.53, 7.96 +/- 2.69 vs 12.35 +/- 3.59, all P < 0.05).
CONCLUSIONElectroacupuncture at the body points and auricular acupuncture play the therapeutic role in the treatment of methamphetamine withdrawal syndrome, anxiety and depression. The longer time the treatment is with electroacupuncture at the body points, the more obvious the efficacy will be on the above symptoms.

7.Chemical constituents from Callicarpa nudiflora and their cytotoxic activities.

The chemical consitituents from cytotoxic fraction of the Callicarpa nudiflora extract were isolated and purified by a combination of HP-20 macroporous resin, silica gel and Sephadex LH-20 column chromatographies. The structures were elucidated on the basis of the spectroscopic data and comparison of their spectroscopic data with reported data. The cytotoxicity was evaluated by the MTT assay. The 50% and 70% EtOH elutions of EtOH-extract showed significant cytotoxic activities, leading to the isolation of twelve compounds, which were identified as luteoloside(1), lutedin-4'-O-β-D-glucoside(2), 6-hydroxyluteolin-7-O-β-glucoside(3), lutedin-7-O-neohesperidoside(4), rhoifolin (5), luteolin-7, 4'-di-O-glucoside (6), forsythoside B (7), acteoside (8), alyssonoside (9), catalpol(10), nudifloside(11), and leonuride(12). Compounds 3-6, 10 and 12 were isolated from this genus for the first time, and compound 9 was isolated from this plant for the first time. The cytotoxicity assay demonstrated that flavonoids 1-6, in various concentrations, showed monolithic proliferation inhibitory activities against Hela, A549 and MCF-7 cell lines. Compounds 3, 5 and iridoid glycoside 11 possessed higher cytotoxicacivities. In short, flavonoids are the main components of cytotoxic extract from C. nudiflora, while phenylethanoid glycosides are the predominant ingredient but inactive to cancer cell lines. In addition, the minor iridoid glycoside expressed weak cytotoxic activity.

8.Preliminary analysis on the treatment of infection caused by pandrug-resistant Acinetobacter baumannii

Objective To analyze the clinical features of pandrug-resistant Acinetobacter baumannii (PDR-Ab) in a hospital and compare the efficacy of different antibiotic treatments on patients with pneumonia caused by PDR-Ab.Methods Data were ret- rospectively collected from all isolated PDR-Ab strains in our hospital from February 2004 to March 2005.The clinical features and outcomes were reviewed.Results A total of 77 strains of PDR-Ab were collected, 45 of which were pathogens causing clini- cal infections (35 strains from lower respiratory tract, 6 from bloodstream, 3 from drainage fluid, and 1 from wounds).Lower respiratory tract was the most common source of PDR-Ab.More than 90% of the isolated PDR-Ab strains produced OXA-23 type?-lactamase.Cefoperazone-sulbactam plus minocyeline showed good efficacy for patients with PDR-Ab pneumonia.The total clinical cure rate was 68.4%.Bacterial eradication rate was 42.1%.The factors influencing bacterial clearance were pro- longed mechanical ventilation prior to positive culture (17.5 d vs 5.5 d).mixed infection (100% vs 12.5%) and lower GCS score (9.1?0.7 vs 13.2?2.1).Concomitant septic shock (OR=13.8) and APACHEⅡscore (OR=2.1) were independent factors of clinical outcome.Conclusions Nosocomial infections caused by PDR-Ab are not untreatable.Our analysis suggests that cefoperazone-sulbactam plus minocycline may be an effective treatment for lower respiratory tract infections caused by PDR-Ab in our hospital.

10.Clinical observation on acupoint injection therapy for chronic gastritis of gastric blood stasis type.

OBJECTIVETo objectively evaluate the clinical effect of acupoint injection therapy for chronic gastritis of gastric blood stasis type.
METHODSOne hundred and two cases arranged by registration order were randomly divided into an acupoint injection group and a medicine group, 51 cases in each group. The acupoint injection group was treated with acupoint injection of compound Danshen injection, and Zusanli (ST 36) and Weishu (BL 21) were selected, and the medicine group with oral administration of Omeprazole. After 2 weeks of treatment, the clinical effect and improvement of endoscopic gastric mucosal lesions were observed.
RESULTSThe clinical total effective rate of 96.1% (49/51) in the acupoint injection group was better than 76.5% (39/51) in the medicine group (P < 0.01). The symptom score decreased significantly after treatment and the gastric mucosal lesion was significantly improved in both of the two groups (all P < 0.05), and the acupoint injection group was superior to the medicine group (all P < 0.05).
CONCLUSIONAcupoint injection has outstanding effect for treatment of chronic gastritis of gastric blood stasis type and this therapy is worth generalizing and applying.
